Background. Endometriosis is the most prevalent gynecological disease with elusive etiology. The mysterious entity and the lack of noninvasive diagnostic methods affect women’s lives negatively. This study is aimed at finding the relationship between miR-340-5p, 92a-3p, and miR-381-3p and the pathogenesis of endometriosis in endometrial mesenchymal stem-like cells (eMSCs) of endometriosis and assessing their potential as a noninvasive biomarker in plasma. Methods. Peripheral blood and eMSC specimens were collected from suspected women of endometriosis before laparoscopy. Total RNA was isolated from plasma and cultured eMSCs to synthesize complementary DNA. The expression of miR-340-5p, miR-92a-3p, and miR-381-3p was analyzed by RT-qPCR. To understand these miRNAs’ role, we also did a bioinformatic analysis. Results. There was a downregulation of miR-340-5p, miR-92a-3p, and miR-381-3p in plasma, and the upregulation of miR-340-5p and the downregulation of miR-92a-3p and miR-381-3p in eMSCs of women with endometriosis. There was a positive concordance between the expression of miR-92a-3p and miR-381-3p in plasma and eMSCs. Our study also showed three genes, Solute Carrier Family 6 Member 8 (SLC6A8), Zinc Finger Protein 264 (ZNF264), and mouse double minute 2 (MDM2), as common targets of these miRNAs. Conclusions. This study has been one of the first attempts to examine the expression of miR-340-5p, miR-92a-3p, and miR-381-3p in both plasma and eMSCs and revealed their possible role in endometriosis based on in silico analysis. Biomarkers pave the way to develop a new therapeutic approach to the management or treatment of endometriosis patients. Our result as a first report shows that combined levels of miRNAs 340-5p and 381-3p may have the potential to be utilized as diagnostic biomarkers for endometriosis.

AbstractPhosphoketolase (PKET) pathway is predominant in cyanobacteria (around 98%) but current opinion is that it is virtually inactive under autotrophic ambient CO2 condition (AC-auto). This creates an evolutionary paradox due to the existence of PKET pathway in obligatory photoautotrophs. We aim to answer the paradox with the aid of bioinformatic analysis along with metabolic, transcriptomic, fluxomic and mutant data integrated into a multi-level kinetic model. We discussed the problems linked to neglected isozyme, pket2 (sll0529) and inconsistencies towards the explanation of residual flux via PKET pathway in the case of silenced pket1 (slr0453) in Synechocystis sp. PCC 6803. Our in silico analysis showed: (1) 17% flux reduction via RuBisCO for Δpket1 under AC-auto, (2) 11.2–14.3% growth decrease for Δpket2 in turbulent AC-auto, and (3) flux via PKET pathway reaching up to 252% of the flux via phosphoglycerate mutase under AC-auto. All results imply that PKET pathway plays a crucial role under AC-auto by mitigating the decarboxylation occurring in OPP pathway and conversion of pyruvate to acetyl CoA linked to EMP glycolysis under the carbon scarce environment. Finally, our model predicted that PKETs have low affinity to S7P as a substrate.

Abstract Backgroud: ZBTB protein is an important member of the C2H2 zinc finger protein family. As a transcription factor, it is widely involved in the transcriptional regulation of genes, cell proliferation, differentiation, and apoptosis. However, the role of ZBTB7A in uterine corpus endometrial carcinoma (UCEC) is unclear.Methods: In our work, we assessed the importance of ZBTB 7A in UCEC. Firstly, Using Oncomine and Tumor Immunoassay Resource (TIMER) databases to evaluate the expression of ZBTB7A. Secondly, we explored the co expression network of ZBTB7A through the cBioPortal online tool, Metascape, and LinkedOmics. TIMER was also used to explore the relationship between ZB TB7A and tumor immu ne invasion, and to detect the correlation between the ZBTB7A and the marker genes related to immune infiltration. Finally, CKK8,migration, ChIP assays were introduced to partly validate ZBTB7A function in endometrialcancer cells.Results: We found t he ZBTB7A expression in TIMER was associated with various cancers, especially UCEC. The decreased expre ssion of ZBTB7A was markedly related to the stage and prognosis of UCEC. Furthermore, ZBTB7A was also related to the expression of various immune markers s uch as Neutrophils, Dendritic cell, T cell (general), Th1, Th2, and Finally, we verified that ZBTB7 A repressed E2F4 transcription and inhibited cell s proliferation and migration . These results indicate that ZBTB7A may play a vital role in regulating immune cellinfiltration in UCEC, and is a valuable prognostic marker.Conclusions:In summary, we demonstrate that ZBTB7A is notably downregulated in UCEC, play s a vital role in regu lating immune cell infiltration, possesses diagnostic and pr ognostic values and attenuated E2F4 transcription and cell proliferation , migration in vitro.

: Glioma predominantly targets glial cells in the brain and spinal cord. There are grade I, II, III, and IV gliomas with anaplastic astrocytoma and glioblastoma multiforme as the most severe forms of the disease. Current diagnostic methods are limited in their data acquisition and interpretation, markedly affecting treatment modalities and patient outcomes. Circulating extracellular vesicles (EVs) or “magic bullets” contain bioactive signature molecules such as DNA, RNA, proteins, lipids, and metabolites. These secretory “smart probes” participate in myriad cellular activities, including glioma progression. EVs are released by all cell populations and may serve as novel diagnostic biomarkers and efficient nanovehicles in the targeted delivery of encapsulated therapeutics. The present review describes the potential of EVbased biomarkers for glioma management.

Background: Stroke is one of the most common deadly diseases with an estimated 780,000 new cases globally, of which ischemic stroke accounts for over 80% of all cases. Ferroptosis is a new form of programmed cell death that plays a vital role in many diseases, including ischemic stroke and heart diseases. The role of the ferroptosis-related gene in the diagnosis, prognosis, or therapy of ischemic stroke was not fully clarified.Methods: Ferroptosis-related differentially expressed genes (DEGs) in ischemic stroke were identified by bioinformatic analysis of the GSE16561 and GSE22255 datasets. Subsequently, receiver operator characteristic (ROC) monofactor analysis was performed to evaluate the diagnostic value of ferroptosis-related biomarkers in ischemic stroke.Results: A total of 10 ferroptosis-related DEGs were identified in ischemic stroke vs. normal control. GO and KEGG analysis revealed that these 10 ferroptosis-related DEGs were mainly enriched in response to oxidative stress, HIF-1 signaling pathway, ferroptosis, lipid, and atherosclerosis. Moreover, the random forest model suggested three ferroptosis-related biomarkers, namely, PTGS2, MAP1LC3B, and TLR4, for ischemic stroke. Interestingly, the expression of PTGS2, MAP1LC3B, and TLR4 was upregulated in ischemic stroke. ROC monofactor analysis demonstrated a good performance of MAP1LC3B, PTGS2, and TLR4 in the diagnosis of ischemic stroke. The expression and diagnostic value of MAP1LC3B, PTGS2, and TLR4 in ischemic stroke were also verified using GSE22255. We also revealed the transcription factor regulation network and co-expressed protein network of ferroptosis-related biomarkers. Several potential therapeutic compounds corresponding to MAP1LC3B, PTGS2, and TLR4 were also identified for ischemic stroke, including Zinc12503187 (Conivaptan), Zinc3932831 (Avodart), Zinc64033452 (Lumacaftor), Zinc11679756 (Eltrombopag), Zinc100378061 (Naldemedine), and Zinc3978005 (Dihydroergotamine).Conclusion: Our results suggested MAP1LC3B, PTGS2, and TLR4 as potential diagnostic biomarkers for ischemic stroke, providing more evidence about the vital role of ferroptosis in ischemic stroke.

The relationship between intestinal microbiota and human health is widely recognized. The advent of molecular and genetic diagnostic methods has made it po-ssible to make truly revolutionary discoveries in the field of understanding the role of the digestive tract microbiome. Many studies have proved the association of the composition of microbiota with the development of a number of diseases of the digestive tract, such as inflammatory bowel diseases, functional gastroin-testinal disorders and some types of cancer. The food and pharmaceutical industry has presented a huge selection of food products, dietary supplements and medicines containing prebiotic and probiotic components. The use of agents that affect microbiota in the treatment of intestinal diseases in real clinical practice should be carried out strictly in accordance with the indications, in recommended doses.

Purpose of the work was to investigate the mediating role of psychological capital in the relationship between subjective perceptions of stress and person’s life satisfaction.Methods. Psycho-diagnostic methods “Stress Perception Scale-10”, “Psychological Capital”, “Life Satisfaction Scale” were used. Statistical analysis included calculation of descriptive statistics, correlations between variables (r-Pearson), finding differences between samples (ANOVA) and mediator analysis.Results. On the basis of a cross-sectional research of a sample of 460 people, the hypothesis that life satisfaction negatively correlates with the level of subjective perception of stress and positively with psychological capital was confirmed. Subjective perception of stress, in turn, negatively correlates with psychological capital. The hypothesis of the mediating role of psychological capital was partially confirmed, as the relationship between the perception of personal stress and life satisfaction remained statistically significant, although it decreased after psychological capital was added as a mediator. This result indicates that the cognitive assessment of stressors remains an important variable for predicting life satisfaction, even when regulating dispositional characteristics. Psychological capital as an integral personal resource is by a third statistically significant in determining the relationship between person’s perception of stress and life satisfaction. Affecting directly, subjective perception of stress explains 18.9% of the life satisfaction variance; at the same time the model mediated by psychological capital adds another 8.6%.Conclusion. Psychological capital is one of the integral personal resources, which provides a mechanism of response, processing and recovery from stressors, supporting the subjective well-being. It explains a third of the variance of the relationship between subjective perception of stress and life satisfaction. Psychological capital provides a partial mediating effect when people encounter stressors to maintain their well-being.Key words: life satisfaction, subjective well-being, psychological capital, perception of stress, mediator analysis. Метою роботи було вивчення опосередкованої ролі психологічного капіталу у взаємозв’язку між суб’єктивним сприйняттям стресу та задоволеністю життям особистістю.Методи. Використовувались психодіагностичні методики «Шкала сприйняття стресу-10», «Психологічний капітал», «Шкала задоволеності життям». Статистичний аналіз містив розрахунок описових статистик, кореляцій між змінними (r-Пірсона), знаходження відмінностей між вибірками (ANOVA) та медіаторний аналіз.Результати. На основі кроссекційного дослідження вибірки з 460 осіб підтверджено гіпотези, що задоволеність життям негативно корелює з рівнем суб’єктивного сприйняття стресу та позитивно – з психологічним капіталом. Суб’єктивне сприйняття стресу негативно взаємозв’язане з психологічним капіталом. Частково підтвердилась гіпотеза про опосередковану роль психологічного капіталу, оскільки взаємозв’язок між сприйняттям стресу особистістю та задоволеністю життям залишився статистично значущим, хоча і зменшився після того, як був доданий психологічний капітал як медіатор. Цей результат указує на те, що когнітивна оцінка стрес-факторів залишається важливою змінною для прогнозу задоволеності життям, навіть за умов контролю диспозиційної характеристики. Психоло-гічний капітал як інтегральний особистісний ресурс на третину статистично значуще визначає взаємозв’язок між сприйняттям стресу особистістю та задоволеністю життям. Суб’єктивне сприйняття стресу безпосереднім впливом пояснює 18,9% дисперсії задоволеності життям, а модель з опосередкуванням психологічним капіталом додає ще 8,6%.Висновки. Одним з інтегральних особистісних ресурсів, що забезпечує механізм реагування, переробки та відновлення після стресових факторів, підтримуючи суб’єктивне благополуччя, є психологічний капітал. Він пояснює третину дисперсії взаємозв’язку між суб’єктивним сприйняттям стресу та задоволеністю життям. Психологічний капітал забезпечує частковий медіаторний ефект, коли люди зіштовхуються зі стресовими факторами для підтримки свого благополуччя.Ключові слова: задоволеність життям, суб’єктивне благополуччя, психологічний капітал, сприйняття стресу, медіаторний аналіз.

The review article summarizes the results of studies on the pathogenesis, as well as the clinical and prognostic role of coronary artery calcification (CAC) in coronary artery disease. The modern views of cardiologists, surgeons and general practitioners on the comorbidities manifested by CCA are presented. The modern ideas on the relationship between atherogenesis, CCA and bone resorption are described; groups of informative biological markers reflecting the severity of process are identified. Modern diagnostic methods for the detection and study of CCA are highlighted, their advantages and limitations are indicated. The effect of atherosclerosis treatment on coronary calcification and osteopenia are discussed. Further prospects and lines of research in this area are presented.

Abstract. Background: Lowered eye blink rate may be a clinically useful indicator of acute, imminent, and severe suicide risk. Diminished eye blink rates are often seen among individuals engaged in heightened concentration on a specific task that requires careful planning and attention. Indeed, overcoming one’s biological instinct for survival through suicide necessitates premeditation and concentration; thus, a diminished eye blink rate may signal imminent suicidality. Aims: This article aims to spur research and clinical inquiry into the role of eye blinks as an indicator of acute suicide risk. Method: Literature relevant to the potential connection between eye blink rate and suicidality was reviewed and synthesized. Results: Anecdotal, cognitive, neurological, and conceptual support for the relationship between decreased blink rate and suicide risk is outlined. Conclusion: Given that eye blinks are a highly observable behavior, the potential clinical utility of using eye blink rate as a marker of suicide risk is immense. Research is warranted to explore the association between eye blink rate and acute suicide risk.

Parenting behaviors – specifically behaviors characterized by high control, intrusiveness, rejection, and overprotection – and effortful control have each been implicated in the development of anxiety pathology. However, little research has examined the protective role of effortful control in the relation between parenting and anxiety symptoms, specifically among adults. Thus, we sought to explore the unique and interactive effects of parenting and effortful control on anxiety among adults (N = 162). Results suggest that effortful control uniquely contributes to anxiety symptoms above and beyond that of any parenting behavior. Furthermore, effortful control acted as a moderator of the relationship between parental overprotection and anxiety, such that overprotection is associated with anxiety only in individuals with lower levels of effortful control. Implications for potential prevention and intervention efforts which specifically target effortful control are discussed. These findings underscore the importance of considering individual differences in self-regulatory abilities when examining associations between putative early-life risk factors, such as parenting, and anxiety symptoms.
