The first color photograph of the whole earth taken by an astronaut aboard the Apollo 11 spacecraft in 1969 appeared in posters, popular magazines and films, and a staggering number of advertising campaigns promoting the sale of an enormous variety of consumer goods and services. Like any iconic visual image repeatedly used in a variety of diverse contexts for different purposes, the image of the blue planet shimmering under the flow of its delicate atmosphere against the backdrop of the interstellar dark seemed, over time, to lose much of its original significance and emotive power. But for some of us, myself included, it still serves as sign and symbol of two astonishing facts: life on earth is a self-organizing and self-perpetuating whole that evolved from a single organism to a level of enormous complexity and incredibly beauty; our species emerged from this evolutionary process as fully conscious and self-aware beings in the vast cosmos. Many scientists are convinced that biological life must have arisen on other planets and that the process of evolution on some of them resulted in the emergence of intelligent life-forms and advanced technological civilizations. The most ardent promoters of this idea are physicists and molecular biologists, and public acceptance of their views resulted in the creation of the Search for Extraterrestrial Intelligence (SETI) project. The intent of those involved in this project, as many Americans learned for the first time after seeing a movie based on the late Carl Sagan’s book Cosmos , is to intercept intragalactic or intergalactic communications between advanced civilizations on other planets. But what the book and movie failed to mention is that many evolutionary biologists are convinced that the odds that these civilizations exist anywhere else in the vast cosmos are slim to none. The skeptics argue that even if life exists on many other planets, this does not mean that the process of evolution on any of them would result in life-forms that have the capacity to acquire and use a complex symbol system like the human language system.

When Charles Darwin and Alfred Russell Wallace proposed their theory of evolution by natural selection, the concepts of evolution and speciation were not new. Darwin introduced The Origin with “An Historical Sketch,” in which he summarized the work of 34 previous authors who had speculated on evolution and the origin of species. What was new about Darwin and Wallace’s proposition was natural selection as the mechanism of evolutionary change. Darwin further proposed that natural selection was a unifying process that accounts for adaptation, for speciation, and hence for the diversity of life on earth. Darwin and Wallace proposed natural selection as a process that caused evolution. Adaptations are features of organisms that were shaped by this process. The modern version of Darwin and Wallace’s theory allows for other agents of evolution, such as genetic drift, migration, and mutation, but adaptation remains a product of natural selection alone. The virtue of their proposal is that it allows us to develop testable hypotheses about cause-and-effect relationships between features of the environment and presumed adaptations. Natural selection immediately became a source of controversy, although the nature of the controversy has shifted over time. First, there has been considerable debate about the definition of adaptation (e.g., Reeve and Sherman 1993). We do not wish to add to or summarize this debate because we feel that Darwin got it right the first time. Besides defining a cause-and-effect relationship between selection and adaptation, Darwin emphasized that we should not expect organisms to be perfectly adapted to their environment. In fact, this emphasis was a large component of his argument against divine creation. For example, Darwin recognized, through his experience with artificial selection, that different aspects of morphology were in some way “tied” to one another so that selection on one trait would cause correlated changes in others that were not necessarily adaptive. He also recognized that organisms were subject to constraints that might limit their ability to adapt. Finally, he argued that how organisms evolved was a function of their history, so that the response to selection on the same trait would vary among lineages. A more telling criticism considers the application of cause-and-effect reasoning to the interpretation of features of organisms as adaptations, and hence to the empirical study of adaptation.

Introduction: Hepatocholangiocarcinoma (HCC-ICC) is a rare tumor presenting the histologic characteristics of both hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (ICC). As there is no consensus on it management, the therapeutic strategy rests on the specific treatments for HCC or ICC. Programmed cell death 1 (PD-1) inhibitors showed encouraging results in the second line treatment of HCC after sorafenib but it efficacy in HCC-ICC has never been reported. Methods and results: We present the case of a 72-year-old male patient treated for metastatic HCC-ICC due to a viral hepatitis C cirrhosis in progression after two lines of treatment. Tumor was characterized by a PDL-1 status of 85%. Patient received pembrolizumab at doses of 200 mg every 21 days by intravenous infusion. After one injection he was presented an immediate clinical benefit, a partial response was observed after two months of treatment and a complete response two months later. This response was maintained over time along with toxicity-free tumor control after 18 months treatment. Conclusion: To our knowledge, we reported for the first time the efficacy of a PD1 inhibitor treatment in a patient presenting metastatic HCC-ICC due to viral cirrhosis and overexpressing PDL-1 after failure of two lines of treatment.

Abstract This article continues the discussion on the nature of the early language of Buddhism and the language that the Buddha spoke, arguing that the received Pāli transmission evolved out of an earlier Middle Indic idiom, which is identified as a koine. Evidence for this koine can be found by examining correspondence sets within Pāli and its various varieties and by examining parallel, cognate correspondence sets between Pāli and other Prakrits which have survived. This article compares 30 correspondence sets transmitted in the Dhammapada recensions: the Gāndhārī Prakrit verses, the partially Sanskritized Pāli and Patna Dhammapada Prakrit verses, and the fully Sanskritized verses of the Udānavarga. By comparing cognate words, it demonstrates the existence of an underlying inter-language which in many cases can be shown to be the source of the phonological differences in the transmission. The paper includes a discussion on the two major factors of dialect change, evolution with variation over time, and the diffusionary, synchronic influence of dialect variation; it concludes that both are important, with dialect variation – and the phonological constraints of indigenous speakers who adopted MI as a second language – providing the pathways on which the natural evolutionary process was channeled.

Abstract Under Article 27.3(b) of the Trade-Related Aspects of Intellectual Property Rights (TRIPS) Agreement of the World Trade Organization (WTO), all members are required to extend private property rights to life forms. Using official WTO documents, this article analyzes the negotiating positions of WTO members on life patents during a review of Article 27.3(b) which commenced in 1999 and is currently ongoing. Initially, developing countries raised serious ethical concerns regarding life patents, creating a clear North-South divide. However, over time the position of Brazil and India moved away from the ethics of life patents to the prevention of bio-piracy, a position supported by China. Russia too is supportive of life patents. A group of small developing countries have, however, continued to question the morality of life patents despite this “BRIC wall,” changing the dynamics of the negotiations from a North-South divide to one which now includes a South-South divide.

An artist who finds themselves in the last days of a war in the enemy’s defeated capital may not just fix its objects dispassionately. Many factors influence the selection and depicturing manner of the objects. One of the factors is satisfaction from the accomplished retribution, awareness of the historical justice triumph. Researchers think such reactions are inevitable. The article offers to consider from this point of view the drawings created by Soviet artists in Berlin in the spring and summer of 1945. Such an analysis of the German capital’s visual image is conducted for the first time. It shows that the above reactions were not the only ones. The graphics of the first post-war days no less clearly and consistently express other feelings and intentions of their authors: the desire to accurately document and fix the image of the city and some of its structures in history, the happiness from the silence of peace, and the simple interest in the monuments of European art.The article examines Berlin scenes as evidences of the transition from front-line graphics focused on the visual recording of the war traces to peacetime graphics; from documentary — to artistry; from the worldview of a person at war — to the one of a person who lived to victory. In this approach, it has been important to consider the graphic images of Berlin in unity with the diary and memoir texts belonging to both artists and ordinary soldiers who participated in the storming of Berlin. The combination of verbal and visual sources helps to present the German capital’s image that existed in the public consciousness, as well as the specificity of its representation by means of visual art.

Охарактеризованы комбинации белков, связанных с изменением иммунных процессов, нарушением реологии крови, в том числе риском коагулопатии, противоопухолевых защитных механизмов, инсулинового сигнального пути, с изменением характеристик клеточного деления и качества новообразованной ткани. Таким образом, возрастная динамика основных процессов запускает каскад реакций, проявляющихся в замыкании «патологических биохимических кругов», которые формируют предпосылки к развитию заболеваний и, с течением времени, клинические проявления. For the first time proteins are described, reliably increasing and decreasing in urine with age in the range of 20 to 60 years. The combinations of proteins associated with changes in immune processes, violation of blood reology, including the risk of coagulopathy, anticancer defense mechanisms, insulin signaling pathway, changes in cell characteristics are characterized division and quality of the newly formed fabric. Thus, the age dynamics of the main processes triggers a cascade of reactions manifested in the closure of «pathological biochemical circles» that form the prerequisites for the development of diseases and, over time, clinical manifestations.

Abstract. The hyperarid core of the Atacama Desert, Chile, is possibly the driest and most life-limited place on Earth, yet endolithic microorganisms thrive inside halite pinnacles that are part of ancient salt flats. The existence of this microbial community in an environment that excludes any other life forms suggests biological adaptation to high salinity and desiccation stress, and indicates an alternative source of water for life other than rainfall, fog or dew. Here, we show that halite endoliths obtain liquid water through spontaneous capillary condensation at relative humidity (RH) much lower than the deliquescence RH of NaCl. We describe how this condensation could occur inside nano-pores smaller than 100 nm, in a newly characterized halite phase that is intimately associated with the endolithic aggregates. This nano-porous phase helps retain liquid water for long periods of time by preventing its evaporation even in conditions of utmost dryness. Our results explain how life has colonized and adapted to one of the most extreme environments on our planet, expanding the water activity envelope for life on Earth, and broadening the spectrum of possible habitats for life beyond our planet.

Abstract Background Despite the high disease burden of RSV in older adults and children, there is currently no approved vaccine. Ad26.RSV.preF, an experimental RSV vaccine, has demonstrated immunogenicity and tolerability in first-in-human clinical studies. The aim of this study was to assess the potential of the Ad26.RSV.preF vaccine to protect against RSV infection and disease in an established RSV human challenge model, used for the first time to evaluate a vaccine. Methods We conducted a randomized, double-blind, placebo-controlled, human challenge study (NCT03334695). Healthy adults received 1 × 1011 vp Ad26.RSV.preF vaccine (active) or placebo (pbo) intramuscularly. After 28 days, volunteers were challenged intranasally with a low-passage clinical strain of RSV-A (0.8 mL of Memphis 37b) and then quarantined for 12 days. Nasal washes were collected twice daily throughout quarantine, starting 2 days post-challenge (viral load [VL] by qRT-PCR and quantitative cultures). Disease severity was recorded thrice daily using symptom diary cards. Results Fifty-three volunteers (active, n = 27; pbo, n = 26) were challenged with RSV-A. Quantitative viral assessments were consistently lower in active than pbo. The primary endpoint of the study was met: the area under the curve (AUC) for RSV VL over time (via qRT-PCR) was significantly lower in active pbo (P = 0.012). Median peak VL was lower for active (0 log10 copies/mL) than pbo (5.4 log10 copies/mL). Median AUC for RSV VL over time (quantitative culture) was lower for active than pbo (0 vs. 109, P = 0.002). Disease severity was lower for active than pbo, with a median AUC total symptom score of 35 (active) vs. 167 (pbo) (P = 0.002). Overall, RSV infection (defined by qRT-PCR alone or combined with symptoms) and disease severity over time were lower in active vs. pbo. Conclusion RSV infections, VL, and RSV disease severity were consistently lower in healthy adults receiving Ad26.RSV.preF vs. placebo, demonstrating promising protection from RSV infection and disease. This was the first time that antiviral prevention was observed against RSV after active immunization. Ad26.RSV.preF warrants further evaluation in field trials for efficacy against natural RSV infections in populations considered at risk of severe RSV disease. Abstract Backgroud Streptococcus agalactiae is a common colonizer of the rectovaginal tract and lead to infectious diseases of neonatal and non-pregnant adults, which also causes infectious disease in fish and a zoonotic risk as well. Lysine crotonylation (Kcr) is a kind of histone post-translational modifications discovered in 2011. In yeast and mammals, Kcr function as potential enhancers and promote gene expression. However, lysine crotonylation in S. agalactiae has not been studied yet. Methods In this study, the crotonylation profiling of fish pathogen, S. agalactiae was investigated by combining affinity enrichment with LC MS/MS. The Kcr modification of several selected proteins were further validated by Western blotting. Results In the present study, we conducted the proteome-wide profiling of Kcr in S. agalactiae and identified 241 Kcr sites from 675 screened proteins for the first time. Bioinformatics analysis showed that 164 sequences were matched to a total of six definitively conserved motifs, and many of them were significantly enriched in metabolic processes, cellular process, and single-organism processes. Moreover, four crotonylation modified proteins were predicted as virulence factors or to being part of the quorum sensing system PTMs on bacteria. Conclusions These data provide a promising starting point for further functional research of crotonylation in bacterial virulence in S. agalactiae.
