<p class="abstrak3"><span lang="IN">The study </span><span>was </span><span lang="IN">aimed to examine effect of protecting soybean meal using mahogany leaf extract on rumen fermentation products in vitro. The material used was cow rumen fluid, basal ration consisting of concentrate and elephant grass with a ratio of 60:40%, and mahogany leaves. The research was carried out in three stages: mahogany extraction, protein protection using mahogany extract, and in vitro stages. The test was conducted in vitro </span><span>based on</span><span lang="IN"> a completely randomized design (CRD). The treatments consisted of 4 kinds of soybean meal protection with 0% tannin concentration (</span><span>T</span><span lang="IN">0); 1.5% (</span><span>T</span><span lang="IN">1); 3% (</span><span>T</span><span lang="IN">2); and 4.5% (</span><span>T</span><span lang="IN">3). </span><span>D</span><span lang="IN">ata obtained were analyzed by analysis of variance and tested using orthogonal polynomials. </span><span>R</span><span lang="IN">esults showed that  addition of protected soybean meal with mahogany leaf extract had a cubic</span><span>al</span><span lang="IN"> effect on partial VFA, methane gas and post-rumen dissolved protein, a quadratic effect on protozoa, and a linear effect</span><span>on </span><span lang="IN">N-NH<sub>3</sub>, SPM, and RUDP. Giving extra mahogany leaves at a level of 1.5% produced a fermented product that was not different from the control while giving a level of 3% got the highest fermentation product. Giving mahogany leaf extract at a level of 4.5% resulted in the highest SPM, and RUDP but there was a decrease in soluble protein, which indicated the occurrence of overprotection. The addition of 3% mahogany leaf extract effectively increased rumen fermentation products, RUDP, and soluble protein without disturbing the activity of rumen bacteria.</span></p>

A study has been conducted to examine the effect of Mg and Co supplementation on rumen fermentation products. The study was conducted in an in vitro experiment, applying completely randomized design, 3x3 factorial. The first factor was three levels of Mg (0; 7.5 and 15.0 mM), and three levels of Co (0; 0.03 and 0.06 mM) as the second factor, total 9 treatments each repeated three times, comprising 27 experimental units all together. In vitro incubation lasted for 4 hours. Variables measured were the concentrations of VFA, N-NH3 and protein synthesis of microbial rumen. Data were subject to analysis of variance and orthogonal polynomials test. The results showed an interaction effect between Mg and Co on the concentration of VFA, N-NH3 and protein synthesis of microbial rumen. The increasing supplementation of Mg at 0.06 mM Co increased VFA concentration; the highest concentration of N-NH3 was achieved by rumen fluid supplemented with 15.0 mM of Mg and 0.03 mM of Co. The highest protein synthesis of microbial rumen was achieved by the rumen fluid supplemented with 8.18 mM of Mg with no supplementation of Co.

An experimental study aimed at assessing the reduction of gas methane production in   thin-tailed sheep through the use of Indigofera zollingeriana leaf extract was conducted from May to September 2019 at Animal Nutrition and Feed Laboratory, University of Jenderal Soedirman Purwokerto. A One Way Classification of Completely Randomized Design (CRD) experiment was employed. The treatment included Indigofera zollingeriana leaf extract with levels of 0%, 0.40%, and 0.80% of feed dry matter and each treatment was repeated six times so that there were 18 experimental units. The feed provided consisted of 60% concentrate and 40% ammoniated rice straw. The concentrate consisted of coconut cake and rice bran with a ratio composition of 1:2. The material used was rumen fluid obtained from three thin-tailed sheep, sampled from the Sokaraja slaughterhouse immediately after the sheep were slaughtered. In vitro incubation was carried out for four hours. The variables measured included protozoan population, methane gas production and bacterial populations. The data obtained were then analyzed using analysis of variance and continued with the Orthogonal Polynomial test. The use of 0.80% Indigofera zollingeriana leaf extract can reduce the protozoan populations, methane gas production and can increase the bacterial populations of sheep rumen fluid.

AbstractEnteric methane production contributes significantly to the greenhouse gas emission globally. Although, buffaloes are integral part of livestock production in Asian countries, contributing milk, meat and draft power, the contribution of enteric methane to environmental pollution attracts attention. The present study investigated the efficacy of garlic (Allium sativum) oil in reducing enteric methane production from buffaloes (Bubalus bubalis) by in vitro rumen fermentation. Garlic oil (GOL) was tested at four concentrations [0 (Control), 33.33 µl (GOL-1), 83.33 µl (GOL-2) and 166.66 µl (GOL-3) per litre of buffered rumen fluid] in 100-ml graduated glass syringes and incubated at 39℃ for 24 h for in vitro rumen fermentation study. Supplementation of GOL-1 increased (p < 0.05) total gas production in comparison with GOL-3; however, it remained comparable (p > 0.05) with control and GOL-2. Graded doses of garlic oil inclusions reduced (p < 0.001) methane concentration (%) in total gas and total methane production (ml/g DM), irrespective of concentrations. The feed degradability, volatile fatty acids and microbial biomass production (MBP) were not affected (p > 0.05) by GOL-1, but these tended to decrease in GOL-2 with marked reduction (p < 0.01) in GOL-3. The decrease (p < 0.01) in NH3–N concentration in fermentation fluid in the presence of garlic oil, irrespective of concentration, suggests reduced deamination by inhibiting rumen proteolytic bacterial population. The activities of ruminal fibrolytic enzymes (CMCase, xylanase, β-glucosidase, acetyl esterase) were not affected by lower dose (GOL-1) of garlic oil; however, reduction (p < 0.05) of these enzymes activity in rumen liquor was evident at higher doses (GOL-2 and GOL-3) of supplementation. This study shows positive impact of garlic oil supplementation at low dose (33.33 µl/l of rumen fluid) in reducing enteric methane production, thereby, abatement of environmental pollution without affecting feed digestibility.

Previous studies have shown that Bacillus subtilis natto affects rumen fermentation and rumen microbial community structure, which are limited to detect a few microbial abundances using traditional methods. However, the regulation of B. subtilis natto on rumen microorganisms and the mechanisms of microbiota that affect rumen fermentation is still unclear. This study explored the effects of live and autoclaved B. subtilis natto on ruminal microbial composition and diversity in vitro using 16S rRNA gene sequencing and the underlying mechanisms. Rumen fluid was collected, allocated to thirty-six bottles, and divided into three treatments: CTR, blank control group without B. subtilis natto; LBS, CTR with 109 cfu of live B. subtilis natto; and ABS, CTR with 109 cfu of autoclaved B. subtilis natto. The rumen fluid was collected after 0, 6, 12, and 24 h of fermentation, and pH, ammonia nitrogen (NH3-N), microbial protein (MCP), and volatile fatty acids (VFAs) were determined. The diversity and composition of rumen microbiota were assessed by 16S rRNA gene sequencing. The results revealed LBS affected the concentrations of NH3-N, MCP, and VFAs (p < 0.05), especially after 12 h, which might be attributed to changes in 18 genera. Whereas ABS only enhanced pH and NH3-N concentration compared with the CTR group (p < 0.05), which might be associated with changes in six genera. Supplementation with live B. subtilis natto improved ruminal NH3-N and propionate concentrations, indicating that live bacteria were better than autoclaved ones. This study advances our understanding of B. subtilis natto in promoting ruminal fermentation, providing a new perspective for the precise utilization of B. subtilis natto in dairy rations.

Abstract Vistore® minerals are hydroxychloride minerals that feature high metal content and improved bioavailability. This study was conducted to compare different sources of zinc (Zn) on in vitro rumen fermentation parameters. Three ruminally-cannulated Jersey heifers were adapted to a lactation diet for two weeks before used as donors. Three sources of Zn were tested at 20 ppm: No supplemental Zn (CON), ZnSO4, Vistore Zn, and another Zn hydroxychloride (Vistore-competitor). The concentration of Zn in this study was selected from a titration study (0 to 40 ppm ZnSO4) to identify the minimum concentration of ZnSo4 affecting rumen fermentation. The lactation diet (TMR) was dried and ground to 1mm and used as substrate. Rumen fluid was collected two hours after feeding. Substrate (0.5 g) was inoculated with 100 mL of 3:1 McDougall’s buffer: ruminal flued mixture at 39ºC for 24 h. Each treatment was run in triplicate and in three runs. Data were analyzed with R 3.0. The model included fixed effect of treatment and random effect of run. ZnSO4 reduced (P &lt; 0.05) maximum gas production, DMD (54 vs. 55.9%) and cellulose (27.5 and 40.7%) digestibility. acetate to propionate ration (2.20 vs. 2.24) and NH3-N concentration (6.0 vs. 7.0 mg/dL), increased (P &lt; 0.05) propionate % (27.2 vs 26.7%) compared to control. Vistore had higher pH than control (6.44 vs. 6.40, P = 0.02) but did not affect other parameters compared to CON. Vistore-competitor reduced total VFA production compared to control, ZnSO4, and Vistore (94 vs. 102, 106 and 107 mM, respectively, P = 0.01) but did not affect other parameters. In general, Vistore Zn maintained in vitro ruminal fermentation and digestibility, while ZnSO4 had negative effects on both fermentation and digestibility and Vistore-competitor reduced total VFAs. Results indicate hydroxychloride minerals may stabilize rumen parameters versus sulfate sources but different hydroxychloride sources appear to influence rumen parameters differently.

Feeding goats with calcium salts of fatty acids (CSFA) can supply ruminants with lipids, with minimal effects on ruminal fermentation and fiber digestibility. However, there is a shortage of information on the effect of CSFA on characteristics of rumen fermentation in grassland goats. Thus, the present study aimed to assess the addition of CSFA to concentrate on the parameters of rumen fermentation of grazing goats. Five rumen cannulated goats were distributed in a Latin square 5x5 design (treatments: 0%, 1.5%, 3.0%, 4.5% and 6.0% CSFA. The pH, ammonia N and volatile fatty acids (VFA) content were analyzed in the ruminal fluid at 0, 2, 4, 6 and 8 hours after concentrate supplementation. The pH and ammonia N concentration showed a linear effect with the addition of CSFA. There was no effect observed for the VFA molar concentration after grazing goats were fed with the experimental diet. In conclusion, further research is needed to investigate the addition of CSFA to goat diets because there is evidence that CSFA increases ruminal pH and decreases excess ruminal ammonia without changing the VFA concentration in the rumen fluid.

Abstract Background A need for research searching for alternative rumen enhancers warrants immediate attention. The in vitro fermentation experiment was conducted using factorial arrangement of two factors of roughage to concentrate and seven level of red amaranth leaf powder percentage of total substrate in a Completely randomized design (CRD). Two factors, namely Factor A was two ratio of roughage (R) to concentrate (C) at 60:40 and 40:60 and Factor B was level of red amaranth (Amaranthus cruentus, L) leaf powder (RALP) supplementation at 0, 2, 4, 6, 8, 10, and 12% of total dietary substrate. Results Red amaranth leaf powder (RALP) contained phytonutrients both condensed tannins and saponins in addition with high macro minerals (Ca, K, and Mg). This experiment revealed innovations of the RALP supplementation by enhancing rumen propionate (C3) production, reducing acetate (C2) to (C3) ratio, reducing protozoal population and mitigating methane (CH4) production. Furthermore, rumen dry matter degradation percentages were remarkably enhanced (P < 0.001) by increasing RALP supplementation. Conclusion Plants rich in phytonutrients and minerals such as red amaranth leaf powder (RALP) have a vital and promising role in modulating rumen fermentation, mitigating methane production, as well as increasing substrate degradability.

The effects of adding essential oils (EO) at different levels (125, 250, 500 mg/l) on rumen fermentation and biohydrogenation were examined in a rumen batch culture study. Treatments were: control without EO (CON), control with anise oil (ANO), cedar wood oil (CWO), cinnamon oil (CNO), eucalyptus oil (EUO), and tea tree oil (TEO). Essential oils, each dissolved in 1 ml of ethanol, were added to the culture flask containing 40 ml of buffer solution, 2 ml of reduction solution, 10 ml of rumen fluid, 25 mg of soybean oil, and 0.5 g of the diet. After 24 h of incubation in a water batch at 39&deg;C, three samples were collected from each flask and analyzed for ammonia-N, volatile fatty acids (VFA), and fatty acids (FA). Expect for CNO, the proportions of acetate, propionate, and acetate to propionate ratios were not affected (P &gt; 0.05) by EO addition. Addition of CWO, CNO, and TEO reduced total VFA concentrations (P &lt; 0.05) regardless of dose level. The ammonia-N concentration was greater in cultures incubated with EO regardless of dose level. Compared with the CON, the concentrations of C18:0 and trans C18:1 were reduced (P &lt; 0.05) with EO addition regardless of dose level. Compared with the CON, the concentration of linoleic acid was greater (P &lt; 0.05) when EO were added at 500&nbsp;mg/l. EO tested in this study had no effects on VFA profile but significantly reduced the formation of biohydrogenation products (C18:0 and trans C18:1).

An experiment was aimed to assess the use of the legume leaf as a source of protein feedstuff and levels of synchronization protein-energy (SPE) index in the diet of cattles on ammonia (N-NH3) and microbial protein synthesis (MPS). In vitro techniques was done. The research was used a completely randomized design (CRD), with factorially pattern (2x3), the first factor was the two species of legume (Sesbania leaves and Leucaena leaves) and the second factor was the three level of the SPE index (0.4, 0.5, and 0.6), there were 6 treatment combinations and each was 4 replicates. The results showed that no interaction between legume with SPE index, but each factor was significantly effect (P<0.05) on N-NH3 of rumen fluid and MPS. The research concluded that Leucaena leaf is a legume that is better than Sesbania leaf in terms of their ability toincrease MPS. SPE index is the best in producing MPS at level 0.6.
