ENHANCED TESTOSTERONE IN FEMALE PATIENTS WITH ANOREXIA NERVOSA: ITS NORMALIZATION AFTER WEIGHT GAIN

ABSTRACT In order to elucidate the mechanism of disturbances of gonadal hormones secretion in anorexia nervosa 14 female patients were investigated. A control group also consisted of 14 women of the same age. The serum LH, progesterone, oestrogens: oestrone + oestradiol (Oe1 + Oe2), oestriol (Oe3) and testosterone were determined by radioimmunological methods. In patients with anorexia nervosa the serum testosterone and Oe3 concentrations were dramatically elevated, whereas LH, progesterone and Oe1 + Oe2 were decreased as compared with the control group. Considerable weight gain induced by cyproheptadine treatment caused a normalization of the serum testosterone and Oe3 concentrations in all the patients. A negative correlation between the testosterone level and the deficit in body weight was observed. The mechanism causing the dramatically high serum testosterone concentration in the female patients with anorexia nervosa is discussed.

Abstract. In order to elucidate the mechanism of impaired LH secretion, 60 female patients with anorexia nervosa were investigated. A control group consisted of 14 women of the same age, examined in the follicular phase of the menstrual cycle. The serum LH, FSH, prolactin, TSH, testosterone, dihydrotestosterone, dehydroepiandrosterone, Δ 4-androstenedione, oestrone, oestradiol, oestriol, progesterone, thyroxine, triiodothyronine, reverse T3 and serum sex hormone binding globulin (SHBG) concentrations were measured. The results showed a significant increase in serum dehydroepiandrosterone, testosterone, oestriol and reverse T3 concentrations. However, oestrone, oestradiol, progesterone, SHBG and triiodothyronine levels were significantly lower than those of the control group. The mean serum LH concentration in patients with anorexia nervosa before and after LRH stimulation was significantly lower than that in the control group, but FSH secretion in response to LRH was normal. All hormonal changes in anorexia nervosa disappeared after weight gain during cyproheptadine treatment. Dramatically increased dehydroepiandrosterone levels suggest that the high testosterone in women with anorexia nervosa is derived from adrenal rather than from gonadal steroids. There was no correlation between serum testosterone, dehydroepiandrosterone, oestriol and LH concentrations indicating that steroid hormone disturbances do not cause impaired LH release in anorexia nervosa.

Abstract. The aim of this study was to evaluate the effect of the agonistic LRH analogue (D-Trp6-LRH) on LH, FSH and prolactin secretion, and its possible efficacy in female patients with anorexia nervosa. The study included 18 female patients with anorexia nervosa and 7 healthy women in the same age group. The patients with anorexia nervosa were divided into two groups of 9 women each. The first group consisted of patients in the weight loss phase of the disorder and the second of 9 women who had achieved normal body weight. Additionally, in 6 women with anorexia nervosa serum LH concentration was determined in response to native LRH before and 14 days after the treatment with D-Trp6-LRH. Serum LH, FSH, prolactin concentrations were determined before and 5, 10, 24 and 48 h after administration of D-Trp6-LRH in a dose of 5 μg. The serum LH response to D-Trp6-LRH in the second group did not differ from that of the control group. However, in women with anorexia nervosa in the weight loss phase, LH release in response to administration of the analogue was significantly lower. FSH release after LRH analogue administration in both groups of patients with anorexia nervosa did not differ from that in the control group. The increased LH secretion in response to native LRH after treatment indicates that D-Trp6-LRH in low doses did not inhibit pituitary responsiveness. A dose of 5 μg of D-Trp6-LRH was administered in a therapeutic regimen every 48 h from 1 to 3 months to 7 of 9 patients with anorexia nervosa who exhibited amenorrhoea in spite of normalization of body weight. All the patients treated with the analogue resumed menstruation. The results of this study suggest that D-Trp6-LRH may be of value in the treatment of amenorrhoea in women with anorexia nervosa.

The current study was presented to conclude the influence of different ratio of pomegranate peel powder (PP) in feed of Karadi male lambs, they divided to 4 groups (0% as a control, 1% treatment 1, 2% treatment 2 and 4% as treatment 3), on scrotal circumference (SC), scrotum height (SH), body weight (BW) and blood serum testosterone concentration (T). In this study, sixteen (16) of Karadi male lambs used with 24.30 ± 0.40 kg weight and with 4-5 months old, all lambs have been randomly divided equally into four (4) groups and individually penned in a box for during 63 days. Results in our study showed that SC was significantly different and recorded higher 17.875±0.54 cm in group that fed on 1% PP, also SH recorded higher and significant different in group fed on 1% PP compared with other groups 10.000±0.32 cm. Average blood serum testosterone reduces in all groups compared to control group that fed on 0% of PP (11.418±0.15 n.mol/L). Final body weight in our study had significantly different (P < 0.05) between groups, 1% PP recorded higher weight (34.9 ± 0.58) kg. Each average daily gain, total body gain and initial body weight had not significant different in all groups. Correlation coefficient between SC, SH, blood serum testosterone concentration and final BW computed and ranged between (-0.397 to + 0.914).

Phyllanthus niruri L. (Euphorbiaceae), known as "quebra-pedra" (Portuguese for "stonebreaker"), is an herb used for kidney disorders. In light of its frequent use by the population, the present study aimed to investigate the genotoxic, antigenotoxic and cytotoxic activities of a standardized P. niruri extract in bone marrow rats. Three groups of 12 animals were treated daily by gavage over a period of 30 days, with 50, 150 or 250 mg/kg of P. niruri extract aqueous solution. The control group (n = 12) received tap water. At the end of treatment (day 31), groups were divided into two minor subgroups (n=6/group) and received cyclophosphamide (50 mg/kg, i.p.) or saline 0.9% (i.p.). After 24 hours, we evaluated the frequency of micronucleated polychromatic erythrocytes for each animal (MNPCE) at 1000 PCE. Cytotoxicity was evaluated with the PCE/NCE ratio (NEC = normochromatic erythrocytes). General toxicity was assessed during treatment using the parameters of body weight gain, ration and water consumption. The dry extract did not provoke changes in body weight, weight gain, ration and water intake or changes in the frequency of MNPCE or cytotoxicity in bone marrow. We propose that the P. niruri extract used here showed no genotoxic, antigenotoxic and cytotoxic activities under the experimental conditions.

Abstract Anorexia nervosa (AN) results in gut dysbiosis, but whether the dysbiosis contributes to AN-specific pathologies such as poor weight gain and neuropsychiatric abnormalities remains unclear. To address this, germ-free mice were reconstituted with the microbiota of four patients with restricting-type AN (gAN mice) and four healthy control individuals (gHC mice). The effects of gut microbes on weight gain and behavioral characteristics were examined. Fecal microbial profiles in recipient gnotobiotic mice were clustered with those of the human donors. Compared with gHC mice, gAN mice showed a decrease in body weight gain, concomitant with reduced food intake. Food efficiency ratio (body weight gain/food intake) was also significantly lower in gAN mice than in gHC mice, suggesting that decreased appetite as well as the capacity to convert ingested food to unit of body substance may contribute to poor weight gain. Both anxiety-related behavior measured by open-field tests and compulsive behavior measured by a marble-burying test were increased only in gAN mice but not in gHC mice. Serotonin levels in the brain stem of gAN mice were lower than those in the brain stem of gHC mice. Moreover, the genus Bacteroides showed the highest correlation with the number of buried marbles among all genera identified. Administration of Bacteroides vulgatus reversed compulsive behavior but failed to exert any substantial effect on body weight. Collectively, these results indicate that AN-specific dysbiosis may contribute to both poor weight gain and mental disorders in patients with AN.

Poultry farming holds a special place in ensuring the products that the consumers demand, it provides the population with essential food products,such as eggs and meat,that contain vital micro and macronutrients, proteins, lipids, and vitamins. Therefore, the issues of rational, economically feasible feeding of meat poultry, namely broiler chickens, are an urgent task. It is also essential to find effective methods of their application in order to correct the natural resistance and immune and biological reactivity of birds. The purpose of this research is to study the effect of the biological stimulant-Nucleostim on the growth and development of chickens, hematological, and immunological parameters of the blood of birds.This Biostimulant is a purified bovine spleen extract containing at least 1 mg / ml of low molecular weight peptides (nucleotides and nucleosides) formed as a result of autolysis, using dry whey and diatomite as fillers. Onthe application ofNucleostim, the gain in live weight of chickens was increased by 9.7%. At the end of the experiment, the livability of the chicks of the experimental group treated with Nucleostimcame up to 88%, compared with the 72% of the control group. The use of biostimulant had a stimulating effect on the liver of chickens confirmed by the research results presented in the article, as well as contributed to the development of the thymus in the setting of general dystrophy. Thus, it improved chicklivability and increased body weight gain. The biological stimulant-Nucleostim as an adaptogenic, anabolic, and immunostimulatory agent is promising for finding new drugs that improve the health and productivity of poultry.

The objective of this study was to determine the effects of wood, cigarette and marijuana smoke on the reproductive health of tandoor occupants. A total of 100 male individuals were selected (50 control and 50 tandoor occupants). A standard questionnaire was designed regarding their age, economic status, marital status, fuel type, exposure time (per day), use of mask, addiction and reproductive health. Morning blood samples of 5 mL of the size were taken from all participants. Serums were obtained and analyzed for total serum testosterone concentration. Bio-check (USA) kit was used according to the manufacturer protocol and procedures for testosterone analysis. In control group the mean ± SEM of total serum testosterone was 671.9 ± 20.02 ng/dl where as in tandoor occupants it was 542.7 ± 16.40 ng/dl. There was a significant reduction (P**** < 0.0001) in total serum testosterone concentration in tandoor occupants as compared to control group. Reproductive health problems like, low libido, erection problems, infertility, decreased frequency for shaving and absent morning and nocturnal erection were common in tandoor occupants as compared to control group. Wood, cigarette and marijuana smoke negatively affects testosterone concentration and lowers it significantly. This reduced testosterone concentration then produces ill effects like low libido, erection problems, infertility and absent morning and nocturnal erection.

Abstract Background The aim of this study is to investigate whether the energy matrix values of the nonstarch polysaccharide- (NSP-) degrading enzymes, bioemulsifier (LYSOFORTE®), guanidinoacetic acid (CreAMINO®), or their combinations could be summed. The effects of these additives on the growth performance, carcass traits, and economic value of the broiler chicken diets were evaluated. A total of 525-one-day-old Ross chicks with an initial body weight of 42.96 ± 0.87 g were haphazardly allocated into seven groups with five replicates. The seven experimental treatments are as follows: (1) basal diet with no additives (breeder recommendation), which is the control group, (2) basal diet minus 100 kcal/kg supplemented with 0.02% NSP-degrading enzymes (NSP), (3) basal diet minus 50 kcal/kg supplemented with 0.025% emulsifier (LYSOFORTE®), (4) basal diet minus 50 kcal/kg supplemented with 0.06% guanidinoacetic acid (CreAMINO®), (5) basal diet minus 150 kcal/kg supplemented with a mixture of NSP and LYSOFORTE® (NSPL), (6) basal diet minus 100 kcal/kg supplemented with a mixture of NSP and CreAMINO® (NSPC), and (7) basal diet minus 200 kcal/kg supplemented with a mixture of NSP, LYSO, and CreAMINO® (NSPLC). The experiment lasted for 35 days. Results It was found that the final body weight, body weight gain, and relative growth rate were significantly higher in birds fed diets supplemented with NSPL, NSPC, CreAMINO, and LYSO with the reduced energy matrix value. The overall feed conversion ratio was significantly improved due to the supplementation of NSPC, CreAMINO, NSPL, and LYSO with the reduced energy matrix value compared to the control group. Moreover, no significant effect on the carcass criteria was observed by the different treatments. As a result of the dietary supplementation with NSPL, NSPC, CreAMINO®, and LYSO with the reduced energy matrix value, the net profit, total return, economic efficiency, and performance index were increased and the cost of feed per kg of body weight gain was decreased. Conclusion The energy matrix value of NSPL, NSPC, CreAMINO®, and LYSOFORTE could be established in the diets of broiler chickens to improve the growth performance and economic efficiency.

The efficacy of tulsi (Ocimum sactum) and neem (Azadirachta indica) leaves extract as a growth promoter were studied in broiler. A total of 40 day-old broiler chicks were purchased from Kazi hatchery and after three days of acclimatization the chicks were randomly divided into four groups (n=10). No vaccination schedule was practiced and no antibiotic was added in ration of group A, B, C, and D respectively. Group A served control without any supplements while group B, C and D were supplemented with combination of tulsi and neem extract @ 1 ml, 2ml and 3 ml/liter of drinking water. Live body weight gain was recorded weekly up to 6th weeks and hematological studies were performed at 21st and 42nd day of experiments. At the end of 42nd day of experiment final body weight of group A, B, C and D were 1561± 12.10 g, 1698± 12.87 g, 1608± 12.04 g and 1763± 13.28 g, respectively. The birds of group D utilized their feed more efficiently among the treatment groups (p<0.05). The net body weight gain were, 1533± 11.98 g, 1443± 11.32 g and 1588± 12.10 g in treated groups compared to control group (1393± 11.07 g ) and total net profit per broiler was 19.08, 40.10, 20.68 and 45.07 Taka , respectively. Hematological parameterss (TEC, PCV, Hb and ESR) were not significantly changed among the treated and control group suggesting no side effects of herbal extracts in broiler. This study was conducted under the Department of Physiology and Pharmacology to determine the effects of Neem and Nishyinda leaves supplementation on growth performance and GUT biosis in broilers. A total of 40 day old broiler chicks were divided into four groups A, B, C and D. Group A was considered as control, fed only with commercial broiler ration. Group B supplemented with formulation of 2 gm grinded Neem leaves, Group C with 2 gm grinded Nishyinda leaves and Group D with 1 gm grinded Neem leaves plus 1 gm grinded Nishyinda leaves per liter of water respectively. Observations were recorded for live body weight, weight gain, feed consumption, feed efficiency, hemato-biochemical parameters and GUT biosis specially the parasite of birds in six weeks. Body weights were increased significantly (p<0.05) in all treated groups in respect to the control and highest was recorded in combine Neem plus Nishyinda supplemented groups (Group D). No significant (p>0.05) differences were observed among the groups for PCV and TEC values. Hb content increased and ESR decreased significantly (p<0.05) with combine Neem plus Nishyinda supplemented group (Group D). The serum SGOT and SGPT levels were also decreased with the same kind of supplementation. In treatment group there was significantly decreased of parasitic eggs, but in case of control group no decrease of parasitic eggs. There was no significant pathological change in any internal organs of the broiler of treated groups. Best result was found in the group D. The present study reveals that combine supplementation of Neem plus Nishyinda gives better result over other groups in respect to body weight gain, feed conversion ratio, hemato-biochemical parameter and profitability without making any health hazard of the broilers.
