Effects of supplemental D-methionine in comparison to L-methionine on nitrogen retention, gut morphology, antioxidant status, and mRNA abundance of amino acid transporters in weanling pigs

Abstract A N-balance experiment was conducted to test the hypothesis that D-Methionine (D-Met) has the same bioavailability and efficacy as L-Methionine (L-Met) when fed to weanling pigs. A Met-deficient basal diet containing 0.24% standardized ileal digestible (SID) Met was formulated. Six additional diets were formulated by adding 0.036, 0.072, or 0.108% D-Met or L-Met to the basal diet, and these diets, therefore, contained 77, 87, or 97% of the requirement for SID Met. Fifty-six barrows (10.53 ± 1.17 kg) were housed in metabolism crates and allotted to the 7 diets with 8 replicate pigs per diet. Feces and urine were collected quantitatively with 7-d adaptation and 5-d collection periods. Blood and tissue samples from pigs fed the basal diet and pigs fed diets containing 0.108% supplemental Met were collected on the last day. Results indicated that N retention (%) linearly increased (P < 0.01) as supplemental D-Met or L-Met increased in diets. Based on N retention (%) as a response, the linear slope-ratio regression estimated the bioavailability of D-Met relative to L-Met to be 101% (95% confidence interval: 57 to 146%). The villus height and crypt depth in the jejunum were not affected by Met level or Met source. Total antioxidant capacity or thiobarbituric acid reactive substances concentrations in plasma or tissue samples from pigs fed the control diet or diets containing 0.108% supplemental D-Met or L-Met were not different. Abundance of mRNA for some AA transporters analyzed in intestinal mucosa of pigs also did not differ. Therefore, it is concluded that D-Met and L-Met are equally bioavailable for weanling pigs.

Abstract Two nitrogen balance studies were conducted to evaluate the relative bioavailability values (RBV) of DL-Methionine (DL-Met) and DL-Methionine hydroxy analog calcium salt (MHA-Ca) to L-Methionine (L-Met) as Met sources fed to pigs. In Exp. 1, 42 pigs were assigned to 7 treatments feeding with basal diet (BD) formulated to be deficient in Met (0.22% standardized ileal digestible basis) but adequate in other amino acids. Diets included (1) BD, (2) BD + 0.025% DL-Met, (3) BD + 0.050% DL-Met, (4) BD + 0.075% DL-Met, (5) BD + 0.025% L-Met, (6) BD + 0.050% L-Met, and (7) BD + 0.075% L-Met. Increasing levels of L-Met and DL-Met enhanced N retained (g/d) and N retention (% of intake) linearly (P < 0.01). Using a linear slope-ratio procedure, a product-to-product RBV of DL-Met compared to L-Met was 94% (95% confidence limits: 65 to 123%) based on N retained expressed as g/d and 99% (95% confidence limits: 70 to 128%) for N retention expressed as % of intake. In Exp. 2, 42 pigs were allotted to 7 treatments in another N-balance trial. Diets included (1) BD, (2) BD + 0.025% L-Met, (3) BD + 0.050% L-Met, (4) BD + 0.075% L-Met, (5) BD + 0.030% MHA-Ca, (6) BD + 0.060% MHA-Ca, and (7) BD + 0.089% MHA-Ca. An increase in dietary inclusion rates of L-Met increased (P < 0.01) N retained (g/d) linearly while increasing levels of MHA-Ca had no effects (P > 0.05) on N retained (g/d) and N retention (% of intake). Using linear slope-ratio regression, the RBV of MHA-Ca compared to L-Met was 70% (95% confidence limits: 59 to 81%) on a product-to-product basis or 83% on equimolar basis based on N retained expressed as g/d. Overall, the mean RBV of DL-Met to L-Met of 97% (95% confidence limits cover 100%) indicated that DL-Met and L-Met are equally bioavailable as Met sources in pigs. Compared to L-Met, the RBV of MHA-Ca was lower at 70% (95% confidence limits: 59 to 81%) on a product-to-product basis or 83% on equimolar basis in starter pigs.

ABSTRACT: The purpose of this research was to investigate the progress of the rancidity in Nile tilapia diets containing bacterial biomass of Rubrivivax gelatinosus . Six experimental treatments comprised basal diet (negative control), diet with asthaxanthin (positive control) and 4 diets with different concentrations of the bacterial biomass. The thiobarbituric acid assay for rancidity analyses were accomplished after 6 and 12 months of diets storage. It was concluded that Rubrivivax gelatinosus biomass minimized the racidity in Nile tilapia diets in 32.52 to 44.72% at 6 months and in 37.85 to 52.37% at 12 months of storage.

We determined the effects of increased dietary concentrations of molybdenum and sulfur on the accumulation and tissue concentrations of cadmium in sheep, and compared them with effects on copper. Forty sheep, each weighing approximately 40 kg, were adjusted for 3 weeks to a basal diet of 80% wheaten chaff and 20% lupin seed containing (per kg dry weight) 0·016 mg Cd, 0·45 mg Mo, 3·4 mg Cu, and 1·9 g S. On Day 0 of treatment, 8 sheep were killed and the tissues analysed for trace minerals to provide a baseline value. The remaining sheep were divided into 4 dietary treatment groups: control (basal diet plus 4 mg Cd/kg), +Mo (control diet plus 15 mg Mo/kg), +S (control diet plus 4 g S/kg), +Mo+S (control diet+15 mg Mo+4 g S/kg). The treatment period lasted 80 days, after which sheep were killed for tissue samples. Sulfur alone reduced the accumulation of Cd in liver, kidney, and muscle by 60% compared with control sheep (P < 0·05). Molybdenum alone reduced Cd accumulation by 40% in liver and muscle (P < 0·05) and 30% in kidney (P = 0·09). When provided together (+Mo+S), the effect was equivalent to feeding with Mo alone, showing that Mo blocked the effect of S. Cadmium concentrations in whole kidneys for the 4 respective treatments were 6·40 ± 0· 7, 2·8 ± 0·3, 4·5 ± 0·8, and 5·0 ± 0·5 mg/kg fresh weight. The pre-treatment concentration was 0·7 ± 0·2 mg/kg. For Cu in blood and tissues, the effects of Mo and S treatment were consistent with the thiomolybdate hypothesis, and were quite different from those seen for Cd. Copper concentrations in whole kidney for the 4 treatments were 4·1 ± 0·1, 3·5 ± 0·2, 4·7 ± 0·3, and 22·4 ± 3·9 mg/kg fresh weight. The pre-treatment concentration was 4·1 ± 0·3 mg/kg. The results show that increased dietary levels of Mo and S reduce the accumulation of Cd in tissues, and the mechanisms of action differ from those involving Cu.

Four calves equipped with permanent rumen and T-piece duodenal caninulus were given four diets in a Latin square experiment carried out at two stages of development. DM flow was measured by dual-phase markers. The basal diet of hay and concentrate was calculated to be low In rumen degradable N (RDN) and. tissue N. Additional RDN was provided by ad­ding 6, 12 or 18g urea/kg concentrate. Supplementary urea did not affect OM digestion either in the stomach or In the entire digestive tract. At the older age OM digestion in the stomach and the entire tract was significantly higher. On the basal diet, N retention was low. The flow of microbial N measured by the DAPA technique was not affected by diet and it was calculated that between 3 and 7g "s/d were recycled to the rumen. The addition of supplementary urea increased N retention par­ticularly at the lowest level of supplementation due to a concomitant decrease In urinary N.

Three trials were conducted to evaluate the potential of cassava (Manihot esculenta) foliage (leaves and petioles) as a protein supplement for sheep. In the first trial, nylon bag degradability of cassava foliage was studied. Proximate analysis of cassava foliage was conducted before and after the incubation. Rumen Ammonia Concentration (RAC) and pH were determined at 0, 4 and 8 hours after feeding cassava foliage diet (30%). Proximate composition of sundried cassava foliage was (%) DM 91.25, CP 18.55, NDF, 31.41, ADF, 29.2, EE, 6.6, Ash, 12.95. Nylon-bag dry matter disappearance (DMD) increased significantly (P<0.05) from 15.75% to 75.5% RAC values increased significantly from 8.20mg NH3/100ml at zero hour to 11.41mg NH3/100ml at 8 hours post feeding. Rumen fluid pH was not significantly changed by the 30% cassava foliage diet. In the second trial lasting 56 days, twenty-eight Yankasa/ WAD yearling rams of initial average weight of 18kg were balanced for weight and randomly allotted to seven treatments. All rams except those on control diet were fed cassava foliage or groundnut haulms or both at 1.0% or 1.5% of body weight (BW) as supplements to a basal diet of Gamba hay. Feeding Gamba hay alone (control) resulted in weight loss (-30.5g/day) but with cassava foliage or groundnut haulms supplementation at 1.0% BW, significantly higher gains of 39.2 and 44.6g/day were achieved respectively. At 1.5%BW level of supplementation with cassava foliage or groundnut haulms higher weight gains of 41.2 and 51.7g/day were achieved while feeding the (50.50) combined supplements at 1.0 and 1.50% BW resulted in live weight gains of 65.1 and 69.3g/day respectively. The third trial consisted of a four week grazing trial followed by a metabolic study. Twenty-one Yankasa/WAD yearling rams were randomly allotted to seven treatments and were balanced for live weight. All rams were allowed to graze standing digitaria hay as basal diet, except the rams on control diet. Those on treatment were fed cassava foliage or groundnut haulms or a (50:50) combination of both at 0.5% and 1.0% (BW). The metabolic study consisted of a seven day adjustment period and a seven day total collection period. There was no significant difference (P>0.05) in the live weight gains of rams on the control diet compared with those supplemented except for those supplemented with cassava foliage and groundnut haulms combined at 1% level where the weight gain was significantly higher (P<0.05). Feed intake was not significantly (P>0.05) affected by feeding either supplement. Supplementing digitaria hay with cassava foliage at both 0.5% and 1.0% BW did not significantly increase the nitrogen retention values, but groundnut haulms supplemented at 1.0% BW significantly (P<0.05) increased the nitrogen retention values.

Ten weaned rabbits were fed a basal (control) diet containing 0.12 mg Se/kg. In 10 rabbits the basal diet was supplemented with Se-enriched yeast to increase the Se concentration to 0.50 mg/kg. Rabbits were slaughtered at the age of 11 weeks. Samples of meat, liver and hair of 4 rabbits from each group were taken and analyzed. No effect of Se on growth, feed conversion and dressing out percentage was observed. Loin and hindleg meat, liver and hair of control rabbits contained 93, 98, 521 and 267 &mu;g Se/kg, respectively (average values). In treated rabbits the corresponding Se concentrations were 400, 389, 1 414 and 914 &mu;g/kg. Supranutritional Se supply had no effect on the activity of glutathione peroxidase in meat, and oxidative stability of meat expressed as production of thiobarbituric acid-reactive substances in meat stored for 3 and 6 days. Thus, the enrichment of meat with Se is the main benefit of Se supplementation of rabbit diets.

SummaryThree experiments were conducted to investigate the influence on nitrogen retention of feeding protein supplements to early weaned lambs direct to the abomasum via the oesophageal groove. In Exp. 1 the lambs received a basal diet of barley and a protein mixture consisting of soyabean meal, sunflower meal and fish meal which was given either in a liquid suspension from a bottle or as dry feed incorporated with the barley. The results showed that the regression coefficients of nitrogen retention on intake were 0.48 and 0.33 with liquid and dry feeding respectively. The regressions of urinary nitrogen on intake were with liquid and dry feeding respectively 0.26 and 0.44.In Exp. 2, various sources of protein were compared isonitrogenously as substitute for part of a barley concentrate ration. The nitrogen intakes and retentions were 14·28 and 5·09, 20·16 and 9·39, 20·53 and 8·19, 19·00 and 7·80, and 18·87 and 7·86 g/day for the control diet and fish meal, yeast, soyabean meal and sunflower meal supplemented diets respectively.In Exp. 3 the fish meal supplement was compared with a high lysine, bloodmeal and with casein as liquid supplements to a basal diet of barley concentrate. The intake and retention of nitrogen were for bloodmeal, casein and fish meal respectively 23·41 and 7·48, 22·56 and 9·11, and 21·88 and 8·31 g/day. It appeared that the protein sources high in methionine resulted in the highest nitrogen retention. The implications of these findings in relation to the nutrition of young ruminants is discussed together with an evaluation of the experimental method as a technique to study postruminal digestion.

ABSTRACT: The aim of this study was to measure the nitrogen (N) balance of pigs fed with lysine-limiting diets containing practical levels of wheat bran (WB) or soybean hulls (SH). Twelve pigs with average weights of 57.36±2.01 and 72.68±3.24 kg were used in trials 1 and 2, respectively. In trial 1, treatments were CT1 - control diet and WB - diet with inclusion of 15% WB. In trial 2, CT2 - control diet and SH - diet with inclusion of 6% SH. Fibrous diets increased (P<0.05) the fecal N in 63.54 and 60.55% in relation to CT1 and CT2, respectively. The urinary N was higher (P<0.05) in pigs receiving the WB diet, but was not influenced in the trial with SH. The N retention (NRET) was higher (P>0.05) in pigs ingesting the WB diet; although, when the proportion of NRET was expressed relative to N ingested (NING) there was no difference (P>0.05) between treatments. Inclusion of SH did not affect (P>0.05) the NRET when the result was expressed in absolute or proportional terms. In conclusion, inclusion of practical levels of WB and SH does not significantly affect the metabolic costs involved with nitrogen metabolism. However, only WB was able to contribute to the N balance of pigs, while the relative amount of N added by SH was fully recovered in feces.

Abstract A N-balance study was conducted to compare the effect of D-methionine (D-Met) or L-methionine (L-Met) supplementation on N balance, gut morphology and antioxidant status of weaned pigs. Fifty-six weaned barrows (10.5 ± 1.2 kg initial BW) were allotted to 7 diets in 2 blocks. A Met-deficient basal diet (BD; 0.24% standardized ileal digestible (SID) Met) but adequate in other AA, was supplemented with 3 graded levels (0.036, 0.072, and 0.108%) of D-Met or L-Met. After a 7-d adaptation, feces and urine were collected quantitatively for 5 d to determine N balance. At the completion of the experiment, blood samples were collected from all pigs. Pigs fed the BD and pigs fed the highest level of SID Met (0.34%) of both Met sources were euthanized and tissue samples from liver, kidney, muscle (longissimus dorsi), duodenal and jejunal mucosa were collected. N retention as % of N intake increased (P &lt; 0.001; 67, 72, 73, 74, 71, 74, 74%, respectively) by graded supplemental level of D-Met or L-Met. However, there was no interaction between Met source and supplemental level for all N-balance metrics. Using a slope-ratio regression, the bioavailability of D-Met relative to L-Met was 100.1% (95% confidence intervals: 85-116%) based on N retention (% of N intake). Villus height and crypt depth in the duodenum and jejunum were not affected by Met sources. Pigs fed the D-Met diet had a greater (P &lt; 0.05) total glutathione concentration in liver (4.9 vs. 1.5 µM) vs. BD. However, total antioxidant capacity and concentration of thiobarbituric acid-reactive substances in liver, muscle or plasma samples were not different among treatments. Supplementation with D-Met increased glutathione peroxidase activity in kidney (878 vs. 413 and 229 mU/mL; P &lt; 0.05) compared with BD or L-Met diet, however, activity of glutathione reductase in liver and kidney were not affected by treatments. These data indicate that D-Met and L-Met are equally efficient to support N retention, intestinal morphology and oxidative status in weaned pigs.

A study was carried out to investigate the effect of feeding two varieties of cowpea haulm on  growth performance, nutrient digestibility and nitrogen balance in Red Sokoto bucks. The  experiment consisted of 15 Red Sokoto bucks of average weight of 10kg, which were balanced  for weight and randomly assigned to three dietary treatments with five bucks per treatment in  a complete randomize design. The three treatment diets consisted of Brachairia decumbens  hay as basal diet and concentrate diets which were supplemented with cowpea haulms at 0%  (Control), 10% (SAMPEA14) and 10% (SAMPEA15). The study lasted for 90 days. Total dry  matter intake (TDMI, g/day), total weight gain (TWG, kg), faecal (unit) and urine (unit)  outputs were recorded. Results indicated that the TDMI were different (P<0.05) with higher  intake (237.64 g/day) in bucks fed control diet, followed by 223.00 g/day 10% SAMPEA 15  and 10% haulm of Sampea 14 (206.49g/day) in the concentrate. The TWG and average daily  weight gain increased by 59.77% and 59.76%, respectively with respect to 10% SAMPEA 14  haulm inclusion compared to the control diet. The digestibilities of dry matter (DM), crude  protein (CP), crude fibre (CF), ether extract (EE) and nitrogen free extract (NFE) were  higher (P<0.05) in bucks fed diet constaining 10% SAMPEA 14 inclusion level compared to  the treatment diets. The quantity of N retained by the bucks was higher (P<0.05) with the  inclusion of 10% of SAMPEA 14 (3.79 g/day) compared to those fed 10% SAMPEA 15 (1.18  g/day) and the control diet (0.68 g/day). It is concluded that farmers should include 10%  SAMPEA 14 in concentrate diet of Red Sokoto bucks for optimum performance under smallholder production system.     Une étude a été menée pour étudier l'effet de l'alimentation de deux variétés des haulms de niébé sur la performance de croissance, la digestibilité des nutriments et l'équilibre azoté chez les mâles Red Sokoto. L'expérience s'est composée de 15 bucks rouges de Sokoto de poids moyen de 10kg, qui ont été équilibrés pour le poids et aléatoirement assignés à trois traitements diététiques avec cinq dollars par traitement dans une conception randomize complète. Les trois régimes de traitement se sont composés deBrachairiadecumbenshay comme régime basal et régimes concentrés qui ont été complétés avec des haulms de niébé à 0% (contrôle), 10% (SAMPEA 14) et 10% (SAMPEA 15). L'étude a duré 90 jours. La consommation totale de matière sèche (TDMI, g/day), le gain de poids total (TWG, kg), les sorties fécales (unitaires) et urinaires (unitaires) ont été enregistrées. Les résultats ont indiqué que le TDMI était différent (P<0.05) avec une consommation plus élevée (237.64 g/jour) dans le régime de contrôle alimenté par les mâles, suivi de 223.00 g/jour10% SAMPEA 15 et 10% de transport de Sampea 14 (206.49 g/jour) dans le concentré. Le TWG et le gain quotidien moyen de poids ont augmenté de 59.77 % et de 59.76 %, respectivement en ce qui concerne l'inclusion de 10 % de SAMPEA 14 haulms par rapport au régime de contrôle. Les digestibilités de la matière sèche (le 'DM'), des protéines brutes (le 'CP'), des fibres brutes (le 'CF'), de l'extrait d'éther (le 'EE') et de l'extrait sans azote (le 'NFE') étaient plus élevées (P<0,05) chez les mâles nourris à l'alimentation constaining 10% SAMPEA 14 niveau d'inclusion par rapport aux régimes de traitement. La quantité de N retenue par les mâles était plus élevée (P<0.05) avec l'inclusion de 10% de SAMPEA 14 (3.79 g/jour) par rapport à celles nourries 10% SAMPEA 15 (1.18 g/jour) et le régime de contrôle (0,68 g/jour). Il est conclu que les agriculteurs devraient inclure 10% SAMPEA 14 dans le régime concentré de bucksRed Sokoto pour une performance optimale dans le cadre du système de production des petits exploitants.
