Growth and hepatic composition in the guinea pig after long-term parenteral hyperalimentation

This study examined the feasibility of maintaining male Hartley guinea pigs on long-term hyperalimentation. Data from animals fed ad libitum, but infused with 0.9% saline, indicated that there was minimal effect from catheter implantation, sepsis, or the infusion of large volumes of fluid. This group compared with animals fed and watered ad libitum demonstrated a nearly identical growth rate (6.33 +/- 1.18 and 6.12 +/- 1.32 g/day, respectively), serum chemistry, tissue weights, and hepatic composition and morphology. Animals infused with a total parenteral diet demonstrated growth rates of 4.06 +/- 1.46 g/day for up to 25 days. Loss of infused animals was due in varying degrees to sepsis, mechanical failure, improper placement of the cannula, loss of patency, and death from unknown causes. Morphological analysis of animals fed by total parenteral nutrition revealed an altered distribution and increased size of lipid droplets in hepatic parenchymal and Kupffer cells and glycogen accumulation by the parenchymal cells. Decreased hepatic content of total protein and lipid, as well as cytochrome P450, was also observed. Similarly, serum values of triglyceride were decreased in animals fed by the total parenteral diet. This study indicated that the guinea pig fed by hyperalimentation may be a useful animal model for a number of clinical and basic research applications.

Lipid droplets (LDs) are essential organelle in most eukaryotes, and tracking intracellular LDs dynamics using synthetic small molecules is crucial for biological studies. However, only a limited number of fluorescent markers that satisfy all requirements, such as the selective staining of LDs, high photostability, and sufficient biocompatibility, have been developed. Herein, we report a series of donor-p-acceptor dyes based on the thiophene-containing fused polycyclic scaffold [1]benzothieno[3,2-<i>b</i>][1]benzothiophene (BTBT), in which either or both thiophene rings are oxidized into thiophene-<i>S</i>,<i>S</i>-dioxide to form an electron-accepting building block. Among these dyes, LAQ1 satisfied all the aforementioned requirements, and allowed us capturing ultra-small LDs on the endoplasmic reticulum (ER) membrane by stimulation emission depletion (STED) microscopy with a super-resolution below the diffraction limit of light. Moreover, the extremely high photostability of LAQ1 enabled recording the lipolysis of LDs and the concomitant lipogenesis as well as long-term trajectory analysis of micro LDs at the single particle level in living cells.

Complete androgen insensitivity syndrome (CAIS) is due to complete resistance to the action of androgens, determining a female phenotype in persons with a 46,XY karyotype and functioning testes. CAIS is caused by inactivating mutations in the androgen receptor gene (AR). It is organized in eight exons located on the X chromosome. Hundreds of genetic variants in the AR gene have been reported in CAIS. They are distributed throughout the gene with a preponderance located in the ligand-binding domain. CAIS mainly presents as primary amenorrhea in an adolescent female or as a bilateral inguinal/labial hernia containing testes in prepubertal children. Some issues regarding the management of females with CAIS remain poorly standardized (such as the follow-up of intact testes, the timing of gonadal removal and optimal hormone replacement therapy). Basic research will lead to the consideration of new issues to improve long-term well-being (such as bone health, immune and metabolic aspects and cardiovascular risk). An expert multidisciplinary approach is mandatory to increase the long-term quality of life of women with CAIS.

A modification of the Novikoff-Goldfischer alkaline 3,3'-diaminobenzidine medium for visualizing peroxisomes is described. It makes possible light microscopic as well as electron microscopic studies of a recently described class of peroxisomes, the microperoxisomes. Potassium cyanide (5 x 10–3 M) is included in the medium to inhibit mitochondrial staining, the pH is 9.7 and there is a high concentration of H2O2 (0.05%). Two cell types have been chosen to illustrate the advantages of the new procedure for demonstrating the microperoxisomes: the absorptive cells in the human jejunum and the distal tubule cells in the guinea pig kidney. Suggestive relations of microperoxisomes and lipid are described in the human jejunum. The microperoxisomes are strategically located between smooth endoplasmic reticulum that radiates toward the organelles and contains lipid droplets and "central domains" of highly specialized endoplasmic reticulum which do not show the lipid droplets. The microperoxisomes are also present at the periphery of large lipid-like drops. In the guinea pig kidney tubule there is a striking difference between the thick limb of Henle and distal tubule. The distal tubule has a population of cells with large numbers of microperoxisomes readily visible by light microscopy; these cells are not present in the thick limb of Henle. Other differences between the two are also described.

The Section of Urology of the American Academy of Pediatrics met for three days in conjunction with the 53rd annual meeting of the Academy in Chicago. Papers and panel discussions that would be of interest to the pediatrician are summarized herein according to topic. As there has been an increasing number of basic research papers presented in recent years, the Section, this year, initiated a prize for basic research. Also new to the meeting was the awarding of the first annual Pediatric Urology Medal, which was given to Harry Spence for contributions made to the field. TESTICULAR TORSION Recent reports of unilateral testicular torsion have suggested that adverse effects may occur to the contralateral testicle, if the ischemic testicle is treated by surgical detorsion and left in place rather than removed.1-4 Three authors addressed this problem by investigating animal models. Animal Models In prepubertal rats undergoing experimentally produced testicular torsion, Rabinowitz and associates found histologic changes in the contralateral testicle. The changes in the contralateral testicle were dependent upon the duration of torsion and were significantly greater when the ischemic testicle was detorsed and left in place. The degree of histologic changes in the ischemic testicle, especially the degree of germ cell damage, was the best prediction of long-term damage to the contralateral testicle. In the animal model, these changes could be averted by removing the ischemic testicle. Although not going as far to recommend orchiectomy as the treatment of choice for all cases of torsion, Dr. Rabinowitz did suggest that a testicular biopsy in prepubertal boys with testicular torsion may be a reliable index to subsequent long-term contralateral damage if the ischemic testicle is detorsed and pexed rather than removed.

Recently, pollen grains (PGs) have been introduced as drug carriers and scaffolding building blocks. This study aimed to assess the in-vitro biocompatibility of Pistacia vera L. hollow PGs/Fe3O4 nanoparticles (HPGs/Fe3O4NPs) composites using human adipose-derived mesenchymal stem cells (hAD-MSCs). In this regard, iron oxide nanoparticles (Fe3O4NPs) were assembled on the surface of HPGs at different concentrations. The biocompatibility of the prepared composites was assessed through MTT assay, apoptosis-related gene expression and field emission scanning electron microscopy (FE-SEM) analysis. Compared to the bare HPGs, the HPGs/Fe3O4NPs exhibited a biphasic impact on hAD-MSCs. The composite containing 1% Fe3O4NPs demonstrated no cytotoxicity up to 21 days while higher Fe3O4NPs contents and long-term exposure revealed adverse effects on the hAD-MSCs’ growth. The obtained result was verified by the qRT-PCR and morphological analysis carried out through FE-SEM which suggests that a narrow region below 1% Fe3O4NPs may be the optimum choice for medicinal applications of HPGs/Fe3O4NPs microdevices.

Severe caloric restriction (sCR) increases the risk for acute cardiovascular disease. Less understood are the long-term effects on cardiovascular disease risk after the sCR period has ended. We investigated the effects of sCR on heart structure and function months after refeeding (sCR-Refed). Female Fischer rats (3-months-old) were maintained on (CT) ad libitum or a 60% caloric restricted diet for 2 weeks. Thereafter, all rats received ad libitum chow for 3 months and they were analyzed by precision ultrasound to assess their heart function. After imaging, the animals were sacrificed and the hearts were subjected to ischemia-reperfusion (I/R) using a Langendorff preparation. After 2 weeks of sCR, rats lost 15% of their initial body weight (BW) [% (100*(Final-Initial/Initial)): CT, 1.5±0.8 vs sCR, -15.4±1.1; p<0.001;n=8]. After 3 months of refeeding, there was no detectable difference in BW between CT and sFR-Refed groups. Isolated hearts from the sCR-Refed rats exhibited worse myocardial pathology after I/R compared to CT rats. The parallel orientation of myofibers and striations normally present in cardiomyocytes was lost in sCR-Refed rats. Further analysis revealed uneven blood-filling of the microcirculatory vessels and prominent interstitial edema of the myocardium. Hearts from sCR-Refed rats had more atrophied cardiomyocytes than CT [Atrophied/Total (%): CT, 0.2±0.1 vs sCR-Refed, 50.6±1.1; p<0.001; n=5]. The number of arrhythmic events during a 30 min ischemic interval in isolated hearts doubled after 2 weeks on the sCR diet ( data not shown ) and remained doubled 3 months later [Arrhythmias (% of time): CT, 34±8 vs sCR-Refed, 68±9; p=0.02; n=8]. Ultrasound imaging showed no difference in stroke volume, coronary perfusion pressure and left ventricular mass. However, the thickness of the left ventricular posterior wall was significantly reduced in sCR-Refed rats [(mm): CT, 2.55 ±0.03 vs sCR-Refed, 2.10±0.04; p=0.002; n=4]. These findings indicate heart structure and function remained damaged months after the sCR period ended and BW was restored. These studies have adverse cardiovascular risk implications for who are subjected either voluntarily (crash diets) or involuntarily (very low food security) to periods of inadequate caloric intake.

AbstractWe report on how a long-term study of the reproductive biology of the Critically Endangered Schiedea adamantis (Caryophyllaceae), one of Hawai‘i's rarest plant species, was leveraged for conservation purposes. Our major goals were to provide seeds with the greatest genetic variation possible for reintroduction and to ensure that both female and hermaphroditic plants of this wind-pollinated species were reintroduced in a manner that maximized both outcrossing and seed production. Schiedea adamantis was one of the first Hawaiian plant species listed under the Endangered Species Act (USA). The species has been studied intensively to test hypotheses addressing the evolution of breeding systems. Information on outcrossing levels and the extent of inbreeding depression was integrated into ongoing reintroduction efforts. Population size peaked in 1994, when 267 flowering individuals were found on Lē‘ahi (Diamond Head Crater). By 2016 only 17 flowering individuals were present, with drought and invasive species being possible causes of this decline. Reintroduction attempts in 1998 using genetically diverse seeds were unsuccessful because of drought and a lack of sufficient supplemental irrigation. Additional reintroduction attempts in 2012 and 2014 were more successful because of increased supplemental irrigation. Plants used in reintroductions represent genotypes long since absent in the natural population, and may contain the genetic variability essential for evolutionary responses to climate change and the spread of invasive species. The destruction of many plants reintroduced in 2015 and 2016 by a fire in March 2016 highlights the need for additional restoration areas at Lē‘ahi and elsewhere, and storage of seeds for future use.
