To investigate the antihyperglycemic and antihyperlipidemic effect of Momordica charantia (Karala), the aqueous extract of the Karala fruit was tested on streptozotocin (STZ)-induced diabetic rats. Thirty six albino rats were used in the experiment, 30 diabetic and the remaining six as negative control (T1). Diabetes was induced by administering (injecting) STZ at dose of 55mg/kg body weight. Thirty diabetic animals were randomly divided into five groups such as diabetic control group (T2) without any application of treatment, and groups T3,T4,T5 and T6 were treated with aqueous extract of Karala fruits daily at the doses of 250,    500 and 750mg/kg and glibenclamide (at a dose of 5mg/kg body weight) respectively. The body weight was taken and blood samples were collected from individual animal to determine glucose levels at 15 day interval up to 90 days. In addition, Asparate  Transaminenase(AST), Alanine Transaminenase(ALT), Alkaline Phosphatase(ALP), Total cholesterol (TCh) and Triglyceride (TGA) were determined at day 15 and at the end of the experiment. All three doses of Karala extracts reduced diabetic induced blood sugar and the reduction is comparable with standard glibenclamide (GLM) dose particularly with higher doses Karala extracts (500 and 750mg). Karala also prevented body weight loss due to induced diabetes as did by GLM treatment.. The treatment also resulted in a significant reduction of Asparate Transaminenase(AST), Alanine Transaminenase(ALT), Alkaline Phosphatase(ALP), Total cholesterol (TCh) and Triglyceride (TGA) activities of treated rats when compared to the STZ induced  diabetic rats. Higher doses of Karala (500 and 750mg/kg) are as effective as standard GLM dose on measured variables. This study demonstrated that Karala has hyperglycemia and antihyperlipidemic effect against STZ induced diabetic rats. These findings open the possibility of using Karala extract to treat diabetic animal and human patients although further research is warranted. DOI: http://dx.doi.org/10.3329/jesnr.v5i1.11550 J. <p><strong>Background:</strong> Diabetes is one of the commonest and serious metabolic disorders. Much of the morbidity and mortality associated with diabetes is primarily attributed to sequelae requelac of microvascular and macrovascular changes, in which diabetic dyslipidemia is one of the many modifiable risk factors for coronary artery disease, stroke and peripheral vascular disease.</p><p><strong>Objectives:</strong> The main aim of this study was to evaluate the hypoglycemic and antidyslipidemic effect of selected Indian plants in streptozotocin induced diabetic rats.</p><p><strong>Material and Methods:</strong> Azadirachta indica and Ocimm sanctum leaves, Allium sativum bulbs and Momordica charantia fruits were collected, identified taxonomically and extract was obtained. Male Albino rats was used and divided into 8 groups, each consisting of 6 animals, one group act as a control. Diabetes in rats was induced with streptozotocin. Blood samples were collected and biochemical analysis was done for blood sugar, lipid peroxide and lipid profile. The diabetic group without drug treatment was compared with the control, and diabetic plus drug-treated groups were compared with the diabetic group without drug treatment. Data were analyzed using Student ‘t’ test.</p><p><strong>Results:</strong> Our results revealed that administration of streptozotocin in rats caused increase in the levels of glucose, lipid peroxides, cholesterol and triglycerides with lessening of the HDL-cholesterol. Treatment with aqueous extracts of Momordica charantia, Allium sativum, Azadirachta indica and Ocimum sanctum not only reduced the level of blood glucose but also caused lowering of total cholesterol and triglycerides following an increase in the level of HDL-cholesterol.</p><p><strong>Conclusion:</strong> We concluded that the herbal plants tested possess both hypoglycemic and antidyslipidemic activities and their use as a therapeutic tool in diabetes related complications encourage further investigation in this field.</p>

Background: There is a growing interest in the correlation between antioxidants and periodontal disease. In this study, we aimed to investigate the effect of oxidative stress and the impact of two antioxidants, curcumin and rutin, respectively, in the etiopathology of experimentally induced periodontitis in diabetic rats. Methods: Fifty Wistar albino rats were randomly divided into five groups and were induced with diabetes mellitus and periodontitis: (1) (CONTROL)—control group, (2) (DPP)—experimentally induced diabetes mellitus and periodontitis, (3) (DPC)—experimentally induced diabetes mellitus and periodontitis treated with curcumin (C), (4) (DPR)—experimentally induced diabetes mellitus and periodontitis treated with rutin (R) and (5) (DPCR)—experimentally induced diabetes mellitus and periodontitis treated with C and R. We evaluated malondialdehyde (MDA) as a biomarker of oxidative stress and reduced glutathione (GSH), oxidized glutathione (GSSG), GSH/GSSG and catalase (CAT) as biomarkers of the antioxidant capacity in blood harvested from the animals we tested. The MDA levels and CAT activities were also evaluated in the gingival tissue. Results: The control group effect was statistically significantly different from any other groups, regardless of whether or not the treatment was applied. There was also a significant difference between the untreated group and the three treatment groups for variables MDA, GSH, GSSG, GSH/GSSG and CAT. There was no significant difference in the mean effect for the MDA, GSH, GSSG, GSH/GSSG and CAT variables in the treated groups of rats with curcumin, rutin and the combination of curcumin and rutin. Conclusions: The oral administration of curcumin and rutin, single or combined, could reduce the oxidative stress and enhance the antioxidant status in hyperglycemic periodontitis rats.

Currently, cardiovascular diseases are the leading cause of global mortality, while diabetes mellitus remains an important cause of cardiovascular morbidity. A recent study showed that patients with diabetes mellitus treated with mineralocorticoid receptor antagonists have improved coronary microvascular function, leading to improved diastolic dysfunction. In this study, we evaluated the influence of acute administration of spironolactone on myocardial function in rats with streptozotocin-induced diabetes mellitus, with special emphasis on cardiodynamic parameters in diabetic rat hearts. The present study was carried out on 40 adult male Wistar albino rats (8 weeks old). Rats were randomly divided into 4 groups (10 animals per group): healthy rats treated with 0.1 μmol/L of spironolactone, diabetic rats treated with 0.1 μmol/L of spironolactone, healthy rats treated with 3 μmol/L of spironolactone, and diabetic rats treated with 3 μmol/L of spironolactone. Different, dose-dependent, acute responses of spironolactone treatment on isolated, working diabetic and healthy rat heart were observed in our study. In healthy rats, better systolic function was achieved with higher spironolactone dose, while in diabetic rats, similar effects of low and high spironolactone dose were observed.

The aim of the present study was to investigate the protective effects of Sesbania grandiflora flower (SGF) extract on erythrocyte membrane in Streptozotocin (STZ)-induced diabetic rats. Adult male albino rats of Wistar strain, weighing 190–220 g, were made diabetic by an intraperitonial administration of STZ (45 mg/kg). Normal and diabetic rats were treated with SGF, and diabetic rats were also treated with glibenclamide as drug control, for 45 days. In this study plasma insulin and haemoglobin levels were decreased and blood glucose, glycosylated haemoglobin, protein oxidation, lipid peroxidation markers, and osmotic fragility levels were increased in diabetic rats. Moreover, erythrocytes antioxidant enzymes such as superoxide dismutase, catalase, glutathione peroxide, glutathione reductase, glutathione-S-transferase, and glucose-6-phosphate dehydrogenase activities and non-enzymatic antioxidants such as vitamin C, vitamin E, reduced glutathione (GSH), and oxidized glutathione (GSSG) levels were altered. Similarly, the activities of total ATPases, Na+/K+-ATPase, Ca2+-ATPase, and Mg2+-ATPase were also decreased in the erythrocytes of diabetic rats. Administration of SGF to STZ-induced diabetic rats reduced blood glucose and glycosylated haemoglobin levels with increased levels of insulin and haemoglobin. Moreover, SGF reversed the protein and lipid peroxidation markers, osmotic fragility, membrane-bound ATPases activities, and antioxidant status in STZ-induced diabetic rats. These results suggest that SGF could provide a protective effect on diabetes by decreasing oxidative stress-associated diabetic complications.

Background: Diabetes mellitus (DM) affects the musculoskeletal system through its metabolic perturbations. Exercise modulates blood sugar levels and increases the body’s sensitivity to insulin in patients with DM. Objective: This study aimed to investigate the potential effects of combined quercetin and coenzyme Q10 (CoQ10) supplements with or without exercise on the histological, biochemical and molecular structures of diabetic rat’s skeletal muscle. Method: A total of 64 adult male albino rats were divided into six groups: control, trained nondiabetic, non-trained diabetic, diabetic rats treated with combined CoQ10 and quercetin, diabetic rats with treadmill training, and diabetic rats treated with treadmill training and CoQ10 and quercetin. Blood and skeletal muscle samples were obtained from all groups for routine histological examination and biochemical determination of cytokine levels and protein activities. Quantitative real-time polymerase chain reaction (qRT-PCR) and morphometric analysis of PAS and Bax expressions were also performed. Results: Biochemical analysis revealed improvement in all studied parameters with combined CoQ10 and quercetin than exercise training alone. Combined treatment and exercise showed significant improvement in all parameters especially interleukin 6 and malondialdehyde. Fibronectin type III domain-containing protein 5 (FNDC5) expression and irisin levels increased in all trained groups but combined treatment with exercise significantly increased their levels than exercise alone. Histological analysis revealed improvement after exercise or combined treatment; however, when exercise was combined with CoQ10 and quercetin, marked improvement was observed. Conclusion: the combination of CoQ10 and quercetin could be promising in preserving musculoskeletal function in patients with DM concomitantly with physical exercise.

One of the major obstacles that males with diabetes may confront is subfertility or infertility. Thus, the present study investigated the effect of co-administration of metformin and zinc (Zn) on the testes of streptozotocin-induced diabetic rats. Male albino rats were randomly divided into 4 groups: control group; untreated diabetic group; diabetic + metformin group, in which diabetic rats were treated orally with metformin (250 mg/kg) once daily for 4 weeks; and diabetic + metformin + Zn group, in which diabetic rats were treated orally with metformin in combination with Zn (10 mg/kg) once daily for 4 weeks. Concomitant administration of metformin and Zn produced a significant decrease in serum levels of glucose and insulin and testicular levels of malondialdehyde and tumor necrosis factor α. Additionally, there was a significant increase in serum levels of Zn, testosterone, and follicle-stimulating hormone, as well as testicular total antioxidant capacity and anti-apoptotic protein Bcl-2, when compared with both the diabetic and metformin-treated diabetic groups. Moreover, co-administration of Zn and metformin significantly improved testicular histopathology, with a significant reduction in percent area of collagen fibers and nuclear factor kappa B (p65) immunoreactivity and a significant increase in seminiferous tubule diameter and connexin 43 immunoreactivity as compared with the diabetic and metformin-treated diabetic groups. In conclusion, the combination of Zn and metformin was an efficacious and safe alternative treatment, as it had superior antihyperglycemic efficacy and provided additional benefits over metformin alone in rats with type 2 diabetes.

Postprandial hyperglycemia is a predisposing factor for vascular dysfunction and organ damage.α-glucosidase is a hydrolytic enzyme that increases the glucose absorption rate and subsequently elevates blood glucose levels. Garlic (Allium sativumL.) is a rich source of several phytonutrients, including thiosulfinate (THIO). The aim of this study was to evaluate the ability of THIO, a potent inhibitor of intestinalα-glucosidase, to reduce postprandial blood glucose. Male albino rats were randomly assigned to five different groups (n=10/group). Group 1 served as the control group. Groups 2–5 were injected intraperitoneally with a single dose of streptozotocin (STZ) to induce diabetes. Group 2 comprised untreated diabetic rats. Groups 3 and 4 contained diabetic rats that were given THIO orally (20 mg/kg body weight/day and 40 mg/kg body weight/day, resp.). Group 5 was the positive control having diabetic rats treated orally with acarbose (10 mg/kg body weight/day; positive control). Diabetic rats treated with THIO displayed a significant blood glucose reduction (p<0.001and < 0.01 by analysis of variance, resp.) and a significant elevation in insulin compared with that of untreated rats. THIO is an effective noncompetitive intestinalα-glucosidase inhibitor that promotes hypoglycemic action (p<0.001) in STZ-injected rats. THIO is a promising agent for the management of postprandial hyperglycemia.

Diabetes is characterized by hyperglycaemia with alterations of lipid parameters. The effects of Aloe vera (A.V) gel extract on lipid profiles of streptozotocin-induced diabetic rats were investigated. Seventy-two male albino rats were randomly divided into six groups of 12 rats each; control, diabetic control, 4 ml/kg and 8 ml/kg body weight (b.w.) A.V, 10 mg/kg b.w. glibenclimide and non-diabetic 8 ml/kg b.w. A.V groups. They were treated for 14 and 28 days. The plasma and HDL lipid profiles (triacylglycerols, cholesterol and phospholipids) of the diabetic rats were significantly (p<0.05) increased, while those of the VLDL+LDL, RBC, RBC membrane and organs were observed to be significantly (p<0.05) reduced, when compared to the control after 14 and 28 days of treatment. A.V gel extract significantly (p<0.05) ameliorated the disruptions observed in the lipid profiles of the diabetic and thus provide a scientific rationale for the use of Aloe vera as a therapeutic agent.Bangladesh J Med Biochem 2016; 9(1): 11-26
