Chinese cabbage is one of the most important leafy vegetables widely used in East Asian cuisines. The glucosinolate (GSL) accumulation and transcript levels of 7 transcription factors (Dofl.l, IQD1-1, MYB28, MYB29, MYB34, MYB51, and MYB122, and their isoforms) involved in the biosynthesis of aliphatic and indolic glucosinolates (GSLs) were analyzed at different stages of Chinese cabbage (Brassica rapa ssp. pekinensis) seedlings under light and dark conditions using high performance liquid chromatography and quantitative real time PCR. During seedling development, transcription of almost all transcription factors under light conditions was higher expressed than under dark conditions. Five aliphatic GSLs (progoitrin, sinigrin, glucoalyssin, gluconapin, and glucobrassicanapin) and four indolic GSLs (4-hydroxyglucobrassicin, glucobrassicin, 4-methoxyglucobrassicin, and neoglucobrasscin) were detected. Total GSL contents under light conditions 6, 8, and 10 days after sowing (DAS) were 3.2-, 3.9-, and 6.9-fold higher, respectively than those of dark conditions. Interestingly, total GSL contents 2 {85.4 μmol/g dry weight (DW)} to 10 (7.74 μmol/g DW) DAS under dark conditions were gradually decreased. In this study, our results suggest that light affects the levels of GSL in Chinese cabbage seedlings. These results could be useful for obtaining cabbage varieties rich in GSLs.

Kohlrabi is considered an important dietary vegetable worldwide. In this study, we investigated the growth and accumulation of phenolic compounds (PCs) and glucosinolates in sprouts of pale green and purple kohlrabi (Brassica oleracea var. gongylodes) in response to light and dark conditions. Pale green kohlrabi presented high fresh weight and root length irrespective of light treatment, whereas under dark conditions, it presented higher fresh weight and shoot length than purple kohlrabi. In contrast, the root length of both kohlrabies increased markedly under light conditions compared to that under dark conditions. Thirteen PCs and eight glucosinolates were detected and quantified in 10-day-old pale green and purple kohlrabies. In both kohlrabies, the individual and total phenolic levels were much higher under the light treatment than under the dark treatment. Under light and dark conditions, the total phenolic content was 6362.13 and 5475.04 µg/g dry weight in the pale green kohlrabi, respectively, whereas in the purple kohlrabi, it was 10,115.76 and 9361.74 µg/g dry weight, respectively. Dark conditions favored higher accumulation of glucosinolates than light conditions. Progoitrin, neoglucobrassicin, glucoerucin, and 4-methoxyglucobrassicin were the predominant glucosinolates in both kohlrabies and were present in much higher amounts in the pale green kohlrabi. In pale green kohlrabi under dark conditions, the total glucosinolates content was 4.75 and 2.62 times higher than that of the purple kohlrabi under light and dark conditions, respectively. Among individual glucosinolates, in the pale green kohlrabi under the dark condition, progoitrin was found to have the highest content, which was 90.28 and 54.51 times higher than that in the purple kohlrabi under light and dark conditions, respectively. These results show that the phenolic and glucosinolates levels varied widely, and these variations between the two types of kohlrabi under both light and dark conditions were significant. Our findings suggest that light and dark conditions enhance the accumulation of PCs and glucosinolates, respectively, during the development of kohlrabi seedlings.

One of the most important requirements for a high-performance EM, especially an analytical EM using a fine beam probe, is to prevent specimen contamination by providing a clean high vacuum in the vicinity of the specimen. However, in almost all commercial EMs, the pressure in the vicinity of the specimen under observation is usually more than ten times higher than the pressure measured at the punping line. The EM column inevitably requires the use of greased Viton O-rings for fine movement, and specimens and films need to be exchanged frequently and several attachments may also be exchanged. For these reasons, a high speed pumping system, as well as a clean vacuum system, is now required. A newly developed electron microscope, the JEM-100CX features clean high vacuum in the vicinity of the specimen, realized by the use of a CASCADE type diffusion pump system which has been essentially improved over its predeces- sorD employed on the JEM-100C.

This study was aimed to investigate the ability of N.oleander to remove Cadmium (Cd) from wastewater. A prolonged toxicity test was performed in a single exposure and run for 65 days with various concentrations of Cd. Plants were grown in sand medium and irrigated with simulated wastewater contaminated with Cd, using different concentrations (0, 10, 25, 50, 75 mg/L), which were chosen based on previous preliminary test. The results of physical observation of the plants didn’t show any withering symptoms. The Cd concentration in plants increased, while in water decreased. The results of plant analysis showed that Cd concentration in plant shoots (stems and leaves) was higher than that in roots for almost all exposure doses along the test duration. The concentration of Cd in water decreased significantly from the first week of the test and become (0 mg/l) on day-35 for 10 and 25 mg/l exposure doses, while exceeded the permissible limits for 50 and 75 mg/l exposure doses and were 0.14 and 0.91 mg/l, respectively. Wet weight and dry weight of Oleander decreased with increasing Cd concentration level except for 10 mg/l exposure dose where the plant wet weight and dry weight increased at the end of the test. Bioaccumulation factor (BAF) and Translocation Factor (TF) was found to be greater than 1, indicating that Oleander is a successful hyperaccumulator for Cd.

Denaturing High Performance Liquid Chromatography (DHPLC) is a relatively new method used for screening DNA sequences, characterized by high capacity to detect mutations/polymorphisms. This study is focused on the Transgenomic WAVETM DNA Fragment Analysis (based on DHPLC separation method) of a 485 bp fragment from human EC-SOD gene promoter in order to detect single nucleotide polymorphism (SNPs) associated with atherosclerosis and risk factors of cardiovascular disease. The fragment of interest was amplified by PCR reaction and analyzed by DHPLC in 100 healthy subjects and 70 patients characterized by atheroma. No different melting profiles were detected for the analyzed DNA samples. A combination of computational methods was used to predict putative transcription factors in the fragment of interest. Several putative transcription factors binding sites from the Ets-1 oncogene family: ETS member Elk-1, polyomavirus enhancer activator-3 (PEA3), protein C-Ets-1 (Ets-1), GABP: GA binding protein (GABP), Spi-1 and Spi-B/PU.1 related transcription factors, from the Krueppel-like family: Gut-enriched Krueppel-like factor (GKLF), Erythroid Krueppel-like factor (EKLF), Basic Krueppel-like factor (BKLF), GC box and myeloid zinc finger protein MZF-1 were identified in the evolutionary conserved regions. The bioinformatics results need to be investigated further in others studies by experimental approaches.

A unique method to design a high-throughput and high-resolution ultrathin Czerny–Turner (UTCT) spectrometer is proposed. This paper reveals an infrequent design process of spectrometers based on Coddington’s equations, which will lead us to develop a high-performance spectrometer from scratch. The spectrometer is composed of cylindrical elements except a planar grating. In the simulation design, spot radius is sub-pixel size, which means that almost all of the energy is collected by the detector. The spectral resolution is 0.4 nm at central wavelength and 0.75 nm at edge wavelength when the width of slit is chosen to be 25 μm and the groove density is 900 lines/mm.

The content of two groups of compounds with biological activity (non-hallucinogenic indole compounds and free phenolic acids) were analyzed in extracts of fruiting bodies of four species of Phellinus: P. igniarius, P. pini, P. pomaceus and P. robustus. The presence of indole compounds in methanolic extracts was analyzed by high-performance liquid chromatography and thin-layer chromatography coupled with densitometric detection. Three metabolites (serotonin, tryptamine, and L-tryptophan) were identified. The contents of individual indole compounds ranged from 1.70 (tryptamine in P. robustus) to 8.32 mg x 100 g1 dry weight (L-tryptophan in P. robustus). Four free phenolic acids were detected in methanolic extracts by the HPLC method. The total content ranged from 9.9 mg x 100 g1 DW (P. igniarius) to 32.5 mg x 100 g1 DW (P. robustus).

A simple microplate technique was adopted for toxicity assessment of a number of pesticides including six herbicides (Atrazine, Dichloroprop, Glyphosphate, Chlorsulfuron, MCPA, and Simazine), an insecticide (Dimethoate) and a fungicide (Propiconazol). Growth response of free and immobilized cultures of the green chlorococcal algae Selenastrum capricornumtum to different treatments of these pesticides was tested and compared. The biotests were carried out under conditions optimal for the growth of the test alga. Algal growth was exposed in terms of dry weight, and was employed as the toxicity-response parameter. Dose-response curves were used to calculate the toxicity of the tested compounds in terms of EC50. Based on EC50 values, the responses of both immobilized and free cultures were quite similar for almost all the treatments. The technique facilitated the visual detection of the lowest toxic concentration giving no detectable algal growth (EC100). The technique is quite simple, rapid, practical, accurate, and space saving. It suggested that batteries of immobilized algae could replace free cultures in studies of toxicity testing.

Agriculture in Mali, a country in Sahelian West Africa, strongly depends on rainfall and concurrently has a low adaptive capacity, making it consequently one of the most vulnerable regions to climate change worldwide. Since early-season drought limits crop germination, and hence growth, ultimately yield during rain-fed depending on production is commonly experienced nowadays in Mali. Germination and establishment of key crops such as the staple sorghum could be improved by seed priming. The effects of hydro-priming with different water sources (e.g., distilled, tap, rain, river, well water) were evaluated respectively for three priming time durations in tepid e.g., at 25 °C (4, 8, and 12 h) and by hot water at 70 °C (in contrast to 10, 20, and 30 min.) in 2014 and 2015. Seed germination and seedling development of nine sorghum genotypes were monitored. Compared to non-primed seed treatments, hydro-priming significantly [p = 0.01] improved final germination percentage, germination rate index, total seedling length, root length, root vigor index, shoot length, and seedling dry weight. The priming with water from wells and rivers resulted in significant higher seed germination (85%) and seedling development, compared to the three other sources of water. Seed germination rate, uniformity, and speed were enhanced by hydro-priming also. It is argued that hydro-priming is a safe and simple method that effectively improve seed germination and seedling development of sorghum. If used in crop fields, the above most promising genotypes may contribute to managing early season drought and avoid failure of seed germination and crop failure in high climate variability contexts.

En el presente estudio se ha tratado de obtener una aproximación al perfil ideal del entrenador de alto rendimiento en atletismo. En el mundo del deporte de alta competición, el entrenador debe desarrollar funciones de maestro, técnico y líder, lo cual hace difícil delimitar sus competencias (conocimientos, habilidades y cualidades personales). Por tanto, la principal motivación de este trabajo es establecer el papel que debe desempeñar el entrenador para la preparación física, técnica, táctica, psicológica y moral del deportista de alto nivel. Para ello, se ha considerado como población objeto de estudio al conjunto de entrenadores de alto rendimiento que en el momento de la investigación estuviesen activo, ya que interesaba realizar este estudio desde su propia perspectiva. Los datos recogidos forman parte de una investigación más amplia, llevada a cabo mediante encuesta, para la que se diseñó un cuestionario de 78 preguntas en el que se abarcaban tres temas: el perfil del entrenador y del deportista, así como la figura del formador de entrenadores en el Practicum. En este trabajo nos centramos en el primer tema y el análisis de la información obtenida nos permite concluir que la que práctica totalidad de los entrenadores de atletismo consideran necesario dominar con precisión los aspectos técnico-metodológicos. Además, consideran que su ética profesional debe estar regida por los principios de autonomía y beneficencia, de modo que valores como la honestidad y la justicia deben primar en el desarrollo de sus funciones.Abstract: In the current study, we have tried to get an approximation of the ideal profile of high performance coaches in athletics. In the high-level sports world, the coach must develop the role of teacher, technician and leader,  which makes it difficult to delimit  his/her competencies (knowledge, skills and personal qualities). Therefore, the main motivation of this work is to define the role that the coach plays in relation  to the physical, technical, tactical, psychological and moral preparation of elite athletes. Keeping this aim as an objective, we have considered as the target population the high performance coaches who are active at the time of the investigation, since we were interested  in knowing their own perspective. The data that was collected is a part of a wider investigation, conducted by a survey, for which we designed a questionnaire of 78 questions divided on three topics: the profile of the coach and the athlete, as well as the figure of the coach educator in the Practicum. In this work, we focus on the first topic and the analysis of the information that we obtained allows us to conclude that almost all the track and field coaches find it necessary to dominate the technical and methodological aspects. They also consider that professional ethics must be governed by the principles of autonomy and care that values   like honesty and justice must prevail in the performance of their work.

The metabolic profile of T-2 toxin (T-2) and its modified form T-2-3-glucoside (T-2-3-Glc) remain unexplored in human samples. Therefore, the present study aimed to investigate the presence of T-2, T-2-3-Glc and their respective major metabolites in human urine samples (n = 300) collected in South Italy through an ultra-high performance liquid chromatography (UHPLC) coupled to Q-Orbitrap-HRMS methodology. T-2 was quantified in 21% of samples at a mean concentration of 1.34 ng/mg Crea (range: 0.22–6.54 ng/mg Crea). Almost all the major T-2 metabolites previously characterized in vitro were tentatively found, remarking the occurrence of 3′-OH-T-2 (99.7%), T-2 triol (56%) and HT-2 (30%). Regarding T-2-3-Glc, a low prevalence of the parent mycotoxin (1%) and its metabolites were observed, with HT-2-3-Glc (17%) being the most prevalent compound, although hydroxylated products were also detected. Attending to the large number of testing positive for T-2 or its metabolites, this study found a frequent exposure in Italian population.
