Carbon nanotubes (CNTs) are promising candidates for various applications including biomedical purposes. Owing to their remarkable physical, mechanical, electrical and chemical properties, CNTs have become an area of intense research and industrial activity in recent years. Therefore, toxicity and risk assessment studies are becoming increasingly important. The present study was designed to assess the effects of hydroxyl-functionalized multiwalled CNTs (OH-f MWCNTs) on sperm health and testes of adult Wistar rats. Animals were treated with different doses of OH-f MWCNTs (0.4, 2.0 and 10.0 mg/kg) along with a control group receiving only vehicle. Assessments after 15 alternate intraperitoneal doses revealed dose-related adverse effects on many endpoints tested. Results of the study showed significant impairment of sperm health at 2.0 and 10.0 mg/kg. Histology of testes demonstrated degeneration of germinal epithelium and loss of germ cells in the treatment groups. The exposure resulted in increased oxidative stress in testes in a dose-dependent manner. The findings of the study demonstrate that CNTs are potentially harmful for male reproductive health.

Carbon nanotubes bound to tumor specific antibodies offer specific treatment for cancer cells without affecting surrounding tissue. The present study seeks to affirm the initial results of CNTs in cancer therapy by investigating the toxicological effect in mice injected with CNT-Ab followed by microwave hypothermia. We were particularly interested in evaluating the biodistribution, toxicity, and immune response that may be elicited from CNT-Ab exposure in mice. 4–5 week old mice (C57BL/6) were injected with various concentrations and combinations of multiwalled carbon nanotubes (MWCNT) conjugated with specific prostate-specific membrane antigen (PMSA) antibodies. After 1-week post-injection, mice were sacrificed followed by the collection of blood separated into serum, liver, kidney and other tissues for further analysis. Serum total protein concentration across the treatment groups was varied. No significant changes in albumin levels were detected when compared to the control group (No Treatment). Group YE (.125 mg/ml anti-PSMA-MWCNT + Microwave) was found to have consistently high blood serum analyte levels, indicating impaired liver and kidney function. Likewise, groups YB (Microwave only), YF [.5 mg/ml anti-PSMA-MWCNT (No Microwave)], and YG (.5 mg/ml plain MWCNT + Microwave) seemed to show indications of impaired liver function. Analysis of gene expression revealed a significant impact on the NF-KB inflammatory response pathway. NF-KB gene was up-regulated relative to controls in all treatment groups. These results seem to suggest marginal toxicity from the injection of CNT-Ab followed by microwave hyperthermia in mice subjects.

The study objective was to evaluate targeted reprogramming of peripheral blood mononuclear cells in systemic juvenile idiopathic arthritis (sJIA) modeled in 6-month-old male Wistar rats by co-administration of complete Freund’s adjuvant and lipopolysaccharide. Methods. On day 40 of the experiment, rats were divided into three groups: control, doxycycline, and dexamethasone groups. Blood samples were collected on days 0, 41, and 55. Peripheral blood mononuclear cells were isolated gravimetrically and stained for markers and cytokines. Monocytes (CD3-CD4+) and T-helpers (CD3+CD4+) were differentiated as target cells. IL-4 was considered a marker for the pro-M2 phenotype since IL-4 can activate M2 macrophage polarization; IFN-g was considered a marker for the pro-M1 phenotype. Time-related changes in the intracellular expression of IL-4 and IFN-g were studied using flow cytometry. Data were analyzed using nonparametric statistical tests (Mann-Whitney-Wilcoxon) in the R environment for statistical computing. Results and conclusions. Monocytes (like macrophages) underwent reprogramming during the development of modeled sJIA disease. In monocytes of doxycycline and dexamethasone treatment groups, pro-M2 effects were observed earlier (day 41) than in the control group. Pro-M1 effects were observed in monocytes of doxycycline and dexamethasone groups on day 55, as compared with the control group. Characteristic time-related changes of intracellular cytokine expression were described for different groups. Importantly, the differently directed phenotype polarization was observed in sJIA and treatment groups for both monocytes and T-helpers.

The stability of carbon nanotubes (CNTs) suspension is a key factor in determining their transport, fate, and toxicity in an aquatic environment, which is significantly influenced by CNTs’ nature and water chemistry. Macromolecular dissolved organic matter (DOM) is reported to influence the stability of CNTs aggregation. However, little is known on small polar dissolved organic compound’s effects on CNTs aggregation. Nitrobenzene was selected to investigate its interaction with three different functionalized multiwalled CNTs (MWCNTs). Both the stability of CNTs aggregation and sorption hysteresis were affected by the initial concentration of nitrobenzene and the surface functionalization coverage of MWCNTs. At the initial concentration below 580 mg/L, the thermodynamic index of irreversibility (TII) and turbidity of CNTs suspension had the same tendency, indicating that the underlying mechanism is closely related. A conceptual adsorption–desorption model was proposed to further explain the relationship between the sorption hysteresis and stability of MWCNTs suspension under different initial concentrations of nitrobenzene. This provided data support to further clarify the environmental behaviors and risks of CNTs.
