Effect of fermented whey with a probiotic bacterium on gut immune system

AbstractThe aim of the work presented in this Research Communication was to evaluate the effect of fermented whey (FW) with Lactobacillus rhamnosus RC007 in a mice model. BALB/c mice were divided into three groups: control group: animals received orally 0.1 ml of phosphate buffered saline (PBS); FW group: animals received orally 0.1 ml of FW; whey (W) group: animals received orally 0.1 ml of W without fermentation with probiotic bacterium. After 10 d mice were sacrificed. Small intestines were collected for determination of IL-10; IL-6, TNFα, goblet cells and intraepithelial lymphocytes. Increases of all the cytokines assayed were observed in mice that received FW compared to control and W group. The ratio between the anti and pro-inflammatory cytokines (IL-10/TNFα) increased in the group of mice that received FW. The number of goblet cells and intraepithelial lymphocytes were also increased in animals that received FW. The results showed that FW with L. rhamnosus RC007 was able to stimulate and to modulate mouse immune system. Whey fermented by this probiotic bacterium is an interesting alternative for development of a new food additive for pig production, taking advantage of the beneficial properties of probiotic bacterium and the nutritional properties of whey.

In this paper, it is shown that the modeling process of bronchial asthma (BA ) is accompanied by changes, in comparison with the control group, in the indicators of immune system response: T- & B-lymphocytes, circulating immune complexes in blood of guinea pig males on the 4th, 18th, 25th day of experiment.The aim of the study – determination of some indices of immune system in blood of guinea pigs in the modeling process of BA on 4th, 18th, 25th day of the experiment.Materials and Methods. Experiments were conducted on 40 guinea pigs (males), with body weight 0.25–0.27 kg. Animals were divided into four groups of ten animals in each. Intact guinea pigs were the first group. Animals with experimental BA – the second, third, fourth group respectively on the 4th, 18th, 25th day of the experiment. Experimental BA was reproduced by V. I. Babych method. In blood of intact guinea pigs and animals with experimental BA , the number of T- and B-lymphocytes was determined by the method of E. F. Chernushenko, L. S. Kohosov, determination of the level of circulating immune complexes was carried out by the method of V. Haskova and co-authors. The results of the study were processed by the method of variation statistics using Student's criterion.Results and Discussion. The results of the studies showed unidirectional changes in individual parameters of the immune system, depending on the periods of the formation of BA : an increase in the number of B-lymphocytes and circulating immune complexes, a decrease in the number of T-lymphocytes for all of the studied days of the experiment.Conclusions. The obtained results indicate significant changes in the immune system parameters in the blood of experimental animals with BA and are important for understanding the pathogenesis of BA . These studies provide an opportunity to find the more perfect and effective methods of diagnosis of BA.

The influence of trauma on parameters of humoral immunity changes has been studied. The study includes 52 patients with various forms of traumatic illness, aged 18-69 years (37,91±4,28). The control group consisted of 16 patients who had undergone a routine surgery not related to the pathology of musculoskeletal system. All patients of the main group were divided into 3 groups according to the severity of their condition. Analysis of parameters of cellular link of immune system was performed by defining the pool of B-lymphocytes (CD22+ lymphocytes), determination of main classes' immunoglobulins, and calculation of integrated indicators. The highest expression (immune disorders of II-III grades) of changes of cellular immunity was observed in patients with severe traumatic illness (detailed clinical picture). Surgical intervention, even without traumatic injury significantly affect cellular immunity, but in patients with traumatic illness immunity disorders were significantly higher than in the comparison groups.

The intestinal ecosystem contains a normal microbiota, non-immune cells and immune cells associated with the intestinal mucosa. The mechanisms involved in the modulation of the gut immune system by probiotics are not yet completely understood. The present work studies the effect of a fermented milk containing probiotic bacteriumLactobacillus(Lb.)caseiDN114001 on different parameters of the gut immune system involved with the nonspecific, innate and adaptive response. BALB/c mice received the probiotic bacteriumLb. caseiDN114001 or the probiotic fermented milk (PFM). The interaction of the probiotic bacteria with the intestine was studied by electron and fluorescence microscopy. The immunological parameters were studied in the intestinal tissue and in the supernatant of intestinal cells (IC). Results showed that the probiotic bacterium interact with the IC. The whole bacterium or its fragments make contact with the gut associated immune cells. The PFM stimulated the IC with IL-6 release, as well as cells related to the nonspecific barrier and with the immune cells associated with the gut. This last activity was observed through the increase in the population of different immune cells: T lymphocytes and IgA+ B lymphocytes, and by the expression of cell markers related to both innate and adaptive response (macrophages). PFM was also able to activate the enzyme calcineurine responsible for the activation of the transcriptional factor NFAT. PFM induced mucosal immune stimulation reinforcing the non-specific barrier and modulating the innate immune response in the gut, maintaining the intestinal homeostasis.

The aim of this study was to develop a novel therapeutic method for improving immune system regulation in SLE using escalating dose self-antigen dsDNA immunotherapy. Methods: Female Balb/c mice were given a single intraperitoneal injection of  0.5 ml pristane.. Starting at 12 weeks after injection,. the mice were evaluated for clinical and serological manifestations. Mice with lupus signs (PIL mice) were divided into four groups; positive control group, PIL A (0.01 μg/ml, 0.1 μg/ml, 1 μg/ml ) EDI dsDNA, PIL B (0.1 μg/ml, 1 μg/ml, 10 μg/ml ) EDI dsDNA, and PIL C(1 μg/ml, 10 μg/ml, 100 μg/ml). EDI dsDNA were administered once every week in consecutively. The doses would increase every week. dsDNA were complexed with the cationic polyethylenimine (PEI) before injection. Samples  were analyzed for autoantibodies levels (dsDNA, ANA) and ,TGF-β cytokine  from serum using ELISA and T-Reg, mature dendritic cells  from spleen using flowcytometry.. Results: Escalating dose antigen spesific immunotherapy with self-antigen dsDNA significantly decreased ANA (p=0.02), anti-dsDNA (p=0.03), dendritic cell mature  (p=0.02) compare to positive control, and not significantly decreases Th17 cells (p=0,18) but the result tend to get lower. Desensitization using self-antigen dsDNA was increased T-reg proliferation (p=0.00) and level of TGF-β (p=0.03) significantly compare to positive control. Conclusion: Desensitization using self-antigen dsDNA coupled to PEI was able to modulate T-Reg as a regulator immune respon and inhibit B and T cell functions in lupus mice model.

SummaryIn fish, many factors can affect reproduction during in vitro fertilization, therefore determination of the factors that affect affecting gamete quality is needed. However, few studies have focused on gamete quality and the ploidy status. This study was conducted to elucidate whether oocyte storage can affect ploidy status, survival, and embryo viability in the characid species Astyanax altiparanae. Oocytes were stored in Dulbecco's phosphate-buffered saline (PBS) at 26°C, then aliquots were fertilized immediately after extrusion (control) and also after 60, 120, 180, and 240 min of storage. Fertilization and hatching rates were measured, and the developmental stages were analyzed at each stage before describing the main abnormalities. Ploidy status was analyzed by flow cytometry and blood smear. In the control group, 100% of the samples were diploid. After treatment for 60 min, 95.56 ± 4.44% samples were diploid and 4.44 ± 4.44% were triploid. After 120 min, 94.44 ± 9.62% of the samples was diploid and 5.56 ± 5.56% were triploid; 100% of the samples were diploid after 180 min and, after 240 min, there was no survival. In other treatments, the highest percentage of hatching was after 60 min (88.93 ± 5.15%; P = 0.015), and treatment with 180 min storage resulted in the highest percentage of abnormal larvae (95.76 ± 12.67%; P = 0.012). These results show that oocyte storage can affect ploidy status and may be an interesting parameter for analysis in studies on chromosome set manipulation and micromanipulation.

ABSTRACT The fractional determination of 17-ketosteroids in the daily urine was performed in nine cases of hyperemesis gravidarum and in four control cases, in the first trimester of pregnancy both before and after corticotrophin administration. The excretion of total 17-KS is similar in the two groups. Only in the hyperemesis group does the excretion of total 17-KS increase significantly after corticotrophin administration. The fractional determination reveals no difference between the two groups of patients with regard to the values of the fractions U (unidentified 17-KS), A (androsterone) and Rest (11-oxygenated 17-KS). The excretion of dehydroepiandrosterone is significantly higher in the hyperemesis group than in the control group. The excretion of androstanolone seems to be lower in the hyperemesis group than in the control group, but the difference is not statistically significant. The differences in the correlation between dehydroepiandrosterone and androstanolone in the two groups is significant. The high excretion of dehydroepiandrosterone and low excretion of androstanolone in cases of hyperemesis gravidarum is a sign of adrenal dysfunction.

ABSTRACT Various techniques for sensitising erythrocytes and latex particles with gonadotrophins, particularly with HCG, are described. The haemagglutination inhibition reactions are generally interpreted by means of »erythrocyte settling patterns«. By a new method of evaluating these patterns a relatively precise quantitative determination is possible. Latex agglutination inhibition reactions on slides are particularly suitable as rapid qualitative tests. In cases where the maximum attainable sensitivity of the agglutination inhibition tests is insufficient, e. g. for determining LH concentrations in urine, the hormone in the test fluid has to be concentrated or extracted. An alternative method is a modified haemagglutination inhibition test for large volumes which is applicable to unconcentrated urine. Due to non-specific inhibitions the above-mentioned tests cannot be applied to unprocessed serum. Agglutination inhibition tests with HCG are already well advanced, pregnancy diagnosis being their main application. Now that highly purified HCG is available, a satisfactory specificity for these tests can be attained. If the immune system for HCG is used for estimating LH, it has to meet additional specificity requirements. Furthermore, the measure of cross-reaction and the choice of standard merit special attention. Finally, a literature survey is given of test systems in which LH and FSH were used as antigens.

The objective: the study of immunomodulatory effects of a probiotic, which contains lyophilized Lactobacillus (Lactobacillus rhamnosus) – 13 mg (2,0ґ109 CFU) and lyophilized bifidobacteria (Bifidobacterium lactis) – 4 mg (2,0ґ109 CFU) the level of serum immunoglobulin IgA as a marker of local immunity in the plasma of women of reproductive age with the violation of the biocenosis of the vagina. Patients and methods. The study involved 86 patients of reproductive age with the violation of the vaginal biocenosis, which were divided into two groups according to received treatment. A survey was conducted for all patients in both groups: determine the level of serum IgA, measuring pH of vaginal environment and the quantification of lactobacilli and pathogenic flora with the help of test-system «Florotsenoz» before treatment and in 6 weeks after treatment. The state of vaginal microbiocenosis in both groups before treatment was homogeneous. Patients in both groups as therapy at the first stage of treatment received, if necessary antimicrobial therapy depending on the selected flora. In the second stage (restoration of microflora) patient of the main group received systemic probiotic combined with a complex prebiotic local action, patients in the control group, the probiotic localy in the form of the vaginal candles or tablets. Results. The research stated the increasing level of serum IgA in blood plasma of patients of the main group compared to control group at 20%, normalizing the pH of the vaginal environment in the main group in 94% of cases, which indicates an increase of immunity in mucosal. Conclusion. The inclusion of the systemic probiotic in the scheme of treatment of disorders of biocenosis of the vagina system enhances the increasing of immunity of the mucous membranes, and the vaginal tablets prebiotic of local action restores the own normal microflora of the vagina. Aim. To study the influence of deproteinized dialysate from blood of milk-fed calves on the type of exudative discharge and dynamics of inflammatory reaction after hernia repair with plastics of the anterior abdominal wall with hernioendoprosthesis of polypropylene. Materials and Methods. The study involved 59 patients being on stationary treatment in the surgical department of Kursk Regional Clinical Hospital. Patients were hospitalized for the herniation of small or medium dimension. The patients were divided to two groups: the main (n=30) and control (n=29) groups. After endoprosthetics, the patients of the control group were given complex conservative treatment. The patients of the main group, besides standard treatment, were administered deproteinized dialysate from blood of milk-fed calves intravenously in drips 10 ml + 200 ml of 0.9% sodium chloride solution within 7 days. For cytological examination and determination of the type of cytograms, the traumatic discharge was collected and analyzed using the method of M.F. Kamaev and M.A. Palthsev. Results. Cytomorphometric examination was conducted on the third, fifth and seventh day after endoprosthetics with the aim of studying dynamic changes. Determination of cell composition and also of its changes characteristic of each studied period, was necessary for obtaining further information characterizing inflammatory process in the region of placement of the endoprosthesis. After endoprosthetics in patients who were administered hemodialysate, the inflammatory reaction was less pronounced than in patients who did not receive the preparation. This was associated with a more dynamic change of stages of the inflammatory process. In patients of the main group who received deproteinized dialysate, regenerative type of inflammation first appeared on the fifth day and made 6.9%; by the seventh day the share of patients with the regenerative type rose to 17.5%, while in the patients of the control group no regeneration stage was observed in both periods. Conclusion. Analysis of the efficiency of influence of hemodialysate on the inflammatory reaction in plastics of the anterior abdominal wall with polypropylene endoprosthesis evidences faster course of all stages of inflammation and reduction of its intensity by 10%.

Retrograde menstruation lead to I Kappa B Kinase (IKK) fosforilation in peritoneum macrophage and cause secretion of proinflammatory cytokine interleukin1β then stimulate endometriosis cell to produce Vascular Endothelial Growth Factor which lead to increasing of endometriosis lession seen as endometriosis implant area. Cytokine secretion was inhibited through prevention of NF-κB activation by dragon red fruit rind extract (Hylocereuspolyrhizus). The aim of this reserach is to know the effect of dragon red fuit rind extract with 0,25; 0,5; and 1 mg/g bodyweight dosage toward IL-1β, VEGF expression and implant area in endometriosis mice model. The design of this experiment was randomized post test only control group design.Endometrios mice model were made in 14 days and split into two group, positive control group and treatment group after two week negative control group and postive control group were given Na-CMC 0,5% solution consequetively, and treatment group were given dragon red fruit extract with different dosage. Signification number for IL-1β is p>0,05, signification number for VEGF is p>0,05, and implant area signification number is p>0,05. Administration of dragon red fruit rind extract can decrease IL-1β, VEGF, and implant area.
