Effects of different monochromatic LED light colours on fear reactions and physiological responses in Mulard ducks

Although the effects of monochromatic light on behaviour and performance of birds have been extensively studied, it is not known how rearing Mulard ducks in different monochromatic lights affects their fear reactions, physiological responses to stress and welfare. A total of 108 newly hatched Mulard ducks, representing three replicates, were housed in either blue light (BL), green light (GL), red light (RL) or white light (WL) for 12 weeks. Ducks were exposed to a light/dark schedule of 23 L/1 D, and food and water were provided ad libitum. At the beginning of the 13th week of the rearing period, behavioural measurements of fear were assessed by using a tonic immobility test, open field test and fear of man test. Physiological responses of the birds to stress such as heterophil : lymphocyte ratio, total leukocyte count and plasma concentrations of corticosterone were also assessed. Results demonstrated that birds exposed to both RL and WL displayed higher levels of behavioural indicators of fear including tonic immobility durations (P < 0.01), latency to first immobilisation (P < 0.001) and avoidance of man index (P < 0.001), and higher levels of physiological indicators of stress such as heterophil : lymphocyte ratios (P < 0.001) and plasmas concentrations of corticosterone (P < 0.001), and lower total leukocytic counts (P < 0.01) compared with birds exposed to either BL or GL. Rearing Mulard ducks in BL or GL appeared to reduce their fear reactions and physiological responses to stress and to enhance their ability to cope with the environment and may therefore improve their welfare.

AbstractFluorescent microscopy employs monochromatic light which can affect the cells being observed. We reported earlier that fibroblasts relax their contractile force in response to green light of typical intensity. Here we show that such effects are independent of extracellular matrix and type of cell. In addition, we establish a threshold light that invokes minimal effect on cells. We cultured fibroblasts on soft 2D elastic hydrogels embedded with fluorescent beads to trace substrate deformation. The beads move towards cell center when cells contract, but they move away when cells relax. We use relaxation/contraction ratio, λr, as a measure of cell response to light. The cells were exposed to green (wavelength, λ = 545-580 nm) and red (λ = 635-650 nm) light with a range of intensities. We find red light with intensity less than ~ 57 W/m2 results in λr = 1, i.e., cells maintain force homeostasis. Higher intensities and smaller wavelengths result in widespread force-relaxation in cells with λr > 1. We suggest the use of λ > 650 nm light with low intensity (I ≤ 57 W/m2) for time-lapse imaging of cells and tissues in order to avoid light-induced artifacts in experimental observations.

Light during incubation can influence embryonic and postembryonic development of chickens, but the underlying mechanisms are poorly understood. Previous studies have demonstrated that red and blue lights during incubation had opposite effects on the development of embryonic melatonin biosynthesis; red light results in the highest and blue light in the lowest amplitude of the daily rhythm. Therefore, in this study, we investigated if exposure to monochromatic red (632 nm) and blue (463 nm) light during incubation can differently influence growth, selected biochemical (glucose, cholesterol, triacylglycerols) and endocrine (corticosterone and thyroid hormones) traits and behavioural parameters during postembryonic development in broiler chickens. For analysis, we used 10 and 11 hatchlings incubated in red and blue light, respectively and 10 birds per each group (six males and four females) in 3-weeks-old broilers. During the rapid growth phase (days 18, 20 and 21 of age), higher body weight was recorded in broilers incubated under red compared to blue light, whereas endocrine and metabolic traits did not differ between the treatments. The improved growth rate was related to behavioural traits, mainly because chickens incubated in red light exhibited more passive (resting, standing, preening, dust bathing) and less active behaviours (walking, foraging, fighting, wing-flapping) than the blue-light incubated birds. The time spent for eating and drinking and the results of the tonic immobility test did not differ between both groups. Our results suggest that red and blue monochromatic light during incubation can differently program the postembryonic development of broilers, with possible consequences for their growth and welfare.

This study was conducted to evaluate the effect of monochromatic light on luteinising hormone (LH) and follicle-stimulating hormone (FSH) concentrations, LH and FSH mRNA levels, and mRNA levels of their receptors (LHR and FSHR) in Pigeons (Columba livia). A total of 60 paired adult White King pigeons were randomly divided into four groups; each group was exposed to violet light (VL), green light (GL), red light (RL), or white light for 16 h. Nine paired pigeons from each group were selected for analysis of FSH and LH concentrations and six paired pigeons from each group were selected for analysis of mRNA. The results showed that the concentration of LH in females was lower in the VL group (P<0.05), and in males, it was lower in the VL and GL groups (P<0.05). The concentration of FSH in females was significantly higher in the RL group than in the VL group (P<0.05), whereas no significant difference was observed in males (P>0.05). In females, the LH and FSH mRNA levels were relatively high in the pituitary gland, whereas LHR and FSHR mRNA levels were relatively high in the ovary. In males, the LH, LHR, FSH, and FSHR mRNA levels were high in the pituitary gland, whereas their respective mRNA levels were relatively low in the testis. These results suggest that RL has the potential to improve LH and FSH concentrations and mRNA levels, and indicates that RL could promote reproductive performance of pigeons.

Masalah lalu lintas  merupakan salah satu  masalah yang sangat sulit diatasi dengan hanya menggunakan system waktu (timer). Oleh sebab itu diperlukan suatu system pengaturan otomatis yang bersifat real-time sehingga waktu pengaturan lampu lalu lintas dapat disesuaikan dnegan keadaan di lapangan. Penelitian ini bertujuan mengembangkan suatu simulasi sistem yang mampu mengestimasi panjang antrian kendaraan menggunakan metoda pengolahan citra digital hanya dengan menggunakan satu kamera untuk dijadikan parameter masukan  dalam menghitung lama waktu nyala lampu merah dan lampu hijau. Oleh karena itu, sistem lalulintas sangatlah diperlukan, sebagai sarana dan prasarana untuk menjadikan lalulintas lancar, aman, bahkan sebagai media pembelajaran disiplin bagi masyarakat pengguna jalan raya. Penelitian ini penulis menggunakan sistem pengontrolan berbasis citra digital dimana camera sebagai sensor. Untuk aplikasi dari  semua metode dalam penelitian ini digunakan Microcontroller AurdinoTraffic problems is one of the problems that is very difficult to overcome by only using the system time (timer). Therefore we need an automatic real-time adjustment system so that the time settings for traffic lights can be adjusted according to the conditions on the ground. This study aims to develop a system simulation that is able to estimate the length of the vehicle queue using a digital image processing method using only one camera to be used as input parameters in calculating the length of time the red light and green light. Therefore, the traffic system is very necessary, as a means and infrastructure to make traffic smooth, safe, even as a medium for disciplined learning for road users. In this study the authors used a digital image-based control system where the camera as a sensor. For the application of all methods in this study, Aurdino Microcontroller is used

Abstract Background and objectives Among placental mammals, females undergo immunological shifts during pregnancy to accommodate the fetus (i.e. fetal tolerance). Fetal tolerance has primarily been characterized within post-industrial populations experiencing evolutionarily novel conditions (e.g. reduced pathogen exposure), which may shape maternal response to fetal antigens. This study investigates how ecological conditions affect maternal immune status during pregnancy by comparing the direction and magnitude of immunological changes associated with each trimester among the Tsimane (a subsistence population subjected to high pathogen load) and women in the USA. Methodology Data from the Tsimane Health and Life History Project (N = 935) and the National Health and Nutrition Examination Survey (N = 1395) were used to estimate population-specific effects of trimester on differential leukocyte count and C-reactive protein (CRP), a marker of systemic inflammation. Results In both populations, pregnancy was associated with increased neutrophil prevalence, reduced lymphocyte and eosinophil count and elevated CRP. Compared to their US counterparts, pregnant Tsimane women exhibited elevated lymphocyte and eosinophil counts, fewer neutrophils and monocytes and lower CRP. Total leukocyte count remained high and unchanged among pregnant Tsimane women while pregnant US women exhibited substantially elevated counts, resulting in overlapping leukocyte prevalence among all third-trimester individuals. Conclusions and implications Our findings indicate that ecological conditions shape non-pregnant immune baselines and the magnitude of immunological shifts during pregnancy via developmental constraints and current trade-offs. Future research should investigate how such flexibility impacts maternal health and disease susceptibility, particularly the degree to which chronic pathogen exposure might dampen inflammatory response to fetal antigens. Lay Summary This study compares immunological changes associated with pregnancy between the Tsimane (an Amazonian subsistence population) and individuals in the USA. Results suggest that while pregnancy enhances non-specific defenses and dampens both antigen-specific immunity and parasite/allergy response, ecological conditions strongly influence immune baselines and the magnitude of shifts during gestation.

Abstract This experiment compared performance and physiological responses of the offspring from cows supplemented with Ca salts of soybean oil (CSSO) or prilled saturated fat (CON) during late gestation. Non-lactating, pregnant Angus × Hereford cows (n = 104) that conceived during the same fixed-time artificial insemination protocol, using semen from 2 sires, were used in this experiment. Cows were ranked by pregnancy sire, body weight (BW), and body condition score (BCS). On d 180 of gestation (d -15), cows were randomly assigned to receive (dry matter basis) 415 g of soybean meal per cow daily in addition to 1) 195 g/cow daily of CSSO (n = 52) or 2) 170 g/cow daily of CON (n = 52). Cows were maintained in 2 pastures (26 cows/treatment per pasture), and received daily 12.7 kg/cow (dry matter basis) of grass-alfalfa hay. From d 0 until calving, cows were segregated into 1 of 24 feeding pens thrice weekly and received treatments individually. Cow BW and BCS were recorded, and blood samples were collected on d -15 of the experiment and within 12 h after calving. Calf BW was also recorded and blood sample collected within 12 h of calving. Calves were weaned on d 290 of the experiment, preconditioned for 35 d (d 291 to 325), and transferred to a feedyard where they remained until slaughter. Upon calving, CSSO cows and calves had greater (P &lt; 0.01) plasma concentrations of linoleic acid and total ω - 6 FA compared with CON cohorts. No differences in calf birth BW, weaning BW, and final preconditioning BW were noted (P ≥ 0.36) between treatments. Average daily gain and final BW in the feedlot were greater (P ≤ 0.05) in steers from CSSO cows compared with CON. The incidence of calves diagnosed with BRD that required a second antimicrobial treatment was less (P = 0.03) in calves from CSSO cows, resulting in reduced (P = 0.05) need of treatments to regain health compared with CON (Table 5). Upon slaughter, longissimus muscle area was greater (P = 0.03) in calves from CSSO cows compared with CON. Collectively, these results suggest that supplementing CSSO to late-gestating beef cows stimulated programming effects on postnatal offspring growth and Page 2 of 15 For Peer Review health. Therefore, supplementing late-gestating beef cows with CSSO appears to optimize offspring welfare and productivity in beef production systems.

Artificial lighting is a key factor in Closed Production Plant Systems (CPPS). A significant light-emitting diode (LED) technology attribute is the emission of different wavelengths, called light recipes. Light recipes are typically configured in continuous mode, but can also be configured in pulsed mode to save energy. We propose two nonlinear models, i.e., genetic programing (GP) and feedforward artificial neural networks (FNNs) to predict energy consumption in CPPS. The generated models use the following input variables: intensity, red light component, blue light component, green light component, and white light component; and the following operation modes: continuous and pulsed light including pulsed frequency, and duty cycle as well energy consumption as output. A Spearman's correlation was applied to generate a model with only representative inputs. Two datasets were applied. The first (Test 1), with 5700 samples with similar input ranges, was used to train and evaluate, while the second (Test 2), included 160 total datapoints in different input ranges. The metrics that allowed a quantitative evaluation of the model's performance were MAPE, MSE, MAE, and SEE. Our implemented models achieved an accuracy of 96.1% for the GP model and 98.99% for the FNNs model. The models used in this proposal can be applied or programmed as part of the monitoring system for CPPS which prioritize energy efficiency. The nonlinear models provide a further analysis for energy savings due to the light recipe and operation light mode, i.e., pulsed and continuous on artificial LED lighting systems.

Abstract Background The development of multiple gene expression systems, especially those based on the physical signals, such as multiple color light irradiations, is challenging. Complementary chromatic acclimation (CCA), a photoreversible process that facilitates the control of cellular expression using light of different wavelengths in cyanobacteria, is one example. In this study, an artificial CCA systems, inspired by type III CCA light-regulated gene expression, was designed by employing a single photosensor system, the CcaS/CcaR green light gene expression system derived from Synechocystis sp. PCC6803, combined with G-box (the regulator recognized by activated CcaR), the cognate cpcG2 promoter, and the constitutively transcribed promoter, the PtrcΔLacO promoter. Results One G-box was inserted upstream of the cpcG2 promoter and a reporter gene, the rfp gene (green light-induced gene expression), and the other G-box was inserted between the PtrcΔLacO promoter and a reporter gene, the bfp gene (red light-induced gene expression). The Escherichia coli transformants with plasmid-encoded genes were evaluated at the transcriptional and translational levels under red or green light illumination. Under green light illumination, the transcription and translation of the rfp gene were observed, whereas the expression of the bfp gene was repressed. Under red light illumination, the transcription and translation of the bfp gene were observed, whereas the expression of the rfp gene was repressed. During the red and green light exposure cycles at every 6 h, BFP expression increased under red light exposure while RFP expression was repressed, and RFP expression increased under green light exposure while BFP expression was repressed. Conclusion An artificial CCA system was developed to realize a multiple gene expression system, which was regulated by two colors, red and green lights, using a single photosensor system, the CcaS/CcaR system derived from Synechocystis sp. PCC6803, in E. coli. The artificial CCA system functioned repeatedly during red and green light exposure cycles. These results demonstrate the potential application of this CCA gene expression system for the production of multiple metabolites in a variety of microorganisms, such as cyanobacteria.

Background:The Virgen Macarena University Hospital belongs to the Public Health System of Andalusia and serves 481,296 inhabitants in Seville, Spain. In 2018 the Fracture Liaison Service switched to a multidisciplinary unit.Objectives:To describe FLS, to know the characteristics of patients with emphasis on gender differences and to know the completion of International Osteoporosis Foundation quality standards.Methods:Prospective, observational, analytical, research of usual clinical practice. All the consecutive patients attended from May 2018 to October 2019, ≥50 years, with a fragility fracture (occurred in the previous 24 months) were included. The study was approved by the Ethics Committee, Code 1084-N-16.Results:Our FLS is a type A multidisciplinary Unit, with a high level of intervention in the evaluation, estimation of fracture risk and fall risk, treatment prescription and follow-up of the patients. We included 408 patients, 80% females, one third with ≥80 years. Fragility fractures recorded in 328 women were hip (132, 40%), clinical vertebral (81, 25%) and no hip no vertebral (115, 35%). Those recorded in 82 males were hip (53, 66%), clinical vertebral (20, 24%) and no hip no vertebral (9, 10%), p=0.0001. Males had a higher rate of secondary causes of OP, drinker, and smoking. The most relevant gender difference was the low percentage of patients receiving pre-FF OP treatment. Forty-nine (16%) women versus 9 (7%) males had received it at some point in their life, p=0.04. Two hundred and seventy-one (86%) women vs 48 males (63%) had received it at after their FF in their reference unit, and all them were treated after the FLS evaluation. The probability of a male not receiving prior treatment was 2.5 (95% CI 1.01- 6.51); p=0,04. This probability was 0.64 (0.38-1.09) after the FF. After twelve months of follow-up in FLs, 96% continued treatment, with no differences between men and women. The completion of IOF quality standards was bad (red light) for patient identification items and FLS reference time. Abstract This experiment evaluated the impacts of bovine appeasing substance (BAS) administration on performance, health, and physiological responses of feedlot cattle during a 45-d receiving period. A total of 342 recently-weaned Angus-influenced steers, originating from 16 cow-calf operations, were obtained from an auction yard on d -1 and road-transported (12 h) to the feedlot. Upon arrival on d 0, body weight (BW) was recorded and steers were ranked by BW and source and assigned to receive BAS (Nutricorp, Araras, SP, Brazil; n = 171) or placebo (diethylene glycol monoethyl ether; CON; n = 171). Treatments (5 mL) were topically applied to the nuchal skin area of each animal. Within treatment, calves were allocated to 1 of 24 drylot pens (12 pens/treatment) and received a free choice total-mixed ration from d 1 to 45. Calves were assessed for bovine respiratory disease (BRD) signs and feed intake was recorded from each pen daily. Steer BW was again recorded on d 1, 7, 17, 31, and 45, whereas blood samples were collected from 5 steers/pen concurrently with each BW assessment. Average daily gain was greater (P = 0.05) in BAS vs. CON calves, although final BW did not differ (P = 0.36) between treatments. No treatment effects were detected for feed intake (P = 0.95), resulting in greater (P = 0.05) feed efficiency in BAS vs. CON steers. No treatment effects were detected (P ≥ 0.37) for plasma concentrations of haptoglobin, whereas plasma cortisol concentrations were greater (P = 0.05) in CON vs. BAS steers on d 7 (treatment × day; P = 0.07). Incidence of BRD was greater (P ≤ 0.05) in BAS vs. CON on d 6 to 10 and d 18 to 21 (treatment × day; P &lt; 0.01), although overall BRD incidence did not differ (P = 0.24) between treatments. The number of antimicrobial treatments required per steer diagnosed with BRD symptoms to recover from sickness was greater (P = 0.04) in CON vs. BAS calves. No treatment differences were detected (P ≥ 0.41) for mortality incidence, or proportion of steers removed from the experiment due to extreme sickness. Results from this experiment indicate BAS administration upon feedlot entry improved average daily gain by enhancing feed efficiency. Administration of BAS facilitated earlier detection of BRD and reduced the need for antimicrobial treatments. Collectively, these results suggest BAS administration as a promising strategy to benefit performance and immunocompetence of feedlot receiving cattle.
