Barbary sheep (Ammotragus lervia) are considered vulnerable species by the World Conservation Union (IUCN). The purpose of this study was to describe the basic characteristics of fresh semen, test the efficacy of commercial extender Triladyl, and collect necessary data that may help to create a frozen semen bank for the Barbary sheep in Spain. A total of 21 ejaculates were collected by rectal-probe electroejaculation from one dominant (D) and three minor (M) adult males housed in the Madrid Zoo. After ejaculation, semen volume, concentration, and mass motility were assessed. Remaining raw semen was diluted at 37°C with TRIS-based extender Triladyl (Minitüb, Tiefenbach, Germany) and 20% egg yolk to a final concentration of 200 × 106 sperm per mL. Diluted samples were kept at 5°C for 4 h and then loaded into 0.25-mL French straws, frozen at 5 cm above liquid nitrogen (LN2) for 10 min and then plunged into LN2. Samples were thawed in a water bath at 37°C for 30 s. Post-thaw semen survival was evaluated by sperm motility (%M), quality of movement (Q), normal acrosome status (%NAS), normal sperm morphology (%NOR), membrane integrity (hypo-osmotic test; %HOST), and sperm viability (eosin-nigrosin vital staining; %V), and were compared with the same parameters in the fresh semen. Data between D and M males were analyzed by one way ANOVA. Mean volume of ejaculates, total sperm concentration and mass motility of raw semen were respectively; 5.2 ± 1.56 mL, 2800.0 ± 1290.5 × 106 and 3.4 ± 0.4 for the D male, and 3.5 ± 3.2 mL, 251.2 ± 103.9 × 106, and 1.88 ± 1.4 for M males (P < 0.05). Remaining semen parameters evaluated in raw semen showed no differences between D and M males. However, post-thaw semen quality was significantly (P < 0.05) reduced in all analyzed parameters except %NAS and %NOR in M males groups as compared to the D male (Table 1). It can be concluded that Barbary sheep raw semen collected by electroejaculation is of sufficient quality to be used in an artificial insemination program and can be successfully frozen in commercially available Triladyl extender. However, the post-thaw viability of semen may considerably depend on the male reproductive status in the flock. Table 1. The Bermeya goats are an endangered autochthonous breed distributed in the north of Spain. To ensure their genetic diversity and long-term survival, morphological and functional parameters of the semen must be known in order to preserve the current genetic stock in a germplasm bank. The aim of this work was to establish basic characteristics and post-thaw survival of Bermeya goat's semen obtained by electro-ejaculation, that is not well described in the literature. The semen was collected by electro-ejaculation from 7 bucks, 1 to 7 years old, twice per week, for 9 weeks (n = 83). Fresh semen was evaluated for volume (V), concentration (C), motility, morphology, functional integrity of the sperm (spz) membranes (hypoosmotic swelling test; HOST), and acrosome integrity rate (NAR). Individual and progressive sperm motility were analyzed by means of a computer-assisted sperm analysis system (CASA: SCA 2002�, Microptic, Barcelona, Spain) immediately after dilution with the extender at 37�C, and after cooling to 4�C; five fields per sample (diluted to 204 � 106 spz mL–1) were evaluated under a phase contrast microscope (100�). The NAR and morphological abnormalities of sperm head, midpiece, tail, and cytoplasmic droplets were determined by counting 100 spz under 1000�. For freezing, ejaculates with at least 80% motile spz were diluted at 32�C with Krebs-Ringer solution containing 20% egg yolk and 14% glycerol to a final concentration of 400 � 106 spz mL–1, cooled to 4�C for 90 min, aspirated into 0.25-mL plastic straws (IMV�, L'Aigle, France), frozen at 7 cm above liquid nitrogen (LN2) phase for 10 min, and then plunged into the LN2. Straws were thawed in a water bath at 39�C for 30 s for post-thaw survival analysis. Data were analyzed by the GLM and FREQ procedures (SAS; SAS Institute, Inc., Cary, NC, USA) and expressed as means � standard error. Fresh semen characteristics were: V = 1.7 � 0.1 mL; C = 2619 � 106 � 153 spz mL–1; total and progressive motility were 89.0 � 2.1% and 66.9 � 2.1%, respectively. Percentages of head abnormalities were 4.8 � 0.5; midpiece: 3.8 � 0.7; tail: 4.7 � 1.0; cytoplasmic droplets: 8.3 � 0.7; intact acrosome: 91.8 � 0.6; and membrane integrity: 49.2 � 2.1. At 4�C, the % of total motile spz was 62.6 � 1.6, and the post-thaw survival rate was 46.3 � 1.5. There were only individual differences (P < 0.001) between bucks on sperm concentration, head abnormalities, and cytoplasmic droplets. In conclusion, our results indicate that semen quality is related to each individual animal and that electro-ejaculation allows collection of semen of satisfactory quality to use as fresh and for cryopreservation. However, the validity of our results for possible future sperm banking of endangered Bermeya goats semen must be confirmed by field trials.

ABSTRAK. Penelitian ini bertujuan untuk menentukan karakteristik dan komposisi plasma semen sapi aceh. Sampel semen dikoleksi dari 2 ekor sapi aceh jantan sehat berumur 3,0-3,5 tahun menggunakan vagina buatan. Koleksi semen dilakukan satu kali dalam seminggu selama enam minggu. Evaluasi karakteristik semen meliputi volume, warna, konsistensi, pH, konsentrasi spermatozoa, motilitas, viabilitas, abnormalitas spermatozoa dan komposisi kimia plasma semen. Data kualitas dan komposisi kimia plasma semen yang diperoleh dianalisa secara diskriptif. Hasil penelitian menunjukkan semen segar sapi aceh memiliki volume 3,82 ± 0,47 ml, warna krem keputihan, konsistensi kental, pH 6,84 ± 0,17, konsentrasi 1194 ± 4,80 × 106sp/ml, persentase motilitas 77,28 ± 3,17 % dan abnormalitas 5,98 ± 1,77%. Plasma seminalis semen segar sapi aceh mengandung protein total sebanyak 1,51 ± 0,21 g/100ml, Kalium 75,80 ± 22,63 mg/100 ml dan Magnesium 32,30 ± 17,64 mg/100 ml lebih tinggi, tetapi Natrium 170,00 ± 81,60 mg/100 ml; kalsium 40,00 ± 12,76 mg/100 ml, Fosfor 16,70 ± 6,17 mg/100 ml, dan Mangan 7,08 ± 2,48 mg/100 ml lebih rendah dibandingkan dengan sapi potong. Secara umum kuantitas dan komposisi plasma seminalis semen sapi aceh yang dihasilkan cukup baik, dan memenuhi syarat untuk diproses sebagai semen beku. (The characteristics of semen and chemical composition of the seminal plasma of aceh cattle maintaned in BIBD Saree Aceh Besar) ABSTRACT. This study aim were to determine characteristics and composition of semen plasma Aceh cattle. Semen samples were collected from two healthy Aceh bulls that aged from 3.0 to 3.5 year. Samples were yielded by using an artificial vagina. Samples collecting were conducted once a week for six consecutive weeks. The evaluation of semen characteristics includes volume, color, consistency, pH, sperm concentration, motility, viability, abnormal spermatozoa and seminal plasma chemical composition. Data of semen quality and chemical composition of seminal plasma were analyzed descriptively. The evaluation showed that Aceh Cattle semen had 3.82 ± 0.47 ml volume, beige whitish color, thick consistency, 6.84 ± 0.17 pH, 1194 ± 4.80×106 sp/ml sperm concentration, 77.28 ± 3.17% motility and 5.98 ± 1.77% abnormal spermatozoa. Plasma seminalis fresh semen of Aceh Cattle contained 1,51 ± 0,21 g/100ml total protein; 75,80 ± 22,63 mg/100ml potassium and 32,30 ± 17,64 mg/100ml magnesium higher than the heifer, but 170,00 ± 81,60 mg/100ml sodium; 40,00 ± 12,76 mg/100ml calcium, 16,70 ± 6,17 mg/100ml fosfor, and 7,08 ± 2,48 mg/100ml mangan lower. It was concluded that quality and composition of seminal plasma Aceh cattle semen could be made as frozen semen.

Semen was collected from four rams, using artificial vagina and viability%, motility% and plasma membrane integrity% were measured. Fresh ejaculates (n = 32) were separated by modified swim-up separation using modified human tubal fluid medium. Four fractions of supernatant were collected at 15-minute intervals. The mean volume, mass activity, concentration, motility%, viability%, normal morphology and membrane integrity% (HOST +ve) of fresh semen were 1.0 ± 0.14, 4.1 ± 0.1 × 109 spermatozoa/ml, 85.0 ± 1.3, 89.4 ± 1.0, 85.5 ± 0.7, 84.7 ± 0.5 respectively. There was no significant (P>0.05) difference in fresh semen quality parameters between rams. The motility%, viability% and HOST +ve % of first, second, third and fourth fractions were 53.4 ± 0.5, 68.2 ± 0.3, 74.8 ± 0.3 and 65.5 ± 0.4; 55.5 ± 0.4, 66.2 ± 0.4, 74.5 ± 0.3 and 73.6 ± 0.3 and 66.7 ± 0.5, 66.8 ± 0.5, 65.2 ± 0.4 and 74.7 ± 0.5 respectively. The motility%, viability% and membrane integrity% of separated semen samples differed significantly (P<0.05) between four fractions. The mean motility% and viability% were significantly higher (P<0.05) in third fraction (74.8 ± 0.3%), whereas the mean HOST +ve% was significantly higher (P<0.05) in fourth fraction (74.7 ± 0.5). All quality parameters of separated spermatozoa were significantly (P<0.05) lower than that of fresh semen. ABSTRACT Background Wide variation in fertility rates is observed when using frozen bull semen, even when the bulls have met quality standards for semen production. Therefore, a simple and reliable test to assess the freezing potential of bull semen based on the analysis of fresh semen or blood would be of great value. Attention is now turning to assessment of seminal plasma components such as proteins and elements. In the present study, the concentrations of macro- and microelements in fresh bull semen plasma and in serum and their correlation with quality characteristics of fresh semen and with semen quality after freezing and thawing were determined. Ejaculates were collected from 30 mature bulls, and semen volume, concentration, sperm motility, morphology, tail membrane integrity, plasma membrane permeability and DNA fragmentation were determined on the day of collection and after freezing and thawing. The concentrations of macroelements (Na, Mg, K and Ca) and microelements (Cu, Fe, Zn and Se) were determined in the seminal plasma and serum. The semen samples were classified into satisfactory and unsatisfactory groups according to the fresh semen quality. Results Zinc and Se levels measured in serum were associated with almost all fresh and frozen-thawed semen quality characteristics, while Fe levels were associated only with acrosomal defects in fresh semen. Zinc and Fe levels in fresh seminal plasma were associated with various quality characteristics of fresh and frozen-thawed semen, while Se level in fresh seminal plasma was not associated with any of the semen quality characteristics. Conclusions Microelements were shown to be useful as biomarkers involved in the analysis of bull sperm quality and could be used as an additional tool to predict bull semen quality after freezing and thawing. Our results confirm that the analysis of Zn and Se levels in serum and Zn, Cu and Fe levels in fresh seminal plasma can provide information to discriminate between bull semen samples with spermatozoa with high or low cryotolerance.

Background: New methodologies have been developed seeking to maximize pregnancy rate in female dogs created in commercial kennels, and also in order to maintain the quality of canine semen after dilution, refrigeration or freezing. One of the main factors that generate damage to sperm is oxidative stress, to minimize sperm damage, selenium and antioxidants like vitamin E are administered, by oral administration, seeking to improve the quality of semen. The objective was to study the effect of vitamin E and selenium, by oral administration, in the quality of fresh, refrigerated and frozen semen in adult dogs French Bulldog breed.Materials, Methods & Results: Semen samples were collected from 5 adult dogs, French Bulldog breed, being 2 semen drawing before the daily oral supplementation with vitamin E and selenium (ESE®) and semen drawing at 20, 40 and 60 days after the beginning of oral supplement. The ejaculated samples were diluted in TRIS - fructose citric acid (3.28 g TRIS-hydroxy-methyl-amino-methane, 1.78 g of citric acid monohydrate and 1.25 g of D - fructose, dissolved in 100 mL of distilled water and added of 20% egg yolk and 6% of glycerol. The characteristics evaluated in fresh semen were: volume (mL), color, appearance, concentration (x106 / mL), sperm motility (%), sperm strength (1 to 5) and morphology (%). For refrigerated and frozen semen were analyzed: sperm motility (%), sperm strength (1-5) and morphology (%). Diluted semen samples were centrifuged at: 1500 g/10 min and “pellets” formed by sperm of each ejaculated, detached from the tube wall were diluted homogeneously in the diluent TRIS type up to the final volume of 1.5 mL. After that, packaged in 0.5 mL French straws, kept under refrigeration at 5ºC/4 h, placed in nitrogen vapor at -120ºC/15 min, and dipped in liquid nitrogen at -196ºC and then stored on identified rachis and stored in liquid nitrogen container until the time of thawing in  water bath at 37°C/30 s for semen microscopic analysis. Data from fresh, refrigerated and frozen semen were statistically analyzed by analysis of variance and the average compared by 5% of Tukey test. Fresh semen sperm concentration differed (P < 0.05) between the samples, rising after 40 days after the beginning of oral supplementation with selenium and vitamin E. For the spermatic strength, better score (P < 0.05) was observed at collection 4, in 40 days after the beginning of oral supplementation to dogs. For fresh and refrigerated semen, the total defects, defects of head, acrosome and tail did not differ (P > 0.05) between the samples. Total sperm defects and minor head and tail defects did not differ (P > 0.05) between the samples in post-thawing. Regarding the acrosome defects after thawing, there was a significant reduction (P < 0.05) in samples performed 40 and 60 days after the beginning of oral supplementation with selenium and vitamin E.Discussion: Attention should be paid for what purpose the extenders within the refrigeration or freezing biotech will be used. The managed supplement, by oral administration, containing selenium and vitamin E, influenced beneficially raising the sperm concentration in fresh semen and decreasing the acrosome defects in frozen semen. Oral administration of supplementation with selenium and vitamin E is recommended for improving the quality of fresh and frozen semen in dogs.

Abstract Study question What is the cause of semen quality impairment in oncological patients during fertility preservation programs? The cancer type and stadiation or the resulting inflammatory state? Summary answer The inflammatory state seems to be related to the decrease of sperm concentration, motility, morphology and viability due to the worsening of oxidative stress microenvironment. What is known already Fertility preservation acquired a great importance in the last decades due to increase survival of oncological patients, boost of diagnosis under 40 years and postponement of paternal age. At the time of cryopreservation, only one third of these males are normozoospermic. Tumor itself or other factors, added to psychological reasons, may be involved but there is no clear evidence. An imbalance of ROS (reactive oxygen species) in semen can compromise its quality. However, the correlation between cancer-related generalized stress state and fertility is poorly investigated. Inflammatory conditions induced by infections and pathologies, including cancer, increase ROS. Study design, size, duration Retrospective observational analysis was performed on 45 patients (29.0 ± 6.9 yrs) recruited during their fertility preservation program between 2016 and 2019 with written consent on use of their clinical data for research purpose. Patients presented several oncological diagnoses. Semen samples obtained from multiple collections (N = 58) were analyzed before applying standard freezing protocol. Data on semen parameters, inflammatory indices, hematological values and type/stage of tumors were collected. No exclusion criteria were applied. Participants/materials, setting, methods Routine semen analysis was performed according to the WHO standards. Sperm concentration and motility were evaluated on Makler Chamber, whereas eosin stain and Diff-quick slides were used for viability and morphology, respectively. Lymphoma was present in 72% of cases, leukemia in 8%, seminoma in 7% and other cancers in 13%. Correlations (Pearson/Spearman tests) among principal semen parameters and hematological values (leukocytes, erythrocytes, hemoglobin, RDW, albumin, etc.) were calculated with a P-value &lt;0.05 considered statistically significant. Main results and the role of chance The majority of semen samples showed a severe impairment, with one or more parameters under lower reference limits (WHO): 48.3% had sperm concentration under 15 millions/ml, 43.1% had a progressive motility under 32%, 41.4% had viability under 58% and 91.4% had abnormal morphology (under 4%). The role of potential inflammatory state was analyzed by correlating semen parameters and some hematological values. No correlation was found with cancer type. Negative association resulted between progressive motility (%PR) and leukocytes (p = 0.041) or RDW% (p = 0.015), but positive one with albumin (p = 0.012). Even sperm count, total motility (%PR+NP) and morphology were significantly correlated to RDW% (p = 0.003, p = 0.032, p = 0.034, respectively). These findings suggest a possible role of inflammation and ROS related generation in semen quality impairment. Indeed, albumin exerts a protective action, but leukocytes are known to cause ROS increase. Cancer-induced oxidative stress state may alter red blood cells homeostasis and vitality and increase erythrocytes turnover resulting in high RDW values. It is likely semen is worse when blood values indicate more severe cancer-induced inflammatory condition. Limitations, reasons for caution Significant correlations with type/stage of cancer were not found due to small number of each diagnosis, in spite our study considered 3 years of patients inclusion. Moreover, we lack to analyze the same patient before the cancer onset to avoid the influence of inflammatory state generated by the tumor itself. Wider implications of the findings: Understanding the influence of cancer-induced inflammatory state on semen quality could increase the awareness that clinicians should direct patient to the fertility preservation as soon as possible, even if diagnosis is still ongoing. It should be evaluated whether offering specific treatments may reduce oxidative stress conditions. Trial registration number Not applicable

Abstract Background: Iodine is an essential element for hormones synthesized by the human thyroid gland. Thyroid hormone deficiency affects all body tissues, including multiple endocrine changes that alter growth hormones, corticotrophin, glucocorticoids, and gonadal function. In many countries, subfertility is an important problem affecting about 10–15% of all couples trying to conceive. Male subfertile factors have accounted for 25–30% of all cases in their studies. The aim of this study was to study the correlation between urinary iodine concentration (UIC) and semen quality of fertile male in China.Methods: semen and urine samples were collected from males who attended a survey for the reproductive physiological constants of male and female in China. All participants were fertile and they should be free from diseases of male reproductive system and thyroid related diseases. We studied the correlation between UIC and semen quality parameters in fertile males and the effect of iodine concentration on male reproduction. Participants were divided into six groups based on UIC. We measured semen parameters and UIC. Semen parameters or time-to-pregnancy (TTP) were analyzed by Spearman correlation, linear regression and multivariable logistic regression. Results: 1089 men were recruited in the study. The mean and median age of participants were 27.46±0.12 and 27±3.88 years, respectively. UIC of 274 (25.16%) men was lower than the UIC of WHO recommended. UIC of 405 men located in the region of WHO recommended. 62.35% of them were not located in the optimal region. Semen concentration and total sperm count were found to decrease when UIC increases when the UIC≥100 ug/L. Conclusions: A male with deficient UIC or excessive UIC has more risk of lower semen quality such as sperm concentration and total sperm count. In addition, if a male with deficient UIC or excessive UIC, his wife has longer TTP than the males who had normal UIC. Deficient UIC associated with lower semen quality and TTP than excessive UIC.

The objective of this study was to determine the semen cryopreservation capacity of BBB bulls in Hanoi-Vietnam. Research conducted on the fresh semen collected from 05 BBB bulls. Results showed that semen color was normal (milky white, ivory white, ivory yellow), semen volume ranged from 6.35 mL to 7.48 mL (P <0.05), initial motility of semen ranged from 80.53% to 82.92% (P <0.05), sperm concentration in semen  ranged from 1.02 x 109 sperms/ml to 1.12 x 109 sperms/mL (P <0.05), abnormal sperm ratio ranged from 6.45% to 8.12% (P <0.05), alive sperm ratio ranged from 76.34% to 82.97% (P <0.05), sperm motility after thawing from straw semen ranged from 71.33% to 75.92% (P<0.05). In conclusion, successfully semen collection from 05 breeding BBB bulls at Hanoi Cattle Breeding Joint Stock Company, semen samples had normal color and good quantity and quality, suitable for production of frozen semen; and semen cryopreservation of straws of the 05 bull BBB semen mentioned at -196oC, sperm motility after freezing-thawing reached the economic and technical norms of 675/2014 of the Ministry of Agriculture and Rural Development.

The current study was planned to evaluate the supplementation of Polyvinylpyrrolidone in extender on cryopreservation of Nili-Ravi buffalo bull semen. The semen samples were collected from Nili-Ravi buffalo (Bubalus bubalis) bull kept at SPU Qadirabad, District Sahiwal, Pakistan. Qualifying semen ejaculates having motility >60%, volume >5-6ml and concentration >0.5 billion/ ml were diluted 50 × 106 motile sperm ml approximately at 37°C in Tris-citric acid extender supplemented with different concentrations of PVP (0.01, 0.05, 0.1mM). The extender without PVP was kept as control. Semen was stored at 4°C for a period of 2 h and kept at 4°C for 4h. Semen was filled in 0.5 ml French straws using suction pump at 4°C, plunged and stored in liquid nitrogen (-196°C). Semen straws were rewarmed at 37°C for 30 seconds and assessed for sperm motility, plasma membrane integrity (PMI), dead sperm percentage and the live sperm percentage. The data on the role of PVP on different parameters of semen quality were analyzed by using ANOVA and RCBD. Higher percentage (P< 0.05) of sperm motility (66.1±7.51 and 59.4±10.72) and PMI (72.9±5.39 and 75.7±6.5) was observed in extenders having 0.05 mM and 0.1mM PVP compared to extenders having 1.5mM PVP and control. The percentage acrosomal integrity was observed greater (P< 0.05) in extended semen containing 0.1mM (68.2±0.50) PVP compared to extenders having 0.01 and control.

The purpose of this research was to analysis the value of repeatability and correlation among the traits affecting the production of frozen semen from Holstein’s bull in Indonesia. Repeatability and correlation were calculated based on the data of frozen semen production of 15.699 records from 44 Holstein bulls at Singosari Artificial Insemination Center (SAIC) and 8.935 records from 39 Holstein bulls at Lembang Artificial Insemination Center (LAIC). Repeatability for volume, motility, fresh semen concentration and frozen semen production was evaluated by intraclass correlation method. The repeatability values of LAIC for volume, motility, fresh semen concentration and frozen semen production were 0.60; 0.54; 0.37 and 0.47. The repeatability values of SAIC for volume, motility, fresh semen concentration and frozen semen production were 0.54; 0.30; 0.43 and 0.29. The linear correlation value between volume, motility and fresh semen concentration with the amount of semen produced per collections were 0.41, 0.36, and 0.58. Concentration was the most factors influencing the number of frozen semen produced. The effectiveness of the selection of Holstein's frozen semen producing could be determined by the value of repeatability and the phenotypic correlation among semen quality traits such as volume, motility, concentration and frozen semen production.
