In vitro evaluation of the methane mitigation potential of a range of grape marc products

Grape marc consists of the skins, seeds and stems remaining after grapes have been pressed to make wine. Interest in grape marc for use as a dietary feed additive for ruminants has grown after recent research showed that inclusion of grape marc in the diet of dairy cows reduced their enteric methane (CH4) emissions. In the present research, in vitro fermentations were conducted on 20 diverse grape marcs to evaluate their potential as ruminant feed supplements and, in particular, mitigants of enteric CH4 emissions. The grape marcs, which were sourced from vineyards in south-eastern Australia, contained a range of red and white grape varieties with different proportions of skins, seeds and stalks, and had diverse chemical compositions. For each grape marc, four replicate samples, each of 1 g DM, were incubated in vitro with ruminal fluid. The volumes of total gas and CH4 produced after 48 h of incubation were determined. Total gas production ranged from 21.8 to 146.9 mL and CH4 production from 6.8 to 30.3 mL. White grape marcs produced more (P < 0.05) total gas (81.8 mL) than did red grape marcs (61.0 mL), but had a lower (P < 0.05) percentage of CH4 (25.3% and 30.3% of total gas). Grape marcs with a higher proportion of seeds produced less (P < 0.05) total gas than did the types composed of either skin or stalks; however, the seed types produced the greatest (P < 0.05) percentage of CH4 (49.8% of total gas). It is concluded that grape marcs differ greatly in their potential as mitigants of enteric CH4 emissions for ruminal production systems.

In vitro gas production systems are utilized to screen feed ingredients for inclusion in ruminant diets. However, not all in vitro systems are set up to measure methane (CH4) production, nor do all publications report in vitro CH4. Therefore, the objective of this study was to develop models to predict in vitro CH4 production from total gas and volatile fatty acid (VFA) production data and to identify the major drivers of CH4 production in these systems. Meta-analysis and machine learning (ML) methodologies were applied to a database of 354 data points from 11 studies to predict CH4 production from total gas production, apparent DM digestibility (DMD), final pH, feed type (forage or concentrate), and acetate, propionate, butyrate and valerate production. Model evaluation was performed on an internal dataset of 107 data points. Meta-analysis results indicate that equations containing DMD, total VFA production, propionate, feed type and valerate resulted in best predictability of CH4 on the internal evaluation dataset. The ML models far exceeded the predictability achieved using meta-analysis, but further evaluation on an external database would be required to assess generalization ability on unrelated data. Between the ML methodologies assessed, artificial neural networks and support vector regression resulted in very similar predictability, but differed in fitting, as assessed by behaviour analysis. The models developed can be utilized to estimate CH4 emissions in vitro.

Abstract Grape marc (GM) is a viticulture by-product used as cattle supplement in periods of shortage of conventional feed sources. It contains fats, high concentrations of polyphenols and has been reported to reduce enteric methane (CH4) emissions. In-vitro batch culture was used to study the effects of substitution of mixed hay (MH) for a traditional Chilean variety (Vitis vinifera “País”) of GM on in vitro dry matter disappearance (IVDMD), rumen fermentation parameters (short chain fatty acids, pH, partitioning factor), gas and CH4 production in a 60% forage diet (dry matter, DM). The study was a randomized complete design with 3 treatments and 3 replicates, incubated for 24 h at 39º C. Treatments were: T1 (Control): 20% MH, 40% corn silage, 40% concentrate; T2 = 10% MH, 10% GM, 40% corn silage, 40% concentrate; T3 = 20% GM, 40% corn silage, 40% concentrate. Means were compared with the Tukey test (P &lt; 0.05), and polynomial contrasts. Substitution of MH with GM significantly reduced ammonia nitrogen (NH3-N) by 50% (P &lt; 0.05), although it did not affect IVDMD, gas production or other rumen fermentation parameters (P &gt; 0.05). Total CH4 (mg) linearly decreased (P = 0.013) as concentrations of GM increased. Methane production (mg/g DM incubated) and yield (mg/g DM digested) decreased linearly (P = 0.002 and P = 0.003, respectively) as inclusion of GM increased. Inclusion of GM at 20% reduced CH4 production by 19% and CH4 yield by 16.4%. These results indicate that partial substitution of dietary fiber sources with traditional Chilean País GM in high fiber diets is a viable feeding alternative, and can decrease environmental impact (lower CH4 and ammonia emissions) of ruminant livestock, without negatively affecting rumen fermentation parameters.

Methane (CH4) from ruminal feed degradation is a major pollutant from ruminant livestock, which calls for mitigation strategies. The purpose of the present 4 × 2 factorial arrangement was to investigate the dose–response relationships between four doses of the CH4 inhibitor 3-nitrooxypropanol (3-NOP) and potential synergistic effects with low (LC) or high (HC) concentrate feed proportions (CFP) on CH4 reduction as both mitigation approaches differ in their mode of action (direct 3-NOP vs. indirect CFP effects). Diet substrates and 3-NOP were incubated in a rumen simulation technique to measure the concentration and production of volatile fatty acids (VFA), fermentation gases as well as substrate disappearance. Negative side effects on fermentation regarding total VFA and gas production as well as nutrient degradability were observed for neither CFP nor 3-NOP. CH4 production decreased from 10% up to 97% in a dose-dependent manner with increasing 3-NOP inclusion rate (dose: p < 0.001) but irrespective of CFP (CFP × dose: p = 0.094). Hydrogen gas accumulated correspondingly with increased 3-NOP dose (dose: p < 0.001). In vitro pH (p = 0.019) and redox potential (p = 0.066) varied by CFP, whereas the latter fluctuated with 3-NOP dose (p = 0.01). Acetate and iso-butyrate (mol %) decreased with 3-NOP dose, whereas iso-valerate increased (dose: p < 0.001). Propionate and valerate varied inconsistently due to 3-NOP supplementation. The feed additive 3-NOP was proven to be a dose-dependent yet effective CH4 inhibitor under conditions in vitro. The observed lack of additivity of increased CFP on the CH4 inhibition potential of 3-NOP needs to be verified in future research testing further diet types both in vitro and in vivo.

Abstract País Grape (Vitis vinifera L.) is an ancestral variety used in Chilean wine industry. It has a higher content of proanthocyanidins than commercial varieties such as Carmenère or Pinot Noir, resulting in País grape marc (PGM) with high contents of condensed tannins. As such, PGM inclusion in ruminant diets would have the potential to reduce enteric methane (CH4) emissions and decrease urinary N excretion. The objective of this study was to evaluate the effects of substitution of mixed hay (MH) with PGM in a high concentrate diet [65% dry matter (DM)] on in vitro dry matter disappearance (IVDMD), ruminal fermentation parameters, gas and CH4 production. Treatments were: T1 (Control) = 20% MH, 15% corn silage, 65% concentrate; T2 = 10% MH, 10% PGM, 15% corn silage, 65% concentrate; T3 = 20% PGM, 15% corn silage, 65% concentrate. The study was a randomized complete design with 3 treatment and 3 replicates, incubated for 24 h at 39º C. Data were compared by Tukey test and polynomial contrasts. There was a linear reduction in NH3-N (P = 0.001) as dietary PGM increased. Inclusion of PGM reduced NH3-N by 50% when added at 10% DM, and 71.7% at 20% DM. However, there also was 4% reduction in IVDMD (P ≤ 0.001) and gas production (P = 0.012) in the 20% GM diet. There were no treatment effects (P ≥ 0.05) on CH4 production or yield. Fermentation efficiency determined by the partition factor increased linearly (P = 0.013) as PGM inclusion increased, suggesting that it increases organic matter to be degraded. Based on this study it could be concluded that PGM is an alternative source of fiber for ruminants on concentrate diets, as it can result in improved rumen fermentation efficiency and a substantial reduction in ruminal ammonia nitrogen concentration (NH3-N).

The objective of this study was to determine the effect of pomegranate (Punica granatum L.) seed oil (PSO) on gas and methane (CH4) production, ruminal fermentation and microbial populations under in vitro conditions. Three treatments consisting of a control diet containing 10 mg tallow (CON); the control diet with 5 mg PSO + 5 mg tallow (MPSO) and the control diet containing 10 mg PSO (HPSO) were compared. Ten mg of the experimental fat/oil samples were inserted into a gas-tight 100 mL plastic syringe containing 30 mL of an incubation inoculum and 250 mg of a basic substrate of a hay/concentrate (1/1, w/w) mixture. In vitro gas production was recorded over 0, 2, 4, 6, 8, 10, 12 and 24 h of incubation. After 24 hours, incubation was stopped, and methane production, pH, volatile fatty acids (VFAs) and microbial counts were measured in the inoculant. Gas production at 4, 6, 8, 10, 12 and 24 h incubation, metabolizable energy and in vitro organic matter disappearance increased linearly and quadratically as level of PSO increased. Furthermore, the 10 mg PSO (HPSO) decreased CH4 production by 21.0% compared with the control (CON) group. There were no significant differences in total and individual VFA concentrations between different levels of PSO, except for butyric acid. After 24 h of incubation, methanogenesis decreased in the HPSO compared with the MPSO and CON treatments. In addition, total bacteria and protozoa counts increased with rising PSO levels, while population methanogenesis declined significantly. These results suggested that PSO could reduce methane emissions, which might be beneficial to nutrient utilization and growth in ruminants.

Abstract The aim of the research was to evaluate the effect of three Eucalyptus globulus extracts rich in phenolic compounds, especially flavonoids, on rumen fermentation, methane (CH4) production, organic matter degradability and protozoa population using an in vitro gas production technique. Four concentrations (0, 50, 75 and 100 mg) of three Eucalyptus extracts (ethyl acetate, n-butanol and aqueous) were added to a diet of ruminants (forage: concentrate ratio 60:40) and incubated at 39°C under anaerobiosis with buffered rumen fluid. After 24 h, the fermentation fluid was analysed for ammonia-N and volatile fatty acids (VFA). Organic matter degradability (OMD) and protozoa were also determined; in vitro gas production was also recorded and CH4 concentration was measured. Compared to the control, CH4 production was significantly lower for ethyl acetate extract (P<0.05), but higher for n-butanol and aqueous extracts. Production of ammonia- N was lower in all Eucalyptus extracts (P<0.05). Propionate production (P<0.05) increased for ethyl acetate and n-butanol extracts, whereas no effect was registered for VFA, for all Eucalyptus extracts. Ethyl acetate extract decreased in vitro OMD (P<0.05), whereas n-butanol and aqueous extracts were comparable to the control. Protozoa population decreased (P<0.05) for all extracts in comparison with the control. Eucalyptus ethyl acetate extract might be promising to be used as a potent anti-methanogenic additive. Moreover, the assessment of the right dosage seems to be important to decrease methane production, without reducing feed nutritional value.

Abstract: Enteric methane (CH4) production is the main source of greenhouse gas emissions from livestock globally with beef cattle contributing 5.95% of total global greenhouse gas emissions. Various mitigation strategies have been developed to reduce enteric emissions with limited success. In vitro studies have shown a reduction in CH4 emissions when using garlic and citrus extracts. However, there is paucity of data regarding in vivo studies investigating the effect of garlic and citrus extracts in cattle. The objective of this study was to quantitatively evaluate the response of Angus × Hereford cross steers consuming the feed additive Mootral, which contains extracts of both garlic and citrus, on CH4 yield (g/kg dry matter intake [DMI]). Twenty steers were randomly assigned to two treatments: control (no additive) and Mootral supplied at 15 g/d in a completely randomized design with a 2-wk covariate and a 12-wk data collection periods. Enteric CH4 emissions were measured using the GreenFeed system during the covariate period and experimental weeks 2, 6, 9, and 12. CH4 yield (g/kg DMI) by steers remained similar in both treatments for weeks 2 to 9. In week 12, there was a significant decrease in CH4 yield (23.2%) in treatment compared to control steers mainly because the steers were consuming all the pellets containing the additive. However, overall CH4 yield (g/kg DMI) during the entire experimental period was not significantly different. Carbon dioxide yield (g/kg DMI) and oxygen consumption (g/kg DMI) did not differ between treatments during the entire experimental period. DMI, average daily gain, and feed efficiency also remained similar in control and supplemented steers. The in vivo results showed that Mootral may have a potential to be used as a feed additive to reduce enteric CH4 production and yield in beef cattle but needs further investigation under various dietary regimen.

Legume–grass intercropping systems are a sustainable option to improve nutritional quality of animal feed and decrease livestock greenhouse gas emissions. Thus, the present study evaluated yield, chemical composition and in vitro gas production of silages produced with intercropped palisade grass (Urochloa brizantha.(A.Rich.) R.D.Webster), pigeon pea (Cajanus cajan cv. Super N) and corn (Zea mays. L.). Forage was harvested and placed inside micro-silos, which were opened after 100 days and samples were collected for chemical composition and in vitro gas production analyses. Intercropped silage had higher crude protein, acid detergent fiber, and lignin content than corn silage. Moreover, intercropped silage decreased total gas and methane production. Therefore, intercropped silage showed potential to increase conserved feed nutritional quality and reduce methane emissions in livestock production systems.

We assessed and ranked different dietary strategies for mitigating methane (CH4) emissions and other fermentation parameters, using an automated gas system in two in vitro experiments. In experiment 1, a wide range of dietary CH4 mitigation strategies was tested. In experiment 2, the two most promising CH4 inhibitory compounds from experiment 1 were tested in a dose-response study. In experiment 1, the chemical compounds 2-nitroethanol, nitrate, propynoic acid, p-coumaric acid, bromoform, and Asparagopsis taxiformis (AT) decreased predicted in vivo CH4 production (1.30, 21.3, 13.9, 24.2, 2.00, and 0.20 mL/g DM, respectively) compared with the control diet (38.7 mL/g DM). The 2-nitroethanol and AT treatments had lower molar proportions of acetate and higher molar proportions of propionate and butyrate compared with the control diet. In experiment 2, predicted in vivo CH4 production decreased curvilinearly, molar proportions of acetate decreased, and propionate and butyrate proportions increased curvilinearly with increased levels of AT and 2-nitroethanol. Thus 2-nitroethanol and AT were the most efficient strategies to reduce CH4 emissions in vitro, and AT inclusion additionally showed a strong dose-dependent CH4 mitigating effect, with the least impact on rumen fermentation parameters.

Two experiments were conducted to investigate the effects of adding gallic acid (GA) to ruminant diets on long- and short-term in vitro rumen fermentation and methane (CH4) production, and to test possible interactions between GA and ethanol on fermentation. The first experiment was conducted using the rumen simulation technique (Rusitec), as a completely randomised block design with four replications and the following four doses of GA: 0, 5, 10 and 20 mg GA/g dry matter (DM). Ethanol was used in all treatments to increase the solubilisation of GA in rumen fluid. The experimental period lasted 16 days, of which the first 7 days were for adaptation and the subsequent 9 days were for sampling. The second experiment was a 48-h batch-culture incubation conducted as a completely randomised design with a 4 (GA dose; 0, 10, 20, and 40 mg GA/g DM) × 2 (with or without ethanol) arrangement of treatments. In the Rusitec experiment, addition of GA up to 20 mg/g DM did not affect DM disappearance (DMD), organic matter (OM) disappearance, neutral detergent-fibre disappearance (NDFD), acid detergent-fibre disappearance (ADFD) or starch disappearance (P &gt; 0.05), but crude protein disappearance was linearly decreased (P = 0.04) from 78.3% to 72.0%. Daily gas production and CH4 production expressed as mL/g DM and mL/g DMD were not affected by addition of GA (P &gt; 0.05). Addition of GA up to 20 mg/g DM increased butyrate and isovalerate production (P &lt; 0.05) and tended to increase isobutyrate (P = 0.09) and decrease heptanoate production (P = 0.07). In the batch-culture experiment, adding GA up to 40 mg/g DM linearly increased 48-h DMD, NDFD and ADFD (P &lt; 0.05) and decreased (P &lt; 0.05) CH4 expressed as mL/g DMD, mL/g NDFD and mL/g ADFD. Methane production was decreased after 24 h and 48 h only when GA was added at 10 mg/g DM without ethanol. Fermentation liquid pH and concentration of ammonia-nitrogen (ammonia-N) were also reduced (P &lt; 0.05) with an increasing concentration of GA. Treatments with ethanol notably enhanced 48-h DMD, NDFD, ADFD, gas production (mL/g DM, mL/g OM or mL/g DMD), CH4 production (mL/g DM, mL/g DMD or mL/g NDFD), total volatile fatty acid concentration, the acetate:propionate ratio, acetate, valerate, isovalerate and caproate molar proportions (P &lt; 0.01) and decreased propionate, butyrate and isobutyrate molar proportions (P &lt; 0.01). Significant dose of GA × ethanol interaction was observed only for acetate molar proportion (P = 0.03). In conclusion, our study suggests that the beneficial effects of GA on feed digestion and CH4 production may be short term, while improvements in N metabolism may be sustained over the long term. It may be useful to conduct long-term in vivo studies using a range of diets and doses to verify whether GA can be used as a feed additive to mitigate enteric CH4 production and improve N metabolism of ruminants.
