Barringtonia racemosa L. has many medicinal properties especially the fruit and leaf parts. The fruits are used to relief pain and inflammation, the leaves were proved to control high blood pressure whereas the roots barks are effective to treat chicken pox. Due to its medicinal importance for human health, it is essential to conserve this plant. A comparative study of different types of media was performed to study its effect on callus growth of endosperm explant from B. racemosa. Types of basal media studied including Murashige and Skoog (MS), Lloyd and McCown Woody Plant (WPM) and Gamborg (B5). The endosperm explant of B. racemosa were cultured on different basal MS, WPM and B5 media supplemented with 1.0 mg/L 2,4-D and 1.5 mg/L kinetin. The observation of callus growth and morphological characteristics of callus were done on weekly basis within 4 weeks. After 4 weeks of incubation period, the maximum fresh weight (0.300 ± 0.027g) and dry weight (0.025 ± 0.003g) were recorded from the explants cultured on MS medium followed by WPM and B5 media supplemented with 1.0 mg/L 2,4-D and 1.5 mg/L kinetin. The textures of callus produced from MS were nodular in shape and creamy colour. In conclusion, all the media studied had successfully induced the callus growth of B. racemosa with the present of 1.0 mg/L 2,4-D and 1.5 mg/L kinetin.

The study was undertaken with a view to investigate the effects of different basal media and organic extracts on organogenesis of Dendrobium orchid. Protocorm like bodies (PLBs) were cultured in Knudson C (KC), Vacin and Went (VW), half strength Murashige and Skoog (½MS) and New Phalaenopsis (NP) media supplemented with Sabri banana pulp (Sb), Charcoal (C) and Coconut water (Cw) organic extracts. KC, VW, ½MS and NP media significantly influenced the plantlet regeneration of Dendrobium orchid. Among the media, ½MS showed better performance on fresh weight, number PLBs, shoots and leaves per plantlet, The longest shoots were found in KC medium and maximum length of leaves was found in both KC and ½MS media. In case of different organic extracts Sb showed significantly better performance on fresh weights of PLBs, number of shoot and leaves per explant, length of shoots and leaves. The experimental studies were conducted in 2007-2008, 2008-2009 and 2009-2010 crop seasons, in order to know the agro-economic yield of 'Chinês' and "Macaquinho" taro clones, propagated using huge, extra, large, medium, small and tiny cormels. The harvest was done on average on 202 days after planting, in three crop seasons. Based on the joint analysis of variance carried out, it was observed that taro clones showed significant differences in the yield of fresh and dry weight of leaves, cormels, and commercial and non-commercial comels; besides, there were significant differences in yield of a crop season to another and the size of the cuttings induced significant differences in yield. In the conditions that the experiments were conducted, and considering the highest average yield of fresh weight of commercial cormels (28.69 t.ha-1) and highest net income (US $14,741.14) correspondent to the three crop seasons, it is recommended to cultivate 'Macaquinho' clone using small cuttings in propagation.

<p>Wood fiber waste of arenga produced from starch palm industry. It is not handled properly so that could be potentially cause problems for the surrounding community. Wood fiber waste of arenga is potentially used as a medium for hydroponics. The waste can be utilized to get a new composition of hydroponic media. This study aims to determine the effect of a mixture of sand with arenga fiber waste. This research was carried out by using a mixture of sand and arenga wood fiber waste as a medium. This is an experimental study using a completely randomized design. This study aims to determine the effect of a mixture of sand and arenga fiber waste for kailan growth. The results showed that the mixture of sand and arenga wood fiber waste has no effect on kailan growth. The combination of mixtures of sand and arenga fiber waste showed no interaction. Observations on plants such as plant height, number of leaves, leaf greenness levels, the fresh weight of crop stover and stover dry weight of the plant, showed a row of the highest results in the media mix washed sand, washed sand and rice husk</p>

Pruatjan (Pimpinella pruatjan Molk.) is an Indonesian endangered plant which has various medicinal properties such as aphrodisiac, diuretic, and tonic. The plant is commonly harvested from its natural habitat, therefore it becomes endangered. Regeneration of pruatjan through organogenesis has been studied, but its shoot multiplication was very low (5 shoots per explant). The study aimed to investigate the best regeneration technique of pruatjan through somatic embryogenesis. This research was conducted at the tissue culture laboratory, Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development in 2004-2005. Callus formation of pruatjan was induced from the petioles and leaves in Driver and Kuniyaki’s (DKW) based medium containing 2,4-D combined with picloram at the level of 0.5, 1.0, 1.5, and 1.5 ppm. Embryogenic calli were then transferred into embryo development medium in two ways. First, they were directly transferred into media containing IBA/NAA at the level of 0.5, 1.0, and 1.5 ppm. Second, they were indirectly transferred into media containing 2.0 ppm 2,4-D and 0.3% casein hydrolysate prior to the IBA/ NAA media. Parameters evaluated were fresh weight, dry weight, time initiation of embryogenic callus formation, and total number of embryos. The result showed that calli of pruatjan were successfully induced from the petioles and leaves. The best calli were induced from the leaves in the DKW medium containing 2.0 ppm 2,4-D and 0.5 ppm picloram. Embryo development of the calli was best if they were first grown in the media containing 2.0 ppm 2,4-D and 0.3% casein hydrolysate then transferred to the IBA/NAA media. The total number of somatic embryos was counted up to 103 on the medium containing 1.5 ppm IBA. This study indicated that pruatjan somatic embryogenesis regeneration required three different media, i.e. for callus induction, development and maturation, and for germination.

Abstract The aim of this study was to determine the effect of fresh and dry weight, as well as the number and length of roots and number of crowns of different types of plants, on the yield of strawberry grown for the spring and autumn harvests. Three different types of frigo strawberry plants (waiting bed plants, plug plants, A+ plants) and two strawberry cultivars (‘Honeoye’ and ‘Elsanta’) were compared in the experiment. The plants were grown in polypropylene bags in an unheated glasshouse in the years 2002-2005. The correlation coefficients were calculated to estimate the relationship of the strawberry plants’ parameters to the yield quantity. An analysis of regression for the plant parameters that most significantly correlated to the strawberry yield was carried out, which determined that the fresh and dry weight of the strawberry plants, crown number, as well as length of roots were positively correlated to the quantity of the strawberry yield. The highest positive correlation coefficient was found for the fresh weight of a whole plant and the lowest one for the number of roots.

Nicotiana tabacum callus growth (fresh weight) was measured after culture in the light (16-hour photoperiod) or in darkness for four different culture media, differing in iron chelate type or concentration. All media contained MS basal medium supplemented with 30 g·L–1 sucrose, 2 mg·L–1 IAA, 0.2 mg·L–1 KIN, and 7 g·L–1 agar, pH 5.8. Three of the media contained iron-metalosate (Albion Laboratories), an organic iron chelate, at 100, 200, and 400 micromolar concentrations, and the fourth medium contained 100 μm Fe-EDTA. Twenty-five culture tubes were prepared for each of the 4 different media concentrations and 2 light treatments (8 treatments total). A 1-cm3 callus explant was used for each treatment and cultured for 56 days at 20°C. About 20-fold increases in callus fresh weight were observed for cultures incubated in light or in darkness. In addition, callus growth was not significantly affected by iron chelate type, suggesting the potential utility of this organic chelator in tissue culture media to alleviate potential problems of light-induced EDTA instability and subsequent IAA inactivation. These cultures are being maintained to examine the influence of iron chelate type on organogenesis.

<p>Indonesian society has a consumption pattern dominated staple grains group. To reduce dependence on other countries for imports of foodstuffs, need to diversification their meal using existing biodiversity. One of the crops that can be developed is the cocoyam (Xanthosoma spp.) potential as a source of carbohydrate. Cocoyam can be grown in the lowland to the mountains with a height of up to 1300 meters above sea level. Therefore it is necessary to find the most appropriate planting site so that plants can grow and develop optimally. This study aimed to determine the growth of three types of new cocoyam at a different altitude variation. This study was conducted in two different regions of altitude, is Tawangmangu (1200 m above sea level) and Klaten (98 m above sea level). Observations using three different types of cocoyam, the tuber white, purple and yellow. Each type grown in both altitude with 10 replications each. The observations were obtained include fresh weight, dry weight, number of leaves, chlorophyll and carotenoid levels were analyzed by unpaired t test and ANOVA test at 95% confidence level. The results of this study indicate that tuber white, purple and yellow tuber are planted in the lowlands (Klaten) and highlands (Tawangmangu) had significant results in the measurement of plant height, fresh weight, dry weight, and chlorophyll and carotenoid levels have significantly different results, but not the parameters of the number of leaves. Height and weight of the fresh plant<br />in Klaten higher than in Tawangmangu, while the white tuber dry weight in Tawangmangu higher than other treatments. The levels of chlorophyll and carotenoid yellow tuber is the highest in Klaten than others.</p>

The study aimed to find the media composition and watering frequency through drip irrigation technology, which is appropriate to support the growth of vanilla plants in polybags. The research was conducted in the STIPER Dharma Wacana Metro experimental garden from April to July 2020. The first factor is the composition of the planting medium, namely soil + manure, soil + manure + rice husks, soil + manure + sand, and soil + manure + cocopeat. The second factor is watering frequency, namely 1, 2, and 3 times a day. Each treatment was repeated three times. The variables observed included plant height (cm), number of leaves (strands), shoot fresh weight (g), root fresh weight (g), and biomass dry weight (g). The use of mixed media of soil, manure, and cocopeat is the best media composition by producing a root wet weight of 8.0 g and a dry weight of 7.2 g. The frequency of watering three times a day resulted in the best height of vanilla plants being 51.8 cm compared to the frequency of watering once and twice a day. There was no interaction between the composition of the growing media and the frequency of watering on vanilla plants growth in polybags.

Microgreens are vegetables that can be harvested about 7-14 days after germinationwhich has a higher nutrient and vitamin content than commonly grown vegetables. Thisstudy aims to determine the interaction between the type of light and the duration oflighting and the effect of the types of red, blue, yellow LEDs and red, blue, yellow LEDlighting on the growth of kangkung microgreen. This research is a box experiment using aSplit Plot Experiment Design with 9 treatments and 3 replications. The results showed thatthere was an interaction between the type of lamp and the duration of lighting to thegrowth of kangkung microgreen. The combination of C3T1 treatment (Yellow LED + 6hours) produced the highest plant length, namely 6.47 cm at 7 DAS, the number of leaveswas 2.00 for all treatments. The different types of lamps in kangkung microgreen had asignificant effect on growth, where the blue LED light produced the highest number ofleaves, namely 2.00 leaves at 14 DAS. The highest fresh weight per box was 18.36 g. While the Yellow LED produced the highest plant length of 9.42 cm at 14 DAS, the highestnumber of leaves was 2.00 pieces 14 DAS, the highest root length was 7.22 cm, thehighest dry weight per box was 1.03 g. The difference in lighting time onkangkung microgreen has a significant effect on the growth, where the 6 hours lightingduration produces the highest root length, namely 7.96 cm, the highest fresh weight perbox is 23.17 g, the highest dry weight per box is 1.87 g. The length of 12 hours of lightingresulted in the highest plant length, namely 10.41 cm at 14 DAS, the highest number ofleaves was 2.00 leaf blade. Based on the results of this study it can be recommended thatthe cultivation of kangkung microgreen in the box experiment can be using lighting for 6hours and pay attention to the microelements that affect the microgreen for better growth.Keywords : Microgreen, Kangkung, Red, Blue, Yellow, LED lights

The present study was taken up to know the morphological and germination physiognomies of karonda (Carissa carandas) seeds influenced by seed storage period at ambient conditions (26 ⁰C temperature and 55 % RH). Seeds were subjected to store at 10, 20, 30, 40, 50 and 60 days at room temperature (26⁰C). Germination percentage and various morphological characteristics viz., vigour index, fresh weight of shoot and root, dry weight of shoot and root, root to shoot ratio were recorded. Among stored seeds 10 days old seeds recorded highest germina-tion percentage (57 %) and 60 days old seeds recorded lowest germination (20.33 %). At the end (90 days after sowing) 10 days old seeds again possessed higher fresh weight of root (0.300 g) and shoot (1.240 g), dry weight of root (0.103 g) and shoot (0.487 g), root to shoot ratio (0.212) and vigour index (1772.70 cm) among stored seeds. These physiological observations were quite similar with the freshly harvested seed which found maximum values for all the parameters owing to higher moisture content. Karonda seeds showed good viability upto10 days thereafter its value declined and it reached minimum after 60 days of storage.
