Abstract Effect of cobalt manganese ferrite nanoparticles (M-NPs) (Co0.5Mn0.5Fe2O4) on vanadium hazards was assessment in the present study. Four groups of adult male albino rats [control group and three variably treated groups with ammonium metavanadate accompanied with or without cobalt M-NPs] were studied. The oral administration of ammonium metavanadate (Am.V) (20 mg/kg b.wt.) demonstrated the facility of vanadium to distribute and accumulate in the distinctive body organs and ordered as kidney > liver > lung > brain > spleen. Also, Am.V administration induce a significant disturbance in many physiological parameters (RBS, cholesterol, triglyceride, aspartate transaminase, alanine transaminase, Alb., bilirubin, Alk.Ph., urea, creat., Hb%, red blood cell count and packed cell volume) which might be expected to the liberation of free radicals according to the vanadium intoxication or its ability to disturb many body metabolisms. On the other hand, the intraperitoneal administration of 5% M-NPs in parallel with Am.V orally administration showed the ability of M-NPs to reduce Am.V dangerous impacts, which might be resulted from the essentiality of M-NPs metals to the body metabolism and to its free radicals scavenging properties. So, M-NPs could reduce Am.V hazardous effects.

The study was designed to prove the hypothesis that lipopolysaccharide (LPS)-induced fever elicits the changes in surfactant specific proteins, potentially related to thermal tachypnea. In adult rats fever was induced by intraperitoneal administration of LPS at a dose 100 µg/kg of body weight; control group received saline. Respiratory parameters, arterial blood gases and pH and colonic body temperature (BT) were recorded. Five hours later, surfactant proteins (SP) A, B, C and D were evaluated in bronchoalveolar lavage fluid (BALF) and lung tissue (LT). LPS evoked monophasic thermic response (at 300 min 38.7±0.2 vs. 36.4±0.3 °C, P0.05) and an increase in minute ventilation due to changes in breathing rate and tidal volume. LPS-instilled animals had higher levels of SP-A and SP-D in LT (P0.05 and 0.01), and higher SP-D in BALF (P0.01) than controls. SP-B increased in LT and SP-C in BALF of animals with LPS (both P0.05 vs. controls). The changes in all surfactant specific proteins are present in LPS-induced fever. Alterations of proteins related to local immune mechanisms (SP-A, SP-D) are probably a part of general inflammatory response to pyrogen. Changes in proteins related to surface activity (SP-B and SP-C) might reflect the effort of the body to stabilize the lungs in thermal challenge.

Abstract Background Repairing urinary bladder (UB) defect by enterocystoplasty remains the gold standard surgical bladder reconstruction procedure to increase the capacity and compliance of dysfunctional bladders. However, many complications were recorded. Aim of the work This work aimed to compare the consequences of reconstruction of urinary bladder defect using untreated small intestinal submucosal (SIS) matrix versus seeded and unseeded decellularized SIS matrix. Material and Methods Fifty female albino rats were used in this study. The animals were divided into three groups: Group I (Control) included ten adult rats from which ileal tissue was obtained. Group II included ten adult rats in which their UB defect was repaired by untreated cellular SIS. Group III included twenty adult rats that were subdivided into two subgroups, 10 rats each; Subgroup IIIA where rats had their UB defect repaired by acellular SIS and subgroup IIIb where rats had their UB defect repaired by acellular SIS seeded with adipose mesenchymal stem cells (AMSCs).Ten young rats were used for preparation of AMSCs. Morphometric and statistical analysis were also performed. Results In rats where UB defect was repaired by untreated cellular SIS, the graft area showed loss of epithelial polarity, presence of intraepithelial cysts and occasional extension of urothelium to the outer surface forming fistula. There were areas of metaplasia with the appearance PAS positive cells. In the lamina propria, there was areas of lymphocytic infiltration together with significant increase in the collagen fiber deposition (p < 0.05). There was a significant decrease thickness of muscle layer as compared to control (p < 0.05). In rats where UB defect was repaired by acellular SIS, urothelium in the graft area showed occasional squamous metaplasia and often the urothelium extended to the deeper layers forming Brunn's nest. There was minimal muscle regeneration in the graft area. However, in rats where UB defect was repaired by acellular SIS seeded with AMSCs, the urothelium in the graft area was nearly similar to control group with uniform urothelium thickness, minimal collagen fibers deposition and thick muscle layer that showed no significant difference from the control (p > 0.05). Conclusion Acellular SIS seeded with AMSCs showed better results compared to non-seeded and cellular SIS in reconstructing urinary bladder defects.

Background Hypothyroidism is a metabolic disorder affecting the functions of many tissues in the body including the testis. Testis is rich in the polyunsaturated fatty acids content and lacks strong intrinsic antioxidant system making it prone to such oxidative stress. L-carnitine (LC) regulates long chain fatty acids metabolism; and is considered a valuable antioxidant factor. Aim It was to evaluate the effect of hypothyroidism induced by propylthiouracil (PTU) on rats’ testes and the possible protective role of LC. Methods Forty-eight adult male albino rats were used in this work. The animals were divided into three groups with sixteen animals in each. Group 1 (Control): Animals were kept without medications. Group 2 (PTU-treated): was subjected to administration of PTU; while group 3 (PTU and LC) received both PTU and LC. By the end of the experiment “30 days”, blood samples were taken for hormonal assay; then animals were anaesthetized and sacrificed. Specimens were homogenized for biochemical analysis; epididymal content of each rat was obtained immediately for semen analysis. Testes’ specimens were harvested, prepared and examined by light microscope examination. Results Induced hypothyroidism was noticed to cause histopathological, morphometric and biochemical changes in rat’s testes. LC protected the testicular specimens against such changes; it also improved the seminal quality and quantity as well as testicular structure and biochemistry. Conclusion Hypothyroidism could result in hazards to the structure of testis. Fortunately co-administration of LC might reduce such hazards.

Background:Use of dietary supplements to treat illnesses has increasedtremendously in recentyears.Adrenal gland is one ofthemost commonly damaged endocrine gland in the body, not only by chemical or radiation injuries, but also as a result of differenttypes of stress.Search is underway for use ofnatural foods for protection of adrenal gland from different types ofinsults.Objective: To determine the protective effects of L-arginine on streptozotocin (STZ)-induced adrenal gland injury in albino rats,andto compare its efficacy to insulin.Material and Methods: This prospective experimental study was done at BMSI, JPMC, Karachi. Forty male, healthy albino rats,90-120 days old were segregated into 4 groups. Group A was marked as control, group B was administered STZ, group C and Dwere treated with STZ along with insulin and L-arginine respectively. At the end of study period, i.e., 6 weeks, animals weresacrificed under ether anaesthesia. Tissue from the left adrenal gland was processed for frozen sectioning to observe fat content ofthe adrenal cortex by applying OilRed O stain.Results: Oil Red-0 stained frozen sections revealed closely aggregated fat globules in adrenal cortex of STZ treated group B ascompared to control. Moderate betterment was seen in group C and in group D Oil Red O stained frozen sections as compared toSTZ treated group B.Conclusion: The results ofthe study demonstrated adrenal cortex injury by STZ which ameliorated with concomitant use of insulinandL-arginine. Background: Ovulation induction with clomiphene citrate in women with infertility has been practiced more than 40% years but in infertile patients this treatment plan proved to be ineffective with multiple complication. Body weight plays an important role modulating reproductive development and functioning. Aim: To observe the effects on body weight of female albino rat after use of clomiphene citrate and letrozole for consecutive 1-4 estrous cycles Method: Eighty four adult female Albino rats were equally divided into three groups for this research. Body weight of each rat was measured before and after the experiment. Vaginal smear cytology of each rat was performed to study different phases of estrous cycle. Control group A was given normal saline orally , In Experimental group B rats were given letrozole (Femara) at dose 5mg/kg orally and in Experimental group C rats were given clomiphene citrate at dose 100ug/kg orally. Results: Significant weight gain is observed in rats taking clomiphene citrate as compared to letrozole Conclusion : Comiphene citrate directly affects the body weight which indirectly reduces the ovulation induction and pregnancy rate. Letrozole is good alternate for ovulation induction and for CC resistant patients. Keywords: Estrous cycle, body weight, citrate and letrozole

To investigate the antihyperglycemic and antihyperlipidemic effect of Momordica charantia (Karala), the aqueous extract of the Karala fruit was tested on streptozotocin (STZ)-induced diabetic rats. Thirty six albino rats were used in the experiment, 30 diabetic and the remaining six as negative control (T1). Diabetes was induced by administering (injecting) STZ at dose of 55mg/kg body weight. Thirty diabetic animals were randomly divided into five groups such as diabetic control group (T2) without any application of treatment, and groups T3,T4,T5 and T6 were treated with aqueous extract of Karala fruits daily at the doses of 250,    500 and 750mg/kg and glibenclamide (at a dose of 5mg/kg body weight) respectively. The body weight was taken and blood samples were collected from individual animal to determine glucose levels at 15 day interval up to 90 days. In addition, Asparate  Transaminenase(AST), Alanine Transaminenase(ALT), Alkaline Phosphatase(ALP), Total cholesterol (TCh) and Triglyceride (TGA) were determined at day 15 and at the end of the experiment. All three doses of Karala extracts reduced diabetic induced blood sugar and the reduction is comparable with standard glibenclamide (GLM) dose particularly with higher doses Karala extracts (500 and 750mg). Karala also prevented body weight loss due to induced diabetes as did by GLM treatment.. The treatment also resulted in a significant reduction of Asparate Transaminenase(AST), Alanine Transaminenase(ALT), Alkaline Phosphatase(ALP), Total cholesterol (TCh) and Triglyceride (TGA) activities of treated rats when compared to the STZ induced  diabetic rats. Higher doses of Karala (500 and 750mg/kg) are as effective as standard GLM dose on measured variables. This study demonstrated that Karala has hyperglycemia and antihyperlipidemic effect against STZ induced diabetic rats. These findings open the possibility of using Karala extract to treat diabetic animal and human patients although further research is warranted. DOI: http://dx.doi.org/10.3329/jesnr.v5i1.11550 J. In this study, African Catfish (Clarias gariepinus) were cultured in water contaminated with phthalate, benzene and cyclohexane (10 μg/mL), respectively, over a period of 65 days. They were, thereafter, used as protein source to formulate feed for albino rats (Wistar strain) for 28 days. The weights of the body and selected tissues of the rats were monitored and standard enzyme assays were conducted for some enzymes in the serum. Results showed that rats in cyclohexane group gained 58% whereas the control group gained 46% of body weight. Liver weight (absolute and relative) of test rats were found to be significantly lower than that of control (p < .05). Enzyme activity of serum of test rats was found to be significantly higher than that of control (p < .05). Experimental evidence suggests leakage of enzymes from the liver to the serum, thus the elevated serum enzyme activity and that the contaminants may damage the liver.

Objective: The objective of this study was to evaluate the protective effect of Erdosteine on complete freund’s adjuvant (CFA) induced arthritic rats. Methods: Wistar Albino rats of 100–250 g were divided into five groups (n=6) and administered with 0.1 ml of CFA subcutaneously into the left hind paw except the negative control group. The standard group received methotrexate (MTX) 0.075 mg/kg body weight orally. Besides, the test groups received Erdosteine orally at a dose 10 mg/kg and 20 mg/kg bodyweight for 12 days. The changes in body weight, paw volume, hematological parameters, radiographical, and histological findings were the indicators to evaluate the efficacy of the test product. Discussion: Significant change in the body weight, paw volume, radiographical, hematological, and histological parameters were observed which supports the remarkable reduction of the arthritic development in the standard and test groups compared to the untreated group. However, the test group (Erdosteine) with the dose 20 mg/kg shows to be more potent than the test group (Erdosteine) with a dose 10 mg/kg and the standard group (MTX) to reduce the arthritic effect. Results: The test group with 20 mg/kg Erdosteine showed much better outcome than the standard group at significant (p<0.05). Therefore, Erdosteine acting as an anti-inflammatory and anti-oxidant is effective at a dose 20 mg/kg in treating the progression of rheumatoid arthritis in rats.

Abstrak Latihan fisik yang berlebihan dapat meningkatkan produksi radikal bebas di dalam tubuh. Peningkatan radikalbebas ini kalau tidak dinetralisasi oleh antioksidan di dalam tubuh, maka terjadilah stres oksidatif yang dapatmenyebabkan terjadinya peroksidasi lipid, sehingga menghasilkan produk senyawa F2-isoprostan yang dapatberdampak terhadap berbagai masalah kesehatan. Tujuan penelitian ini adalah untuk menentukan pengaruhpemberian susu coklat terhadap kadar F2-isoprostan. Penelitian ini merupakan eksperimental pre and post test controlgrup design terhadap 36 siswa atlet, yang memenuhi kriteria inklusi dan ekslusi. Subjek dibagi dua kelompok yaituyang diberikan perlakuan susu coklat dan kontrol susu putih. Susu diminum setelah latihan fisik rutin atlet sebanyaksatu kali sehari selama 15 hari. Kadar F2-isoprostan dinilai sebelum dan sesudah secara ELISA. Data dianalisisdengan paired sample t-Test, bermakna bila p<0,05. Hasil penelitian didapatkan rerata kadar F2-isoprostan sebelumperlakuan pada kelompok susu coklat adalah 65,98±14,61 pg/ml dan sesudah perlakuan adalah 45,55±13,74 pg/ml.Rerata kadar F2-isoprostan sebelum perlakuan pada kelompok kontrol susu putih adalah 54,24±20,44 pg/ml dansesudah perlakuan adalah 48,80±20,23 pg/ml. Kesimpulan yang diperoleh ada pengaruh yang bermakna dari reratakadar F2-isoprostan sebelum dan sesudah perlakuan baik pada kelompok perlakuan susu coklat dan kelompok kontrolsusu putih.Kata kunci: latihan fisik, radikal bebas, F2-isoprostan, susu coklatAbstract Excessive physical exercise can increase the production of free radicals in the body. The increase in freeradicals if not neutralized by antioxidants in the body and it caused oxidative stress which can cause lipid peroxidation,resulting in F2-isoprostane product compounds that can affect a variety of health problems. The objective of this studywas to determine the influence of milk chocolate on F2-isoprostane levels. This study was an experimental pre andpost test control group design to the 36 student athletes, who meet the inclusion and exclusion criteria. The subjectswere divided in two groups: the treatment given milk chocolate and white milk is given control, drunk after a routinephysical exercise athlete, once a day for 15 days. F2-isoprostane levels were assessed before (pre) and after (post) byELISA, the data were analyzed by Paired Sample t-Test, significant if p <0.05. The results showed average levels ofF2-isoprostane before treatment group was 65.98 ± chocolate milk 14.61 pg / ml and after treatment was 45.55 ±13.74 pg / ml. Average F2-isoprostane levels before treatment in the control group was 54.24 ± white milk 20.44 pg /ml and after treatment was 48.80 ± 20.23 pg / ml. There is a significant influence on the mean levels of F2-isoprostaneboth before and after treatment in the treatment group and the control group of milk chocolate and white milkKeywords: physical exercise, free radicals, f2-isoprostane, milk chocolate

Liver is a vital organ present in vertebrates, which has a wide range of functions including aiding of digestion, detoxification and protein biosynthesis. But the ability of the liver to perform these functions can be compromised by numerous substances it is daily exposed to, including certain medicinal agents which when taken in over doses. Liver damage, just like many other diseased conditions can lead to oxidative stress, especially when the body’s antioxidant system is overwhelmed by the free radicals thus generated. A major component of this antioxidant system are the natural antioxidant enzymes superoxide dismutase and catalase manufactured in the body, which provide an important defense against free radicals usually generated in diseased conditions. Most synthetic anti-hepatotoxicity drugs available present serious side effects and are generally out of reach of the common man. Consequently, the effect of administration of aqueous extract of Anacardium occidentale stem bark on the activities of superoxide dismutase and catalase in some tissues of acetaminophen-induced hepatotoxic rats was investigated. There was a significant (p < 0.05) reduction in the activities of superoxide dismutase and catalase in the serum, liver, kidney and heart of the hepatotoxic rats. However, treatment of hepatotoxic rats with aqueous extract of Anacardium occidentale stem bark led to a significant (p < 0.05) increase in the activities of superoxide dismutase and catalase in the serum, liver, kidney and heart of acetaminophen-induced hepatotoxic rats.
