Carbohydrase enzymes improve performance of broilers fed both nutritionally adequate and marginal wheat-based diets

SummaryA study was conducted to examine the effects of a multi-carbohydrase enzyme complex on the nutritive value of wheat in diets differing in nutrient density. It was hypothesised that response to enzyme inclusion would be greater in diets with lower nutrient density. The study was conducted using 1008 Ross 308 male broiler chicks (four treatments with seven replicate pens of 36 chicks). A 2 × 2 factorial arrangement of treatments was employed. Factors were adequate or low nutrient density with or without enzyme supplementation. The wheat-soybean meal based positive control (PC) diet was formulated to be nutritionally adequate in energy and digestible amino acids according to local industry recommendations. A negative control (NC) was formulated to have 80 kcal/kg less ME and 1.5% less digestible amino acids as compared to the PC. A multi-carbohydrase complex containing 19 carbohydrase activities derived from Penicillium funiculosum was added in both the PC and NC diets (Rovabio® Excel LC, Adisseo Asia Pacific Pte Ltd., Singapore). Birds fed the NC had 3.7 points (P < 0.05) poorer FCR than the PC. Across the diet type, enzyme supplementation increased body weight by 3.2% (P < 0.05) and improved FCR by 5.2 points (P < 0.01). There was no nutrient density x enzyme interaction (P > 0.05), indicating that performance improvement was independent of nutrient density. Apparent ileal digestibility of crude protein followed a similar trend, showing a 4.9% enhancement (P < 0.01) with the inclusion of the enzyme product in either diet. Enzyme supplementation reduced ileal viscosity by 39.0% (P < 0.05). It was concluded that multi-carbohydrase could overcome the negative effect in broiler performance brought by nutrient reduction, however, there was no indication that nutrient density affected bird response to supplementation of multi-carbohydrase.

This experiment evaluated the effects of supplemental protease in maize-based diets formulated with reduced density of digestible amino acids (dAA) on net energy (NE) utilisation and nutrient digestibility in broilers. A total of 312, one-day-old, male broilers (Ross 308) were randomly distributed into 24 floor pens and fed one of three treatments, with eight pen replicates per treatment throughout the starter (1 to 14 d of age) and grower (15 to 28 d of age) phases. Dietary treatments consisted of a positive control (PC) reference diet, a negative control diet (NC; dAA density 60 g/kg less than PC diet), and a NC diet with supplemental protease (200 mg/kg). All diets contained supplemental phytase (200 mg/kg) and xylanase (200 mg/kg). The reduction in dAA density between the PC and NC did not affect (P>0.05) NE, but protease supplementation in the NC diet increased (P<0.05) NE by 0.27 MJ/kg, compared with those receiving the NC diet without protease. The reduction in dAA for broilers fed the NC and PC diets did not (P>0.05) affect nitrogen, starch, or DE in the jejunum or ileum. Likewise, no differences (P>0.05) in jejunal (nitrogen, starch, and DE) and ileal (starch and DE) digestibility values were observed between those offered the NC diets without or with protease, but a small difference (P<0.05) in ileal nitrogen digestibility was observed.

Three experiments were carried out to verify the effects of the enzyme phytase, alone or combined with an enzyme complex, in diets deficient in available phosphorus (AP), calcium (Ca), and metabolizable energy (ME) on broiler performance, ME, and dietary amino acid digestibility. A total of 1,538 male Cobb 500 broilers were allocated to the three experiments, each of which consisted of five treatments: positive control (PC; basal ration); negative control 1 (NC1; PC minus 0.15% of AP, 0.16% of Ca, and 68 kcal kg-1 ME); negative control 2 (NC2; PC minus 0.15% of AP, 0.16% of Ca, and 101 kcal kg-1 of ME); NC1 plus phytase; and NC2 plus phytase plus enzymatic complex. Body weight gain (WG) and feed intake were measured from 1-21 days and from 1-42 days, and the corrected feed conversion rate (FCR) for mortality was calculated. In the second and third experiments, the apparent ME corrected for nitrogen balance (AMEn) and standardized digestibility of amino acids, respectively, were determined, for the diets supplemented with phytase and the enzymatic complex. In the first experiment, enzyme supplementation increased (p < 0.05) WG at 21 days and 42 days relative to the negative controls. Phytase inclusion improved (p < 0.05) FCR at the initial phase compared to the NC1 diet. In the second experiment, enzyme supplementation did not affect (p > 0.05) AMEn. In the third experiment, both enzyme treatments improved (p < 0.05) the digestibility of amino acids in the supplemented diets compared to the deficient diets. Supplementation with phytase and carbohydrases preserves the performance of broilers fed diets deficient in AP, Ca, and ME and improves amino acid digestibility.

Ross 308 broiler chicks (n = 240) aged 1 day were assigned to five groups for eight replicates (six chicks for each) (3♂ and 3♀). Basal dietary groups were supplemented by 2000, 4000, and 6000 mg/kg cinnamon (CN) for 21 days. Basal diet alone was used as a negative control, and basal antibiotic diet (Colimox) was used as a positive control. At 10, 14, and 21 days of age, chicks that received 2000 mg CN and Colimox had a higher body weight, resulting in an increase in body weight gain. CN also resulted in the maximum improvement in the feed conversion ratio and feed efficiency over 1–21 days at the level of 2000 mg/kg. At days 10, the maximum relative breast weight was 2000 mg/kg of CN. Mean serum albumin concentrations, duodenal villus height, and goblet cell density increased (p < 0.05) by 2000 mg/kg of CN, and mean serum globulin and total protein concentrations and crypt depth increased (p < 0.05) by 6000 mg/kg of CN compared with control. Increased cecal Escherichia coli number was CN dose-dependent. In conclusion, dietary inclusion of 2000 mg/kg CN can be applied as an alternative to in-feed antibiotics for broiler starter diet.

Individual and group housed crossbred pigs 45 to 105 kg and 65 to 95 kg in experiments 1 and 2, respectively, were given basal diets with L-threonine 0.6, 1.2 and 1.8 g/kg. Positive and negative control diets contained total threonine 5.7 and 4.5 g/kg, respectively. To prevent other amino acids being limiting, the negative control diet was supplemented with lysine, methionine, tryptophan, isoleucine, histidine and valine. The positive control diet was supplemented with lysine and methionine. The requirement for total threonine of growing-finishing pigs for maximum growth performance was about 5.6 g/kg in a diet containing net energy 9.4 MJ/kg. This figure corresponds with about 4.7 g/kg apparent faecal digestible threonine and 4.3 apparent ileal digestible threonine. There was no difference between the growing and the finishing pigs. The requirement for ileal digestible threonine, relative to ileal digestible lysine requirement, was about 64%. It is concluded that dietary protein can be reduced by 2 percentage units without any adverse effect on growth performance, if limiting amino acids are sufficiently supplemented. This study was conducted to investigate the effects of in ovo and post-hatching supplementation with L-Glutamine (Gln) on hatching characteristics, performance, small intestinal morphology, and muscle development of broilers. At day 18 of incubation, 960 fertilized eggs were allocated to four treatments with six replicates. Eggs were i) not injected (negative control) (NC), ii) subject to the standard incubation procedure (PG), iii) injected with 1 ml sterile solution with 0.9% salt (positive control) (PC), and iv) injected with 1% Gln solution (IG). On hatching, the SC chicks were fed with 1% Gln for seven days. The remaining chicks were fed a commercial starter feed. After hatching, there were six replicates of 28 birds in each treatment. Hatchability and yolk sac weight were lower and yolk-free chick weight (YFCW), whole gastrointestinal tract (GIT) and breast muscle weights were higher at hatching for chicks from the IG treatment. At 42 days old, feed conversation ratio (FCR) was lower in birds that had Gln added to their diet than for the other treatments. The FCR was also lower in IG birds than birds in the NC group. The GIT weight, villus height, villus width and crypt depth of the birds receiving dietary supplementation of Gln were greater than those of birds in PC and IG. Thus, in ovo injection of Gln improved hatching characteristics except for hatchability. Further, in ovo and dietary Gln administration reduced FCR by stimulating digestive system development.

The physiological and immune responses of adult shrimp (Litopenaeus vannamei) undergoing the acute phase of the necrotizing hepatopancreatitis (NHP) disease and the efficiency of oxytetracycline (OXI) and florfenicol (FF) to eliminate the pathogen were evaluated. Four shrimp groups were considered: three groups infected with necrotizing hepatopancreatitis bacteria (NHP-B) (two treated with antibiotics and a positive control) and one group non-infected (negative control). Hemolymph concentration of glucose, lactate, acylglycerides, cholesterol, total protein, aminotransferases, superoxide dismutase, and the transcriptional expression of several immune related genes were monitored at the acute phase of the disease, and at 15 and 20 days after administration of antibiotics (daa). Shrimp from the positive control registered a mortality of 100%. NHP-B infection affected the immu-nophysiological response of shrimp; herein, most of the parameters were significantly up regulated in infected shrimp before the use of antibiotics, compared to the negative control. Increased transcriptional levels of clotting protein, lipopolysaccharide and β-1-3-glucan binding protein (LGBP), serine protease, peroxinectin, lysozyme, heat shock proteins (HSP) 60 and 70 were detected in shrimp treated with OXI. At 20 daa NHP-B was still detected in FF-treated shrimp, but not in OXI-treated shrimp. It is hypothesized that despite none of the antibiotics per se eliminated the bacterium, both had a negative effect on its virulence. OXI seems to have a greater effect, allowing shrimp to integrate a better immune response at 15 daa.

Fifty-four samples of wheat, representing duplicate samples of nine varieties grown in three environments, were included in chick bioassays using rapidly growing broilers, complete diets, and enzyme supplementation to remove the deleterious effects of non-starch polysaccharides. An insoluble ash marker was included in the diets, which allowed calculation of apparent digestibilities of CP and AA in the grain from analysis of the ileal contents obtained after sacrifice of the chicks at 17 d of age. The wheat cultivars contained from 13.5 to 16.8% CP and differed in the percentage of specific AA, with Pro, Glu and Phe making up proportionately larger parts of high CP samples than low CP samples. Without enzyme supplementation, the digestibility of CP varied from 83 to 88% and that of specific AA from 76 to 94%, determined in part by the class and variety of wheat. Xylanase enzyme supplementation improved the digestibility of protein by an average of 4%, and reduced the differences in digestibility between wheat samples. These results suggest that variation between wheat samples should be taken into consideration when formulating broiler feed, especially when formulation is for specific AA. Enzyme supplementation may provide a partial alternative to using high-quality protein sources or supplementing with synthetic AA. Abstract Three experiments were conducted to determine the effect of three fermented amino acids (AA) with their respective biomass compared to crystalline AA on the growth performance of swine and poultry. In experiment 1, 315 barrows (DNA 200 × 400, initially 11.3 ± 0.69 kg) were allotted to 1 of 4 dietary treatments with 5 pigs per pen and 15 or 16 pens per treatment. Dietary treatments included a negative control (16% standardized ileal digestible [SID] Tryptophan:lysine [Trp:Lys] ratio), positive control (21% SID Trp:Lys ratio from crystalline Trp), or diets containing Trp with biomass to provide 21 or 23.5% SID Trp:Lys ratios, respectively. Pigs fed the positive control or low Trp with biomass diet had increased (P &lt; 0.05) ADG compared to pigs fed the negative control diet, with pigs fed the high Trp with biomass diet intermediate. Pigs fed the low Trp with biomass diet had increased (P &lt; 0.05) G:F compared to the negative control diet, with others intermediate. In experiment 2, 1,320 1-d-old male broilers (Cobb 500, initially 45.2 g) were allotted to one of four dietary treatments with 33 birds per pen and 10 pens per treatment. Dietary treatments included a negative control (58/58% Threonine:lysine [Thr:Lys] ratio), positive control (65/66% Thr:Lys ratio from crystalline Thr), or diets containing Thr with biomass to provide 65/66 or 69/70% Thr:Lys ratios in starter and grower diets, respectively. Broilers fed the positive control or Thr with biomass diets had increased (P &lt; 0.05) ADG compared to broilers fed the negative control diet. Broilers fed the positive control or the low Thr with biomass diet had increased (P &lt; 0.05) G:F compared to the negative control and high Thr with biomass treatments. In experiment 3, 2,100 one-day-old male broilers (Cobb 500, initially 39.4 g) were allotted to one of four dietary treatments with 35 birds per pen and 15 pens per treatment. Dietary treatments included a negative control (59/63% Valine:lysine [Val:Lys] ratio), positive control (75/76% Val:Lys ratio from crystalline Val), or diets containing Val with biomass to provide 75/76 or 84/83% Val:Lys ratios in starter and grower diets, respectively. Broilers fed the positive control or Val with biomass diets had increased (P &lt; 0.05) ADG, ADFI, and G:F compared to those fed the negative control diet. In conclusion, Trp, Thr, or Val with their respective biomass appear to be equally bioavailable and a suitable alternative to crystalline AA in swine and poultry diets.

ABSTRACTSix diets based on maize and soya-bean meal were prepared. The basal diet had a crude protein (CP) concentration of 171 g/kg. The same ingredients, together with maize starch, were used to give a diet with CP 131 g/kg. This diet was supplemented with all (positive control) or with three of the four amino acids lysine, threonine, tryptophan and methionine. Each diet was given at two rates of intake. From the results, the optimum ratio between these four amino acids at different planes of intake was calculated. Amino acid digestibility in the basal diet was measured at three intakes, using pigs fitted with T cannulas at the terminal ileum.The results showed that the optimum ratios between the four amino acids (expressed in terms of digestible amino acids) were not affected by the rate of food intake. The optimum ratio between the four amino acids (i.e. when they are equally limiting) was lysine 1·00, methionine + cystine 0-61, threonine 0·64 and tryptophan 0·20. Digestibility was not affected by the level of intake.
