Lactylates are an important group of molecules in the food and cosmetic industries. A series of natural halogenated 1-lactylates – chlorosphaerolactyaltes (<b>1</b>-<b>4</b>) – were recently reported from <i>Sphaerospermopsis</i> sp. LEGE 00249. Here, we identify the <i>cly</i> biosynthetic gene cluster, containing all the necessary functionalities to generate and release the natural lactylates. Using a combination of stable isotope-labeled precursor feeding and bioinformatic analysis, we propose that dodecanoic acid and pyruvate are the key building blocks in the biosynthesis of <b>1</b>-<b>4</b>. We additionally report minor analogues of these molecules<b> </b>with varying alkyl chains. The discovery of the <i>cly</i> gene cluster paves the way to accessing industrially-relevant lactylates through pathway engineering.

Cells were incubated with 12-(1-pyrene)-dodecanoic acid (P12), a long-chain fatty acid to which a pyrene ring has been attached covalently. This acid was transported across the plasma membranes of cells and subsequently incorporated into their neutral lipids and phospholipids. Irradiation of these pyrene-containing cells for short periods (0.5-4 min) with ultraviolet light at 366 nm resulted in eventual cell death. Similar irradiation had no effect on cells that had not been exposed to P12. The time of the period of irradiation necessary for inducing the toxic process was related to the quantity of P12 incorporated, the latter being a function of the respective metabolic activity of the individual cell type. The degree of incorporation of P12 into a cell, and consequently its acquired sensitivity to killing by ultraviolet irradiation at 366 nm, was affected by the incubation temperature and addition of non-fluorescent fatty acid, albumin or other serum proteins. Different degrees of incorporation of P12 into various cell types were used for selective killing and elimination of cell populations by irradiation at 366 nm. The combined procedure of preincubation with P12 followed by ultraviolet irradiation thus permitted selection of cell types with a greater resistance to this procedure.

Abstract Purpose This study was aimed to screen differential metabolites between gastric cancer (GC) and paracancerous (PC) tissues and find new biomarkers of GC. Methods GC (n = 28) and matched PC (n = 28) tissues were collected and LC-MS/MS analyses were performed to detect metabolites of GC and PC tissues in positive and negative models. Principal component analysis (PCA) and orthogonal projections to latent structures-discriminate analysis (OPLS-DA) were conducted to describe distribution of origin data and general separation and estimate the robustness and the predictive ability of our mode. Differential metabolites were screened based on criterion of variables with p value < 0.05 and VIP (variable importance in the projection) > 1.0. Receiver operating characteristic (ROC) analysis was performed to evaluate the diagnostic power of differential metabolites. Kyoto Encyclopedia of Genes and Genomes (KEGG) was performed to search for metabolite pathways and MetaboAnalyst was used for pathway enrichment analysis. Results Several metabolites were significantly changed in GC group compared with PC group. Thirteen metabolites with high VIP were chose and among which 1-methylnicotinamide, dodecanoic acid and sphinganine possessed high AUC values (AUC > 0.8) indicating an excellent discriminatory ability on GC. Pathways such as pentose phosphate pathway and histidine metabolism were focused based on differential metabolites demonstrating their effects on progress of GC. Conclusions In conclusion, we investigated the tissue-based metabolomics profile of GC and several differential metabolites and signaling pathways were focused. Further study is needed to verify those results.

Hypertension and Type 2 diabetes are co-morbid diseases that lead to the development of nephropathy. sEH (soluble epoxide hydrolase) inhibitors are reported to provide protection from renal injury. We hypothesized that the sEH inhibitor AUDA [12-(3-adamantan-1-yl-ureido)-dodecanoic acid] protects the kidney from the development of nephropathy associated with hypertension and Type 2 diabetes. Hypertension was induced in spontaneously diabetic GK (Goto–Kakizaki) rats using AngII (angiotensin II) and a high-salt diet. Hypertensive GK rats were treated for 2 weeks with either AUDA or its vehicle added to drinking water. MAP (mean arterial pressure) increased from 118±2 mmHg to 182±20 and 187±6 mmHg for vehicle and AUDA-treated hypertensive GK rats respectively. AUDA treatment did not alter blood glucose. Hypertension in GK rats resulted in a 17-fold increase in urinary albumin excretion, which was decreased with AUDA treatment. Renal histological evaluation determined that AUDA treatment decreased glomerular and tubular damage. In addition, AUDA treatment attenuated macrophage infiltration and inhibited urinary excretion of MCP-1 (monocyte chemoattractant protein-1) and kidney cortex MCP-1 gene expression. Taken together, these results provide evidence that sEH inhibition with AUDA attenuates the progression of renal damage associated with hypertension and Type 2 diabetes.

The ophthalmic treatment of diabetic retinopathy is aimed at preserving vision and requires an interdisciplinary approach. The main treatments used for proliferative retinopathy and diabetic maculopathy include laser photocoagulation, intra-vitreal, and peri-ocular drug injections, or a combination of both. In advanced cases of diabetic retinopathy (DR), vitrectomy and retinal surgery may help preserve vision. Cataract surgery in diabetic patients is extremely successful, but overall these patients are usually considered to be at risk of more complications than the general population. Those patients with pre-existing DR at the time of surgery should be regarded as a high risk group and will require careful pre-operative assessment and post-operative review. In this group, intervention with laser photocoagulation and intra-vitreal pharmacotherapy may be necessary.

Perfluorinated chemicals (PFCs) can be absorbed on fine particulate matter (PM2.5) and used as stain, water and grease repellent in a wide range of consumer products. Among the PFCs, perfluorooctane sulfonate (PFOS) and perfluoro octanoic acid (PFOA) are widely detected in human blood and serum and are of concern due to their potential toxicity. In the present experiment, fine particulate matter (PM2.5) from some northern (Beijing, Xian) and southern (Hong Kong, Guangzhou and Xiamen) cities of China were collected and analyzed for perfluoro butanoic acid (PFBA), perfluoro hexanoic acid (PFHxA), perfluoro octanoic acid (PFOA), perfluoro nonanoic acid (PFNA), perfluoro decanoic acid (PFDA), perfluoro undecanoic acid (PFUdA), perfluoro dodecanoic acid (PFDoA), perfluoro hexanesulfonate (PFHxS) and perfluoro octanesulfonate (PFOS) using liquid chromatography mass spectrometry (LC-MS/MS). The total PFCs ranged from 121.2 to 192.2pg/m3, leading by Guangzhou followed by Xian, Beijing, Xiamen and Hong Koung. Among the nine measured PFCs compounds, the level of PFHxS was below the limit of detection in all the sampling cities. The other eight PFCs (PFOS, PFDoA, PFUdA, PFDA, PFNA, PFOA, PFHxA and PFBA) were detected in all the sampling locations except PFDoA in Hong Kong samples. Human exposure estimated to PFCs for adults showed PFOS as the dominant inhaled compound representing 1.59, 1.15, 1.0 and 1.0 ng/day exposure for Hong Kong, Guangzhou, Xiamen, Beijing and Xian respectively. The aim of the study was to evaluate the effect of Spirulina platensis, formulation containing microalgal extract, post-extraction residue, and formulation without algal extract (containing only emulsifier) on the content of FAs in the eggs of laying hens. The experiment was conducted on 90 laying hens (ISA Brown) as a completely randomized design. Hens were assigned to five experimental groups (six replicates). The FAs content in eggs was determined after 30, 60, 90, and 120 days of the experiment. There were no statistically significant differences in FA profiles after 30 days of the experiment. It was shown that after 60, 90, and 120 days of the experiment, the investigated additives had a significant impact on the content of such acids as: dodecanoic acid (C12:0), C15:0, nonadecanoic acid (C19:0), myristoleic acid (C14:1 n-5), α-linolenic acid (ALA, C18:3 n-3), DPA, C20:2 n-6, and decosahexaenoic acid (DHA C22:6 n-6). There were also significant differences in total PUFA n-3, PUFA n-6, and n-6/n-3 ratio in eggs. The obtained results suggest that the use of algae extract and emulsifier in laying hens nutrition has the greatest impact on the FA profile in the eggs.
