SummaryThe effects of 2, 4, 7 and 10% supplements of tallow to a basal diet low in fat on the composition of volatile fatty acids (VFA) in the rumen and on the secretion in milk of fat and its component fatty acids are reported. There were no significant effects of the tallow on rumen VFA or on the secretion of total milk fat. The yields in milk of stearic (18:0) and oleic (18:1) acids were positively correlated and those of palmitic (16:0) and myristic (14:0) acids negatively correlated with their respective dietary intakes. Apart from butyric acid which increased, the yields of other short-chain acids synthesized within the mammary gland decreased with increased intake of tallow. Possible factors influencing the output of total milk fat through a balance between increased transfer to milk of dietary fatty acids and decreased intramammary synthesis of fatty acids are discussed.

SummaryEffects of 0, 1·7, 3·3 or 5·0 kg/day of a protected soya bean – tallow supplement, incorporated into a hay:concentrate diet (25:75) and fed ad libitumto Friesian cows, on intake and digestion of fatty acids, on output of milk fatty acids and on blood lipoprotein composition were measured.Most of the increased intake of fatty acids, approximately 1 kg/day, was accounted for by increased intakes of C16:0, C18:0 and C18:1. At low intakes, amounts of all fatty acids apparently digested were linearly related to their respective intakes. At high intakes of C16 and C18 acids, curvilinear relationships were established.Yield of total milk fat was related positively to dietary intakes of total fatty acid and carbohydrate and negatively to live-weight change. Yields of short and intermediate chain acids in milk, synthesized within the mammary gland, were negatively correlated and yields of C18 fatty acids positively correlated with respective dietary intakes of these acids. Decreased proportions of C4–16 and increased proportions of C18:0 and C18:1 fatty acids in milk were associated with increased protected tallow in the diet. Yields of C16:1 and C18:1 were positively related to corresponding outputs of saturated acids and negatively to weeks of lactation. The proportion of C18:1 in milk was positively related to the corresponding proportion of C18:0.The increased intake of fatty acids resulted in increased concentrations of very low density lipoproteins (VLDL, d < 1·019 g/ml), low density lipoproteins (LDL1 + LDL2, 1·019 < d < 1·06 g/ml), high density lipoproteins (d > 1·060 g/ml) and serum free fatty acids. Most of the increase in low density lipoproteins was accounted for by a very large increase in LDL1, whose proportion increased from 17 to 75% (2 to 22% of total serum lipid). The proportion of triglyceride in the combined low density lipoprotein fraction decreased from 11 to 2% whilst phospholipids increased from 29 to 36%. These changes were attributed to the increased proportion of LDL1 present.The proportions of VLDL and LDL triglyceride taken up by the mammary gland averaged 0·79 and 0·34 respectively. The proportion of VLDL+LDL triglyceride taken up by the gland decreased with increased amounts of fatty acid digested. Yields of C18 fatty acids in milk tended to be positively related to apparent uptakes of VLDL triglyceride and to VLDL C18 fatty acids, but negatively related to apparent uptakes of LDL triglycerides and LDL C18 fatty acids. It is suggested that the increased LDL1 resulted from the utilization of VLDL triglyceride for milk fat formation.Protected lipid feeding increased the proportion of C14:0, C16:0 C16:1 and C18:1 and decreased the proportions of C14:1 and C18:0 fatty acids in jugular serum triglycerides. Similar changes were observed in jugular VLDL triglycerides. Differences in the compositions of VLDL and LDL triglycerides across the mammary gland were observed and attributed either to selective uptake or to interchange of fatty acids between triglycerides and free fatty acids.

SummaryThree experiments were conducted to determine the effect of feeding polyunsaturated fish oils to dairy cows receiving rations of high, medium or low roughage content. In the rumen, the decreases in the proportion of acetic acid and increases in propionic acid induced by the oils became greater as the amount of oil given was increased but the magnitude of the response to any dose depended upon the composition of the basal diet. The effects on volatile fatty acids (VFA) proportions of small amounts of the oils (125–150 ml/day) were greatest with the low-roughage rations. With large doses of oil (375–450 ml/day) the responses were variable but it is concluded that, in general, changes in VFA proportions are least with low-roughage rations.The fat content of milk was more sensitive to dietary oil supplementation than were the rumen VFA proportions. When the unsaturated oils were given there was a decrease in milk fat percentage and an increase in the proportion of unsaturated fatty acids in the fat; there was also increased incorporation of fatty acids with more than 18 carbon atoms in the milk fat. The metabolism in the rumen tended to become adapted to the feeding of 150 ml/day of oil, the VFA pattern returning during the second and third week of supplementation to that observed before the addition of oil.

The objective of the present article was to summarise the effects of five common dietary fatty acids (C16:0, C18:0, C18:1, C18:2 and C18:3) on the major milk fat groups (<C16, C16 and C18). Forty published papers were reviewed to evaluate the effect of adding free fat or oil supplements rich in C16 and C18 fatty acids on the response of milk fat secretion and composition. From those 40 studies, 21 were used to investigate the effect of total dietary concentration of C16:0, C18:0, C18:1, C18:2 and C18:3 on milk secretion or concentrations of milk <C16, C16 and C18 fatty acid groups. The results indicated that C16 supplementation increased total milk fatty acids, mainly by increasing milk C16 yield, without affecting milk <C16 and C18 yield. Supplements rich in unsaturated fatty acid decreased total milk fatty acid by inhibiting secretion of milk fatty acids shorter than C18, with linoleic acid being the most inhibitory. Mixtures of feed fatty acid (C16:0 + C18:0 and C16:0 + C18:1) did not significantly affect total milk fatty acid yield. According to regression of milk C16 yield on dietary fatty acid, endogenous C16 contributes ~80% of total milk C16, but this proportion varies with the level and type of dietary fatty acid fed. Milk mid-infrared analysis can be used to routinely measure the presence of milk <C16 fatty acid, the concentration of which provides a good indicator of inhibition of milk fatty acid secretion. In contrast, measurement of total milk fat content is less effective as a diagnostic tool due to the masking effect of the exogenous supply of C16 and C18 dietary fatty acids.

1. The effects, on the secretion in milk of fat and its component fatty acids, of supplementing a basal diet low in fat with red palm oil at three levels are reported.2. The secretion of total fat and of the fatty acids contained in the oil supplement was positively correlated with the dietary intakes, except for linoleic acid which was hydrogenated in the rumen before its absorption and secretion in milk.3. Evidence for the synthesis de novo of palmitic acid and for the possible depression of intramammary de novo synthesis by high levels of dietary fat is discussed.

The rumen contains abundant microorganisms that aid in the digestion of lignocellulosic feed and are associated with host phenotype traits. Cows with extremely high milk protein and fat percentages (HPF; n = 3) and low milk protein and fat percentages (LPF; n = 3) were selected from 4000 lactating Holstein cows under the same nutritional and management conditions. We found that the total concentration of volatile fatty acids, acetate, butyrate, and propionate in the rumen fluid was significantly higher in the HPF group than in the LPF group. Moreover, we identified 38 most abundant species displaying differential richness between the two groups, in which Prevotella accounted for 68.8% of the species, with the highest abundance in the HPF group. Functional annotation based on the Kyoto Encyclopedia of Gene and Genome (KEGG), evolutionary genealogy of genes: Non-supervised Orthologous Groups (eggNOG), and Carbohydrate-Active enzymes (CAZy) databases showed that the significantly more abundant species in the HPF group are enriched in carbohydrate, amino acid, pyruvate, insulin, and lipid metabolism and transportation. Furthermore, Spearman’s rank correlation analysis revealed that specific microbial taxa (mainly the Prevotella species and Neocallimastix californiae) are positively correlated with total volatile fatty acids (VFA). Collectively, we found that the HPF group was enriched with several Prevotella species related to the total VFA, acetate, and amino acid synthesis. Thereby, these fulfilled the host’s needs for energy, fat, and rumen microbial protein, which can be used for increased biosynthesis of milk fat and milk protein. Our findings provide novel information for elucidation of the regulatory mechanism of the rumen in the formation of milk composition.

Glutamate, succinate, propylene glycol, or glycerol were added to a basic concentrate at 3.3% of air-dry feed. Each concentrate was fed both ad libitum and in restricted amounts to four cows in early lactation. Dietary intake, milk yield and composition, molar proportions of rumen volatile fatty acids and blood glucose, ketones and plasma free fatty acids were used as criteria of effect of these supplements. Propylene glycol in the diet resulted in a lower intake of concentrate compared with glycerol (11.44 versus 14.30 kg/day) and significantly decreased (P < 0.05) rumen butyrate and plasma beta-hydroxybutyrate. Glutamate supplementation prevented the fall in milk fat content which occurred when the other three supplemented concentrates were fed ad libitum, and this effect may have been related to the constancy in the ratio of acetate to propionate in the rumen fluid.

The objective of the present study was to describe the intake of dietary fatty acids among healthy 15-year-old boys and girls and to relate the intake of specific fatty acids and the fatty acid composition of the serum cholesterol esters to serum lipid, apolipoprotein (Apo) and insulin concentrations respectively. Fifty-two girls and forty-two boys were randomly selected from the official population register. Unexpectedly, significant inverse associations were found between the dietary content of saturated fatty acids with a chain length of four to fifteen C atoms, mainly derived from milk fat, as well as the corresponding fatty acids in the serum cholesterol esters, on the one hand and the serum concentrations of cholesterol and ApoB on the other. The estimated dietary intake of 4:0–10:0, 12:0 and 14:0 respectively, were all significantly inversely related to the serum cholesterol (r-0.32,r-0.31,r-0.30, all P<0.05) and ApoB (r-0.42,r-0.42, andr-0.40, all P<0.05) concentrations in girls and 12:0 to the ApoB concentration (r-0.55, P<0.01) in boys. The proportions of 12:0 and 15:0 in the serum cholesterol esters were negatively correlated with the serum cholesterol concentrations in both girls (r-0.34,r-0.32, P<0.05) and boys (r-0.53, P<0.01;r-0.32, P<0.05) and with the ApoB concentrations among boys (r-0.61, P<0.01;r-0.43, P<0.05). It is conceivable that milk fat contains or is associated with some component in the diet, or some other characteristics of the food intake, which counterbalances the expected positive relationships between saturated fat intake and lipid levels.

SummaryThe effects of the isocaloric replacement of part of the dietary concentrate mixture by cottonseed oil on the yield and composition of the milk fat and on the pattern of rumen fermentation was investigated in 2 feeding experiments with a total of 8 cows in mid-lactation. The concentrate mixtures were given with high- or low-roughage diets that supplied 9·1 or 1·8 kg of hay/day.In expt 1 the yield of milk fat was not altered by the addition of 10% cottonseed oil to the concentrate mixtures given either with the high- or with the low-levels of dietary roughage. On the low-roughage treatment, dietary cottonseed oil increased milk yield but reduced the fat content of the milk. The change from the high- to the low-roughage diets containing no cottonseed oil resulted in reductions in the yield and percentage of fat in the milk. In expt 2 the yields of milk and milk fat were similar irrespective of whether the concentrate mixture contained 5 or 10% cottonseed oil.In expt 1 the inclusion of 10% cottonseed oil in the concentrate mixture reduced the yields and percentages of the medium-chain fatty acids (12:0, 14:0 and 16:0) and increased the yields and percentages of the C18 fatty acids in the milk fat. In expt 2, when the concentrate mixture contained 5% cottonseed oil, the yields and percentages of all the fatty acids in the milk fat were similar to the values obtained when the concentrates contained 10% cottonseed oil. For any given concentrate mixture, the change from the high- to the low-roughage treatments in both expts 1 and 2 resulted in increases in the percentage of oleic acid in the milk fat. The highest concentration of trans-octadecenoic acid was observed in the milk fat of the cows when they were given the high-roughage diet with the concentrate mixture containing 10% cottonseed oil.In expt 2 the level of cottonseed oil in the concentrate mixture did not influence the pattern of rumen fermentation as measured by the concentrations of the various volatile fatty acids in the rumen liquor. However, the change from the high- to the low-roughage diets reduced the proportion of acetic and increased the proportions of propionic and n-valeric acids in the total volatile fatty acids in the rumen liquor.

AbstractIt may be desirable to increase the level of conjugated linoleic acid (CLA) in milk as a health benefit in human nutrition. The purpose of this work was to separate the effects of linoleic and linolenic acids on CLA production in dairy cows and to determine to what extent endogenous synthesis contributes to cis-9, trans-11 CLA concentration in milk fat. Eight lactating cows and four non-lactating duodenal fistulated cows were used in a 4 ✕ 4 Latin-square design. All cows received a basal diet of grass silage that was supplemented with one of four concentrates, which were designed to differ in their linoleic and linolenic acid contents. The oil components of the concentrates were produced from mixtures of olive, linseed, rape, soya and sunflower oils to produce the four treatments: low linoleic/ low linolenic acid (LL), low linoleic/high linolenic acid (LH), high linoleic/low linolenic acid (HL) and high linoleic/ high linolenic acid (HH). Milk cis-9, trans-11 CLA contents were 0·8, 0·9, 0·9 and 1·1 g/100 g fatty acid methyl esters (P < 0·05) and yields were 5, 7, 7 and 8 g/day (P < 0·05) for the LL, LH, HL and HH treatments, respectively. The yields of trans-C18:1 fatty acids in milk were 19, 22, 21 and 23 g/day (P < 0·05), respectively. Taking the data for the cis-9, trans-11 CLA content and flow of duodenal fluid from the fistulated cows and representing this in terms of dietary intake by the lactating animals, the amounts of cis-9, trans-11 CLA produced in the rumen were calculated to be 0·8, 0·9, 1·2 and 1·1 g/day (P < 0·05) and for trans-C18:1 fatty acids 58, 58, 66 and 69 g/day (P < 0·05). Increasing linoleic and/or linolenic acids in the diet can increase the cis-9, trans-11 CLA content of cows’ milk. Only diets high in linoleic acid increased cis-9, trans-11 CLA production in the rumen. On all four diets, more than 80% of cis-9, trans-11 CLA in milk was produced endogenously by Δ9-desaturase from trans-11 C18:1 in the mammary gland. Cows on the same diet have different milk fat cis-9, trans-11 CLA concentrations that may be partially explained by differences in Δ9-desaturase activity between cows. Increasing the activity of Δ9-desaturase in the mammary gland may offer greater potential for enhancing the cis-9, trans-11 CLA content of milk fat than increasing cis-9, trans-11 CLA production in the rumen.

Nine lambs fitted with rumen cannulae were used in two 5 × 5 Latin squares to measure the effect of alfalfa hay, C4- and C5-branched- and straight-chain volatile fatty acids (C4/C5 VFAs), cell wall from alfalfa hay (ALFCW) and a soybean protein preparation on intake and rumen digestion of a basal corncob diet with urea included at 30 g kg−1. Relative to the control, alfalfa hay decreased (P < 0.05) intake of the basal diet but increased (P < 0.05) total dry matter intake. ALFCW and soybean protein did not affect intake of the basal diet and total dry matter (P > 0.05). C4/C5 VFAs increased intake of basal diet by sheep in one square (P < 0.05) but had no effect (P > 0.05) on intake with sheep in the other square. Alfalfa hay, ALFCW and soybean protein increased (P < 0.05) total VFA concentration in rumen fluid while only alfalfa hay and C4/C5 VFAs consistently increased isobutyrate, isovalerate and valerate levels in rumen fluid (P < 0.05). In comparison with the control diet, soybean protein elevated (P < 0.05) mean rumen ammonia-N levels whereas C4/C5 VFAs depressed them; however, values exceeded 17 mg dL−1 rumen fluid on all treatments. Most measures of digestion kinetics of fiber from corncobs were unaffected by supplementation (P > 0.05). Alfalfa hay and ALFCW increased (P < 0.05) rate of passage of digesta from the reticulo-rumen. It was concluded that alfalfa hay has a reduced role in improving intake of corncob diets when there is also sufficient NPN in the diet.
