For today, the actual problem of veterinary medicine is the diagnosis of animal poisoning with organophosphorus pesticides, which are used as insecticides and acaricides, fungicides, herbicides, rodenticides, nematocides. One of the highly effective nonspecific organophosphorus insecticides is diazinon, falling into the body causing poisoning of various species of birds. The basis of the toxic action of diazinon is the ability to block the enzyme acetylcholinesterase. As a result, acetylcholine is accumulated in cholinergic synapses, excessive stimulation of the nerves and muscles is noted, and in the future, a violation of the normal passage of nerve impulses develops. Complex research was carried out of Domestic Turkeys (Meleagris gallopavo) in which spontaneous poisoning with diazinon was recorded. Biochemical examination of the serum was carried out using the biochemistry analyzer «Cobas Integra 400 plus». The activity of the serum acetylcholinesterase was determined by the Method of Hestrin, 1949. In the stabilized blood, the hemoglobin content was determined by the hemoglobin-cyanide methods, the number of red blood cells and leukocytes was counted in Gorjaev's count chamber and the leukogram - after microscopy of blood smears, painted according to the Romanovsky-Gimza methods. The breeding of birds was carried out using the Shores method. The organs were fixed in 10% neutral formalin, 96% ethanols and Carnoy’s solution. The histological sections were made using a sown and freezing microtome, stained hematoxylin and eosin. The histological study of the preparations and their photographing was performed using microcope Leica DM 2500, and camera Leica DFC 450 C., Application Suite Version 4.4.0. Clinically ill turkeys poisoned by diazinon showed oppression, tremor and paresis of wings. Later, the coordination of movements was disturbed: sick birds fell on their backs, marked swimming movements of the pelvic limbs, cramps and spastic contractions of the limb muscles. From the oral cavity was allocated a rare mucous mass. After acute attacks and spastic contractions of the muscles, muscle weakness developed. In the terminal periods of the disease, there were also spasms of various muscle groups, paralysis, difficulty and frequent breathing accompanied by wheezing, cyanosis of the skin. During the conducted biochemical examination of serum, a marked decrease in the level of acetylcholinesterase, an increase in the level of AST, cholesterol and uric acid was observed. In the pathoanatomical study, expressed discirculatory processes in the form of acute congestive hyperemia, stasis, perivascular edema and hemorrhages, as well as necrotic changes in cardiomyces, hepatocytes, nephrocytes and brain neurons were found. Quality positive reaction on diazinon was received as a result of the bird crop content research by the method of thin-layer chromatography paper manifestation of iodine bismuth.

The activity of the acetylcholinesterase of the serum of blood in the experimental birds was determined by the method of Hestrin. The section of birds that were killed was carried out by the method of Shore. Sections of organs were fixed in 10% neutral formalin, 96% ethanol and Carnoua liquid. The histo-cuttings were made by means of a stent microtome, stained with hematoxylin and eosin. The histological study of the preparations and their photographing was performed using a Leica DM-2500 (Switzerland) microscope, a Leica DFC450C camera and the Leica Application Suite Version 4.4 software. For the production of ultrathin sections, as well as for conducting electron microscopy, pieces of organs were fixed in Millonig’s buffer pH 7,36. Half-thin and ultrathin cuts were made on ultramicrotome LKB-2188 Ultrotome NOVA. The specimens were viewed and photographed in a Tesla BS-500 transmissive electron microscope, at an accelerating voltage of 60, 90 kV. In the pathoanatomical study, expressed discirculatory processes in the form of acute congestive hyperemia, stasis, perivascular edema and hemorrhages, as well as necrotic changes in cardiomyces, hepatocytes, nephrocytes and brain neurons. An electron microscopic study also found destruction of synapses in the cerebral cortex, cerebellum and medulla oblongata. When autopsy selected material (feed the masses crop of birds) for chemical-toxicological research. As a result the research the extract of selected content crop of birds by the method of thin-layer chromatography paper manifestation of iodine bismuth quality received positive reaction on the compound diazinon.

The article represents data on chemical properties and main aspects of the biological action of the diazinon - organophosphate pesticide that is widely used as a drug with marked acaricidal and insecticidal properties. Analyzed published data on metabolism, diazinon accumulation in various organs and systems, and ways of removing from the body. The article shows the median lethal dose (LD50) diazinon for various kinds of animals. The special features of the pathogenesis of poisoning, the main mechanism of action of toxic organophosphorus pesticides is the phosphorylation and inhibition of acetylcholinesterase, resulting in a marked accumulation of acetylcholine in the cholinergic synapses, excessive stimulation of nerves and muscles, disruption passage of nerve impulses. The article detailed the clinical signs of acute poisoning by organophosphate compounds. These data indicate that the effects on the body organophosphorus pesticides includes muscarinic effects (sialorrhea and excessive secretion of sweat glands, bronchorrhea, increased motility of the gastrointestinal tract, accompanied by spastic contractions of the intestinal wall, vomiting and gastroenteritis), nicotine action (miofibrillation, rigidity pectoral muscles, paralysis of respiratory muscles with the development of sudden hypoxemia) and central effects (arising from the impact of the central nervous system and are accompanied by violation of its function). The analysis pathmorphology of poisoning of the organophosphate pesticide. These data indicate that structural changes diazinon poisoning is less specific than clinical signs and characterized by: the development of the circulatory disorders (acute congestive hyperemia recorded and hemorrhage), the appearance of dystrophic and necrotic changes parenchymal elements of the brain and spinal cord, liver, kidneys and so on. Also recorded alterations changes ganglion cells and spinal cord autonomic ganglion, proliferation of glial cells, and for subacute and chronic poisoning – disintegration of the myelin and nerve fibers axial cylinder. Described diazinon influence on organs of the immune system and ability to induce endocrine disorders. There are published data on the potential mutagenic, carcinogenic and teratogenic effects of the diazinon. The range of problems researd structural changes by the origin and development of clinical evidence for the effect of different doses diazinon and certain aspects of forensic veterinary diagnostic poisoning mammals and birds of organophosphate pesticides.

В условиях непрерывного воздействия неблагоприятных факторов окружающей и производственной среды на человека особую актуальность приобретает изучение механизмов, поддерживающих гомеостаз организма. Длительное поступление фторидов в организм приводит к формированию хронической фтористой интоксикации, патогенез которой вызывает многочисленные споры и дискуссии. До сих пор недостаточно внимания уделяется изучению висцеральной патологии, обусловленной нарушениями иммунного статуса в условиях воздействия на организм соединений фтора. Практически отсутствуют исследования по изучению иммунной реактивности, определяющей морфофункциональный характер ответной реакции печени на ранних стадиях развития фтористой интоксикации. Цель работы - изучение действий патофизиологических механизмов иммунной реактивности печени при субхроническом действии на организм соединений фтора. Методика. Опыты проведены на 210 лабораторных крысах-самцах массой 180-220 г., разделенных на 2 группы: контрольную (n=80) и группу животных с субхроническим действием фторида натрия (n=130). Экспериментальные животные в течение 12 нед имели свободный доступ к водному раствору фторида натрия (концентрация 10 мг/л, что составляет суточную дозу фтора 1,2 мг/кг массы тела). Для изучения иммунологических и биохимических показателей забирали кровь из хвостовой вены через 1, 3, 6, 9, 12 нед от начала эксперимента. Для оценки состояния гуморального звена иммунитета определяли уровень сывороточных иммуноглобулинов (IgA, IgG, IgM) иммуноферментным анализом с помощью наборов реактивов ЗАО «Вектор-Бест» (Новосибирск). Уровень сывороточных цитокинов: TNF-α, IL-1β, 2, 4, 6, 10 определяли на анализаторе Multiskan EX методом иммуноферментного анализа с использованием наборов «Вектор Бест» (Новосибирск). Подсчет общего количества лейкоцитов произведен классическим способом в камере Горяева, анализ лейкоцитарной формулы - в окрашенных мазках периферической крови. Метаболические изменения оценивали по активности ферментов в ткани печени: щелочной фосфатазы (ЩФ), аланин- и аспартатаминотрансфераз (АЛТ, АСТ), лактатдегидрогеназы (ЛДГ), гаммаглутамилтранспептидазы (γ-ГТ). Активность ферментов определяли унифицированными методами с помощью наборов реактивов ЗАО «Вектор-Бест» (Новосибирск) на фотометре PM-750 (Германия). Гистологические исследования печени осуществляли после декапитации крыс, проводимой под эфирным наркозом. Результаты. Показано, что субхроническое воздействие фторида натрия сопровождается формированием внутриклеточных и внутрисосудистых повреждений печени. Активация медиаторов воспаления и развитие иммунологических нарушений в динамике эксперимента способствуют формированию системной воспалительной реакции, которая приводит к появлению стойких морфологических нарушений в печени и изменению активности ферментов основных метаболических путей. Заключение. Полученные результаты могут быть использованы при разработке и проведении профилактических мероприятий в условиях воздействия на организм высоких концентраций фтора с последовательным применением детоксикационной, иммуномодуляторной и органопротекторной коррекции. Studying mechanisms, which maintain the body homeostasis, is particularly important in the conditions of continuous impact of adverse environmental and manufacturing factors. Long-term exposure to fluorides leads to chronic fluoric intoxication, the pathogenesis of which is a subject of multiple controversy and discussions. Not enough attention is still paid to elucidating the visceral pathology associated with fluorine-induced immune disorders. There are virtually no studies of immune reactions that define the morphofunctional nature of the liver response to early stages of fluoric intoxication. Aim. To study pathophysiological mechanisms of hepatic immune reactivity in subchronic exposure of the body to fluorine compounds. Methods. Experiments were performed on 210 male rats weighing 180-220 g. The animals were divided into two groups: 1) control (n=80) and 2) subchronic exposure to sodium fluoride (n=130). The rats had free access to a 10 mg/l aqueous solution of sodium fluoride (daily dose, 1.2 mg/kg body weight) for 12 weeks. Blood was withdrawn from the caudal vein at 1, 3, 6, 9, and 12 weeks of the experiment for immunological and biochemical tests. Histological study of the liver was performed after decapitation of rats under ether anesthesia. Results. The subchronic exposure to sodium fluoride was associated with intracellular and intravascular damage of the liver. Activation of inflammatory mediators and development of immunological disorders during the experiment contributed to a systemic inflammatory reaction, which resulted in persistent morphological injuries of the liver and changes in enzyme activities in major metabolic pathways. Conclusion. The study results can be used for development and implementation of preventive measures against the effects of high fluorine concentrations, which would include a successive use of detoxification, immunomodulation and organ protection.

Introduction. The most susceptible to lead is the hematopoietic system of hematopoietic organs due to lead inhibition of heme and globin synthesis and cytotoxic effect on the membrane of Mature red blood cells. The aim of study was to evaluate the informative value of the study of erythrocyte and reticulocyte parameters determined on modern hematological analyzers in patients working in contact with lead during medical and biological monitoring. Materials and methods. 45 employees of the lead battery processing plant and 30 persons of control group were examined. The level of lead in the blood was determined by atomic absorption spectrometry, δ-ALA in the urine-by the reaction of pyrol formation with acetylacetone in terms of gram of creatinine, the study of hematological parameters was performed on a Sysmex HT-2000i analyzer. Statistical processing of the results was performed using the program STATISTICA 10.0. Results. Significant changes in erythrocytic (RDW) and reticulocytic (RET, IRF, LFR, MFR, HFR, RET-He) parameters, erythropoietin in workers in contact with lead compared to the control group, changes in MCV, MCH, RDW, RET indicators in the group working in dynamics after 2 years were revealed. Associations of hematological parameters with biomarkers of exposure and effect (lead level in blood and ALA in urine) were revealed. Conclusions. Assessment of erythrocyte (MCV, MCH, RDW) and reticulocyte parameters (RET% and their distribution by maturity) in dynamics during periodic medical examinations of workers in contact with lead allows us to detect the development of hematological disorders at early stages.

The objective of present study was to analyze and compare the nephroprotective effects of petroleum ether extracts of seeds of Nigella sativa and Silybum marianum in Swiss albino mice. Petroleum ether extracts of seeds of Nigella sativa and Silybum marianum were administered (200 mg kg–1) orally for 14 days following one dose of cisplatin injection (16 mg kg–1 i.p.) on 4th day. Cisplatin toxicity was identified in mice by loss of body weight, reduced triglycerides levels, elevated blood glucose, blood urea nitrogen and creatinine levels (P < 0.05). Silybum marianum and Nigella sativa significantly enhanced the body weight (9%) of diabetic rats. Silybum marianum had significantly reduced the elevated blood glucose (–106%), and BUN levels (–9%) as compared to the Nigella sativa administration. Silybum marianum extract also significantly elevated the levels of triglycerides (257%) which were much reduced after the cisplatin injection. Histological study showed that the cisplatin treated group demonstrated the necrosis signs in the glomerulus of Bowman’s capsule, hyalinization and severe cellular damage. The animals administered with extracts showed nearly normal kidney architecture, however, the kidney tissues of Silybum marianum-treated animals showed some improvement with slight signs of cellular damages.

A 13-month-old boy with hepatoblastoma and hemihypertrophy is presented; this is the fifth known case with this association. Histological study showed that hemihypertrophy was related to cellular hyperplasia but not to hypertrophy of individual cells. Karyotypes of leukocytes and of skin fibroblasts from both sides of the body were normal. The association of hemihypertrophy and hepatoblastoma stresses the dysontogenetic origin of this neoplasm and signals the possibility of this tumor in children with asymmetry. Our patient showed cystathioninuria, which was related to the high content of cystathionine in his tumor. The presence of cystathioninuria in 7 of 10 patients with hepatoblastoma so far examined, shows the usefulness of urinary amino acid screening in the diagnosis of this rare childhood neoplasm.

The paper presents the results of an experimental study of the dose-dependent nature of functional changes in the body systems under chronic administration of uranyl acetate dihydrate in doses of 0.5 and 5.0 mg/kg per element for 18 weeks. The study was performed on 45 male outbred rats. It has been shown that uranyl acetate dihydrate in a dose of 0.5 mg/kg had no significant effect on hematological parameters. At the same time, activation of bactericidal activity of neutrophils, a decrease in the immunoregulatory index, and an increase in the blood concentration of tumor necrosis factor (TNF-α) have been revealed. The toxicant administered to rats in a dose of 5 mg/kg led to a decrease in the absolute number of erythrocytes, hemoglobin, hematocrit, platelets, the release of myelocytes into the blood, basophilia, monocytosis, the appearance of leukolysis cells and plasmatization of lymphocytes. On the part of the immune system, an increase in the biocidal capacity of neutrophilic granulocytes, TNF-α production, an increase in the number of CD8+ cells, and a reduction in the CD4+/CD8+ ratio have been found. Uranyl acetate dihydrate had a dose-dependent effect only on the number of cytotoxic T-lymphocytes, T-cells with the CD4+CD8+ phenotype, on the immunoregulatory index, and on the level of TNF-α. Hyperglycemia and glucosuria were also dose-dependent. An increase in glucose in the blood and urine indicated a violation of carbohydrate metabolism and kidney function. There was a decrease in the concentration of thyroxine, testosterone and an increase in the level of insulin. Uranyl acetate dihydrate led to the development of insulin resistance. The level of hormones did not depend on the dose of the toxicant administered to the animals.

Objective: The study was aimed to evaluate glucose homeostatic and pancreas protective potential of Tecomella undulata root extract in streptozotocin induced diabetic rats.Methods: The ethanolic root extract was prepared by following standard soxhlation methods. The experimental design was divided in to control and treated groups for 28 days of comparative experimental schedule. The body and organ weights, serum biochemistry, histo-pathology, hematology and toxicity profiles were assayed by following standard methods and protocols.Results: The treatment of ethanolic extract of root of T. undulata was significantly (p ≤ 0.001) reduced glucose levels at 7day, 14day, 21day and 28 days in comparison to standard drug of metformin. Correspondingly, lipid profile i.e. total cholesterol, HDL, VLDL, LDL and triglyceride were also altered significantly. Whereas, body and organs weight and hematological parameters were not shown significant changes. Subsequently, toxicity profile i.e. hepatic and renal parameters were remained under normal ranges. Corresponding, the treatment of ethanolic root extract caused normalcy of histoarchitecture of pancreas in comparison to standard drugs.Conclusion: The results of study illustrated that Tecomella undulata root extract possessing particular kind of phytocompounds which caused glucose homeostatic and pancreas protective potential in diabetic rats.

Psoriatic arthritis (PsA) is a heterogeneous disease manifested by peripheral arthritis, dactylitis, spondylitis, and enthesitis. PsA is often undiagnosed by dermatovenerologists because of the difficulty in identifying a variety of clinical signs. The early diagnosis of PsA and the accurate assessment of all its symptoms are necessary for the timely choice of optimal therapy.Objective: to assess the detectability of clinical signs of PsA in patients with psoriasis in dermatological practice.Patients and methods. The investigation enrolled 103 patients (47 men and 56 women) (mean age, 44.0±13.7 years) with psoriasis (its mean duration, 10.7±10.2 years), the average prevalence and severity according to the Body Surface Area (BSA) and the Psoriasis Area and Severity Index (PASI) were 9.3±13.6% and 15.4±12.5 scores, respectively. All the patients completed the Psoriasis Epidemiology Screening Tool (mPEST) and were examined by a dermatovenerologist and a rheumatologist. The diagnosis of PsA was based on the Classification Criteria for Psoriatic Arthritis (CASPAR). The investigators evaluated arthritis, dactylitis, enthesitis, and inflammatory back pain (IBP) according to the rheumatological standards: IBP by the Assessment of SpondyloArthritis International Society (ASAS) criteria, and enthesitis by the Leeds Enthesitis Index (LEI).Results and discussion. Sixty-one (59.2%) of the 103 patients with psoriasis were found to have PsA on the basis of the CASPAR criteria and the rheumatologist's examination. The dermatovenerologist diagnosed arthritis in a significantly smaller number of cases than did the rheumatologist: in 15 (24.6%) and 35 (57.4%) of the 61 patients (p<0.001), respectively. The dermatovenerologist and the rheumatologist demonstrated no significant differences in their clinical evaluation of dactylitis: it was detected in 37 (60.7%) and 40 (65.6%) of the 61 patients, respectively (p=0.32). Based on patient complaints and mPEST findings, the dermatovenerologist recorded pain in the calcaneal region in 32 (52.5%) patients. The rheumatologist identified ulnar, knee, and calcaneus enthesitis in 11 (18%), 8 (13.1%), and 25 (41%) patients, respectively. Based on complaints and mPEST findings, the dermatovenerologist detected back pain in 30 (49.2%) of the 61 patients. The rheumatologist diagnosed IBP in 21 (70%) of these 30 patients and mechanical back pain in 9 (30%). Thus, IBP was noted in 34.4% of PsA patients. Tendonitis was undiagnosed by the dermatovenerologist; the rheumatologist identified wrist tendonitis in 13 (21.3%) of the 61 patients with PsA.Conclusion. Dermatovenerologists frequently underestimate damage to the spine and entheses in patients with psoriasis. The introduction of the ASAS criteria for IBP and methods for assessing enthesitis in dermatological practice can improve the early diagnosis of axial lesion in PsA in patients with psoriasis.

Preeclampsia (PE) is a turmoil in the pregnancy appeared with the onset of hypertension and considerable amount of proteinuria. Extra serum iron is a causative component of oxidative stress concerned in the pathogenesis of preeclampsia. This study is a case control, conducted in 2018, and aimed to determine the iron status in preeclamptics as compared to normotensive pregnancies in Gaza strip. About 100 pregnant women with gestational age between 26 to 36 weeks. Fifty of them were preeclamptics and an equal number were without preeclampsia, aged between 18 to 35 years. Interview questionnaires were used to take sociodemographic and clinical data. Anthropometric evaluation and biochemical analysis were conducted. The SPSS version22 was used for data analysis. There was no statistically difference in the gestational age, gestational number and hemoglobin levels in the cases and controls (p≥ 0.05). In contrast, the body mass index (BMI), systolic, diastolic blood pressure (BP), and uric acid (UA) levels were significantly higher in preeclamptics (P<0.001). As well, the majority of cases were have +2 proteinuria on dipstick testing. Further, serum iron and ferritin levels were significantly higher in preeclamptics. On the other hand, ferritin levels had significant direct correlations with gestational number, previous preeclampsia, BMI, systolic BP, diastolic BP, UA, and proteinuria (P<0.05). Likewise, Iron had significant direct correlation with proteinuria (P<0.05). Preeclamptics have higher hematological parameters levels (iron & ferritin) as compared to normotensive women. However, Iron status of preeclamptic women should be assessed before giving iron supplements as these may cause more harm than benefit.
