Brucellosis and Q fever are neglected zoonoses of global health importance, with unknown true prevalence in occupationally vulnerable settings, partly due to misdiagnosis for other febrile conditions and poor access to primary health care. We examined the seroprevalence of these diseases and associated factors amongst pastoralists and their cattle in Sokoto State, a hub of cattle and pastoral populations in Nigeria. Serum samples randomly collected from 137 pastoralists and 366 cattle from 27 herds in three selected Local Government Areas (LGAs) in the state were analysed for antibodies to Brucella abortus using Rose Bengal Plate Test (RBT) and competitive Enzyme-Linked Immunosorbent Assay (cELISA) as well as antibodies to Coxiella burnetti using indirect ELISA. Consenting pastoralists’ knowledge, perception and practices about the diseases were assessed using a semi-structured questionnaire. Data were analysed using descriptive statistics and bivariate analysis at p ≤ 0.05 level of significance. Brucellosis adjusted individual seroprevalence were 0.83% (95%CI: 0.04–4.59%) and 0% among pastoralists; 2.28% (95%CI: 1.16–4.43%) and 5.70% (95%CI: 3.68–8.74%) in cattle by RBT and cELISA, respectively. Adjusted herd-level seroprevalence for brucellosis were 23.20% (95%CI: 11.07–42.54%) and 42.00% (95%CI: 25.27–61.11%) by RBT and cELISA, respectively. For Q fever, higher seroprevalence of 62.57% (95%CI: 54.04–70.46%) and 2.98% (95%CI: 1.57–5.58%) were recorded amongst the pastoralists and their cattle, respectively. with adjusted herd-level seroprevalence of 40.36% (95%CI: 22.57–63.17%). The LGAs of sampling were significantly (OR: 0.2; 95%CI: 0.02–1.00) associated with Q fever infection, though marginal. The majority of the pastoralists had poor knowledge, perception and practices towards the diseases. This is the first study establishing the presence of brucellosis and Q fever at the human-animal interface in Sokoto State, Nigeria. The pastoralists’ poor knowledge, perception and practices about these diseases are worrisome and are important factors for consideration in disease control.

Abstract Background Reproductive problem is one of the main constraints of livestock genetic improvement efforts in tropical countries. The aim of this study was to determine the prevalence of major infectious causes of reproductive problems of dairy cattle in selected dairy farms in central Ethiopia. Overall 86 serum samples were collected from October 2018 to February 2019 from animals with history of reproductive problems. The collected serum was tested for antibody titer against Brucella species, Neospora caninum, Bovine Viral Diarrhea (BVD), Infectious Bovine Rhinotracheitis (IBR) and Q-fever using rose-bengal and enzyme-linked immunosorbent assay (ELISA) tests. Result Among the animals with the history of reproductive disordered; abortion, still birth and repeat breeding cases were found in 61.6%, 19.8% and 18.6%, respectively. The prevalence of IBR, BVD, Neospora caninum and Coxiella brunetti was found to be 79.1%, 38.4%, 3.5% and 1.2%, respectively. The combined infection of both BVD and IBR were detected in 21% of animals. Out of the total animals examined in this study, 95.9% of Jersey breeds were found seropositive to IBR than Boran-Friesian crosses (57.7%). The incidence of BVD was significantly higher in Boran-Friesian crossbred cattle than in Jersey which was found to be 69.3% and 14.3, respectively. The prevalence of IBR and BVD was directly proportional with age of the animal and parity. Conclusion Vaccination against IBR and BVD is not practiced in Ethiopia, the rising level of those diseases in dairy sector needs regular surveillance and control program.

Abstract Background: Brucellosis is contagious bacterial disease of major socio-economic and public health importance globally and it is also one of the priority zoonotic diseases in Ethiopia. Across-sectional epidemiological study was carried out from April 2017 to April 2018 to estimate the sero prevalence of bovine and human brucellosis and to assess the associated risk factors of brucellosis in Seka Chokorsa and Shebe Sonbo districts of Jimma zone. Results: The overall prevalence of cattle brucella infection at individual and herd level were 5.9% (95%CI: 4.1%-8.1%) and 26.6 (95%CI: 19.1%-35.3%) based on diagnosis using commercial kits of the competitive enzyme-linked Immunosorbent assay (CELISA) from Brucella abortus antibodies. Univariate logistic regression analysis at individual animal level showed that animals from large herd size and households which had a practice of introduction of new animals in their herds were 3.7 and 2.3 times more likely to be seropositive, respectively. The same scenario has been observed at herd level. Molecularly, five Brucella abortus was identified as the species affecting cattle in the study areas. From the total human serum samples tested only one serum was found to be positive for CFT and thus the prevalence is 0.42 %,( 95%CI: 0.01%-2.38%). From 238 respondents, 90% of them drink raw milk and milk products, and 70% of them also eat raw meat. Furthermore, slaughtering, assisting during delivery and poor management of aborted material is a common practice that favor transmission of the pathogen in the area. Conclusions: In conclusion, the results of this study showed that brucellosis is an important and widely distributed disease in cattle in the study areas. The finding of the infection in human indicates the public health importance of brucellosis in the area. The risk behaviors and practices observed suggests the need to design all-inclusive health programmes, such one-health approach aimed at controlling brucellosis spread in the study area.

Q fever is a zoonotic disease that occurs worldwide and is caused by the obligate intracellular bacterium Coxiella burnetii. Infected animals are usually asymptomatic, but infection can cause abortion and stillbirth in ruminants. The main purpose of this study was to evaluate prevalance of Coxiella burnetii infection in aborted and nonaborted sheep serum samples in Eastern Anatolia region by using enzyme-linked immunosorbent assay (ELISA). The determine of prevalance in sheep flocks from four provinces (Elazig, Malatya, Tunceli, Bitlis) and tested for anti-C.burnetii antibody detection, by means of Chekit Q fever Elisa kit. 350 serum samples obtained from flocks belonging aborted sheep showed that a total of 56 (16%) were detected seropositivity, whereas 171 serum samples obtained from nonaborted sheep flocks in 13 of the 171 (7.60%) for C.burnetii in seropositivity were observed. Coxiellosis should be considered an important cause of sheep with abortion history and nonaborted in Elazig and neighboring provinces.

AbstractQ fever is a disease of high zoonotic potential, but interest in its causative agent is rather low although it causes some public health problems in Hungary. The prevalence of Q fever is highly variable by country. The main reservoirs of the disease are the same domestic ruminant species everywhere, but the epidemiological profile depends on the features of the specific reservoir. The aim of this large-scale study was to demonstrate the importance of Q fever in different species as a possible source for human infection in most regions of Hungary. A total of 851 serum samples from 44 dairy farms, 16 sheep flocks, 4 goat farms and 3 zoos located in different parts of Hungary were tested. The presence of antibodies to Coxiella burnetii was surveyed in dairy cattle (n = 547), goats (n = 71), sheep (n = 200) and zoo animals (n = 33). The animal species tested in Hungary showed different seroprevalence values of C. burnetii infection. Seropositivity by the enzyme-linked immunosorbent assay was found in 258 out of 547 (47.2%) cows and in 69 out of 271 (25.5%) small ruminants, among them in 47 out of 200 (23.5%) sheep and in 22 out of 71 (31.0%) goats. Antibodies to C. burnetii were not detected in zoo animals. Seropositivity was demonstrated in 44 out of 44 (100%) dairy cattle farms, with at least one serum sample found to be positive on each farm. The seropositivity rate of small ruminant farms was 55.0% (11 positive out of 20 tested), with 9 out of 16 (56.3%) sheep flocks and 2 out of 4 (50.0%) goat herds showing seropositivity.

ABSTRACTIn the peak of the 2009 Q fever outbreak in the Netherlands, we introduced a diagnostic algorithm for acute Q fever with an enzyme-linked immunosorbent assay for immunoglobulin M antibodies toCoxiella burnetiiphase II antigens (MII screen) as an initial step. Subsequently, an immunofluorescence assay or PCR was performed depending on the MII screen outcome, date of onset of disease, and inpatient or outpatient setting. The impact of MII screen on the number of immunofluorescence assays performed and the contribution of PCR to diagnosis were retrospectively evaluated in 825 patients referred in a 17-day period. Acute Q fever was diagnosed in 256 patients. The introduction of MII screen reduced the number of immunofluorescence assays performed by more than 80%. In 103 patients, PCR analysis contributed to the diagnosis of acute Q fever. Q fever diagnostics were hampered by the fact that for a high number of patients the date of onset of disease was not provided and the requested follow-up serum samples were not received.

Bovine reproductive diseases are endemic in Indonesia, but comprehensive information about their infectious causes is not available. Therefore, our aim for this study was to detect several infectious agents that cause reproductive diseases in Indonesian beef and dairy cows. A total of 152 cow serum samples collected by Faculty of Veterinary Medicine of Brawijaya University and Veterinary Disease Investigation Centre as a part of the mandatory and regularly surveillance system from three provinces during 2019–2020 were used. The samples were then sent to Indonesian Research Centre for Veterinary Science (IRCVS) for further detection of seven reproductive diseases by enzyme-linked immunosorbent assay (ELISA). Seven reproductive diseases to be tested in parallel are neosporosis, chlamydiosis, brucellosis, Q fever, bovine viral diarrhea (BVD), infectious bovine rhinotracheitis (IBR), and BHV-4 infection. The dominant reproductive diseases in Indonesian cows were BVD (45.69%), chlamydiosis (31.58%), IBR (20.53%), neosporosis (11.84%), and BHV-4 infection (10.53%). The seroprevalence of IBR, BHV-4 infection, neosporosis, and brucellosis varied significantly P < 0.05 between dairy and beef cattle. The most dominant reproductive diseases in aborted cows were chlamydiosis (45%), BVD (41%), and neosporosis (10%). The conclusion drawn from this study is that the dominant reproductive diseases in Indonesian cows are BVD, chlamydiosis, IBR, neosporosis, and BHV-4 infection. Chlamydiosis, BVD, and neosporosis are common among aborted cow. Chlamydiosis, neosporosis, and BHV-4 infection should be included in the national priority list in Indonesia. Control and preventive measures should be focused on high-risk areas and animals like stray cat and dog.

A commercially available enzyme-linked immunosorbent assay (ELISA) for the diagnosis of Q fever (PanBio Coxiella burnetiiimmunoglobulin M [IgM] ELISA, QFM-200) was compared to the indirect fluorescent antibody test (IFAT) for C. burnetii IgM and the complement fixation test (CFT). The ELISA demonstrated 92% agreement with the reference method (IFAT), and gave a sensitivity of 99% (69 of 70 samples) and a specificity of 88% (106 of 121). Specificity can be increased with confirmation by IFAT. CFT was found to have a specificity of 90% (107 of 119), although it was lacking in sensitivity (73%; 51 of 70). No cross-reactivity was observed in the ELISA with serum samples from patients with mycoplasma (n = 6), chlamydia (n = 5), or legionella (n = 4) infections, although 2 of 5 patients with leptospirosis and 1 of 4 samples containing rheumatoid factor (RF) demonstrated positive results in the ELISA. Results indicate that the performance of the PanBio C. burnetii (Q fever) IgM ELISA (F = 187) is superior to that of CFT (F = 163), and consequently the ELISA should be a useful aid in the diagnosis of acute Q fever.

Abstract A cross-sectional survey was carried out to estimate the seroprevalence of Coxiella burnetii in extensively grazed cattle and sheep from central Italy and to identify the related risk factors. Data on notified human Q fever cases in the area were also collected and described. A two-stage cluster sampling was performed. A total of 5083 animals (2210 cattle; 2873 sheep) belonging to 186 farms (92 herds; 94 flocks) were tested for the presence of antibodies against C. burnetii using a commercial enzyme-linked immunosorbent assay kit. The prevalence at the animal-level resulted three times higher in sheep compared to cattle (37.8% vs. 12.0%; χ2 = 270.10, P < 0.001). The prevalence at the herd-level was also higher in sheep than in cattle (87.2% vs. 68.5%; χ2 = 9.52, P < 0.01). The multivariate analysis showed a higher risk of seropositivity for cattle aged 67–107 months (OR 2.79, 95% CI 1.86–4.18), cattle >107 months of age (OR 2.07, 95% CI 1.36–3.14) and mixed breed cattle (OR 1.74, 95% CI 1.11–2.72). A herd size >92 animals was recognized as herd-level risk factor in cattle (OR 6.88, 95% CI 1.67–28.37). The risk of being seropositive was double in sheep belonging to flocks >600 animals (odds ratio (OR) 2.04, 95% CI 1.63–2.56). Sheep were confirmed to be the most exposed species. Nevertheless, the prevalence observed in cattle also suggests the potential involvement of this species in the circulation of the pathogen in the area. Seven confirmed human Q fever cases were reported. In five out of seven cases there was at least one exposed herd within a 5 km buffer. Even though the source of the infection was not identified, the possibility of C. burnetii circulating in the livestock and human population in the study area cannot be overlooked. The integration between veterinary and human surveillance will be crucial to understand the spread of this zoonosis and to support the adoption of appropriate control measures.

Mango is one of the major fruit crops of Terai region of Nepal; however, farmers are experiencing poor productivity. Therefore, a study was conducted to determine the factors affecting the productivity and major constraints of the mango production in Saptari district of Nepal. Pre-tested semi-structured questionnaire was administered among randomly selected 106 farmers from the district of Nepal. Face to face interview was scheduled to obtain the data from sampled farmers from March 26 to May 25, 2020. Multiple regressions were used to access the various factors affecting the productivity of the mango. The regression model depicted that the total number of productive trees and training on commercial mango production was found statistically significant at 1% level of significance. A unit change in the total number of productive trees was found to change the productivity by 0.94 units. Additionally, one-unit change in the trainings regarding commercial mango farming caused the change in productivity by 0.53 units. Further, incidence of diseases and pests, poor access to market, lack of irrigation facility, incidences of natural hazards and modicum of fertilizers on orchard were the major production constraints of mango in Spatari district of Nepal. Therefore, the study has suggested indispensable need training on commercial mango cultivation practices in Saptari district of Nepal.

Contagious bovine pleuropneumonia (CBPP) is an infectious and highly contagious respiratory disease of cattle and water buffalo, which is caused by the Mycoplasma mycoides subspecies mycoides small colony. It induces significant economic losses and leads to a serious food security problem, negatively influencing peoples' livelihoods in affected countries. The disease has been reported in different parts of Ethiopia with prevalence ranging from 1.78 to 96%. However, there is not enough epidemiological information about CBPP in the northwestern part of the country, particularly in North Gondar Administrative Zone. This cross-sectional study, therefore, was conducted in four selected districts (Metema, Alefa, Quara, and Dembia) of North Gondar Administrative Zone to detect the incursion and estimate the seroprevalence of CBPP and to identify the potential predisposing factors associated with the spread and occurrence of CBPP in the area. A total of 751 serum samples were collected from 41 herds (villages) having no history of CBPP vaccination. Information like age, sex, breed, herd size, and management were collected during blood sample collection. Data related to agro-climatic zone, trade trekking route from or through CBPP endemic/epidemic zone, transhumance grazing route, vehicle route, and bordering with CBPP epidemic/endemic zone were obtained from district agricultural offices. The serum was screened for CBPP using competitive enzyme-linked immunosorbent assay (c-ELISA) test. The animal level and herd level apparent seroprevalences were 12.92% (95% CI: 10.70–15.52) and 65.85% (95% CI: 49.57–79.10), respectively. The true animal level and herd level prevalences were estimated at 20.13% (95% CI: 16.64–24.21) and 65.22% (95% CI: 48.64–78.72%), respectively. At the animal level, trekking route from or through CBPP endemic/epidemic zone to the study area [OR = 4.77 (95% CI: 1.92–11.84) compared to without trekking route] was identified as the most important risk factor for CBPP spread and seropositivity. In general, epidemiological evidence of the present study indicates that CBPP is a prevalent disease, and animal trekking is an important risk factor for spread of the disease in the study area. This needs due attention from the government and other concerned bodies for its prevention and control to mitigate its economic impact.
