Objective. The aim of this study was to histologically examine the tissue reaction of three different calcium silicate cements in the closure of perforations in rat incisor teeth. Material and Methods. An experimental lateral root perforation with pulp exposure was performed in 32 lower incisors of 16 male Wistar albino rats. They were randomly assigned into three test groups (each including eight teeth) that were filled either by Biodentine (BD) or MicroMega mineral trioxide aggregate (MM-MTA) or EndoSequence root repair material putty (ESRRM putty), besides eight unperforated incisors from the other four rats (control group). The inflammatory response and healing process were evaluated histologically and scored after one and four weeks. Differences among groups were tested by Kruskal–Wallis tests at P≤0.05. Results. In the first week, BD produced more inflammatory response in the pulpal (score 3) than other materials (score 2). Only ESRRM putty showed odontoblast-like cells in 50%, 25% dentine-like deposit, 25% evidence of bone deposition in the drilling site (score 2), and minimum periodontal ligament (PDL) necrosis and disorganization (25%, score 2). After one month, all groups had healthy pulpal tissue, but 25% of ESRRM putty retained score 1 inflammatory response, and 50% of the BD case had an incomplete palisading odontoblast layer (score 3). A thick and regular dentine bridge deposition was seen in the ESRRM putty group in comparison with MM-MTA and BD cases. The cortical plate healing in all ESRRM putty samples was complete (score 3), while an incomplete closure was seen in MM-MTA and BD groups (score 2). Both the MM-MTA and ESRRM putty groups had fully organized PDL (score 2), while in 50% of BD cases, a necrotizing area and disorganized PDL with inflammatory cells infiltration were still present. Statistically significant differences in the scores of any histologic parameters among the three tested materials were observed neither in the 1st nor in the 4th weeks of the experimental period. Conclusion. Better tissue compatibility and repair of pulpal and periodontal tissue have been detected after lateral perforation in the root of rat incisors when treated with ESRRM putty than MM-MTA and BD. However, the difference was not significant.

Abstract Background Repairing urinary bladder (UB) defect by enterocystoplasty remains the gold standard surgical bladder reconstruction procedure to increase the capacity and compliance of dysfunctional bladders. However, many complications were recorded. Aim of the work This work aimed to compare the consequences of reconstruction of urinary bladder defect using untreated small intestinal submucosal (SIS) matrix versus seeded and unseeded decellularized SIS matrix. Material and Methods Fifty female albino rats were used in this study. The animals were divided into three groups: Group I (Control) included ten adult rats from which ileal tissue was obtained. Group II included ten adult rats in which their UB defect was repaired by untreated cellular SIS. Group III included twenty adult rats that were subdivided into two subgroups, 10 rats each; Subgroup IIIA where rats had their UB defect repaired by acellular SIS and subgroup IIIb where rats had their UB defect repaired by acellular SIS seeded with adipose mesenchymal stem cells (AMSCs).Ten young rats were used for preparation of AMSCs. Morphometric and statistical analysis were also performed. Results In rats where UB defect was repaired by untreated cellular SIS, the graft area showed loss of epithelial polarity, presence of intraepithelial cysts and occasional extension of urothelium to the outer surface forming fistula. There were areas of metaplasia with the appearance PAS positive cells. In the lamina propria, there was areas of lymphocytic infiltration together with significant increase in the collagen fiber deposition (p < 0.05). There was a significant decrease thickness of muscle layer as compared to control (p < 0.05). In rats where UB defect was repaired by acellular SIS, urothelium in the graft area showed occasional squamous metaplasia and often the urothelium extended to the deeper layers forming Brunn's nest. There was minimal muscle regeneration in the graft area. However, in rats where UB defect was repaired by acellular SIS seeded with AMSCs, the urothelium in the graft area was nearly similar to control group with uniform urothelium thickness, minimal collagen fibers deposition and thick muscle layer that showed no significant difference from the control (p > 0.05). Conclusion Acellular SIS seeded with AMSCs showed better results compared to non-seeded and cellular SIS in reconstructing urinary bladder defects.

Summary Introduction Direct pulp capping is an important therapeutic method that has goal to provide formation of dentin bridge and healing process of the pulp. The aim of this study was to investigate the effects of new nanostructural materials based on calcium silicate systems and hydroxyapatite on exposed dental pulp in Vietnamese pigs. Material and Methods The study was conducted on 30 teeth of two Vietnamese pigs (Sus scrofa verus). On buccal surfaces of incisors, canines and first premolars, class V cavities were prepared with a small round bur and pulp horn was exposed. In the first experimental group (10 teeth) the perforation was covered with new nanostructural material based on calcium silicate systems (CS). In the second experimental group, the perforation was covered with compound of calcium silicate systems and hydroxyapatite (HA-CS) (10 teeth). In the control group, exposed pulp was covered with Pro Root MTA® (Dentsply Tulsa Dental, Johnson City, TN, USA) (10 teeth). All cavities were restored with glass ionomer cement (GC Fuji VIII, GC Corporation, Tokyo, Japan). Observation period was 28 days. After sacrificing the animals, histological preparations were done to analyze the presence of dentin bridge, inflammatory reaction of the pulp, pulp tissue reorganization and the presence of bacteria. Results Dentin bridge was observed in all teeth (experimental and control groups). Inflammation of the pulp was mild to moderate in all groups. Neoangiogenesis and many odontoblast like cells responsible for dentin bridge formation were detected. Necrosis was not observed in any case, neither the presence of Gram-positive bacteria in the pulp. Conclusion Histological analysis indicated favorable therapeutic effects of new nanostructural materials based on calcium silicate systems and hydroxyapatite for direct pulp capping in teeth of Vietnamese pigs.

ABSTRACT Objective: To compare the fracture resistance of simulated immature teeth filled with an apical barrier of mineral trioxide aggregate (MTA), Biodentine, and calcium-enriched mixture (CEM). Materials and Methods: Fifty-two single-rooted human maxillary central incisors were used. For standardization, the teeth were sectioned 6 mm above and 9 mm below the cementoenamel junction to simulate immature apex. Simulations of roots into immature apices were carried out using 1.5 mm diameter drills. The specimens were then randomly divided into three experimental groups (n = 13) and one control group (n = 13). In experimental groups, MTA, Biodentine, and CEM were placed to apical 4 mm of the simulated immature roots. The samples were stored at 37°C and 100% humidity for 1 week. A load was applied on the crown of all teeth at 135° to their long axis until fracture. The data were analyzed using one-way analysis of variance and Tukey post-hoc tests. Results: No statistically significant differences were found among MTA, CEM, and Biodentine (P > 0.05), and these groups demonstrated higher fracture resistance than control group (P < 0.05). Conclusions: Using any of the MTA, Biodentine, and CEM as an apical plug and restoring with fiber post and composite resin increases the fracture resistance of immature teeth.

Objective: This study was designed to identify the effectiveness of platelet-rich plasma gel (PRP) and platelet-rich fibrin matrix (PRF) application on the induced open chronic full-thickness cutaneous wounds in goats.Methods: A total of 24 adult apparently healthy local breed bucks were used in this study. PRP gel and PRF matrix were prepared from whole blood at the time of treatment, four square full-thickness skin wounds (4 cm × 4 cm) on the dorsal sides of the back of each animal were created, by daily scratched to interrupt healing process continuation to prolong inflammatory reaction to form chronic wound, this surgical procedure was continued for 8 weeks to ensure the traditional chronicity wounds, after that they were treated by PRP gel in the first group (n=12), while PRF matrix was used in the second group (n=12). Each group was also subdivided into treatment (n=2 wounds) and control group (n=2 wounds) in each animal. The healing process was evaluated by clinical examination during experimental period and histologically in day 7, 14, 28, and 45 post operation.Conclusion: Both groups showed faster tissue repair and epithelialization in treatment groups compared to control groups. The results of this study confirmed PRP gel and PRF matrix application improved and accelerated cutaneous open chronic wound healing.

PURPOSE: To investigate the effects of therapeutic ultrasound on the healing of tendon injuries in malnourished rats. METHODS: After the intended nutritional states had been attained, the animals (N=36) were distributed into three groups, named: G1(N=12), control group; G2(N=12), malnourished rats treated with 3 MHz ultrasound at an intensity of 0.5 W/cm²; and G3 (N=12), normal animals treated with 3 MHz at 0.5 W/cm². The injuries were induced by means of an operation to expose the heel tendon and crush it using Allis forceps. RESULTS: The data obtained relating to leukocyte counts, fibroblasts, vessel neoformation, fibrosis and collagen were subjected to statistical treatment using analysis of variance (ANOVA) and Student's t test. Results of p < 0.05 were obtained for fibrosis and collagen. CONCLUSION: The ultrasound was shown to be effective in repairing the tendon, even in malnourished animals whose healing process was retarded.

Abstract Background After renal trauma, surgical treatment is vital, but sometimes there may be loss of function due to fibrosis. This study aimed to evaluate the effect of autologous omentum flaps on injured renal tissues in a rat model. Methods A total of 30 Wistar albino rats were included and randomly divided equally into a control group and four intervention groups. Iatrogenic renal injuries were repaired using a surgical technique (primary repair 1 group and primary repair 2 group) or transposition of the autologous omentum (omentum repair 1 group and omentum repair 2 group). Blood samples were taken preoperatively and on the 1st and 7th postoperative days in all groups and on the 18th postoperative day in the control and two intervention groups. All rats were sacrificed on the 7th or 18th day postoperatively, and their right kidneys were taken for histopathological evaluation. Results The mean urea level significantly decreased from day 1 to day 7 and from day 1 to day 18 in the omentum repair 2 group (P = 0.005 and P = 0.004, respectively). There were no other significant changes in urea or creatinine levels within the intervention groups (P > 0.05). There was no significant correlation between the urea and creatinine levels and the histological scores (P > 0.05). The primary repair 1 and 2 groups had significantly higher median granulation and inflammation scores in the kidney specimen than the control and omentum repair groups (P < 0.05). The omentum repair 2 group had significantly lower median granulation and inflammation scores in the surrounding tissues than the primary repair 2 group (P < 0.05). The completion score for the healing process in the kidney specimen was significantly higher in the omentum repair groups than in the primary repair groups (P < 0.05). The omentum repair 2 group had significantly lower median granulation and inflammation scores in the surrounding tissues than the primary repair 2 group (P < 0.05). Granulation degree in the kidney specimen was strongly and positively correlated with the inflammation degree (r = 0.824, P < 0.001) and foreign body reaction in the kidney specimen (r = 0.872, P < 0.001) and a strong and negative correlation with the healing process completion score in the kidney (r = − 0.627, P = 0.001). Inflammation degree in the kidney specimen was strongly and positively correlated with the foreign body reaction in the kidney specimen (r = 0.731, P = 0.001) and strongly and negatively correlated with the healing process completion score in the kidney specimen (r = − 0.608, P = 0.002). Conclusion Autologous omentum tissue for kidney injury repair attenuated inflammation and granulation. Additionally, the use of omental tissue to facilitate healing of kidney injury may theoretically lead to a more effective healing process and reduced fibrosis and tissue and function loss.

Aim: The present study aimed to elucidate the effects of tramadol on the testicular functions of adult male rats due to the chronic usage of tramadol and the effect of its withdrawal. Method: Adult male albino rats were classified into the following 3 groups: (I) a control administered with normal saline and (II) tramadol-treated rats (40 mg/kg b.w. orally) for 21 successive days; and (III) like the rats in the second group but kept for 4 weeks after the last tramadol dose to study the effect of tramadol withdrawal. At the end of the experimental period, blood was collected and specimens from testis were taken for histopathological, biochemical, and molecular studies. A reverse transcription-polymerized chain reaction after RNA extraction from specimens was detected for the anti-apoptotic and pro-apoptotic genes in testicular tissues. Also, malondialdehyde (MDA) was measured in tissues homogenate and antioxidant enzymes activities were evaluated. Results: The results of this study demonstrated histological changes in testicular tissues in groups II and III compared to the control group, accompanied with increased apoptotic index and proved by increased B-cell lymphoma-2 (Bcl-2) associated-X-protein and caspase-3 expression, whereas anti-apoptotic Bcl-2 markedly decreased. Moreover, in tramadol-abused and -withdrawal groups, the MDA level increased, while the antioxidant enzymes activity decreased and revealed oxidative stress, indicating that tramadol is harmful at the cellular level and can induce apoptotic changes in testicular tissues. The withdrawal effect showed signs of improvement, but it did not return to normal levels. Conclusions: It could be concluded that the administration of tramadol causes abnormalities on testicular tissues associated with oxidative stress, which confirmed the risk of increased oxidative stress on testicular tissues due to tramadol abuse.

This study was conducted to prove whether ellagic acid can be used as a regulator of incision wound healing process in male albino rats as experimental animals. A total of 24 male healthy rats based on physical examination, aged at 3 months old and weight at 150-180 grams used in this study. Rats were divided into three groups and each group has 8 replications, group of control (P0) has no incision wound and no medication, treatment group (P1) has incision wound and treated with Betadine® and (P2) has incision wound and treated with 2,5 % ellagic acid. The variable that observed are histopathology changes (collagen, PMN, angiogenesis and fibrosis) from the male albino rats. The samples collection was taken at day 15 and data were collected and analyzed by Analysis of Variance (ANOVA) test. The result of collagen and PMN cells score in the control group (P0) were 1.80 ± 0.16 and 1.6 ± 0.08 with mild angiogenesis and no fibrosis. The results in the treatment (P2) group, where the collagen and PMN scores were 1.60 ± 0.08 and 1.5 ± 0.8, accompanied by mild angiogenesis and fibrosis. Meanwhile, in the treatment (P1) group, the results of scoring of collagen and PMN were lower than the other groups, 0, 80 ± 0.08 and 0.6 ± 0.16 accompanied by moderate angiogenesis and fibrosis. There was a significant difference of collagen scoring, PMN cells number, fibrosis and angiogenesis among the treatment groups (p<0.05). P1 treatment group has significant different with treatment groups P0 and P2 (p<0.05), but there is no significant difference between treatment groups P0 and P2 (p>0.05).  From the results, it can be concluded that the topical application of ellagic acid ointment was proved to provide healing process on incision wound. Topical administration of ellagic acid ointment for 14 days was able to improve the skin condition and promoted the wound healing.Keywords: Ellagic acid ; wound healing ; collagen ; angiogenesis ; fibrosis ; anti-inflammatory ; antioxidant ; antibacterial.

Aim and Objective: The study was conducted to see the vascular and follicular changes induced by dexamethasone (syntheticcorticosteroid) during involution of hyperplastic thyroid gland in albino rats. Study Design: A comparative histological study done in PostGraduate Medical Institute Lahore in 1998 Procedure: 54 adult rats were taken and divided at random into two control groups having 27 ratsand experimental groups containing 27 rats. Control group was given normal diet along with 2 microgram of Potassium iodide intraperitonealyfor 21 days while experimental groups further subdivided into group A having 3 rats and group B and C containing 12 rats each. All theexperimental groups were treated with Thiourea for 21 days. Group A was sacrificed on day 22 after withdrawal of thiourea. Group B was givenPotassium iodide intraperitonealy after stoppage of TU on day 21 and were sacrificed on days 22,26,30 and 50 in 4 sub groups (B1-B4) tostudy the involution process. Group C was injected dexamethasone from 22 to 50 days after withdrawal of TU and sacrificed on same daysin 4 subgroups (C1-C4) as sub groups B. Results: The results of experimental group. A showed increase in thyroid and relative tissue weight.Histologically this group exhibited significant increase in stromal congestion with tall follicular cells lining the small sized follicles having scantycolloid. The results of experimental groups B and C demonstrated increase in thyroid and relative tissue weight but microscopically subgroupsB revealed early and complete involution whereas subgroups C showed significantly persistent hyperplastic changes in the form of stromalcongestion, vessels wall remained well defined and tall follicular cells lining small empty follicles. Conclusion: It was concluded thatdexamehasone did retain hyperplastic changes during involution evident by stromal congestion and small sized regular follicle lined by tallfollicular cells, so it should be carefully used in thyroid diseases.

Background: The initial inflammatory reaction of pulp capping materials on the dental pulp has an intimate relation in promoting the future cellular differentiation and biomaterial mineralisation. So, analysis of immediate pulpal tissue reaction in vivo, is also important for evaluation of ultimate efficacy any pulp capping agent. To observe immediate inflammatory response of Human Dental Pulp capped with Mineral Trioxide Aggregate (MTA), Biodentin and Portland Cement (PC). Methods: A total of 70 permanent premolars teeth planned to be extracted for orthodontic alignment of occlusion were used as study sample. The teeth were divided into 3 experimental groups, MTA (n=20), Biodentin (n=20) Portland cement (n=20) and control group (n=10). After having an occlusal exposure of approximately 1.5 mm in diameter; in group A, pulp of teeth was capped with 2-mm-thick layer of ProRoot White MTA (Dentsply) and in group B, with sterile Biodentin (Septodont) according to the manufacturer’s recommendations. Whereas in group C, pulp of teeth was capped with sterile Portland Cement (PC). After placing the experimental material in each group, all teeth restored with glass i‹xiomer cement. After 24 hours the teeth were extracted, fixed in 10% buffered formalin solution, then decalcified by 10% nitric acid and embedded in paraffin. Finally, sectioned into 2 to 3-micron-thick serial sections in the linguo-buccal plane and stained with hematoxylin-eosin. After then the amount of pulp inflammation (type, intensity, and extension) were determined by using a predetermined evaluation criterion under an optical microscope at 40a magnification. Ten intact teeth, which received no exposure and pulp capping but extracted due to orthodontic purpose were also collected and treated as the control group (group D); undergone same histologic preparation and evaluation. Significantstatistical differences among the experimental groups were to be found (p<0.05). Results: Histologically, all the three tested materials produced immediate pulpal tissue reaction. ‘Biodentin’ found to be most immediate pulpal tissue reactive (reactive in 100% cases) and ‘Portland Cement’ showed least immediate tissue reaction (only in 30.0% cases). whereas, MTA produced immediate tissue reaction only in 50.0% cases. Immediate pulpal inflammatory reaction in response to tested material found to be statistically significant different between ‘Biodentin’ and ‘Portland cmient’ (p=0.01), also between ‘Biodentin’ and ‘MTA’ and (p=0.001); but there was no statistically significant difference between ‘MTA’ and ‘Portland cement’ (p =0.197). Conclusion: Considering the maximum immediate pulpal tissue reaction (Inflammation), Biodentin is expected to produce most favorable ultimate bioactivity (biomaterial mineralization) after pulp capping.
