Objective. Selenium, as an antioxidant, has been implicated in the development of autoimmune thyroiditis (AIT). Many studies showed selenium supplementation could decrease thyroid autoantibodies in patients with AIT. However, the underlying mechanisms have not been well determined. Therefore, we performed a clinical study to investigate the possible mechanism of beneficial effects of selenium treatment on AIT patients. Methods. Forty euthyroid patients with AIT were randomized into two groups. Group I was treated with 200 μg/day selenium supplementation, and group II received a placebo over a 3-month period. Thyroid stimulating hormone (TSH), thyroid peroxidase antibody (TPOAb), antithyroglobulin antibody (TgAb), malondialdehyde (MDA), total antioxidant capacity (TAC), and superoxide dismutase (SOD) were measured before and 3 months after treatments. Additionally, twenty healthy volunteers also served as a control group for the evaluation of such parameters in basic condition. Results. Totally, 32 patients (group I, n = 18; group II, n = 14) completed the clinical study and were incorporated into the statistics. MDA level was higher and SOD activity and TAC were lower in patients compared to healthy individuals. After 3 months, TPOAb titer significantly decreased within group I (P<0.001) but did not change within group II (P=0.001). There were also no statistically significant changes in TSH and TgAb titers within the two groups (all P>0.05). Additionally, decreased MDA level (from 6.8 ± 1.3 nmol/ml to 4.9 ± 0.7 nmol/ml; P<0.001) and increased TAC (from 10.0 ± 1.9 mmol/l to 12.9 ± 3.1 mmol/l; P=0.003) and SOD activity (from 72.3 ± 10.3 U/ml to 84.3 ± 13.2 U/ml; P=0.007) were simultaneously observed after 3 months’ selenium treatment. Moreover, there was a negative correlation between TAC and TgAb/TPOAb and a positive correlation between MDA and TgAb/TPOAb in AIT patients. Conclusions. Our findings support the hypothesis that selenium treatment could decrease TPOAb titer via enforcing the defense against oxidative stress in euthyroid patients with AIT, which may be a potential underlying mechanism.

Background: Incoming anaesthesia created by the use of many drugs with different physicochemical properties is a source of stress and trauma for the body. This event increases the oxidative response and changes the balance between oxidant/antioxidant capacity in the organism in favor of oxidant capacity. This situation is defined as oxidative stress. For these reasons, studies are conducted to determine the effects of general anaesthetic agents on oxidant and antioxidant systems in the organism. In this study, it was aimed to determine the effects of isoflurane and sevoflurane used for general anaesthesia in humans and animals on lipid peroxidation and antioxidant defense system in calves.Materials, Methods & Results: The study included 14 calves of different breeds, ages, sexes, and weighing, average 2 weeks old. The cases randomly were divided into 2 groups, the isoflurane group (group I), and the sevoflurane group (group II), and each group included 7 animals. Before general anaesthesia, 0.04 mg/kg atropine was administered intramuscularly to all animals for premedication. At 15 min after atropine administration, isoflurane was administered at an inspiratory concentration of 3-5% in group I, and sevoflurane was administered at an inspiratory concentration of 5-7% in group II, via a face mask for 15 min for the induction of anaesthesia. Endotracheal intubation was performed in all cases at the 15min of the induction period following the onset of general anaesthesia symptoms. After the induction, anaesthesia was continued at an inspiratory concentration of 1.5-3% in the isoflurane group and inspiratory concentration of 2.5-4% in the sevoflurane group. Blood samples were taken just before anaesthesia, just before skin incision, at the end of anaesthesia and surgery, and at the 24h postoperatively. The malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), and glutathione (GSH) levels were measured spectrophotometrically in samples. In group I, MDA and antioxidant parameters SOD, CAT, GSH-Px, GSH did not observe a significant change in their concentrations through the study (P > 0.05). In group II, MDA value decreased significantly before incision (P < 0.05), at the end of anaesthesia and surgery compared to the pre-anaesthesia level (P < 0.001), and then although increased significantly at 24th-h postoperatively, the value was still lower than the pre-anaestesia level (P < 0.05). It was determined that SOD activity increased significantly after sevoflurane compared to pre-anaesthesia (P < 0.05) however, the increases in SOD activity detected during sevoflurane were not statistically significant (P > 0.05). During the study, a statistically insignificant increase was observed in the concentrations of CAT, GSH-Px, GSH compared to pre-anaesthesia (P > 0.05). Pre-anaesthesia values of all measured biochemical parameters did not differ significantly between groups (P > 0.05). Before skin incision, at the end of anaesthesia and surgery, and at the 24h postoperatively MDA was lower (P < 0.05) and SOD activity was higher (P < 0.05) than in group I in group II. There was no statistically significant difference between the two groups in terms of CAT, GSH-Px, GSH levels between the other measurement times (P > 0.05). Discussion: An important advantage of sevoflurane compared to currently available anaesthetics is that it provides rapid induction due to its low solubility in blood and tissues, and rapid recovery due to its low solubility in fat. This feature is proof that the side effects of sevoflurane anaesthesia are minimal. The findings of this study show that sevoflurane exposure decreases lipid peroxidation and enhances antioxidant defense. The potential effect of sevoflurane on oxidative stress may lead to its preferred clinical use of sevoflurane compared to isoflurane.

In adults, selenium supplementation decreases thyroid peroxidase antibody (TPO Ab) concentrations in patients with autoimmune thyroiditis (AIT). Our aim in this study was to investigate if selenium supplementation decreased TPO Ab and thyroglobulin antibody (Tg Ab) concentrations in children with AIT. Forty-nine patients (33 females) with newly diagnosed AIT and hypothyroidism were randomized to daily oral therapy with levothyroxine alone (group A, n = 18), levothyroxine plus 100 µg sodium-selenite (group B, n = 13), or levothyroxine plus 200 µg sodium-selenite (group C, n = 18). Mean age at diagnosis was 12.2 ± 2.2 years. All 49 patients needed a mean levothyroxine dose of 1.6 ± 0.5 µg/kg body weight to lower TSH to the treatment goal of 1–2 µU/ml, with no significant difference between groups. At study entry and after 12 months, TPO Ab concentrations were comparable in all three groups. Tg Ab concentrations decreased significantly after 12 months in group A and group C (p= 0.03 andp= 0.01), but not in group B (p= 0.06). It is our conclusion that selenium supplementation with sodium-selenite does not decrease TPO Ab concentrations in children and adolescents, neither given in the reduced dose of 100 µg daily nor given in the “adult” supplementation dose of 200 µg daily.

The aim of this study is to investigate the long-term (9 months) effects of variable doses (200/100 μg/day) of l-selenomethionine on autoimmune thyroiditis (AIT) and the parameters affecting the success rate of this therapy. The present study was designed in three steps: (1) 88 female patients with AIT (mean age = 40.1 ± 13.3 years) were randomized into two groups according to their initial serum TSH, thyroid peroxidase antibody (TPOAb) concentrations, and age. All the patients were receiving l-thyroxine to keep serum TSH ≤2 mIU/l. Group S2 (n = 48, mean TPOAb = 803.9 ± 483.8 IU/ml) received 200 μg l-selenomethionine per day, orally for 3 months, and group C (n = 40, mean TPOAb = 770.3 ± 406.2 IU/ml) received placebo. (2) 40 volunteers of group S2 were randomized into two age- and TPOAb-matched groups. Group S22 (n = 20) went on taking l-selenomethionine 200 μg/day, while others (group S21) lowered the dose to 100 μg/day. (3) 12 patients of group S22 (group S222) went on taking l-selenomethionine 200 μg/day, while 12 patients of group S21 (S212) increased the dose to 200 μg/day. Serum titers of TPOAb decreased significantly in group S2 (26.2%, P < 0.001), group S22 (23.7%, P < 0.01) and group S212 (30.3%, P < 0.01). There were no significant changes in group C and group S222 (P > 0.05). TPOAb titers increased significantly in group S21 (38.1%, P < 0.01). A significant decrease in thyroglobulin antibody titers was only noted in group S2 (5.2%, P < 0.01). l-selenomethionine substitution suppresses serum concentrations of TPOAb in patients with AIT, but suppression requires doses higher than 100 μg/day which is sufficient to maximize glutathione peroxidase activities. The suppression rate decreases with time.

The aim of this research was to investigate the potential effect of methanolic extract of Scurrula atropurpurea (MESA) in reducing oxidative stress through observation of superoxide dismutase (SOD) activity and malondialdehyde (MDA) concentration in mice. In this research, 25 male mice were divided into four groups with five replications. Group I (K1, without MESA) was considered as a control group, while Group II (K2), III (K3), and IV (K4) were given 1000, 2000, and 4000 mg MESA/kg BW, respectively. After 14 days, SOD activity and MDA concentration were measured spectrophotometrically. The SOD activity did not significantly increased (P>0.05) in group K1, K2, K3, and K4 with values were 19.469±2.072, 17.4±1.373, 20.53±3.411, and 23.87±2.528 U/ml, respectively. In contrast, MDA concentration was decreased (P>0.05) with values of 492.750±19.418, 492.750±19.418, 457.125±30.095, 454.625±19.080, and 475.875±37.659 ng/mL in groups K1, K2, K3, and K4, respectively. It can be concluded that the administration of  MESA tend to increase SOD activity and decrease MDA concentration in this study, thus might be able to reduce  oxidative stress in mice.

Background: Hind quarter weakness (HQW) is the loss of bilateral motor function of the rear limbs due to dysfunction of neural or muscular system. Animals are mostly presented with focal or generalized pain, varying degrees of paresis, paralysis and inability to urinate. Serum β- endorphin is an endogenous hormone released during stressful and painful events and functions almost exclusively in an inhibitory fashion. Moreover, electrical stimulation has been demonstrated to release endorphins at low frequencies. Methods: The present study was conducted to investigate the possible participation of endogenous opioid i.e. Serum β - endorphins and oxidative stress parameters in pain modulation and its variation in dogs suffering from hind quarter weakness subjected to ultrasound therapy and shortwave diathermy. Dogs were treated using therapeutic ultrasound (group I, n=6) and shortwave diathermy (group II, n=6) in combination with supportive drug therapy continued regularly for one week. Blood was collected on day 0, 3rd and 7th for oxidative stress estimation and regularly from the day of presentation till 7th day of treatment for serum β-endorphin estimation. Result: LPO values decreased significantly (P less than 0.05) from day 0 to 3 in group I and from day 0 to 7 in group II. SOD values showed a significant (P less than 0.05) decrease between day 0 and 7 in group II cases. A continuous progressive increasing trend in GSH was noticed which was significant (P less than 0.05) from day 3 to 7 in both groups. CAT values showed a significant (P less than 0.05) increase from day 0 to 7 in group II and non-significant (P less than 0.05) increase in group I. Serum β-endorphin values showed a significant (P less than 0.05) increase from day 4th to 6th in group I and between day 0, 1st, 3rd and 5th in group II. The variation in values throughout the treatment was suggestive of reduction in stress response and indicates improvement in dogs suffering from HQW.

BACKGROUND: Gamma glutamyltransferase (GGT) was reported recently to be associated with inflammation, oxidative stress and increased amino acid. However, role of GGT in insulin resistance pathomechanism is not exactly known. Therefore correlation of GGT with inflammation, oxidative stress and elevated amino acid, in men with central obesity need to be confirmed.METHODS: A cross-sectional study was designed. Men with central obesity were recruited and selected. Anthropometric parameters, creatinine, hs-CRP, fasting glucose, fasting insulin, glutathione peroxidase (GPx) activity, GGT, plasma total cysteine (tCys) and fatty liver were measured. Subjects were then divided in 4 groups based on waist circumference (WC) and fatty liver: Group I: WC ≤100 cm, without fatty liver; Group II: WC ≤100 cm, with fatty liver; Group III: WC >100 cm, without fatty liver; Group IV: WC >100 cm, with fatty liver. All biochemical characteristics in each group were then statistically analyzed.RESULTS: Seventy-two men with central obesity were selected. Numbers of subjects in each group were: Group I: n=33; Group II: n=5; Group III: n=17; Group IV: n=17. We found significant difference of HOMA-IR between Group I and IV, significant correlation between GGT and HOMAIR, and significant negative correlation between tCys with HOMA-IR in Group IV.CONCLUSION: GGT was significantly correlated with HOMA-IR in men with WC >100 cm and fatty liver. Further investigation with more subjects is necessary to determine clear GGT cut-off to distinguish subjects with fatty liver and insulin resistance.KEYWORDS: GGT, hs-CRP, GPx, tCys, HOMA-IR, insulin resistance

Environmental and occupational exposure to benzene from fuels is a major cause for concern for national and international authorities, as benzene is a known carcinogen in humans and there is no safe limit for exposure to carcinogens. The objective of this study was to evaluate the genotoxic effects of chronic occupational exposure to benzene among two groups of workers: filling station workers (Group I) and security guards working at vehicles entrances (Group II), both on the same busy highway in Rio de Janeiro, Brazil. Sociodemographic data on the workers were evaluated; the concentration of benzene/toluene (B/T) in atmospheric air and individual trans,trans-muconic acid (ttMA) and S-phenylmercapturic acid (S-PMA) were measured; oxidative stress was analyzed by catalase (CAT), glutathione S-transferase (GST), superoxide dismutase (SOD), thiol groups (THIOL) and malondialdehyde (MDA); genotoxicity was measured by metaphases with chromosomal abnormalities (MCA) and nuclear abnormalities, comet assay using the enzyme formamidopyrimidine DNA glycosylase (C-FPG), and methylation of repetitive element LINE-1, CDKN2B and KLF6 genes. Eighty-six workers participated: 51 from Group I and 35 from Group II. The B/T ratio was similar for both groups, but Group I had greater oscillation of benzene concentrations because of their work activities. No differences in ttMA and S-PMA, and no clinical changes were found between both groups, but linearity was observed between leukocyte count and ttMA; and 15% of workers had leukocyte counts less than 4.5 × 109 cells L−1, demanding close worker’s attention. No differences were observed between the two groups for THIOL, MDA, MCA, or nuclear abnormalities. A multiple linear relationship was obtained for the biomarkers MCA and C-FPG. A significant correlation was found between length of time in current job and the biomarkers C-FPG, MCA, GST, and MDA. Although both populations had chronic exposure to benzene, the filling station workers were exposed to higher concentrations of benzene during their work activities, indicating an increased risk of DNA damage.

Background: Disbudding is often practiced in modern farm because of the reasons for the safekeeping of the animal, other animals lessen the risk of injury and less aggressive behavior. Without regard to the method of disbudding leads to disruption behavioral changes, cardiac and endocrine responses related pain of animals. Sedation, local anesthesia and analgesia are performed studies in order to eliminate of cardiac, endocrine, behavioral response which is caused by pain associated with disbudding. In this study, it was aimed to determine the effect on the oxidant and antioxidant system in the calves of disbudding by using the caustic paste with and without painkiller.Materials, Methods & Results: The animal material of the study was created 24 Simmentals calves in different sexes on average 2 weeks (± 2 days) and 50 kg (± 15 kg) live weight. The cases were randomly divided into 2 groups of 12 calves in each group. In the group I (analgesia group); caustic paste+painkiller (2.2 mg-1 flunixin meglumine intravenous, IV) and group II (non-analgesia group); as disbudding applied with caustic pasta application only were divided into 2 groups. In the I group, 15th min before the application, flunixin meglumine at a dose of 2.2 mg-1 was administered IV and the horn blunting was performed by caustic paste method. Blood samples were taken at 15, 30 and 60th min after completion of horn blunting and physiological findings were recorded. In group II, the horns of the calves were disbudding with the same technique (but no analgesia aplication). Blood samples were taken at 15, 30 and 60th min after completion of horn blunting and physiological findings were record. Cortisol, glucose, total oxidant capacity (TOC) and total antioxidant capacity (TAC), 8-hydroxy-2’-deoxyguanosine (8-OHdG), glutathione (GSH), superoxide dismutase (SOD) enzymes measurements (ELISA) were performed in both groups. TOC and TAC tests were used to determine the oxidative stress index in the obtained serum, 8-OHdG measurement which is the most sensitive and most frequent oxidative DNA damage indicator for determination of oxidative damage; GSH, SOD enzymes were analyzed by ELISA using commercial kit for the evaluation of antioxidant potential. For evaluation of pain in animals, cortisol (ELISA) and glucose level in serum were measured with ELISA by using commercial kit. In the group without flunixin meglumine, cortisol was increased significantly from the 15th min (P < 0.05), but there was no increase in the 30 and 60th min in painkiller administered group. Glucose levels were higher in the group not using painkillers than those used at 15th (P < 0.001) and 30, 60th min (P < 0.05). TOC (P < 0.001) and 8-OHdG (P < 0.05) values were significantly higher in the group without painkiller while TAC, GSH and SOD levels (P < 0.05) were significantly decreased.Discussion: It has been observed that sedation (such as xylazine) and local anesthesia (such as lidocaine) use removed pain-induced stress in the disbudding with caustic paste in calves. However, in order to remove the endocrine response, it is necessary to combine the anesthesia protocol with the analgesic (such as flunixin meglumine) combination. According to our results concluded that the painkiller given before the disbudding process was supporting the antioxidant system, reducing the level of cortisol and oxidative stress.

Doxorubicin-induced cardiotoxicity is the leading cause of morbidity and mortality among cancer survivors. The present study was aimed to investigate the cardioprotective potential of methyl gallate; an active polyphenolic nutraceutical, against doxorubicin-induced cardiotoxicity in Wistar rats. Twenty-four female Wistar rats (150–200 g) were divided into four groups (n = 6) which consist of normal control (group I), doxorubicin control (group II), test-A (group III), and test-B (group IV). Group III and group IV animals were prophylactically treated with methyl gallate 150 mg/kg/day and 300 mg/kg/day orally, respectively, for seven days. Doxorubicin (25 mg/kg; single dose) was administered through an intraperitoneal route to group II, III, and IV animals on the seventh day to induce acute cardiotoxicity. On the 8th day, besides ECG analysis, serum CK, CK-MB, LDH, AST, MDA, and GSH were assayed. Following gross examination of isolated hearts, histopathological evaluation was performed by light microscopy. A significant ( p  < 0.05) cardiac injury, as well as oxidative stress, was observed in doxorubicin control rats in comparison to normal control rats. Methyl gallate at both the doses significantly ( p  < 0.05) reduced doxorubicin-induced ECG changes, dyslipidaemia, and elevation of CK, CK-MB, LDH, AST, MDA and increased GSH level. Methyl gallate reversed the doxorubicin-induced histopathological changes in the heart. The present study revealed that methyl gallate exerts cardioprotection against doxorubicin-induced cardiotoxicity in female Wistar rats by suppressing oxidative stress. Our study opens the perspective to clinical studies for consideration of methyl gallate as a potential chemoprotectant nutraceutical in the combination chemotherapy with doxorubicin to limit its cardiotoxicity.
