Abstract Background Cisplatin is an outstanding anticancer drug, but its use has been decreased remarkably due to sever nephrotoxicity. R. vesicarius L. is a leafy vegetable that is evident with anti-angeogenic, anti-inflammatory, anti-proliferative, hepatoprotective, and nephroprotective potential. Therefore, this study was designed to inspect its methanol extract (RVE) for possible nephroprotective effect. Methods Primarily, in vitro antioxidant activity of RVE was confirmed based on 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging aptitude. Thereafter, Swiss Albino male mice were treated with cisplatin (2.5 mg/kg) for 5 successive days to induce nephrotoxicity. Recovery from nephrotoxicity was scrutinized by treating the animals with RVE (25, 50, and 100 mg/kg) intraperitoneally (i.p.) for the next 5 consecutive days. After completion of treatment, mice were sacrificed and kidneys were collected. Part of it was homogenized in sodium phosphate buffer for evaluating malondialdehyde (MDA) level, another part was used to evaluate gene (NQO1, p53, and Bcl-2) expression. Moreover, the hydrogen peroxide (H2O2) neutralizing capacity of RVE was evaluated in HK-2 cells in vitro. Finally, bioactive phytochemicals in RVE were determined using gas chromatography–mass spectrometry (GC-MS). Results RVE showed in vitro antioxidant activity in a dose-dependent fashion with 37.39 ± 1.89 μg/mL IC50 value. Treatment with RVE remarkably (p < 0.05) decreased MDA content in kidney tissue. Besides, the expression of NQO, p53, and Bcl-2 genes was significantly (p < 0.05) mitigated in a dose-dependent manner due to the administration of RVE. RVE significantly (p < 0.05) reversed the H2O2 level in HK-2 cells to almost normal. From GC-MS, ten compounds including three known antioxidants “4H-Pyran-4-one, 2, 3-dihydro-3,5-dihydroxy-6-methyl-”, “Hexadecanoic acid”, and “Squalene” were detected. The extract was rich with an alkaloid “13-Docosenamide”. Conclusion Overall, RVE possesses a protective effect against cisplatin-induced kidney damage.

Canthium parviflorum leaves were analyzed for their proximate and phytochemical composition. The leaves were extracted with methanol (ME) and analyzed for antioxidant activity by radical scavenging method, reducing power, ferric reducing capacity, and in vitro inhibition of Fenton’s reagent induced oxidation in oil emulsion and microsomes. In addition, the effect of high temperature (100°C, 15 and 30 min) and pH (4.5, 7, and 9) on the antioxidant activity of ME was investigated. The leaves were rich in polyphenols, flavonoids β-carotene, glutathione, α-tocopherol, and ascorbic acid. The ME exhibited varying degree of antioxidant activity in a dose dependent manner. The RSA was 68%–500 μg. Reducing potency was 0.34 and FRAP was 1.377. Canthium exhibited greater inhibition of oxidation in microsomes (73%) than in the oil emulsion (21%). Heat treatment resulted in reduction of radical scavenging activity of extract from 68% to 40%. At pH 4.5 and 7 methanol extract exhibited some percent of antioxidant activity which ranged between 18 and 32%. Data indicates Canthium as a good source of antioxidants and methanol extract exhibited good antioxidant activity.

The present study was done to investigate the in vitro antioxidant activity of aqueous extract of Sesbania sesban seeds. The assays such as DPPH, Chelation, ferrous ion, ABTS, Superoxide radical, hydroxyl radical assay, FRAP assay and total antioxidant activity were done to assess the antioxidant potential of the seed extract. The extract was tested at a concentration range of 100 – 500 μg/ml for all the assays and the values were compared with a standard. The results obtained showed that the radical scavenging activity was in a dose dependent manner and found to increase with increase in concentration of the extract. The IC50 value was calculated for the assays and tabulated for inference. Different assays revealed different levels of radical scavenging potential of the extract and exhibited as a better antioxidant source for therapeutic applications.

Background. Liver diseases are a global health problem. Medicinal plants are being increasingly used to manage a wide variety of diseases including liver disorders. The aim of this study was to investigate the antioxidant properties and hepatoprotective activity of polyphenolic extract from the fruits ofTetrapleura tetraptera (T. tetraptera).Results. The extract ofT. tetrapterawas administered at doses of 50 mg/kg and 100 mg/kg for 07per osto rats before the induction of hepatotoxicity with of 2 ml/kg of 1:1 (v/v) carbon tetrachloride (CCl4) and olive oil through intraperitoneal route. Thein vitroantioxidant and radical scavenging properties ofT. tetrapterawere conducted by the FRAP method, the phosphomolybdate method, and the inhibition potential of DPPH, ABTS, OH, and NO radicals. The extraction yield ofT. tetrapterawas 19.35%. This extract contains polyphenols (273.48 mg CAE/g DM), flavonoids (5.2549 mg SE/g DM), and flavonols (1.615 mg SE/g DM). This extract showedin vitroantioxidant activity, an inhibitor power of various free radicals, and radical scavenging potential dose-dependent. The fifty-percent inhibitory concentration of the extract (IC50) for the studied radical varied from 28.16 to 136μg/L. In rats treated with the extract ofT. tetraptera, in a dose-dependent manner, the levels of hepatotoxicity markers such as alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) significantly increased while the enzyme activities of superoxide dismutase (SOD), catalase (CAT), and the level of reduced glutathione (GHS) significantly increased compared to the control group.Conclusions. The extracts from the fruit ofT. tetrapterademonstrate antioxidant activity and hepatoprotective effects.

Antioxidant activity of Hydrophila auriculata leaf extract and silver nanoparticles (AgNPs) were carried out for proving its utility in free radical mediated diseases. The flower extract and silver nanoparticles were screened for in vitro antioxidant activity by oxygen radical scavenging such as DPPH, total antioxidant assay and superoxide radical scavenging activity at different concentrations. The antioxidant potential was found to be dose dependent. AgNPs possess probable antioxidant activity as compared with Hydrophila auriculata leaf extract and near to the standard. It is an obvious indication that the resulting activity of the AgNPs is not due to any capping agents, which are found in minute amounts but is due to the elemental silver.

Objective: The aims of the present investigation were to develop the herbal and/or herbomineral formulations of Hinguleswara rasa and to compare their anti-inflammatory and antioxidant activities, in vitro, with that of standard drug samples.Methods: This study was an interventional investigation in three samples: In the first sample, Hinguleswara rasa (HR1) was prepared as per methodology described in Rasatarangini using Shuddha Hingula (10 g), Shuddha Vatsanabha (10 g), and Pippali (10 g). In the second and third sample, respectively, Hinguleswara rasa was prepared by replacing Shuddha Hingula with Kajjali where Kajjali made from Hingulotha parada and Sodhita parada constitutes two varieties of Hinguleswara rasa, i.e. HR2 and HR3. In vitro antioxidant activity was studied using 2,2-diphenyl-1-picrylhydrazyl, and the absorbance was recorded at 517 nm. For evaluating the in vitro anti-inflammatory studies, the inhibition of albumin denaturation technique was performed.Results: The results showed that the formulation of Hinguleswara rasa has shown dose-dependent activity which was observed in 100 μg concentration. HR1, HR2, and HR3 showed 36.11, 17.22, and 16.11% radical scavenging activity.Conclusion: It could be concluded that the changes made in the formulations did not affect the in vitro anti-inflammatory and antioxidant effects of the herbomineral formulations.

Dibenz[b,f]azepine and its five derivatives bearing different functional groups were synthesized by known methods. The compounds thus synthesized were evaluated for antioxidant potential through different in vitro models such as (DPPH) free radical scavenging activity,ß-carotene-linoleic acid model system, reducing power assay and phosphomolybdenum method. Under our experimental condition among the synthesized compounds dibenz[b,f]azepine (a) and 10-methoxy-5H-dibenz[b,f]azepine (d) exhibited potent antioxidant activity in concentration dependent manner in all the above four methods. Butylated hydroxyl anisole (BHA) and ascorbic acid (AA) were used as the reference antioxidant compounds. The most active compounds like dibenz[b,f]azepine and its methoxy group substituent have shown more promising antioxidant and radical scavengers compared to the standards like BHA and ascorbic acid. It is conceivable from the studies that the tricyclic amines,i.e. dibenz[b, f]azepine and some of its derivatives are effective in their antioxidant activity properties.

Introduction: Diabetes mellitus is an endocrine and metabolic disorder characterized by the defect in insulin secretion or insulin action or both. The association between diabetes mellitus and oxidative stress is well established. Cucurbitaceae is the largest family containing 120 genera mostly grown for its sweet and juicy fruit in warm climates all over the world. In the present work ethanolic extracts of the seeds of Trichosanthes cucumerina Linn (Chichinno), Lagenaria siceraria (Lauka), Cucurbita pepo (Pharsi), Luffa aegiptiaca (Ghiraula), and Benincasa hispida (Kubindo) collected from Western Nepal has been evaluated for their in vitro antioxidant activity and in vivo hypoglycemic effects. Methods: In vitro antioxidant activity was assessed by using DPPH free radical scavenging activities and their IC50 values were calculated. In vivo hypoglycemic effect was examined on normoglycemic rats. The clinical significance of ethanolic extract at the doses of 250 and 500 mg/kg body weight was investigated in 0, 30, 60, 120 and 180 minutes of oral administration. Metformin treated group was used as the positive control. Results: For DPPH radical scavenging action, the IC50 values of Trichosanthes cucumerina, Luffa aegiptiaca, Benincasa  hispida, Cucurbita pepo, Lagenaria siceraria were found to be 60.72, 127.73, 49.63, 98.16 and 52.46 µg/ml respectively which were compared to the IC50 value of ascorbic acid (Positive control) which was found to be 38.11 µg/ml. The extracts of Benincasa hispida, and were having higher antioxidant activity and tested for in vivo hypoglycemic activity.In vivo administration of two doses of ethanolic extract of Lagenaria siceraria reduced the level of blood glucose while the best result was obtained at 250 mg/kg. Conclusions: Present study revealed promising antioxidant and hypoglycemic activity of ethanolic extract of Lagenaria siceraria. The further exploration of Lagenaria siceraria for its effective use in the traditional medicinal system is essential.

The present study was conducted to evaluate the in vitro antioxidant activity of Ficus carica, commonly known as fig. Methanol and ethanol extracts of F. carica leaves were subjected to 2, 2-diphenyl-1-picryhydrazyl (DPPH) free radical scavenging activity assay where ascorbic acid being positive control had an IC50 value of 3.98±0.26 while methanol and ethanol fractions showed an IC50 of 101.76±1.12 and 93.12±1.17 respectively exhibiting their high antioxidant potential. DPPH assay was also performed on high performance liquid chromatography (HPLC) elutions. Most active antioxidant components in ethanol extract were eluted between 17–18 min, and those in methanol were eluted over 14–15 min and upon ultra-high performance liquid chromatography-mass spectrometery (Orbitrap Liquid Chromatography-Mass Spectrometry) were identified to be 13-Docosenamide, (Z)- for ethanol and ficusin for methanol fraction. Thus, it is concluded that these two components are most probable determinants of antioxidant potential of F. carica leaf extracts. © 2021 Friends Science Publishers

The aim of present study was to estimate the in vitro antioxidant potential and antimicrobial activity of hydroalcoholic extract of Vernonia cinerea. Antioxidant activity was assessed by using 2, 2- diphenyl-1-picryl-hydrazyl (DPPH) assay using ascorbic acid as standard antioxidant. The extract was found to scavenge effectively the free radicals. The total flavonoid contents were determined by established methods and were found to be 0.547 mg/100mg in quercitin equivalents. Antimicrobial activity was performed against 2 stains of human pathogenic bacteria by well diffusion method. Hydroalcoholic extract of Vernonia cinerea showed good antimicrobial activity against gram positive bacteria. The antioxidant activities may be attributed to the presence of significant amounts of flavonoid compounds. Results indicated that hydroalcoholic extract of Vernonia cinerea possess significant antioxidant effect in dose dependent manner, followed by the hydroalcoholic extract of Vernonia cinerea possessed good antimicrobial activity.
