Effects of luteectomy on the maintenance of pregnancy, circulating progesterone concentrations and lambing performance in sheep

In sheep, there have been few and conflicting data regarding the necessity of the corpus luteum (CL) for the maintenance of pregnancy. The aims of the present study were to examine the effect of luteectomy on and after Day 50 of pregnancy on maternal plasma progesterone concentrations and the progression of pregnancy, to determine the minimum placental progesterone support required for the maintenance of pregnancy, and to evaluate the effect of luteectomy on lambing performance. In Experiment 1, four ewes luteectomized on Day 50 of pregnancy aborted 2–7 days after surgery, whereas pregnancy progressed and parturition occurred between Days 143 and 149, with live lambs, in three of four ewes and in four ewes luteectomized on Days 60 and 70 of pregnancy respectively. The mean ( SEM) progesterone concentrations on the day before and one day after luteectomy decreased from 4.87 0.85 to 0.42 0.06 ng mL –1 (P<0.01), from 4.57 0.51 to 0.80 0.12 ng mL –1 (P<0.02) and from 6.05 0.52 to 1.67 0.11 ng mL –1 (P<0.01), respectively, for the ewes luteectomized on Days 50, 60 and 70 of pregnancy. The fall in progesterone concentrations was 90%, 80% and 71%, respectively, for the ewes luteectomized on Days 50, 60 and 70 of pregnancy. In Experiment 2, pregnancy progressed in four ewes luteectomized on Day 70 and parturition occurred between Days 146 and 149, with live lambs. The mean progesterone concentrations declined (P<0.01) from 6.9 0.7 ng mL –1 on the day before luteectomy to 2.1 0.3 ng mL –1 the day after surgery. The concentrations of progesterone in blood collected every 3 h during a 24-h period were stable on Days 60 and 80 of pregnancy, but they were lower (P<0.03) on Day 80 than on Day 60 of pregnancy, for each time period examined. In Experiment 3, the gestation length and birthweights of single, twin and triplet lambs were not different between the control intact ewes (n = 111) and the ewes luteectomized on Days 70–80 of pregnancy (n = 71). Lamb mortality was not different between the two groups (7.2% v. 8.4%, control v. luteectomized). In conclusion, these results showed that (1) the sheep CL is necessary to maintain pregnancy until at least Day 60, (2) progesterone withdrawal induced by luteectomy on and after Day 50 of pregnancy must be of a critical magnitude to provoke abortion, (3) after Day 60 of pregnancy, the CL and the placenta together secrete more progesterone than required for pregnancy maintenance, (4) there is no apparent 24- hour rhythm in maternal plasma progesterone concentrations before and after luteectomy, and (5) luteectomy at mid pregnancy has no apparent effect on gestation length, lamb birthweight or lamb mortality.

The concentrations of progesterone and oestradiol-17β in the maternal plasma of Bennett's wallaby, Macropus rufogriseus rufogriseus, were measured daily throughout gestation after reactivation of the diapausing corpus luteum by removal of the suckling pouch young (RPY). Progesterone increased from mean concentrations of 382–424 pmol/l (120–133 pg/ml) during lactation to reach peak concentrations of 908 ± 172 (s.e.m.) pmol/l (285 ± 54 pg/ml) (n = 8) 4 days after RPY and 971 ± 220 and 971 ± 229 pmol/l (305 ± 69 and 305 ± 72 pg/ml) (n = 7) 24 and 25 days after RPY respectively. The mean gestation length (RPY to birth) was 26·8 ± 0·6 (s.d.) days (n = 6, range 25·75–27·50 days). Immediately after birth the plasma progesterone concentration declined to 299 ± 51 (s.e.m.) pmol/l (94 ± 16 pg/ml) (n=6). Oestradiol-17β increased from mean concentrations of 291–553 pmol/l (80–152 pg/ml) during lactation to reach a peak concentration of 967 ± 331 pmol/l (266 ± 91 pg/ml) (n = 9) 1 day after RPY. The concentration declined from 7 days after RPY and fluctuated between mean concentrations of 273 and 480 pmol/l before reaching a minimum of 207 ± 69 pmol/l (57 ± 19 pg/ml) (n = 6) 19 days after RPY. A transient increase to 542 ± 207 pmol/l (n = 7) occurred at 22 days after RPY. Plasma concentrations declined to a low of 156 ± 55 pmol/l (43 ± 15 pg/ml) (n = 6) 5 days after parturition. The mean concentration of plasma 13,14-dihydro-15-oxo-prostaglandin F2α was less than 2·8 nmol/l (1 ng/ml) for all samples from 13 days after RPY until 4 days after parturition. The results suggest that oestradiol-17β may be important in the early stages of blastocyst reactivation to synergize with progesterone in stimulating uterine secretions. 13,14-Dihydro-15-oxo-prostaglandin F2α is unlikely to be involved in the birth process and any luteolytic effect is likely to be from a local production of PGF2α.

ABSTRACT Progesterone was determined and identified in human peripheral blood during the preovulatory period of the menstrual cycle, by combined isotope derivative and recrystallization analysis. The mean concentration of progesterone in 1.095 ml of plasma obtained 9 days before ovulation was 0.084 μg/100 ml. However, the mean concentration of progesterone in 1.122 ml of plasma obtained 4 days before ovulation was 0.279 μg/100 ml. These data demonstrate a source of progesterone secretion other than the corpus luteum. The higher plasma-progesterone concentration 4 days before ovulation may indicate progesterone secretion of the ripening Graafian follicle of the ovary.

Antenatal glucocorticoids are administered to women at risk of preterm delivery to prevent neonatal respiratory morbidity. The effects of exogenous glucocorticoids on the development of lactation are unknown. This study investigated the effects of a single dose of antenatal glucocorticoids on secretory activation in sheep before and after parturition. Pregnant ewes (N=36) were randomised to receive either medroxyprogesterone acetate (MPA) at 118 days of pregnancy and betamethasone at 125 days (BETA group), MPA at 118 days and saline at 125 days (MPA group) or saline at 118 and 125 days (SALINE group). The concentration of lactose, progesterone, cortisol and prolactin in maternal plasma was measured during pregnancy. After term parturition, the concentration of lactose in milk and maternal plasma was measured daily for 5 days. Lambs were weighed at birth and at 5 days of age; milk volume was measured on day 5. The concentration of lactose in maternal plasma increased significantly after betamethasone administration, corresponding to a fall in plasma progesterone. No changes in lactose were observed in MPA or SALINE ewes. Transient decreases in cortisol and increases in prolactin were observed in the BETA group, but not in either the MPA or SALINE group. After parturition, BETA ewes experienced reduced milk yield and lamb weight gain, and delayed increases in milk lactose levels compared with MPA and saline controls. This study demonstrated that, in sheep, antenatal glucocorticoid administration disrupted secretory activation, causing precocious mammary secretion before parturition and compromising postpartum milk production and lamb growth.

ABSTRACT The effect of orally given diethylstilboestroldiphosphate (DES) and 17α-ethinyl-oestradiol-3-methylether (EEM) on plasma progesterone levels was studied. Both compounds were administered for 5 days to 5 women in daily doses of 60 mg (DES) and 30 mg (EEM). The fully informed volunteers were found to have a normal menstrual cycle before the study. The mean corpus luteum phase (corpus luteum phase = days between LH surge and onset of menstruation) of all control cycles lasted 12.8 days. Daily plasma samples were collected for radioimmunoassay (RIA) of progesterone, immunoreactive oestrogens and LH. After a control cycle the first treatment was carried out with DES. The third and the fifth cycle were control cycles again. The EEM-treatment was done in the fourth cycle. Although the effect of the two compounds was different, a dependence of the age of the corpus luteum (CL) could be demonstrated for both. DES-treatment lowered plasma progesterone levels during administration. This effect was only demonstrable if the treatment was begun on the day of the LH-peak. The length of the CL-phase remained unaltered. EEM-treatment if started on the day of the LH surge, suppressed corpus luteum function in the late luteal phase. If the treatment was started later, the effect was less pronounced. The administration of both compounds did not shorten the time between ovulation and the next bleeding. After DES-treatment this interval was not altered. After EEM-treatment the subsequent bleeding was even delayed depending on slowly decreasing levels of plasma oestrogens.

ABSTRACTIn heifers, plasma progesterone levels were low (<0·3μg/l) from the 59th to about the 15th day before puberty (defined as the day when oestrus was first detectable by the fern pattern of dried cervical mucus). Plasma oestradiol-17β levels varied randomly within the range 1 to 4 ng/l over the same period, but then showed three peaks of about 6, 9 and 4 ng/l, respectively 8 days before puberty, on the day of puberty and 4 days later. The pre-pubertal oestradiol-17p peak was followed by a short period of elevated progesterone levels. Whether this oestradiol-17β peak induced ovulation is uncertain; it is however clear that a corpus luteum of normal life-span was not formed. The oestradiol-17β peaks on the day of puberty and 4 days later, when progesterone levels were rising rapidly, were typical both in magnitude and duration of those occurring before and after ovulation in post-pubertal oestrous cycles.

ABSTRACTIn this study, endometrial samples were collected in 14 Nelore cows on days zero (ovulation), five, nine, thirteen and nineteen of the estrous cycle (biopsy group), and in 15 females these collections weren't performed (control group). Biopsies were done on the uterine horn endometrium contralateral to the ovary with corpus luteum. Blood samples were taken at -24, -16, -8, 0 +8, +16 and +24 hours in relation to progesterone drop (<1ng/mL, zero moment) and evaluated for 13, 14-dihydro-15-keto prostaglandin F2-alpha (PGFM) by radioimmunoassay (RIA). Plasma progesterone concentration was determined by RIA every 24 hours. Data were analyzed by ANOVA using the PROC GLM and MIXED of the SAS. The mean value for PGFM during the entire period evaluated was greater in the biopsy group. The mean concentration of PGFM at moment zero was not different between the groups; the mean concentration of PGFM was higher in the biopsy group before and after the drop in progesterone level. The maximum mean concentration observed was not different between the groups; however, the mean minimum concentration was different with high values in the biopsy group. Although the PGFM concentrations were higher in the biopsy group, the biopsy and control groups had similar length of estrous cycle showing that repeated endometrial biopsy on the side contralateral to the ovary with corpus luteum does not affect luteolysis and the length of the estrous cycle.

ABSTRACTPatterns of oestrous cycle activity in the first breeding season were studied in 30 Suffolk × Mule ewe lambs, Twenty-eight reached puberty and the mean date of first oestrus was 27 November (s.d. 13·6 days). Mean live weight at first oestrus was 37·7 (s.d. 3·63) kg. The mean number of oestrous cycles was 3·3 (s.d. 1·6), 0·92 (65) being normal length single cycles (14 to 19 days), 0·03 (2) long single cycles (20 to 26 days) and 0·05 (4) multiple cycles (> 26 days).Plasma progesterone profiles before and after the breeding season were studied in 10 randomly selected animals. All showed profiles suggesting one or two silent ovulations preceding the first oestrus but none showed any indication of silent ovulations following the last oestrus.Plasma progesterone profiles were also studied on the four animals showing multiple cycles and on the two animals that did not reach puberty. These suggested that, in all four animals showing multiple cycles, one or two silent ovulations had occurred between the two overt oestrous periods. Of the two animals that did not reach puberty, one showed three abnormal silent cycles and the other showed no change in progesterone concentration from a low basal level.

Abstract. A satellite database including 16 555 satellite images and ice charts displaying the area of Isfjorden, Hornsund and the Svalbard region has been established with focus on the time period 2000–2014. 3319 manual interpretations of sea ice conditions have been conducted, resulting in two time series dividing the area of Isfjorden and Hornsund into "Fast ice", "Drift ice" and open "Water". The maximum fast ice coverage of Isfjorden is > 40 % in the periods 2000–2005 and 2009–2011 and stays < 30 % in 2006–2008 and 2012–2014. Fast ice cover in Hornsund reaches > 40 % in all considered years, except for 2012 and 2014, where the maximum stays < 20 %. The mean seasonal cycles of fast ice in Isfjorden and Hornsund show monthly averaged values of less than 1 % between July and November and maxima in March (Isfjorden, 35.7 %) and April (Hornsund, 42.1 %) respectively. A significant reduction of the monthly averaged fast ice coverage is found when comparing the time periods 2000–2005 and 2006–2014. The seasonal maximum decreases from 57.5 to 23.2 % in Isfjorden and from 52.6 to 35.2 % in Hornsund. A new concept, called "days of fast ice coverage" (DFI), is introduced for quantification of the interannual variation of fast ice cover, allowing for comparison between different fjords and winter seasons. Considering the time period from 1 March until end of sea ice season, the mean DFI values for 2000–2014 are 33.1 ± 18.2 DFI (Isfjorden) and 42.9 ± 18.2 DFI (Hornsund). A distinct shift to lower DFI values is observed in 2006. Calculating a mean before and after 2006 yields a decrease from 50 to 22 DFI for Isfjorden and from 56 to 34 DFI for Hornsund.

Abstract The objective of this study was to determine if maternal plasma progesterone (P4) during early gestation influences within-litter birth weight variation in pigs. White crossbred females (n = 589) of mixed parity (1–4) were bred at standing estrus designated as day 0 of pregnancy and again 24 h later using single-sire semen from commercial Yorkshire genetics. At day 7 of pregnancy, a single blood sample from each female was collected, processed for plasma and P4 was analyzed using a validated RIA for swine. Dams were allowed to farrow under normal management procedures and standard litter statistics (e.g., number born alive, piglet birth weight, etc.) were recorded. Within-litter birth weight variation (BWV) was determined for each litter using the coefficient of variation of birth weight. Pearson correlation analysis illustrated there was a low (r = 0.0967) but significant (P = 0.0191) positive correlation between P4 and BWV. Linear regression demonstrated that for every ng/ml increase in P4 during early gestation, BWV increased 0.11 ± 0.04%. For this population, the mean BWV was 19.4% with a SD of 7.8%. When analyzing P4 concentrations from extreme groups of females with high or low BWV [i.e. females with 1 SD above, high group (&gt;27.2%; n = 100); or below, low group (&lt; 11.6%; n = 106) the mean BWV] by GLM procedures, high BWV group females had increased (P = 0.0473) concentrations of P4 during early gestation compared to low BWV group (13.9 ± 1.4 vs. 9.0 ± 1.7 ng/ml, respectively). These data demonstrate a positive relationship for early gestation plasma P4 concentrations with greater BWV; thereby, illustrating that high concentrations of P4 during early gestation influence within-litter birth weight variation in pigs. Hepatitis has become a health problem in the world. The hepatitis virus infected many people. According to the teacher of MTsN 02 Bondowoso more than 20 students have hepatitis A viral infection. The purpose of this research was to know the differences of students' knowledge about hepatitis A before and after counseling in MTsN 02 Bondowoso 2015. This study used pre-experimental (pre-post test design). This study used stratified random sampling technique, 127 students from 270 sample involved this research,and 143 students was excluded. We used questionnaires to collect data. The results showed that the mean value of the students 'knowledge about hepatitis A before counseling in MTsN 02 Bondowoso 2015 was 83.96 with the lowest value of 37.5 and the highest value was 100. The mean value of the students' knowledge about hepatitis A after counseling in MTsN 02 Bondowoso 2015 was 93.21 with the lowest value waf 62.5 and the highest value was 100. Paired t test showed that t (-9.07) > t table (1.98), the null hypothesis (H0) was rejected. There was a difference between students' knowledge about hepatitis A before and after counseling in MTsN 02 Bondowoso 2015. This study showed that routine counseling by healthcare provider was important to prevent hepatitis A infection.; Keywords: counseling, knowledge of students, hepatitis
