AbstractThe microbial food fermentation industry requires real-time monitoring and accurate quantification of cells. However, filamentous fungi are difficult to quantify as they have complex cell types such as pellet, spores, and dispersed hyphae. In this study, numerous data of microscopic image intensity (MII) were used to develop a simple and accurate quantification method of Cordyceps mycelium. The dry cell weight (DCW) of the sample collected during the fermentation was measured. In addition, the intensity values were obtained through the ImageJ program after converting the microscopic images. The prediction model obtained by analyzing the correlation between MII and DCW was evaluated through a simple linear regression method and found to be statistically significant (R2 = 0.941, p < 0.001). In addition, validation with randomly selected samples showed significant accuracy, thus, this model is expected to be used as a valuable tool for predicting and quantifying fungal growth in various industries.

AbstractSulfated glycosaminoglycans (GAGs) are a class of important biologics that are currently manufactured by extraction from animal tissues. Although such methods are unsustainable and prone to contamination, animal-free production methods have not emerged as competitive alternatives due to complexities in scale-up, requirement for multiple stages and cost of co-factors and purification. Here, we demonstrate the development of single microbial cell factories capable of complete, one-step biosynthesis of chondroitin sulfate (CS), a type of GAG. We engineer E. coli to produce all three required components for CS production–chondroitin, sulfate donor and sulfotransferase. In this way, we achieve intracellular CS production of ~27 μg/g dry-cell-weight with about 96% of the disaccharides sulfated. We further explore four different factors that can affect the sulfation levels of this microbial product. Overall, this is a demonstration of simple, one-step microbial production of a sulfated GAG and marks an important step in the animal-free production of these molecules.

Myrosinase is a plant defence enzyme catalysing the hydrolysis of glucosinolates, a group of plant secondary metabolites, to a range of volatile compounds. One of the products, isothiocyanates, proved to have neuroprotective and chemo-preventive properties, making myrosinase a pharmaceutically interesting enzyme. In this work, extracellular expression of TGG1 myrosinase from Arabidopsis thaliana in the Pichia pastoris KM71H (MutS) strain was upscaled to a 3 L laboratory fermenter for the first time. Fermentation conditions (temperature and pH) were optimised, which resulted in a threefold increase in myrosinase productivity compared to unoptimised fermentation conditions. Dry cell weight increased 1.5-fold, reaching 100.5 g/L without additional glycerol feeding. Overall, a specific productivity of 4.1 U/Lmedium/h was achieved, which was 102.5-fold higher compared to flask cultivations.

The problem of the SMA RK Deli Murni Bandar Baru school is to predict how many facilities that need to be provided for new students such as chairs, tables and others. This study discusses the prediction of the number of new student registrants at SMA RK Deli Murni Bandar Baru based on the amount of tuition fees using a simple linear regression method. From a commercial point of view, the use of data mining can be used to handle the explosion of data volumes, using computational techniques can be used to produce information needed which is an asset that can increase the competitiveness of an institution. Prediction is almost the same as classification and estimation, except that in the prediction the value of the results will be in the future. This system can be used to predict the number of applicants in the following year to help the school. The advantage is that this simple linear regression method is very simple so that it is easy to calculate and use. Saves the time needed to solve problems, especially those that are very complex.

Starting from the phenomenon occurred especially among Muslim women who were very concerned with fashion issues. Many of them did not know the function of the clothes they wear, and how Islam arranges the clothes. The purpose of this study was to determine the influence of religiosity on the decision of purchasing Muslim clothes in Tembilahan District. This was quantitative research by simple linear regression method analysis. The results showed that the T count was bigger than the T table. The T table was 12,763>2,024 and sig. 0.000 <0.05 means Ha was accepted, and Ho was rejected. The contribution of religiosity to the decision toward purchasing Muslim clothing by looking at the results of the R Square was 81.1%, while the remaining 18.9% influenced by other factors outside of the researcher's discussion.

The aim of this work was to propose a method for the consistency of climatic series of monthly rainfall using a supervised and unsupervised approach. The methodology was applied for the series (1961-2010) of rainfall from weather stations located in the State of Rio de Janeiro (RJ) and in the borders with the states of São Paulo, Minas Gerais and Espírito Santo with the State of Rio de Janeiro. The data were submitted to quality analysis (physical and climatic limit and, space-time tendency) and gap filling, based on simple linear regression analysis, associated with the prediction band (p < 0.05 or 0.01), in addition to the Z-score (3, 4 or 5). Next, homogeneity analysis was applied to the continuous series, using the method of cumulative residuals. The coefficients of determination (r²) between the assessed series and the reference series were greater than 0.70 for gap filling both for the supervised and unsupervised approaches. In the analysis of data homogeneity, supervised and unsupervised approaches were effective in selecting homogeneous series, in which five out of the nine final stations were homogeneous (p > 0.9). In the other series, the homogeneity break points were identified and the simple linear regression method was applied for their homogenization. The proposed method was effective to consist of the rainfall series and allows the use of these data in climate studies.

The study aimed to determine the effectiveness of docosahexaenoic acid (DHA) production by Schizochytrium sp. biomass fed with waste glycerol depending on the concentration of extracellular polymeric substances (EPS) in the culture medium and medium aeration effectiveness. The microalgae from the genus Schizochytrium sp. were proved to be capable of producing EPS composed of glucose, galactose, mannose, fucose, and xylose. The highest EPS concentration, reaching 8.73 ± 0.09 g/dm3, was determined at the stationary growth phase. A high EPS concentration caused culture medium viscosity to increase, contributing to diminished oxygen availability for cells, lower culture effectiveness, and reduced waste glycerol conversion to DHA. The Schizochytrium sp. culture variant found optimal in terms of the obtained technological effects and operating costs was performed at the volumetric oxygen mass transfer coefficient of kLa = 600 1/h, which enabled obtaining dry cell weight (DCW) of 147.89 ± 4.77 g/dm3, lipid concentration of 69.44 ± 0.76 g/dm3, and DHA concentration in the biomass reaching 29.44 ± 0.36 g/dm3. The effectiveness of waste glycerol consumption in this variant reached 3.76 ± 0.31 g/dm3·h and 3.16 ± 0.22 g/gDCW.

Study aims to determine and obtain a clear picture of supportive supervisor, empowerment, and organizational citizenship behavior; and to determine the impact of supportive supervisor and empowerment towards organizational citizenship behavior of employees in PT Setia Makmur Cemerlang. Research used explanatory survey method, while the sample was taken from employees of the company. Research instrument (questionnaires) was used as primary data collection to explain the causal relationship between supportive supervisor and empowerment on organizational citizenship behavior of employees in PT Setia Makmur Cemerlang. Analysis used simple linear regression and multiple linear regression method. Result of this study shows that there is significant influence between supportive supervisor and empowerment towards organizational citizenship behavior.
