AbstractThe purpose of this study was to determine if a methanolic extract of the Pulsatilla patens (L.) Mill. can inhibit the progression of cancer through the modulation of cancer-related metabolic signaling pathways. We analyzed a panel of 13 inducible luciferase reporter gene vectors which expression is driven by enhancer elements that bind to specific transcription factors for the evaluation of the activity of cancer signaling pathways. The root extract of P. patens exhibited strong inhibition of several signaling pathways in HeLa cells, a cervical cancer cell line, and was found to be the most potent in inhibiting the activation of Stat3, Smad, AP-1, NF-κB, MYC, Ets, Wnt and Hdghog, at a concentration of 40 µg/mL. The methanolic extracts of P. patens enhanced apoptotic death, deregulated cellular proliferation, differentiation, and progression towards the neoplastic phenotype by altering key signaling molecules required for cell cycle progression. This is the first study to report the influence of Pulsatilla species on cancer signaling pathways. Further, our detailed phytochemical analysis of the methanolic extracts of the P. patens allowed to deduce that compounds, which strongly suppressed the growth and proliferation of HeLa cancer cells were mainly triterpenoid saponins accompanied by phenolic acids.

The anticancer activities of Rubia cordifolia and its constituents have been reported earlier, but their influence on the crosstalk of complex cancer-related signaling metabolic pathways (i.e., transcription factors; TF) has not yet been fully investigated. In this study, R. cordifolia root extract was subjected to the cancer signaling assay based bioactivity-guided fractionation, which yielded the following compounds viz., three anthraquinones, namely alizarin (1), purpurin (2), and emodin (3); two lignans, namely eudesmin (4) and compound 5; and two cyclic hexapeptides, namely deoxybouvardin RA-V (6), and a mixture of 6+9 (RA-XXI). The structures of the isolated compounds were determined by NMR spectroscopy and HRESIMS. The isolated compounds 1, 2, 3, 6, and a mixture of 6+9 were tested against a panel of luciferase reporter genes that assesses the activity of a wide-range of cancer-related signaling pathways. In addition, reference anthraquinones viz., chrysophanol (11), danthron (12), quinizarin (13), aloe-emodin (14), and α-lapachone (15) were also tested. Among the tested compounds, the cyclic hexapeptide 6 was found to be very active against several signaling pathways, notably Wnt, Myc, and Notch with IC50 values of 50, 75, and 93 ng/mL, respectively. Whereas, the anthraquinones exhibited very mild or no inhibition against these signaling pathways. Compound 6 being the most active, we tested it for stability in simulated intestinal (SIF) and gastric fluids (SGF), since the stability in biological fluid is a key short-coming of cyclic hexapeptides. The anticancer activity of 6 was found to remain unchanged before and after the treatment of simulated gastric/intestinal fluids, indicating that RA-V was stable. As a result, it could be bioavailable when orally used in therapeutics and possibly a drug candidate for cancer treatment. The mechanism for the preferential inhibition of these pathways and the possible crosstalk effect with other previously reported signaling pathways has been discussed.

The present study was aimed to investigate the histochemical and phytochemical studies of four important medicinal plants. Leaf, stem and root sections were examined for alkaloid, protein and phenolic tests. Methanolic extracts of leaf, stem and root were analysed for the phytochemical screening of major bioactive constituents. Histochemical analysis reveals the presence of alkaloids, phenols and proteins in epidermis, cortex and vascular bundles of root, stem and leaf. Preliminary phytochemical analysis showed the presence of alkaloids, tannins, phenols, flavonoids, steroids, saponins, terpenoids, coumarins, quinone and proteins. The root extract of Catharanthus roseus recorded maximum content of phenols compared to other extracts. Costus pictus root and leaf extracts exhibited a significant amount of flavonoids and tannins respectively. Further studies were focused on isolation and characterization of each plant compounds.

Fagonia schweinfurthii Hadidi (family zygophyllaceae) is plant of desert region .In Gujarat it is found on the coastal area of Dwarka, Bet Dwarka and Rann of Kachcha. It is locally called dhamasia or dhamaso. Plants parts including leaves, roots, rhizomes, stems, barks, flowers, fruits, grains or seeds which can be employed in the control or treatment of infectious disease which causes damage to the respiratory system, urinary tract, gastrointestinal and bilary systems and on the skin. The plant parts contain chemical components that are medically active. Tribal people living in desert region use this plant to cure number of ailments such as skin eruptions, skin diseases, anti- pyretic, in pain relief, in heal sores, ear infection, venereal diseases etc. In the new era of science and technology there is increasing need to validate the claims of traditional knowledge database for safe, easily available, cheap, side- effect free healthcare provisions. In the present study preliminary phytochemical analysis of plant parts of Fagonia schweinfurthii Hadidi was done by the standard method of Sofowora, 1982; Trease & Evans, 1989 method, which confirms the presence of alkaloids, tannin, saponin, terpinoid and steroids. High Performance Thin Layer Chromatography method was developed using mobile phase containing Chloroform: methanol: acetic acid in ratio 70:30:0.2 for detecting phyto-active constituents from the methanolic extracts of the leaf, stem and root of Fagonia schweinfurthii Hadidi. Methanolic extract of Fagonia schweinfurthii Hadidi of leaf, stem, fruit and root detected 8, 8, 7 and 4 numbers of peaks at 254 nm and at 366 nm 10, 12,  13 and 12 numbers of peaks from leaf, stem, fruit and root extract of Fagonia schweinfurthii Hadidi were detected.

Sideritis niveotomentosa Hub. -Mor. is a local endemic species belonging to the Lamiaceae family. In this study, GC/MS analysis, total antioxidant capacity and anticancer effects of different extracts obtained from S. niveotomentosa were investigated comparatively. Total phenolic contents of extracts were determined by the Folin–Ciocalteu method, total flavonoid contents by aluminum chloride method, and also the free radical scavenging activities of the extracts by DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay. The cytotoxic effect of the extracts was studied via MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) assay on DLD1, HL60 and ARH77 cell lines. Pro-apoptotic gene expression levels were also tested in the most sensitive cell line ARH77 by Real-Time PCR. The expression levels of 4 pro-apoptotic genes, APAF, BAX, CASP3, and HRK were found to be upregulated in ARH77 cells that were treated extracts. Results showed that methanolic extracts contain more phenolic content than acetone extracts, consistent with DPPH results. As a result, Sideritis niveotomentosa extracts, especially methanolic extracts, are rich in phenolic content and have a strong radical scavenging effect. In addition, the extracts showed selective effects on cell lines. This study is pioneering in terms of future studies, and the findings provide hope for future experimentation.

Genetically uniform plant material, derived from Lychnis flos-cuculi propagated in vitro, was used for the isolation of 20-hydroxyecdysone and polypodine B and subjected to an evaluation of the antifungal and antiamoebic activity. The activity of 80% aqueous methanolic extracts, their fractions, and isolated ecdysteroids were studied against pathogenic Acanthamoeba castellani. Additionally, a Microtox® acute toxicity assay was performed. It was found that an 80% methanolic fraction of root extract exerts the most potent amoebicidal activity at IC50 of 0.06 mg/mL at the 3rd day of treatment. Both ecdysteroids show comparable activity at IC50 of 0.07 mg/mL. The acute toxicity of 80% fractions at similar concentrations is significantly higher than that of 40% fractions. Crude extracts exhibited moderate antifungal activity, with a minimum inhibitory concentration (MIC) within the range of 1.25–2.5 mg/mL. To the best of our knowledge, the present report is the first to show the biological activity of L. flos-cuculi in terms of the antifungal and antiamoebic activities and acute toxicity. It is also the first isolation of the main ecdysteroids from L. flos-cuculi micropropagated, ecdysteroid-rich plant material.

Abstract Background Strobilanthes heyneanus (Nilgirianthus heyneanus) belongs to the family Acanthaceae that contains many species with potential for diverse medicinal uses. It is also called ‘Karun kurinji’ and is commonly found in the South-West regions of India. The species are commonly used in rheumatic complaints, sprain of the ankle, and hernia. The objectives of the study were to evaluate the antioxidant activity, phytochemical analysis, and antibacterial activities of the root extract of S. heyneanus. Results The radical scavenging and reduction assays such as DPPH● radical and OH● radical scavenging assays, as well as phosphomolybdenum reduction and Fe3+ reducing power assays, were determined for the root extract. The highest DPPH● radical scavenging activity was 88.23 ± 1.32 at 120 μg/mL concentration, and the calculated IC50 was 38.52 μg/mL concentration. The highest OH● radical scavenging activity was 51.28 ± 1.06 at 120 μg/mL concentration, and the calculated IC50 was 51.28 μg/mL concentration. The highest ABTS●+ radical scavenging activity was 91.28 ± 1.12 at 30 μg/mL concentration, and the calculated IC50 was 33.92 μg/mL concentration. The highest phosphomolybdenum reduction was 87.43 ± 0.90 at 120 μg/mL concentration, and the calculated RC50 was 24.74 μg/mL concentration. The highest Fe3+ reduction was 89.38 ± 0.98 at 120 μg/mL concentration, and the calculated RC50 was 31.06 μg/mL concentration. The antibacterial activity of S. heyneanus showed the highest zone of inhibition of 24 mm for Salmonella typhi with 500 μg/mL concentration. The cytotoxicity limits of the root extracts were tested by MTT assay using human dermal fibroblast cell lines, reflecting > 90% cell viability at a concentration of 500 μg/mL. Conclusions The current study showed that the root extract of S. heyneanus has better antioxidant properties and potential anti-bacterial compounds. The phytochemical analysis of the root extract showed the presence of alkaloids, steroids, terpenoids, phenols, flavonoids, tannins, saponins, glycosides, and carbohydrates, which are responsible for the antibacterial root extract of S. heyneanus synergistically.

BACKGROUND: The prognosis of lung cancer patients is poor without useful prognostic and diagnostic biomarker. To search for novel prognostic and diagnostic markers, we previously found homeobox-A13 (HOXA13) as a promising candidate in lung cancer. OBJECTIVE: To determine the precisely clinical feature, prognostic and diagnostic value, possible role and mechanism of HOXA13. METHODS: Gene-expression was explored by real-time quantitative-PCR, western-blot and tissue-microarray. The associations were analyzed by Chi-square test, Kaplan-Meier and Cox-regression. The roles and mechanisms were evaluated by MTS, EdU, transwell, xenograft tumor and luciferase-reporter assays. RESULTS: HOXA13 expression is increased in tumors, and correlated with age of patients. HOXA13 expression is associated with unfavorable overall survival and relapse-free survival of patients in four cohorts. Interestingly, HOXA13 has different prognostic significance in adenocarcinoma (ADC) and squamous-cell carcinoma (SCC), and is a sex- and smoke-related prognostic factor only in ADC. Importantly, HOXA13 can serve as a diagnostic biomarker for lung cancer, especially for SCC. HOXA13 can promote cancer-cell proliferation, migration and invasion in vitro, and facilitate tumorigenicity and tumor metastasis in vivo. HOXA13 acts the oncogenic roles on tumor growth and metastasis by regulating P53 and Wnt/β-catenin signaling activities in lung cancer. CONCLUSIONS: HOXA13 is a new prognostic and diagnostic biomarker associated with P53 and Wnt/β-catenin signaling pathways.

The genus Phyllanthus (family Euphorbiaceae) is considered one of the important medicinal and ornamental plants. A phytochemical analysis of the extracts was performed to search for the active ingredient. Results of the investigation of the hepatoprotective activity of Phyllanthus atropurpureus Boj. Hort. Maurit. revealed that the activities of alcoholic extracts of its aerial parts and roots were quite similar to those of silymarin. Both of them improve the parameters of CCl4-induced liver injury including serum aspartate aminotransferase and alanine aminotransferase. Among the extracts tested, the root extract showed maximum activity compared to the aerial parts extract and to silymarin.
