<span>Pollution of water environment with heavy metals is becoming one of the most severe environmental and human health hazards. Lead (Pb) is a major pollutant and highly toxic to human, animals, plants, and microbes. </span><span lang="IN">Toxic metals are difficult to remove from the environment, since they cannot be chemically or biologically degraded and are ultimately indestructible. Biological approaches based on metal-resistant microorganisms have received a great deal of attention as alternative remediation processes. </span><span>This study aim to isolat</span><span lang="IN">e</span><span> and characterize Pb resistant of heterotrophic bacteria in Cilalay Lake, </span><span lang="IN">West Java, </span><span>Indonesia. The water samples were collected </span><span lang="IN">along</span><span> three points around Cilalay Lake. </span><span lang="IN">Water physical and chemical </span><span>determination was performed using the Water Quality Checker</span><span lang="IN">. </span><span>The bacterial isolates were screened on T</span><span lang="IN">r</span><span>ipton</span><span lang="IN">e</span><span> Glucose Yeast (TGY) agar plates. </span><span lang="IN">Afterwards s</span><span>elected isolates were grown on Nutrient Agar media 50% </span><span lang="IN">with </span><span>supplemented Pb 100 ppm by the standard disk. Population of resistant bacteria was counted. The result from metal resistant bacteria indicated that all isolates w</span><span lang="IN">ere</span><span> resistant. The most abundant type of resistant </span><span lang="IN">bacteria </span><span>to lead was Gram negative more than Gram positive. Identified have metal resistant bacteria could be useful for the bioremediation of heavy metal contaminated sewage and waste water</span>

This transmission electron microscopy (TEM) procedure was designed to examine a gram positive spore-forming bacillus in colony on various solid agar media with minimal artifact. Cellular morphology and organization of colonies embedded in Poly/Bed 812 resin (P/B) were studied. It is a modification of procedures used for undecalcified rat bone and Stomatococcus mucilaginosus.Cultures were fixed and processed at room temperature (RT) under a fume hood. Solutions were added with a Pasteur pipet and removed by gentle vacuum aspiration. Other equipment used is shown in Figure 3. Cultures were fixed for 17-18 h in 10-20 ml of RT 2% phosphate buffered glutaraldehyde (422 mosm/KgH2O) within 5 m after removal from the incubator. After 3 (30 m) changes in 0.15 M phosphate buffer (PB = 209-213 mosm/KgH2O, pH 7.39-7.41), colony cut-outs (CCO) were made with a scalpel.

Biocides are widely used in medicine and various industries to protect against a number of harmful microorganisms. Organic quaternary ammonium and guanidine-containing compounds, the biological action of which is based on membrane-toxic properties, are used as bactericidal preparations. The aim of this work was to study the bactericidal and fungicidal activities of the synthesized oligomeric alkylsubstituted guanidinium bromides with different radicals -C3H7, -C7H15, -C10H21, against different isolates of heterotrophic bacteria and microscopic fungi. Methods. The synthesis of alkyl-substituted guanidiniumcontaining oligomers was performed in two stages. In the first stage, alkyl-substituted guanidine was obtained by the reaction of guanidine, previously converted by alkali from the salt form to the base form by the base and alkyl bromides (Alk=-C3H7 (propyl), -C7H15 (heptyl), -C10H21 (decyl)) in methanol at a temperature of 50°C and a molar ratio of 1:1. The second carried out the reaction between aromatic oligoepoxide DER-331 and alkyl-substituted guanidine in methanol at a temperature of 50°C for 2–3 hours and a molar ratio of 1:2. Bacteria were grown on meat-peptone agar for 48 hours at a temperature of 28±2°С. Test cultures of micromycetes were cultured on agar beer wort (6°B), incubated for 14 days in a thermostat at a temperature of 28±2°C. Antimicrobial activity of newly synthesized alkyl-substituted guanidinium-containing oligomers was determined by standard disco-diffusion method (method of disks on agar) and fungicidal activity was determined by the method of holes in agar. Results. Oligomeric alkylsubstituted guanidinium bromides with different radicals composed -C3H7, -C7H15, -C10H21- synthesized by the reaction of guanidine alkyl bromides with aromatic oligoepoxydes. It was found that alkyl-substituted guanidinium-containing oligomers at a concentration of 1–3% inhibited the growth of Escherichia coli 475, Pseudomonas aeruginosa 465, Klebsiella pneumonia 479, Pseudomonas pseudoalcaligenes 109, Staphylococcus aureus 451, E. faecalis 422, Rhodococcus erythropolis 102, Bacillus subtilis 138 and most of the studied micromycetes – Aureobasidium pullulans F-41430, Paecilomyces variotii F-41432, Penicillium funiculosum F-41435, Penicillium ochrochloron F-41431, Scopulariopsis brevicaulis F-41434, Trichoderma viride F-41437, Candida albicans F-41441, Aspergillus flavus F-41442, Aspergillus niger F-41448, Penicillium sp. F-41447. Conclusions. Antimicrobial and fungicidal properties significantly depend on the length of the alkyl radical, with increasing of its length the diameter of the zone of bacterial and micromycetes growth retardation increases.10.15407/microbiolj82.06.054

<p>An important link in solving the problem of healthy food is the intensification of the livestock, poultry and fish farming, which is possible only in the adoption and rigorous implementation of the concept of rational feeding of animals. In the implementation of this concept required is the application of probiotic preparations. Currently, there is an increased interest in spore probiotics. In many ways, this can be explained by the fact that they use no vegetative forms of the bacilli and their spores. This property provides spore probiotics a number of advantages: they are not whimsical, easily could be selected, cultivated, and dried. Moreover, they are resistant to various factors and could remain viable during a long period. One of the most famous spore microorganisms, which are widely used in agriculture, is <em>Bacillus subtilis</em>. Among the requirements imposed to probiotic microorganisms is mandatory – antagonistic activity to pathogenic and conditional-pathogenic microflora. The article presents the results of the analysis of antagonistic activity of collection strains of <em>B. subtilis</em>, and strains isolated from commercial preparations. We studied the antagonistic activity on agar and liquid nutrient medias to trigger different antagonism mechanisms of <em>B. subtilis</em>. On agar media, we applied three diffusion methods: perpendicular bands, agar blocks, agar wells. We also applied the method of co-incubating the test culture (<em>Escherichia coli</em>) and the antagonist (or its supernatant) in the nutrient broth. Our results demonstrated that all our explored strains of <em>B. subtilis</em> have antimicrobial activity against a wild strain of <em>E. coli</em>, but to varying degrees. We identified strains of <em>B. subtilis</em> with the highest antagonistic effect that can be recommended for inclusion in microbial preparations for agriculture.</p><p><em><br /></em><em></em></p>

The objective of this research was to evaluate waters quality in Pari island waters for aquaculture purpose based on bacteriological information conducted in Mei and September 2011. Microbiological parameters analyzed were total density of bacteria for coliforms, E.coli, pathogenic, heterotrophic, halotoleran, phosphate-nitrate-ammonia breaker, and total cells. Method to analyze coliform bacteria was filtration, identification of pathogenic bacteria using biochemical test, density analises for heterotrophic bacteria, analises for phosphate-nitrate-ammonia breaker bacteria using pour plate, and total cell using Acridine Orange Epifluorescence Microscopy. Results showed that the abundance of total coliform cell was about 1000-7000 colony forming unit (cfu)/100 ml. The abundance of heterotrophic, halotolerant, phosphate-nitrate-ammonia bacteria in seawater was (3.6-4.3)x105 cfu/ml, (1.1-1.3)x105 cfu/ml, (0.5-3.44)x103cfu/ml; and (3.6-6.7)x105 cfu/ml, (1.6-2.7)x105 cfu/ml, (0.6-5.22)x103 cfu/ml in sediment, respectively. The total cell of bacteria was (0.05-2.1)x107cells/ml. The dog-conch (Strombus turturella) and blood-clamps (Anadara granosa) can survive in Pari Island and there was a significant increase in sea grass litter with growth average of 0.67 mm/day and 0.90 mm/day. During snails and clamps growth, there were found several genus of pathogenic bacteria such as Salmonella, Vibrio, Aeromonas, Pseudomonas, Citrobacter, Proteus, Shigella, Hafnia, and Yersinia. The results showed that Pari island waters was suitable for developing shellfish aquaculture dog conch and blood clamps. Keywords: bacteria, parameter, shellfish, aquaculture.

Pembuatan media ini bertujuan untuk menghasikan media pembelajaran fisika berbasiskan komputer dengan pokok bahasan listrik dinamis menggunakan macromedia flash 8. Pada pembuatan media pembelajaran fisika ini, software yang digunakan menggunakan software utama Macromedia Flash Professional 8 dalam pembuatan gambar dan animasi. Selain software utama,  diperlukan software pendukung antara lain Nero 7 Ultra Edition digunakan untuk membakar CD atau membuat software dalam kepingan CD dan paint digunakan jika software utama Macromedia Flash  Professional 8 tidak dapat difungsikan. Berdasarkan tujuan dan hasil kegiatan pembuatan media maka proses pembuatannya meliputi proses prancangan dan dua tahap penyelesaian yaitu perubahan tata letak isi media dan penyempurnaan tampilan. Hal ini dilakukan agar media ini memiliki estetis yang tinggi dan kesesuain antara desain dan isi materi. Kemudian dilakukan penyimpanan atau pembakaran file ke dalam bentuk Cd outorun dengan softwere Nero 7 Ultra Edition agar media ini dapat diinstalasikan secara langsung pada komputer pada saat CD dimasukkan kedalam CDRoom. Hasil akhir sebuah softwere pembelajaran fisika dengan bantuan media komputer pada pokok bahasan listrik dinamis  berbasiskan macromedia flash profesioanl 8 adalah media pembelajaran fisika dalam bentuk CD outorun yang asalah datu file utamanya berekstensi .exe.

This paper presents a strategy in which mixed biological cultures are exposed to oscillating concentration levels, to improve the potential for coexistence of desired bacterial species. A mechanistic mathematical model is constructed to investigate and illustrate this strategy. This paper is focused on competition between nitrifying, denitrifying and aerobic heterotrophic bacteria in a CSTR with sludge recycle. For nitrifying and aerobic heterotrophic cultures, the effect of sinusoidal oscillations in DO levels with an amplitude of 1.0 mg/l is a 16% specific growth rate reduction compared to that at a constant DO level. The denitrifiers growth rate is increased by an average of 59%, compared to the constant DO level situation. A similar strategy has been tested in a pilot plant. It is concluded that the influence on specific growth rates is a function of the amplitude of the oscillations. The effects are greatest when concentrations fluctuate around the half saturation concentration of the rate limiting component(s).
