Toxic liver damage is the most problematic for pharmacotherapy and, accordingly, the normalization of the patient’s condition. Most often, hepatotoxins include alcohol, drugs, household and industrial toxins, the effect of which on the liver leads to the development of hepatocyte necrosis. The peculiarity of drug toxic hepatitis is a wide range of clinical manifestations, the absence of unambiguous diagnostic methods, frequent poor prognosis, which makes toxic hepatitis one of the most difficult problems in clinical practice. A promising drug for the treatment of toxic liver damage is a multicomponent herbal remedy, which is currently produced by a domestic manufacturer in the form of a dietary supplement. The aim of the work is to study the effect of the herbal remedy Pancreo-Plant on the histological structure of the liver of rats, under conditions of subacute ethanol-tetrachloromethane intoxication. Materials and methods. A model of poisoning was formed and liver samples from nonlinear white rats were examined. The slides were examined under the light microscope Granum, photographing of microscopic images was carried out with Granum DCM 310 digital video camera. The photographs were processed on a computer using the Toup View program. Results. Micropreparations obtained from liver samples of rats under the condition of subacute ethanol-tetrachloromethane intoxication, were investigated. It has been proven that Pancreo-Plant helps to reduce necrotic manifestations, hydropic degeneration of hepatocytes, manifestations of steatosis, and signs of inflammation; it also stimulates carbohydrate metabolism in hepatocytes and improves regenerative manifestations. Conclusions. It was found that the investigated agent based on medicinal plant materials, when using the model of subacute ethanol-tetrachloromethane intoxication, exhibits a significant hepatoprotective effect. It normalizes the histological structure of hepatocytes, restores the processes of glycogen accumulation, and prevents the development of fatty degeneration of the liver.

Introduction. In Russia, the problem of toxic hepatitis is of great importance and relevance. The etiology of toxic hepatitis (industrial toxicants, drugs, ethanol) and, as a consequence, pathogenesis may have significant differences at the molecular genetic level. The aim of the study was to analyze the expression of genes involved in the response to toxic liver damage of various etiologies. Material and methods. Toxic hepatitis was modulated in male albino mongrel rats weighing 180-200 grams assigned to four groups (control group, carbon tetrachloride, paracetamol, ethanol). After 24 and 72 hours of paracetamol administration, rats were anesthetized and the mRNA levels of the Chek1, Gclc, Gstm1, Gstp1, Gstt1, Nfe2l2, Nqo1, Ripk1 genes in the liver homogenate were examined. Results. As a result of the analysis of the genes expression studied, the expression profile was found TO be differed depending on the etiology of toxic hepatitis. With carbon tetrachloride poisoning, an increase in the expression of the Nqo1 genes (p = 0.001), Gstm1 (p = 0.037) and a decrease in the expression of the Nfe2l2 genes (p = 0.004), Ripk1 (p = 0.004) was observed. With the liver damage by paracetamol and its metabolites, opposite to the expression of the Gstm1 gene (p = 0.001) decreased, and the expression of the Nfe2l2 (p = 0.009), Gclc (p = 0.001), Chek1 (p = 0.011) genes increased. During alcohol intoxication, there were no statistically significant changes in the expression profiles of the genes studied. Conclusion. the results obtained may indicate the involvement of various molecular genetic mechanisms in the process of response to toxic liver damage, depending on the etiology.

Objective. To study the incidence and peculiarities of intrahepatic cholestasis (IHC) syndrome in patients with toxic liver damage in acute and chronic intoxications with pesticides and to substantiate rational methods of diagnosis and treatment. Material and methods. The incidence has been analysed with the description of IHC syndrome peculiarities in acute and remote periods in 238patients with acute poisoning with pesticides: 162 — herbicides based on 2,4-dichlorophenoxyacetic acid (2,4-D), 62 — phosphororganic pesticides (POP), 14 — synthetic pyrethroids (SP) and 70 — with chronic intoxication with pesticides (CIP). Clinical-instrumental, biochemical and statistical methods of the study have been used. Results and conclusions. Along with neurological disorders, the toxic liver damage was observed in 35,8 % of cases of acute poisoning with 2,4-D-based herbicides, in 51,6 % of cases — with POP poisoning, in 64,2 % of cases — with SP poisoning and in 84,2 % of cases — with CIP. IHC syndrome was detected in 22,8 % of cases in patients with pesticide intoxication against toxic hepatitis and in 18,0 % of cases — without signs of hepatitis. The therapeutic efficacy of ursodeoxycholic acid has been established when it is included in the combined therapy of patients with IHC upon intoxication with pesticides. Introduction. The formation of toxic liver damage is carried out with the participation of various mechanisms of pathological processes. Chemicals, drugs and alcohol play a significant role. Timely and correct selection of correction drugs will help reduce the risk of toxic organ damage. The aim of the study is a comparative assessment of the hepatoprotective activity of oxymethyluracil in the early stages of exposure to various toxic agents. Materials and methods. The studies were carried out on male outbred white rats with a single injection of hepatotoxicants (carbon tetrachloride, paracetamol, ethanol) and a correction drug - oxymethyl uracil - followed by the study of biochemical parameters 1 and 3 days after the introduction of the chemical agent. Results. As a result of the studies carried out, it was found that specific metabolic changes are noted already one day after exposure to toxicants in the body of animals. Correction of tetrachloromethane and paracetamol intoxication with oxymethyl uracil leads to the normalization of the functional state of hepatocytes. Conclusion. Oxymethyl uracil exhibits hepatoprotective properties already in the early stages of acute toxic liver damage (1-3 days), being most effective in tetrachloromethane and paracetamol intoxication.

Цель исследования - изучение влияния резекции печени (РП) на аммиакэкскретирующую функцию почек при хроническом тетрахлорметановом гепатите. Методика. Опыты выполнены на 265 беспородных белых крысах (самках) массой 180-220 г. Хронический гепатит воспроизводили подкожным введением 50% раствора тетрахлорметана (CCl) на оливковом масле (0,1 мл/100 г массы тела, через сутки, c двумя двухнедельными перерывами между 6, 7 и 13-14 инъекциями). На 65-е сут. (последние) введения тетрахлорметана, удаляли часть левой доли печени (15-20% массы органа). На 3-и, 7-е и 14-е сут. после РП или лапаротомии («ложнооперированные» животные) в почках, артериальной и венозной крови, моче исследовали содержание аммиака, глутамина и мочевины. Результаты. Прогрессирование эндогенной аммиачной интоксикации после РП на фоне тетрахлорметанового гепатита сопровождается повышенной экскрецией аммиака почками. Однако это не устраняет артериальную гипераммониемию и не предотвращает накопление почками аммиака. Инкреция глютамина из почек в кровоток прекращается. К 14-м сут. послеоперационного периода возрастает потребление глютамина из артериальной крови, что приводит к его накоплению в почках. Стимулируя выведение мочевины из организма с мочой, РП одновременно активирует её образование в почках, с дальнейшим поступлением как в кровоток, так и в мочу. В зависимости от сроков послеоперационного периода это сопровождается изменением скорости реабсорбции мочевины в почках. Заключение. Полученные результаты свидетельствуют, что при РП на фоне тетрахлорметанового гепатита почки не предотвращают прогрессирование эндогенной аммиачной интоксикации, патологическое накопление аммиака и глутамина её клетками, но сохраняют способность принимать участие в регуляции повышенного содержания мочевины в артериальной крови. Mechanical (resection) or toxic (hepatitis) liver damage alone has an ambiguous effect on renal ammonia excretion during development of endogenous ammonia intoxication. The aim. The study investigated the effect of liver resection (LR) on renal ammonia excretion in chronic tetrachlorocarbon (CCl)-induced hepatitis. Methods. Experiments were conducted on 240 mongrel white rats (females) weighing 180-220 g. Chronic hepatitis was induced by subcutaneous injection of 50% solution of carbon tetrachloride (CCl) in olive oil (0.1 ml/100g body weight per day with two two-week breaks between injections 6-7 and 13-14). LR with removal of a part of the left lobe (15-20% of body weight) was performed on the 65th (last) day of CCl injections. The animals were examined on the 3rd, 7th and 14th day after LR or laparotomy (sham operation). Contents of ammonia (AM), glutamine (GN), and urea were measured in the kidney, arterial (AB) and venous ( v.renlis ) blood, and urine. Results. Progression of endogenous ammonia intoxication after LR associated with CCl-induced hepatitis and increased renal excretion of Am involves three mechanisms: 1) excretion of Am that is delivered to kidneys in the free form with AB; 2) stimulation of renal tubule secretion of Am that had formed in kidneys by deamidation of «arterial» Gn; and 3) contrary to rules, partial reabsorption of Am from collecting tubules into the blood. However, this does not eliminate arterial hyperammonemia or prevent accumulation of Am in kidneys. The stimulatory effect of LR in CCl-induced hepatitis on Gn incretion from kidneys to the circulation stops by the 14 day after surgery, and the accompanying increased consumption of Gn from AВ results in Gn accumulation in kidneys. LR stimulates urea excretion with urine and simultaneously activates kidney formation of urea, which further enters the bloodstream and urine. Depending on the postoperative period this is associated with changes in the rate of urea reabsorption in kidneys. Conclusions. In RP associated with CCl-induced hepatitis, kidneys cannot prevent progression of endogenous ammonia intoxication and pathological accumulation of ammonia and glutamine in kidney cells but retain the ability to participate in the regulation of the increased urea level in AB.

The aim of this study was to study the effect of combined MMSC and HSC transplantation on liver regeneration under conditions of toxic carbon tetrachloride damage. Materials and methods. The study was performed on white male mice with toxic liver damage by intraperitoneal administration of carbon tetrachloride at a dose of 50 µl per mouse once. An hour after modeling liver damage, placental MMSCs and HSCs were administered intravenously at a dose of 4 million cells/kg and 330 thousand cells/kg, respectively, suspended in 0.2 ml of 0.9% NaCl solution. Control group animals were given 0.9% NaCl solution-0.2 ml intravenously. On days 1, 3, and 7 after cell transplantation, changes in inflammatory activity in the liver were evaluated, and mitotic and apoptotic indices were determined. On the 7th day after the introduction of cells, the activity of DNA repair enzymes of the PARP family was analyzed. Results. Combined MMSC and HSC transplantation leads to a decrease in the index of inflammatory activity in the liver due to a decrease in necrosis, hepatocyte dystrophy, and a decrease in infiltration. As a result of the study, an increase in the activity of PARP repair enzymes was found, which led to a decrease in programmed cell death. Also, cotransplantation of MMSCs and HSCs was accompanied by increased mitotic activity of hepatocytes. Conclusion. Cotransplantation of MMSCs and HSCs under conditions of toxic liver damage reduces the inflammatory response, stimulates the mitotic activity of hepatocytes, and increases the activity of enzymes of the DNA repair system. Activation of the liver's reparative system, in turn, reduces the programmed death of hepatocytes.

Bacteria in the genus Xanthomonas infect a wide range of crops and wild plants, with most species responsible for plant diseases that have a global economic and environmental impact on the seed, plant, and food trade. Infections by Xanthomonas spp. cause a wide variety of non-specific symptoms, making their identification difficult. The coexistence of phylogenetically close strains, but drastically different in their phenotype, poses an added challenge to diagnosis. Data on future climate change scenarios predict an increase in the severity of epidemics and a geographical expansion of pathogens, increasing pressure on plant health services. In this context, the effectiveness of integrated disease management strategies strongly depends on the availability of rapid, sensitive, and specific diagnostic methods. The accumulation of genomic information in recent years has facilitated the identification of new DNA markers, a cornerstone for the development of more sensitive and specific methods. Nevertheless, the challenges that the taxonomic complexity of this genus represents in terms of diagnosis together with the fact that within the same bacterial species, groups of strains may interact with distinct host species demonstrate that there is still a long way to go. In this review, we describe and discuss the current molecular-based methods for the diagnosis and detection of regulated Xanthomonas, taxonomic and diversity studies in Xanthomonas and genomic approaches for molecular diagnosis.

Quercetin is a flavonoid that is found in many plant materials, including commonly eaten fruits and vegetables. The compound is well known for its wide range of biological activities. In this study, 5-O-acyl derivatives of quercetin were synthesised and assessed for their antiproliferative activity against the HCT116 colon cancer and MDA-MB-231 breast cancer cell lines; and their radical scavenging activity against the 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical species. Four derivatives were found to have improved the antiproliferative activity compared to quercetin whilst retaining radical scavenging activity.

Objective: This study aimed to identify cases of leishmaniasis in the Nuba Mountain area, which is situated in a unique geographical site located in the centre of Sudanese leishmania belt. Wide range of investigations are available for detection of leishmania cases, but still the most reliable and easy test used as screening and epidemiological tool in field studies needs to be evaluated. The most commonly used conventional diagnostic methods direct microscopy and culture have some drawbacks in diagnosing subclinical cases of leishmaniasis. Materials and methods: In this study, comparative properties of various immune-diagnostic tools with Polymerase Chain Reaction used in sub-clinical leishmaniasis isolates were explored. The immune-diagnostic tools involved in this study include- Leishmanin Skin Test (LST), Enzyme Linked Immunosorbent Assay (ELISA) and Direct Agglutination Test (DAT). The study was conducted in the Green Valley village (Rashad Province, South Kordofan State) with a population of 332. Most of the villagers presented with sub-clinical form of leishmaniasis with minor symptoms and signs without the features found in clinical form of visceral leishmaniasis such as fever, diarrhoea, epistaxis, enlarged lymph nodes, spleen and liver. In this study we collected demographic, clinical and epidemiological data using special questionnaire. Leishmanin skin test (LST), ELISA, DAT and PCR for parasite DNA detection were used. Result: The final positive cases detected by PCR were 32 out of 332 belong to L. donovani species. The final positive cases detected by LST were 51.2% of the total population under study, while 11 out of the 37 tested samples were positive by ELISA. All of the 332 villagers showed negative readings by DAT with exception of three individuals who were positive with very high titers. Conclusion: DNA etxtraction and amplification with primers can be a good screening tool in subclinical leishmaniasis isolates. Objective:сonstructive principles development of early diagnosis of bone and cerebral metastases of peripheral non-small cell lung cancer (PNSCLC), is based on the targeted preventive and personalized application of radiological and nuclear diagnostic methods.Materials and methods.There was performed the retrospective analysis of medical cards of 1473 patients with peripheral non-small cell lung cancer for the evaluation of the characteristics and frequency of bones and brain metastases. The frequency and features of bone and brain metastases depending on clinical and morphological features of the primary lung tumor were studied. 118 patients with PNSCLC with bone and brain metastases, regardless of clinical symptoms, were examined using a wide range of modern radiological and nuclear methods.Conclusion:the original algorithm has been developed on the study of the diagnostic significance of medical imaging methods. The application of this algorithm will significantly improve the early diagnosis of bone and cerebral metastases and improve the staging of PNSCLC.
