AbstractThe odonates are insects that have a wide range of reproductive, ritualized territorial, and aggressive behaviors. Changes in behavior are the first response of most odonate species to environmental alterations. In this context, the primary objective of the present study was to assess the effects of environmental alterations resulting from shifts in land use on different aspects of the behavioral diversity of adult odonates. Fieldwork was conducted at 92 low-order streams in two different regions of the Brazilian Amazon. To address our main objective, we measured 29 abiotic variables at each stream, together with five morphological and five behavioral traits of the resident odonates. The results indicate a loss of behaviors at sites impacted by anthropogenic changes, as well as variation in some morphological/behavioral traits under specific environmental conditions. We highlight the importance of considering behavioral traits in the development of conservation strategies, given that species with a unique behavioral repertoire may suffer specific types of extinction pressure.

SummaryWe propose an approach to conservation centred on achieving positive future trajectories of dynamic change, applied to all locations and species, and based on societal inclusiveness. Strategies to facilitate change. We take an Anthropocene perspective, in which human society and biodiversity have been inextricably linked for over 10,000 years, and continuing biodiversity change is inevitable. The challenge is to identify circumstances under which change is acceptable or beneficial, without being tied to specific historic baselines. We outline a Resist-Accept-Facilitate (RAF) framework that could be applied to all conservation activities, from high-level planning and measurement (indicators) through to practical land and species management, to ensure that the facilitation of future biodiversity benefits receives as much attention as the resistance of change.Everywhere is important. Different places are important for different things, such as particular species or ecosystem services, and people vary in how they value these features. We suggest a perspective whereby we evaluate what every area is most important for, and what they could be most important for in future by considering possible trajectories of biodiversity and ecosystem change. We propose zoning any region of interest, such as the UK, and applying the RAF framework in an inclusive manner to develop conservation strategies that are appropriate in each location and zone. This RAF approach will reconcile different conservation philosophies (such as traditional management, land-sharing/sparing, rewilding, novel ecosystems, ecosystem services, human wellbeing) because different conservation outcomes will emerge in different zones.Enabling species to move. Genes and species undertake changes to their abundances and distributions in response to climatic and other environmental changes. We suggest that trans situ conservation be developed as a complement to traditional in situ (sites in the wild) and ex situ (in captivity) conservation. Trans situ conservation primarily involves Accept and Facilitate interventions within the RAF framework. It requires consideration of the connectedness and permeability of regions (facilitating colonisation for a majority of species and genes) and the value of new populations of colonising species and novel communities that arise from species range shifts. Trans situ conservation also considers the potential to translocate (assisted colonisation) species and genes that are unable to shift their distributions without direct intervention, highlighting the transnational needs of globally-threatened species rather than locally-rare ones whose future is secure elsewhere.For everyone. The justification for conservation commonly focuses attention on the benefits that individuals and society derive from the natural world, yet the benefits are not shared equitably. The RAF approach asks people from a wide range of backgrounds ‘what they want’ from local landscapes so as to inform the development of more inclusive approaches to conservation both now and in the future. We identify ways in which the processes of conservation could incorporate an increased diversity of perspectives, whilst continuing to be informed by data and professional expertise.

DNA methylation mediates organisms’ adaptations to environmental changes in a wide range of species. We investigated if a such a strategy is also adopted by Fusarium graminearum in regulating virulence toward its natural hosts. A virulent strain of this fungus was consecutively sub-cultured for 50 times (once a week) on potato dextrose agar. To assess the effect of subculturing on virulence, wheat seedlings and heads (cv. A416) were inoculated with subcultures (SC) 1, 23, and 50. SC50 was also used to re-infect (three times) wheat heads (SC50×3) to restore virulence. In vitro conidia production, colonies growth and secondary metabolites production were also determined for SC1, SC23, SC50, and SC50×3. Seedling stem base and head assays revealed a virulence decline of all subcultures, whereas virulence was restored in SC50×3. The same trend was observed in conidia production. The DNA isolated from SC50 and SC50×3 was subject to a methylation content-sensitive enzyme and double-digest, restriction-site-associated DNA technique (ddRAD-MCSeEd). DNA methylation analysis indicated 1024 genes, whose methylation levels changed in response to the inoculation on a healthy host after subculturing. Several of these genes are already known to be involved in virulence by functional analysis. These results demonstrate that the physiological shifts following sub-culturing have an impact on genomic DNA methylation levels and suggest that the ddRAD-MCSeEd approach can be an important tool for detecting genes potentially related to fungal virulence.

Macadamia is a recently domesticated Australian native nut crop, and a large proportion of its wild germplasm is unexploited. Aiming to explore the existing diversity, 247 wild accessions from four species and inter-specific hybrids were phenotyped. A wide range of variation was found in growth and nut traits. Broad-sense heritability of traits were moderate (0.43–0.64), which suggested that both genetic and environmental factors are equally important for the variability of the traits. Correlations among the growth traits were significantly positive (0.49–0.76). There were significant positive correlations among the nut traits except for kernel recovery. The association between kernel recovery and shell thickness was highly significant and negative. Principal component analysis of the traits separated representative species groups. Accessions from Macadamia integrifolia Maiden and Betche, M. tetraphylla L.A.S. Johnson, and admixtures were clustered into one group and those of M. ternifolia F. Muell were separated into another group. In both M. integrifolia and M. tetraphylla groups, variation within site was greater than across sites, which suggested that the conservation strategies should concentrate on increased sampling within sites to capture wide genetic diversity. This study provides a background on the utilisation of wild germplasm as a genetic resource to be used in breeding programs and the direction for gene pool conservation.

Microglia, besides being able to react rapidly to a wide range of environmental changes, are also involved in shaping neuronal wiring. Indeed, they actively participate in the modulation of neuronal function by regulating the elimination (or “pruning”) of weaker synapses in both physiologic and pathologic processes. Mounting evidence supports their crucial role in early synaptic loss, which is emerging as a hallmark of several neurodegenerative diseases, including multiple sclerosis (MS) and its preclinical models. MS is an inflammatory, immune-mediated pathology of the white matter in which demyelinating lesions may cause secondary neuronal death. Nevertheless, primitive grey matter (GM) damage is emerging as an important contributor to patients’ long-term disability, since it has been associated with early and progressive cognitive decline (CD), which seriously worsens the quality of life of MS patients. Widespread synapse loss even in the absence of demyelination, axon degeneration and neuronal death has been demonstrated in different GM structures, thus raising the possibility that synaptic dysfunction could be an early and possibly independent event in the neurodegenerative process associated with MS. This review provides an overview of microglial-dependent synapse elimination in the neuroinflammatory process that underlies MS and its experimental models.

The longhorn beetle, Callipogon (Eoxenus) relictus Semenov, is the only remnant species found in the Palearctic region, while all other Callipogon species are distributed mainly in Central America and partly in South America. This species has been placed in the first category (as ‘critically endangered’) of the Red Data Book in Russia and designated as one of the top-priority target species among all endangered invertebrate species for restoration in South Korea since 2006. Although its restricted distribution in Northeast Asia with a high conservational value has been highlighted, genetic features of C. relictus from different geographic regions remain unexplored. We first investigated the level of genetic diversity and phylogeographic patterns of C. relictus to evaluate the current conservational status and the feasibility of the implementation of a restoration program. The average genetic divergence of mitochondrial gene COI based on Kimura-2-parameter distance among the four regions in Russia, China, North Korea, and South Korea was 2.2%, which lies within the range of intraspecific levels. However, two separate clades with 3.8% divergence were identified, despite no geographical clustering of haplotypes. The linear pattern of the haplotype network with a high level of haplotype and nucleotide diversities suggests that the wide range of currently fragmented populations might be the remnant of genetically diverse populations in the past. This study will provide crucial information on the genetic characteristics and phylogeographic history of C. relictus, which will help to establish conservation strategies for this cherished insect species in Northeast Asia.

A total of 187 different species of arboviruses and other viruses in vertebrates were identified at the Evandro Chagas Institute (IEC) from 1954 to 1998, among more than 10,000 arbovirus strains isolated from humans, hematophagous insects, and wild and sentinel vertebrates. Despite intensive studies in the Brazilian Amazon region, especially in Pará State, very little is known about most of these viruses, except for information on date, time, source, and method of isolation, as well as their capacity to infect laboratory animals. This paper reviews ecological and epidemiological data and analyzes the impact of vector and host population changes on various viruses as a result of profound changes in the natural environment. Deforestation, mining, dam and highway construction, human colonization, and urbanization were the main manmade environmental changes associated with the emergence and/or reemergence of relevant arboviruses, including some known pathogens for humans.

This work presents an exploratory development of J and CTOD estimation procedures for welded fracture specimens under bending based upon plastic eta factors and plastic rotation factors. The techniques considered include: i) estimating J and CTOD from plastic work and ii) estimating CTOD from the plastic rotational factor. The primary objective is to gain additional understanding on the effect of weld strength mismatch on estimation techniques to determine J and CTOD fracture parameters for a wide range of a/W-ratios and mismatch levels. Very detailed non-linear finite element analyses for plane-strain models of SE(B) fracture specimens with center cracked, square groove welds provide the evolution of load with increased load-line displacement and crack mouth opening displacement which are required for the estimation procedure. The results show that levels of weld strength mismatch within the range ±20% mismatch do not affect significantly J and CTOD estimation expressions applicable to homogeneous materials, particularly for deeply cracked fracture specimens. The present analyses, when taken together with previous studies, provide a fairly extensive body of results which serve to determine parameters J and CTOD for different materials using bend specimens with varying geometries and mismatch levels.

Heat transfer enhancement is a subject of major concern in numerous fields of industry and research. Having received undivided attention over the years, it is still studied worldwide. Given the exponential growth of computing power, large-scale numerical simulations are growing steadily more realistic, and it is now possible to obtain accurate time-dependent solutions with far fewer preliminary assumptions about the problems. As a result, an increasingly wide range of physics is now open for exploration. More specifically, it is time to take full advantage of large eddy simulation technique so as to describe heat transfer in staggered parallel-plate flows. In fact, from simple theory through experimental results, it has been demonstrated that surface interruption enhances heat transfer. Staggered parallel-plate geometries are of great potential interest, and yet many numerical works dedicated to them have been tarnished by excessively simple assumptions. That is to say, numerical simulations have generally hypothesized lengthwise periodicity, even though flows are not periodic; moreover, the LES technique has not been employed with sufficient frequency. Actually, our primary objective is to analyze turbulent influence with regard to heat transfers in staggered parallel-plate fin geometries. In order to do so, we have developed a LES code, and numerical results are compared with regard to several grid mesh resolutions. We have focused mainly upon identification of turbulent structures and their role in heat transfer enhancement. Another key point involves the distinct roles of boundary restart and the vortex shedding mechanism on heat transfer and friction factor.
