Kandungan aluminium yang tinggi pada tanah Ultisol merupakan salah satu masalah utama dalam budidaya tanaman kedelai mulai dari fase perkecambahan hingga pertumbuhan tanaman. Perlakuan priming pada benih diyakini mampu memperbaiki perkecambahan pada lingkungan yang kurang menguntungkan. Penelitian dilakukan untuk mengetahui pengaruh priming pada perkecambahan benih kedelai yang dikecambahkan dalam media masam dan mengandung aluminium. Penelitian disusun dalam Rancangan Acak Lengkap (RAL) pola faktorial. Faktor pertama adalah jenis priming : tanpa priming, H2O (air), KNO3 1%, KNO3 2%, GA3 50 ppm, GA3 100 ppm, dan PEG 6000 7,5%. Faktor kedua adalah varietas kedelai yaitu: Anjasmoro, Burangrang, dan Grobogan. Data dianalisis sidik ragamnya dan dilanjutkan dengan uji Beda Nyata Jujur (BNJ) 5% menggunakan program statistika R Studio. Hasil penelitian menunjukkan priming PEG 7,5% dan varietas Burangrang secara mandiri merupakan perlakuan terbaik untuk meningkatkan vigor benih kedelai berdasarkan variabel waktu muncul kecambah, kecepatan perkecambahan, dan panjang hipokotil. Implikasi penelitian ini adalah PEG 7,5% dapat digunakan untuk perbaikan perkecambahan pada tanah masam dengan kandungan aluminium yang relatif tinggi. High aluminium content in Ultisol is the main problem in soybean cultivation from germination to planting growth phases.Priming treatment is believed to improve seed germination in a poor-growing environment. This study aimed to determine the effect of priming of soybean seeds germinated under acid medium and aluminium stress. The experimental design used was Completely Randomized Design (CRD) with factorial pattern. The first factor was priming type : no priming, H2O (water), 1% KNO3, 2% KNO3, 50 ppm GA3, 100 ppm GA3, and 7.5% PEG 6000. The second factor was soybean variety : Anjasmoro, Burangrang, and Grobogan.  The data obtained were analyzed using the R Studio statistic program with a 5% HSD test. The study resulted that each PEG 7.5% priming and Burangrang  variety independently was the best in increasing soybean seed vigor based on sprout time emergence, germination speed and hypocotyl length. This research implicates that PEG 7.5% can be used to improve seed germination on acid soils with high aluminium content.

Aim: This study was conducted to develop liquid enhancer containing KCl, TU, GA, and SA for germination of drought-stressed Oryza sativa subsp. indica cv. MR284 seed. Study Design: All experiments were conducted in a completely randomized design. Two steps were involved in the development process which are to select an ideal concentration for each KCl, TU, GA, and SA, and to find an ideal combination of chemicals from the selection of ideal concentrations acquired in step 1 to form liquid enhancer. There were 20 treatments for step 1 and 9 treatments for step 2. All of these treatments with 6 replicates. Place and Duration of Study: Department of Biology, Faculty of Science, University Putra Malaysia, between June 2018 and December 2018. Methodology: The sterilized rice seed cv. MR284 was stressed in the -1.2 Mpa PEG 6000 solution for three days and germinated in the KCl, TU, GA, and SA solution in a series of concentration for 10 days, in a controlled room. Seed germination was observed daily. Results: In the first step, drought-stressed rice seed showed the best germination performance in the 30 mM of KCl, 2.0 mM of TU, 0.24 mM GA, and 0.5 mM SA. Meanwhile, in the second step, the drought-stressed rice seed showed the best germination performance in the combination of 30 mM KCl + 2.0 mM TU + 0.24 mM GA + 0.5 mM SA. The best germination performance was evaluated by the highest germination percentage (%), germination index, seed vigor, leaf length, root length and biomass. Conclusion: Therefore, the combination treatments of 30 mM KCl + 2.0 mM TU + 0.5 mM SA was found to be the most effective and simplest liquid enhancer formula that has an ability to enhance seed germination of drought-stressed rice cv. MR284 seed.

The suitable establishment of a crop depends on seed quality, among another factors. However, with high production costs many producers use uncertified seeds, to reduce expenses with this input at sowing time. The objective was to evaluate germination, vigor and health of soybean seeds, diseases incidence in cvs. NS 5445 IPRO and BMX Ativa RR, whose seeds were of certified and uncertified origins, as well as yield components and grain yield of soybeans, with or without fungicides application. The experiments were conducted in the 2015/16 and 2016/17 crop season, in Erechim-RS. Two experiments were carried out in a completely randomized design (DIC): one in the laboratory, under a 2 x 2 factorial scheme (cultivar x origin); and another in the field in a homogeneous area, in a 2 x 2 factorial scheme (with/without fungicide application and certified/uncertified seed) for two cultivars, both with four replications. The variables evaluated were: germination and seed health, yield (kg ha-1), thousand grains weight (g), number of grains per plant, and incidence of foliar fungal diseases. The main fungi identified in seeds were Aspergillus sp., Penicillium sp. and Fusarium sp. The highest incidence percentage were obtained in uncertifed seeds, at two harvests. Both cultivars and origins presented the minimum germination required for commercialization, however, certified seeds had better performance for seed vigor. As for normal seedlings, there was a significant difference only for the 2016/17 crop season, with the highest percentage obtained in certified seeds (52.0% for NS 5445 and 73.5% for BMX Ativa). Best productivity was achieved with cultivation of certified seeds associated with fungicides in both crops and cultivars. For thousand grains weight (TGW) there was no difference in origin, but only for cultivar and fungicides apply. The number of grains per plant was higher in plants from certified seeds and that received fungicides, being cv. BMX Ativa the most responsive for the two harvests analyzed. The main diseases found in the two harvests were: Asian rust, powdery mildew, mildew, septoriosis and cercosporiosis. However, seed origin, in both crops, did not differ in the incidence of Asian rust, powdery mildew and mildew, but for septoriosis and cercosporiosis, considered soybean late season diseases, seed origin is a determining fator.

Setting storage space temperature and time saving becomes an important application in supporting the development of plantations to provide seeds as needed and sustainable. The purpose of this research is to delay the rapid of seed germination in Magnolia champaca by assessing the effect of various suspend temperatures and times saving on seed germination of Magnolia camphaca . The research was conducted in Plant Physiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University in February until June 2016. The method was used completely randomized design pattern Factorial by 3 factors with 12 treatments and 3 replications with the temperature: temperature ± 31°C, temperature ± 25 °C, temperature ± 18 °C, temperature ± 4 °C and the retention of time 0 week, 2 weeks , 4 weeks and 6 weeks. The parameters include the observation of germination rate, germination, vigor index and sprout morphology. The results of the research the treatment of interaction of temperature and time saving significantly affected on germination rate, germination and seed vigor index of Magnolia champaca. Germination speed, germination and vigor index were both produced by the treatment of storage at a temperature of ± 18 °C and a temperature of ± 25 °C at all times to keep (2 weeks, 4 weeks, 6 weeks) compared to control treatment. A normal sprout produced in temperature treatment ± 18 °C and ± 25 °C temperature ranges between 90-93%. An abnormal sprout produced in control treatment (0 week) by 25%. Measurement of moisture content and physiological maturity level of Magnolia champaca seed needs to be done before it is stored and germinated to test the viability of seeds of sprouts.

ABSTRACT: The differentiation of seedling growth and development is the result of the vigor level of the seed lot. Because of this, knowing the seedling growth curve is essential for the evaluation of the seedling length test, to be carried out at the moment when any differences in vigor really manifest, thus being able to reduce the time of evaluation of the test. In this research, the objective was to determine the most efficient time interval to perform the seedling length test in soybean seeds, depending on the vigor levels. The evaluated characteristics were percentage of seed germination, first germination count, emergency, days for emergence, aerial, root and total length (evaluated every 12 hours), aerial, root and total dry mass in completely randomized design. High vigor seeds originated seedlings with higher growth rates (cm. h-1), with vigor being one of the factors that interfere in the seedling length test. After 96 hours of the seedling length test, there was a statistical differentiation between the three vigor levels tested. The soybean seedling length test between lots, with different vigor, can be performed 96 hours after sowing.

Purple seed stain disease, caused by (Cercospora kukuchii), is a major concern in soybean (Glycine max (L.)) in Mississippi, USA, due to its effects on seed quality, reducing soybean seed grade and potential market price at elevators. Therefore, investigating the effects of purple seed stain (PSS) on seed quality (germination and vigor) and seed composition (nutrition) is critical. The objective of this study was to investigate the effects of PSS on seed harvest index, seed germination, seed vigor, and seed composition components (protein, oil, fatty acids, and sugars). A field experiment was initiated in 2019 in Stoneville, MS, at the Delta Research and Extension Center (DREC) on a Commerce silt loam soil (fine-silty, mixed, superactive, nonacid, thermic Fluventic Epiaquepts). Soybean variety Credenz 4748 LL was used. The results showed that infected (symptomatic) seed had a 5.5% greater Seed Index (based on 100 seed weight) when compared to non-infected (non-symptomatic, as control) seed. Non-infected seed had greater percent germination and seedling vigor when compared to infected seed. Germination was 30.9% greater and vigor was 58.3% greater in non-infected seed. Also, the results showed that infected seed with PSS had higher protein content and some amino acids. No changes in total oil and fatty acids. Sucrose and stachyose were lower in infected seed than in non-infected seed. The research showed that PSS impacted seed health and seed quality (germination and vigor) and seed composition (protein, sugars, and some amino acids). Purple stained seed should be avoided when planting and should be managed properly as low germination is a potential risk. Planting population should be adjusted accordingly due to lack of germination and vigor if PSS is present. This research help growers for purple seed management, and scientists to further understand the potential negative impact on seed quality and nutrition. Further research is needed before conclusive recommendations are made.

<p>This study was conducted to determine seed germination on the mechanisms of salinity stress in <em>B. rapa </em>var. <em>parachinensis </em>(Hong kong choy sum). The type of experiment was a completely randomized design with three salinity treatment NaCl, MgCl2, and KCl and minimum four replications was used. Salinity treatment consisted of 0 (control), 50, 100, 150, 200, mM concentrations. The experiment was carried out under 70% relative humidity at 23°C with a photoperiod of 8:16 h light/dark. The germinated seeds were counted every day and germination percentage, seed vigor, seed tolerance, length of seedling and biomass seedling was measured. The data were analyzed by one-way ANOVA multiple comparison test followed by Duncan’s multiple range test.</p>

Germination is considered a critical step in the seedling production process aiming the recovery of environments. Thus, understanding the germination behavior of native species with potential for economic use contributes to the development of the forest seed and seedling chain. Considering the limited information on the tree species Pouteria torta subesp. torta, this study aimed to evaluate its seed germination and formation of normal seedlings using different temperatures and substrates. The experiment was conducted in germination chamber in a research laboratory using two substrates (paper and vermiculite) and six temperatures (20, 25, 30, 35, 20-30 and 25-35 °C) in a completely randomized design in a 2x6 factorial arrangement. Daily evaluation was conducted to verify primary root emission, seed vigor, and seedling formation. The results indicated that the primary root protrusion process began after 20 days of testing under the best conditions, with germination time lasting longer than two months. The results obtained also showed that there was interaction between factors, with use of the vermiculite substrate and the temperatures of 35 and 25-35 °C producing the best germination rates and germination speed index, and use of the paper substrate and the temperatures of 30 and 35 °C showing the best mean germination time. Considering the analyzed variables (germination, germination speed index, and formation of normal seedlings), the temperatures of 35 and 25-35 °C and the vermiculite substrate are the most suitable for the production of Pouteria torta subesp. torta seedlings.

Increased productivity of cocoa needs high quality of cocoa seeds which are generally provided by certified seed gardens located far from smallholders farm, where seed delivery takes long time and may reduce physiological quality of seeds. One effort to maintain the seed quality is by treatment the seeds with polyethylene glycol6000 (PEG 6000). This study was aimed to study the interaction of PEG 6000 concentration and storage period on cocoa seed quality, and to determine the best concentration of PEG 6000 and storage period to maintain cocoa seed quality. The research was conducted in seed storage room, seed laboratory and green houseof PPPPTK, Cianjur. The experimental design used was a factorial completely randomized design and each combination treatment repeated three times. The first factor was concentration of PEG 6000 of 0%, 20%, 40%, and 60%, and the second factor was the storage period of 3 weeks, 6 weeks, 9 weeks, and 12 weeks. The results showed that concentration of PEG 60% with three weeks storage period was able to minimize the number of moldy seeds. The concentration of PEG 20%, 40%, and 60% were able to prevent seed germination in storage until six weeks. Concentration of PEG 6000 20% was able to maintain moisture content, seed germination, and germination rate.Key words:cocoa seed, storage, recalcitrant, PEG

Eighty aluminium-accumulating species were detected among 1324 species tested from the Australian–New Guinea flora. Accumulation was most strongly developed in certain dicotyledons (69 spp.) and Filicales (11 spp.). A high aluminium content of the organs of accumulating plants appears to be associated with normal metabolism. Recorded accumulators are mainly restricted to what are usually regarded as the more primitive groups in dicotyledons and Filicales, suggesting that, accumulation is a physiological relic in these groups. Obligate accumulators are confined to leached acid soils from a variety of parent materials, in comparatively high-rainfall areas. The re-deposition of aluminium in vegetable debris beneath Australian rain-forest communities dominated by accumulators (e.g. Ceratopetalum apetalum D. Don.) may be an important ecological and pedogenic factor. The significance of accumulation is discussed along biochemical, ecological, and taxonomic lines. There is evidence that accumulation is an adjunct of specialization (i.e. relatively narrow physiological tolerances), chiefly among tree species able to exploit impoverished soils in relatively primitive, generally tropical, mesic habitats. Accumulation seems a useful special character to supplement other data in the clarification of some taxonomic problems.

Abstract High-vigor soybean seeds can result in the rapid and uniform emergence of seeds under a wide range of field conditions. We aimed to explore the quantitative trait loci of seed vigor traits and superior allelic and carrier materials from the soybean in northeast China. A total of 257 improved lines and 104 soybean landraces were used to evaluate seed vigor, using normal seedling fresh weight, normal seedling length, main root length, hypocotyl length, and germination percentage, in 2018 and 2019. In this study, the different degrees of LD was detected not only among syntenic markers but also among nonsyntenic ones, suggesting that there had been historical recombination among linkage groups. A total of 19 and 10 simple sequence repeat (SSR) markers associated with seed vigor were detected in improved lines and soybean landraces in 2018 and 2019, respectively. Our results also showed five SSR markers co-associated with two or three vigor traits. In addition, 41 superior alleles and their carrier materials were mined. Based on the elite alleles detected, the best cross combinations for improving seed vigor traits were proposed. The results demonstrate that association mapping is a valuable foundation for molecular breeding for soybean seed vigor.
