Background: Hemibagrus wyckii Bagridae is one of the most important economic fish species that lives in the rivers and reservoir in Riau Province, Indonesia. The present study aimed to determine the reproductive performance of H.wyckii under culture conditions. Methods: A total of 10 female and 10 male fish were selected, and weight, length, characteristics of egg and sperm, and hatchery performance were measured. Eggs were fertilized using the dry method. Egg weight and egg diameters were measured for 50 eggs per female. Egg size (50 eggs for each fish) was measured using an Olympus microscope (CX40). Then, saline solution was added over the eggs, followed by the addition of pooled sperm from 10 males. Results: Average relative fecundity, egg weight and egg diameter were 2060±512 eggs/kg fish, 29.86±1.21 mg and 2.67±0.26 mm, respectively. The fertilization rate and hatching rate were 60.91±4.68% and 42.91±2.92% respectively. Sperm characteristics such as volume per fish (mL), pH, concentration (per mL), motility (%) and duration of motility (second) were 0.82±0.20, 7.15±0.12, 3.68±0.15, 72.77±1.46 and 47.5±4.84, respectively. Conclusion: The study results and scientific observations regarding reproductive performance suggest that H. wyckii can be considered a new candidate species for aquaculture.

The proper culture conditions for producing cellulase of Bacillus amyloliquefaciens S1, isolated from the cecum of goose was optimized by single-factor experiment combined with orthogonal test. The properties of the cellulase were investigated by DNS method. The appropriate doses of B. amyloliquefaciens S1 were obtained by adding them to goose feed. It indicated that the suitable culture conditions of producing cellulase were the culture temperature of 37°C, the initial pH of 7.0, the incubation time of 72 h and the loaded liquid volume of 75 ml per 250 ml. The effects of each factor on producing cellulase by B. amyloliquefaciens S1 were as follows: initial pH > incubation time = culture temperature > loaded liquid volume. The optimum reaction temperature and pH were 50°C and 7.0, respectively. This enzyme is a kind of neutral cellulase that possesses resistance to heat and acidity. It showed high activity to absorbent cotton, soya bean meal and filter paper. By adding different doses of B. amyloliquefaciens S1 to the goose feed, it was found that the egg production, average egg weight, fertilization rate and the hatching rate were promoted both in experiment 1 (1.5 g kg −1 ) and experiment 2 (3 g kg −1 ). Also the difference of egg production, fertilization rate and hatching rate between experiment 1 and control group was obvious ( p  < 0.05), and the average egg weight was significantly increased in experiment 2 ( p  < 0.05).

Almaco jack, Seriola rivoliana as well as some related species is of great interest in marine fish aquaculture. However, there are few studies about their reproduction in captivity. In this research work, reproductive performance and egg quality in two groups of adult Seriola rivoliana, caught in the wild and domesticated-F1 analyzed and compared, reared under optimal maturation conditions in a commercial private Laboratory. A total of 28 wild adult (>5 kg) were caught at La Paz Bay, Baja California Sur, Mexico, and 30 adult domesticated-F1 broodstock (>5 kg), were obtained from an original stock of 1,000 juveniles (3.5 g body weight) produced at Kona Blue (Hawaii, USA) sea farm. Fishes were transported to the Rancheros del Mar commercial private hatchery, where they were grown to adult size. Both groups were evaluated during eight months (May to December 2012) and compared in terms of reproduction performance (total number of spawning events, monthly spawning frequency, total number of eggs, total number of eggs per mL, and fertilization rate), egg biochemical composition (total proteins, total lipids, total carbohydrates, and triacylglycerides) and egg diameter. Results indicated that wild caught broostock showed a better reproductive performance in terms of fertilization rate, total number of spawning, monthly spawning frequency and total number of eggs produced. However, biochemical composition and egg diameter did not show statistical differences (P < 0.05) between two groups. The reproductive performance of broodstock and quality of eggs analyzed in this study are important traits to improve the aquaculture management of this species.

ABSTRACT The intensification of catfish culture requires a regular and sustainable good seeds quality. Microbial-based aquaculture with probiotic application is expected to improve the reproductive performance of catfish. The aim of this study was to evaluate the reproductive performance of catfish given probiotics Bacillus sp. NP5 with different concentrations through feed. This study used a Completely Randomized Block Design (CRBD) with the administration of probiotic Bacillus sp. NP5 at 2% concentration of 108 CFU/mL and 1010 CFU/mL in feed given and control (without probiotics), then repeated four times. Catfish broodstocks with an average size of 955 ± 261.27 g were kept in a net sized 1 m×1 m×1 m with seven broodstocks per net. Parameters observed included the Gonadosomatic Index (GSI), fecundity, number of matured eggs, frequency distribution of egg diameter, fertilization rate, and hatching rate. The result showed that the GSI value of female catfish broodstock was significantly different in sixth week as the highest value was obtained from 1010 CFU/mL probiotics with 5.19%. Moreover, the fecundity of all treatments showed significantly different results (P<0.05) as the highest value was on the concentration of 1010 CFU/mL Bacillus sp. NP5 with 137,123 ± 32,635 eggs/broodstock. This study concludes that the administration of probiotics Bacillus sp. NP5 with 1010 CFU/mL in feed given to catfish broodstock increases the fecundity (137,123 eggs/broodstock), GSI (5.19%), number of matured eggs percentage (64.50%), and hatching rate (82.53%).  Keywords: Bacillus sp. NP5, catfish, reproductive performance   ABSTRAK   Intensifikasi budidaya ikan lele menuntut penyediaan benih yang berkualitas secara teratur dan berkelanjutan. Budidaya berbasis mikroba diharapkan dapat meningkatkan kinerja reproduksi ikan lele. Tujuan penelitian ini adalah mengevaluasi performa reproduksi ikan lele yang diberi probiotik Bacillus sp. NP5 dengan konsentrasi berbeda melalui pakan. Penelitian ini menggunakan rancangan acak kelompok lengkap (RAKL) dengan pemberian probiotik Bacillus sp. NP5 pada konsentrasi 108 CFU/mL dan 1010 CFU/mL sebanyak 2% dari pakan yang diberikan serta kontrol (tanpa pemberian probiotik), masing-masing dengan empat ulangan. Induk ikan lele dengan ukuran 955 ± 261.27 g dipelihara pada jaring hapa ukuran 1 m × 1 m × 1 m sebanyak tujuh ekor per jaring hapa. Parameter yang diamati meliputi indeks kematangan gonad (IKG), fekunditas, jumlah telur matang, sebaran frekuensi diameter telur, derajat pembuahan, dan derajat penetasan. Hasil penelitian menunjukkan bahwa nilai IKG induk betina ikan lele berbeda nyata pada minggu keenam dengan nilai tertinggi diperoleh pada perlakuan probiotik 1010 CFU/mL yaitu sebesar 5.19%. Selain itu, fekunditas semua perlakuan menunjukkan hasil yang berbeda nyata (P<0.05) dengan nilai tertinggi pada perlakuan pemberian probiotik Bacillus sp. NP5 dengan konsentrasi 1010 CFU/mL sebesar 137,123 ± 32,635 telur/induk. Kesimpulan, pemberian probiotik Bacillus sp. NP5 dengan konsentrasi 1010 CFU/mL melalui pakan kepada induk ikan lele mampu meningkatkan nilai fekunditas 137,123 telur/induk, GSI 5,19%, persentase jumlah telur matang 64,50% dan derajat penetasan telur 82,53%.   Kata kunci: Bacillus sp., ikan lele, performa reproduksi

Black ghost knifefish, Apteronotus albifrons, is a South America-introduced ornamental fish species that has been widely cultivated in Indonesia. Some farmers breed this fish with different sex ratios, but the optimum sex ratio remains unclear. This study aimed to evaluate the spawning behavior and reproductive performance of black ghost knifefish with different sex ratios. The treatments in this study were arranged in a completely randomized design consisting of different sex ratios between males and females, namely: A (one male : three females), B (two males : three females), and C (three males : three females). Each treatment was done in triplicate. The broodstock were maintained in an aquarium (80 cm x 40 cm x 40 cm) and fed with bloodworm twice a day. The water was changed every day as much as 60% of the total volume. During the experiment, the parameters of spawning behavior, number of fish spawning, number of eggs, fertilization rate, hatching rate, and daily spawning frequencies were observed. The observation was done for seven days. The study results showed that black ghost knifefish spawned at night (11 pm - 2 am). The mating and spawning occurred between one male and one female. Competition between males was observed in the treatments indicated by aggressive movements of a male toward the others, such as sudden approaching, chasing, and driving away the others. Fish in treatment-B spawned consistently from day-1 to day-5. Fish in treatment-A spawned from day-2 to day-4, while fish in treatment-C spawned only on day-7. The reproductive performance parameters showed no significant difference in all treatments except the hatching rate parameter of treatment-B. The study concludes that better reproductive performance of black ghost knifefish, A. albifrons can be achieved with a spawning ratio of two males and three females. Further research on individual and mass spawning methods with the best spawning ratio of the fish is required.

<p>This experiment was conducted to determine the dietary Vitamin E requirement for reproduction of broodstock zebrafish Danio rerio.  Four isonitrogenous (39% crude protein) and isocaloric (3,260 kcal digestible energy/kg diet) practical diets, namely diets A, B, C, and D with different levels of Vitamin E were fed to zebrafish broodstock.  The broodstock were cultivated in aquaria.  Diet A contained low dosage of Vitamin E (5 mg Vitamin E /kg diet), while diets B (62 mg Vitamin E /kg diet), C (125 mg Vitamin E /kg diet), and diet D (187 mg Vitamin E /kg diet), combined respectively with 1% n-3 fatty acids and 2% n-6 fatty acids.  Fish were fed ad satiation for 60 days using these diets.  During feeding period, gonad maturation stages were examined.  The dietary with different level of Vitamin E affected egg size, chemical content, total number of larvae, normal larvae, and survival rate of larvae produced, fecundity, fertilization rate, and hatching rate.  On the other hand, fish fed on experimental diets did not show any significance differences in the gonad somatic index, fecundity, fertilization rate, and hatching rate.  Fish fed on diet D produced the highest fecundity (633.65 ± 71.86 eggs/grams of fish), fertilization rate (92.3 ± 7.7%), and hatching rate (80.31 ± 9.8%).  The total lipid content of eggs were significant, ranging from D (39.7%), C (33.5%), B (32.1%), and A (29.5%).  At a dosage of 1% of dietary n-3 fatty acids and 2% n-6 fatty acids, zebrafish require 187 mg Vitamin E/kg feed in the diet for reproduction.  .</p> <p>Keywords:  vitamin E, reproductive performance, Danio rerio</p> <p> </p> <p>ABSTRAK</p> <p>Penelitian ini ditujukan untuk mengetahui kebutuhan Vitamin E pada pakan induk ikan zebra (Danio rerio) untuk reproduksi.  Empat pakan perlakuan yang iso-protein (39%) dan iso-kalori (3.260 kcal digestible energy/kg pakan), dinamakan pakan A, B, C, dan pakan D dengan kandungan Vitamin E yang berbeda diberikan kepada induk ikan zebra.  Induk ikan dipelihara pada akuarium.  Pakan A mengandung Vitamin E terendah (5 mg Vitamin E/kg pakan), sedangkan pakan B mengandung Vitamin E 62 mg Vitamin E/kg pakan, pakan C asam mengandung Vitamin E 125 mg /kg pakan, dan pakan D mengandung Vitamin E 187 mg /kg pakan.  Semua pakan perlakuan mempunyai kombinasi asam lemak n-3 berbanding n-6 sebesar 1:2.  Ikan diberi pakan secara ad satiation selama 60 hari pemeliharaan.  Selama masa pemberian pakan, tingkat kematangan gonad diperiksa secara teratur.  Perbedaan kandungan vitamin E dalam pakan mempengaruhi fekunditas, derajat pembuahan telur, serta derajat penetasan telur.  Pakan perlakuan tidak mempengaruhi nilai gonad somatik indeks dan kelangsungan hidup larva umur 3 hari.  Pemberian pakan D menghasilkan fekunditas (633.65 ± 71.86  telur/gram induk), derajat pembuahan telur (92.3 ± 7.7%), serta derajat penetasan telur (80.31 ± 9,8%) terbaik.  Total kandungan lemak pada telur berbeda nyata, yaitu pada pakan D (39.7%), C(33.5%), B(32.1%), dan pakan A(29.5%).  Pada kandungan 1% asam lemak n-3 dan 2% asam lemak n-6, ikan zebra membutuhkan 187 mg Vitamin E/kg pakan untuk reproduksi yang normal. </p> <p>Kata kunci:  Vitamin E, penampilan reproduksi, Danio rerio</p>

Udang windu transgenik merupakan udang hasil rekayasa dengan mengintroduksikan gen antivirus yang diisolasi dari udang windu untuk menghasilkan fenotipe yang lebih baik. Domestikasi udang transgenik telah dilakukan dan berhasil memijah/bertelur, tetapi umumnya telurnya infertil yang disebabkan tidak terjadinya pembuahan di tambak pemeliharaan. Udang betina tidak kawin ditandai tidak membawa spermatofor di telikumnya. Upaya untuk mendapatkan telur fertil udang dengan inseminasi buatan (IB) perlu dilakukan. Tujuan penelitian untuk mengevaluasi performa reproduksi udang betina transgenik dan mutu larva yang dihasilkan pasca IB menggunakan sumber spermatofor yang berbeda. Penelitian ini dirancang dengan tiga perlakuan yaitu: IB menggunakan spermatofor udang windu jantan transgenik (SJT), spermatofor udang windu jantan alam Sulawesi Selatan (SulSel) (SJS) dan spermatofor udang windu jantan alam Aceh (SJA). IB dilakukan pada udang windu betina transgenik setelah dua hari moulting. Hasil penelitian menunjukkan bahwa udang windu betina transgenik pasca IB perlakuan SJT menghasilkan total telur fertil sebanyak 766.949 butir, perlakuan SJS 535.644 butir dan perlakuan SJA 678.016 butir dengan daya tetas telur fertil yaitu: pada SJT, SJS, dan SJA masing-masing adalah 53,5%; 53,7%; dan 55,0%. Uji vitalitas larva dengan perendaman dalam larutan formalin 150-200 mg/L, perendaman air tawar: 5-15 menit, dan pengeringan 3-9 menit menghasilkan sintasan larva udang yang relatif sama pada ketiga perlakuan. Nilai morfologi larva perlakuan SJT, SJA, dan SJS adalah masing-masing 85,0; 84,5; dan 75,0. Dari hasil penelitian ini mengindikasikan bahwa performa reproduksi udang windu betina transgenik dan mutu larva yang dihasilkan pasca IB tidak dipengaruhi oleh sumber spermatofor induk udang windu jantan Penaeus monodon.Transgenic tiger shrimp, Penaeus monodon has been developed in the last decade to equip shrimp with immunity against viral diseases. However, the effort to produce large quantities of specific pathogen resistance (SPR) tiger shrimp seed is hampered by several constraints in the domestication process. The successfulness of domesticated broodstock in producing larvae is very low due to low fertilization rate. An artificial insemination (AI) offers a solution to increase fertilization rate in crustacean. This study was aimed to evaluate the reproductive performance of female transgenic tiger shrimp broodstock and their larval quality after artificially inseminated with males from different sources. The spermatophores of male from different sources i.e. transgenic male spermatophore (SJT), wild male from South Sulawesi (SJS), and wild male from Aceh (SJA) were collected through electric shock and inseminated to female transgenic broodstock two days after moulting. The results showed that the total numbers of fertile eggs produced from SJT, SJS, and SJA treatment were 766,949 pcs; 535,644 pcs; and 678,016 pcs, respectively and not significantly different (P>0.05). Similar to the number of fertile eggs, the hatching rate of eggs of SJT (53.5%), SJS (53.7%), and SJA (55.0%) also did not indicate any significant differences (P>0.05). On the larval vitality test by soaking the larvae in formalin and freshwater as well as by air exposure at different duration showed no significant difference on the survival rate (P>0.05) as indicated by score value at each treatment of 85.0, 84.5, and 75.0 for SJT, SJS, and SJA, respectively. In conclusion, the reproductive performance of female transgenic tiger shrimp and their larval quality were not affected by the different sources of spermatophores inseminated artificially during the spawning cycle.

This study was designed to evaluate the effects of astaxanthin in broodfish diets on reproductive performance, egg quality and larvae quality parameters of clownfish (Amphirion ocellaris). Five treatments were tested with 5 levels of astaxanthin (Carophyll Pink 10% CWS) of 0, 50, 100, 150 and 200 mg/kg added to the feed. Each treatment was repeated in triplicate and the supplemental feeding trials were arranged for 13 months. The results showed that there were significant differences in hatching rate of egg, malformed rate and survival rate of larvae in 3 days post-hatch (p<0.05) among the feeding trials of astaxanthin supplements. The highest hatching rate of egg and survival rate and the lowest malformed rate of larvae were observed in the treatment that was supplemented with astaxanthin 150 mg/kg feed, respectively 92.14 %; 93.57 % and 0.55 %. However, the astaxanthin supplementary diets did not affect the re-maturation and spawning period, spawning frequency, fecundity, egg diameter and larval size of nemo fish among the treatments. The results also suggested that astaxanthin requirement for clownfish broodstock to improve reproductive performance was 150 mg/kg feed.

The experiments investigated the reproductive performance of the domesticated broodstock of the silver perch and the relationship between various degrees of oil globule fragmentation and egg quality. Six years old of second generation broodstock (n=3) were evaluated based on the fecundity, fertilisation rate, hatching rate, the degree of oil fragmentation of egg, and the quality of embryos and larvae produced. The fragmentation were grouped into three categories: un-fragmented (cat-1), moderately fragmented (cat-2), and highly fragmented (cat-3). The results showed that the broodstock had a relatively high fecundity (132,400 ± 7,22), fertilization rate (94.27 ± 1.28%), and hatching rates (87.94 ± 1.23%). The survival rate of larvae at 12 days post hatching (dph) in cat-1 (71.3 ± 0.9%) was higher than cat-2 (66.7 ± 0.9%) whereas cat-2 was higher than cat-3 (61.3 ± 0.3%). The eggs was dominated by cat-1 (78.11 ± 2.44%) which was significantly higher than cat-2 (21.26 ± 2.45%) and cat-3 ones (0.40 ± 0.21%). The survival rate of embryo at 20 hours post spawning (hps) and hatching rate of cat-1 (95.33 ± 0.00% and 93.33 ± 0.00%) and cat-2 (90.00 ± 0.00% and 85.00 ± 0.00%) were significantly higher than cat-3 (72.33 ± 1.76% and 60.33 ± 0.00%). The total length (TL) of the larvae of cat-1 and cat-2 (8.44 ± 0.21 mm and 8.35 ± 0.23 mm respectively) were significantly higher than larvae of cat-3 (7.09 ± 0.14 mm). No significant difference was found in the larval deformities among any categories. In conclusion, the reproductive performance of six year-old broodstock silver perch showing acceptable performance and egg categorisation based on oil globule fragmentation can be used as a useful tool to indicate eggs quality of silver perch.

The G2 trangsenic Mutiara catfish (Clarias sp.) (MTG) is a Mutiara catfish that is inserted with the CgGH gene (Clarias gariepinus Growth Hormone) through the transgenesis. The effect of transgenesis stimulates gonad growth of G2 transgenic Mutiara catfish (Clarias sp.) faster than non-transgenic fish. Study aimed to analyze the maturity of the gonads and the spawning ability of female G2 transgenic Mutiara catfish (Clarias sp.) to obtain superior broodstock candidates. Experimental method with completely randomized design (pair of parents used as treatment and repeated four times) for spawning was used for this study. Three pairs of parent G2 were crossed semi-artificially as treatment A (female 1 MTG G2 crossed with male 1 MTG G2), B (female 2 MTG G2 crossed with male 2 MTG G2) and C (female 3 MTG G2 crossed with male 3 MTG G2). The results showed that the performance of female G2 transgenic Mutiara catfish (Clarias sp.) (treatment A, B and C) was higher given non-transgenic with an average relative fecundity of 82,438 eggs / kg of broodstock, an average egg diameter of 1.76 mm and an average egg weight 1.75 mg. These indications suggest that GH transgenesis increases gonadal maturity. The gonad maturity profile reached the stage of complete maturity (full ripe) compared to Sangkuriang catfish (Clarias gariepinus) (immature gonads). Therefore it is necessary to compare the reproductive performance of G2 transgenic Mutiara catfish (Clarias sp.) with non-transgenic fish (Sangkuriang) as candidates for superior broodstock of catfish.
