ABSTRACT: Haemonchus contortus is the major gastrointestinal parasite of sheep raised in tropical and subtropical areas worldwide. This trial aimed to evaluate the influence of H. contortus infection on the bone marrow response of Santa Ines (SI) and Ile de France (IF) suckling lambs experimentally infected with H. contortus. Fourteen SI lambs and 12 IF lambs were randomized in four groups: infected SI (n=8), non-infected SI (n=6), infected IF (n=8) and non-infected IF (n=4). Lambs of infected groups were submitted to 27 infections, conducted every two days, from 14 to 68 days of age, and each lamb received a total of 5400 H. contortus infective larvae (L3). Ten blood samples were obtained during the experimental period to be used for erythrocyte and leukocyte counts, packed cell volume and total plasma protein estimation. Additionally, it was carried out a differential leukocyte count. Lambs from control groups did not shed eggs in faeces all over the experiment, while infected Santa Ines and Ile de France lambs presented means of 2963 EPG (Eggs Per Gram of faeces) and 8175 EPG in the last sampling (P<0.05), respectively. Infected Santa Ines lambs had an increase in eosinophil release, however differences (P<0.05) on circulation number in comparison with infected Ile de France lambs were identified only in the last sampling (54 days post first infection). The mild H. cortortus infection did not produce significant changes in the blood variables of the Ile de France and Santa Ines suckling lambs.

The objective of the study was to determine the parasitological and immune response in horses naturally re-infected with gastrointestinal nematodes (GIN) after an- thelmintic treatment in the humid tropic of Mexico. The study was conducted in Tenosique, Tabasco, Mexico. A total of 30 horses were sampled monthly for nine months. Fecal samples were obtained to determine the number of nematode eggs per gram (EPG) of feces. Stool cultures were performed to obtain and identify infective larvae (L3) genera. Blood samples were also obtained to determine the packed cell volume (PCV), the total plasma protein (TPP), and the differential leukocyte count (DLC). Serum enzyme-linked immunosorbent assays (ELISA) were performed to determine IgA levels against adult cyathostomins and Strongylus nematode antigen. The analyses considered the age and gender of the horses, and the season (Cold, rainy and dry). The number of EPG and TPP were higher in females (1224 ± 1269 and 7.6 ± 0.6 gdL−1, respectively) than males (623 ± 671 and 7.4 ± 0.5 gdL−1, respectively), and no differences were observed with animal age (p > 0.05). The number of EPG increased in the rainy season. The main nematodes involved were of the subfamily Cyathostominae (97.4%) and to a lesser degree of species Strongylus equinus, S. edentatus, S. vulgaris, and Oxyuris equii. The season affected the leukocytes, neutrophils, and lymphocytes counts but not eosinophils. Variables such as gender, animal age, and environmental conditions are important elements for a diagnosis of GIN and the application of treatments for its control.

AbstrakPenyakit paru obstruktif kronik (PPOK) ditandai oleh adanya hambatan aliran udara yang irreversibel dan bersifat progresif. Asap rokok, polusi udara, dan infeksi berulang pada saluran napas akan mengaktivasi makrofag alveolus dan melepaskan mediator inflamasi yang merangsang progenitor granulositik dan monositik di sumsum tulang sehingga mempengaruhi hitung jenis leukosit pada darah tepi. Faktor lain yang dapat mempengaruhi hitung jenis leukosit pada pasien PPOK adalah adanya penyakit penyerta. Desain penelitian ini adalah retrospektif deskriptif terhadap data rekam medik 69 orang yang dirawat di bagian paru dan penyakit dalam RSUP dr. M. Djamil Padang. Hasil penelitian menunjukkan pasien PPOK tanpa penyakit penyerta (n=9) memiliki nilai rata-rata hitung jenis basofil 0±0%, eosinofil 1,22±1,2%, neutrofil batang 3,33±2,5%, neutrofil segmen 79,56±9,26%, limfosit 13,67±6,55%, dan monosit 2,22±2,44%. Pada pasien PPOK dengan penyakit penyerta infeksi (n=41) didapatkan nilai rata-rata hitung jenis basofil 0±0%, eosinofil 1,02±1,59%, neutrofil batang 1,98±2,63%, neutrofil segmen 81,07±8,44%, limfosit 12,83±6,68%, dan monosit 3,1±2,71%. Pada pasien PPOK dengan penyakit penyerta non infeksi (n=19) didapatkan nilai rata-rata hitung jenis basofil 0±0%, eosinofil 2,16±5,65%, neutrofil batang 2,16±1,77%, neutrofil segmen 79,0±10,44%, limfosit 14,16±8,03%, dan monosit 2,53±1,87%. Penelitian ini memperlihatkan pasien PPOK tanpa penyakit penyerta, dengan penyakit penyerta infeksi, dan dengan penyakit penyerta non infeksi mengalami neutrofilia dan limfositopenia.Kata kunci: PPOK, hitung jenis leukositAbstractChronic Obstructive Pulmonary Disease (COPD) characterized by airflow obstruction that is irreversible and progressive. Cigarette smoke, air pollution, and recurrent infections in the respiratory tract can activates alveolar macrophages to release inflammatory mediators that stimulate granulocytic and monocytic progenitors in the bone marrow that can affect leukocyte counts in peripheral blood. Other factors that can also affect leukocyte count in COPD patients is the presence of comorbidities. The design of this study was descriptive retrospective from medical record of 69 people with COPD who were treated at the lungs and internal medicine department of dr. M. Djamil Hospital Padang. The result of this study show in COPD patients without comorbidities (n=9) average value of basophil counts 0±0%, eosinophils 1.22±1.2%, neutrophils rod 3.33±2.5%, neutrophils segment 79.56±9.26%, lymphocytes 13.67±6.55%, and monocytes 2.22±2.44%. COPD patients with infectious comorbidities (n=41) obtained average value of basophil counts 0±0%, eosinophils 1.02±1.59%, neutrophils rod 1.98±2.63%, neutrophils segment 81.07±8.44%, lymphocytes 12.83±6.68%, and monocytes 3.1±2.71%. In COPD patients with non-infectious comorbidities (n=19) obtained average value of basophil counts 0±0%, eosinophils 2.16±5.65%, neutrophils rod 2.16±1.77%, neutrophils segment 79.0±10.44%, lymphocytes 14.16±8.03%, and monocytes 2.53±1.87%. This study shows that COPD patients without comorbidities, with infectious, and with non-infectious comorbidities obtained neutrophilia and lymphocytopenia.Keywords:COPD, differential leukocyte count

Objective: Leukocytes contribute to the development of abdominal aortic aneurysm (AAA). We evaluated whether associations of differential leukocyte counts with AAA persist after accounting for traditional risk factors of AAA. Approach and Results: Among 11 217 adults from the Atherosclerosis Risk in Communities Study, we evaluated associations of differential leukocyte counts at baseline (1987–1989) with incident AAAs over a median follow-up of 22.5 years, using Cox proportional hazards regression. Each differential leukocyte count was categorized into 5 groups—below normal, tertiles within the normal range, and above normal, with the first tertile serving as the referent. We identified 377 incident AAAs through 2011, using hospital discharge diagnoses, linked Medicare records, or death certificates. At baseline, higher neutrophil, monocyte, and eosinophil counts were associated with higher risk of AAA, independent of smoking, other differential leukocyte counts, and other traditional risk factors. The association with incident AAA was the strongest for above normal neutrophil count, with an adjusted hazard ratio (95% CI) of 2.17 (1.29–3.64). Below normal neutrophil, lymphocyte, eosinophil and basophil counts were associated with higher risk of AAA with adjusted hazard ratio (95% CI) between 1.86 (1.04–3.35) and 1.62 (1.10–2.39). Conclusions: Higher neutrophil, monocyte, and eosinophil counts in midlife are associated with higher risk of AAA, even after accounting for traditional risk factors such as smoking, obesity, and atherosclerosis. This suggests the need to identify nontraditional risk factors and treatment strategies to mitigate the residual risk of AAA conferred by midlife inflammation. Whether immunosuppression is associated with higher risk of AAA needs further investigation.

Abstract Background and objectives Among placental mammals, females undergo immunological shifts during pregnancy to accommodate the fetus (i.e. fetal tolerance). Fetal tolerance has primarily been characterized within post-industrial populations experiencing evolutionarily novel conditions (e.g. reduced pathogen exposure), which may shape maternal response to fetal antigens. This study investigates how ecological conditions affect maternal immune status during pregnancy by comparing the direction and magnitude of immunological changes associated with each trimester among the Tsimane (a subsistence population subjected to high pathogen load) and women in the USA. Methodology Data from the Tsimane Health and Life History Project (N = 935) and the National Health and Nutrition Examination Survey (N = 1395) were used to estimate population-specific effects of trimester on differential leukocyte count and C-reactive protein (CRP), a marker of systemic inflammation. Results In both populations, pregnancy was associated with increased neutrophil prevalence, reduced lymphocyte and eosinophil count and elevated CRP. Compared to their US counterparts, pregnant Tsimane women exhibited elevated lymphocyte and eosinophil counts, fewer neutrophils and monocytes and lower CRP. Total leukocyte count remained high and unchanged among pregnant Tsimane women while pregnant US women exhibited substantially elevated counts, resulting in overlapping leukocyte prevalence among all third-trimester individuals. Conclusions and implications Our findings indicate that ecological conditions shape non-pregnant immune baselines and the magnitude of immunological shifts during pregnancy via developmental constraints and current trade-offs. Future research should investigate how such flexibility impacts maternal health and disease susceptibility, particularly the degree to which chronic pathogen exposure might dampen inflammatory response to fetal antigens. Lay Summary This study compares immunological changes associated with pregnancy between the Tsimane (an Amazonian subsistence population) and individuals in the USA. Results suggest that while pregnancy enhances non-specific defenses and dampens both antigen-specific immunity and parasite/allergy response, ecological conditions strongly influence immune baselines and the magnitude of shifts during gestation.

Abstract 1. Thirty-seven normal subjects were given subcutaneous injections of epinephrine, ranging from 0.25 to 0.5 mg., and the effects on the leukocytes were noted. 2. The neutrophils rose steadily for the three and one-half hours during which blood counts were made. The small lymphocytes rose in the first half hour, then fell below normal and finally returned towards normal. The eosinophils rose at first and then fell below normal for the remainder of the period. 3. The three doses of epinephrine used produced effects which differed quantitatively but not qualitatively. The most readily identified effect of the smallest dose was the one-half hour rise in lymphocytes or the one-half hour rise in total count. A dose of 0.5 mg. is satisfactory for work of this kind. 4. Subjects with a history of allergy showed a greater tendency than the remainder to exhibit a one-half hour rise in the eosinophil count. 5. The changes in the leukocyte count produced by epinephrine are similar to, but not identical with, those produced by adrenocortical hormone or adrenocorticotropic hormone.

The maternal and developmental toxicity of combined exposure to restraint stress and caffeine was assessed in mice. On gestational Days 0–18, three groups of plug-positive females (n = 13–15) were given by gavage caffeine at 30, 60, and 120 mg/kg/day. Three additional groups received the same caffeine doses and were restrained for 2 hr/day. Control groups included restrained and unrestrained plug-positive mice not exposed to caffeine. All animals in the group concurrently exposed to 120 mg/kg/day of caffeine and restraint died during the experimental period. In the remaining groups, cesarean sections were performed on Day 18 of gestation, and the fetuses were weighed and examined for external, internal, and skeletal malformations and variations. Although maternal and embryo/fetal toxicity were observed at all caffeine doses, the adverse maternal and developmental effects were significantly enhanced in the groups concurrently exposed to caffeine and restraint. It was especially remarkable at 60 and 120 mg/kg/day. The results of this study suggest that maternal and developmental toxic effects might occur if high amounts of caffeine were consumed by women under a notable stress during pregnancy.

ABSTRACT Hematology has become important for making clinical diagnoses in snakes because maintenance techniques in captivity have been improving and increasing their life expectancy. The emergence of diseases in captivity requires the creation of parameters to understand the species in their healthy state. The possible association between the onset of neoplasia, immunosuppression, and viral infection are also important factors to consider. Thus, hematology is a fundamental tool for observing the responses of animals to diseases and treatments. The present study aims to study hematology between two species of snakes and compare the profiles observed with published results. Blood samples were collected from 16 male and female snakes: seven Corallus hortulanus from the family Boidae and nine Python bivittatus from the family Pythonidae belonging to the Biological Museum of the Butantan Institute, São Paulo, Brazil. Complete blood count results including blood smear, total erythrocyte count, leukocytes, and differential leukocyte count were evaluated. The data obtained in this study could help with the diagnosis, the treatment of snakes in captive conditions and in of nature conservation programs.

Hemograms and acute-phase proteins in adult male New Zealand White rabbits that had been experimentally infected orally with sporulated oocysts of Eimeria stiedai were evaluated over a 28-day period. Fifty animals were used, divided into two groups: group A infected with 1 × 10(4) sporulated oocysts of E. stiedai and group B inoculated with distilled water. On the seventh day after infection, the infected animals presented anemia and leukocytosis with neutrophilia and monocytosis. Protein fractionation by means of electrophoresis identified 19 acute-phase proteins with molecular weights ranging from 24 to 238 kD. Ceruloplasmin, transferrin and haptoglobin showed high levels on the seventh day after infection, with gradual increases in their concentrations until the end of the experimental period. Thus, from the data of the present study, E. stiedai is considered to be a pyogenic etiological agent for which the infection level can be monitored through the leukocyte count and serum concentrations of ceruloplasmin, transferrin and haptoglobin, and these can be recommended as complementary tests.

Evaluation of the effect of hydroalcoholic extracts of Cassia occidentalis leaves in neutrophil adhesion test in Rats. The effect of oral administration of hydroalcoholic extract of Cassia occidentalis leaves on neutrophil has been studied and is compared with control group on rats. The differential leukocyte count (DLC) was performed by fixing the blood smears and staining with leucofine and percent neutrophils in each sample was determined. After the initial counts, blood samples were incubated with 80 mg/ml of nylon fibers for 10 min at 37ºC. Supplementation with hydroalcoholic extract of Cassia occidentalis leaves significantly decreased neutrophil adhesion. The results indicate that the leaf of Cassia occidentalis is endowed with protected neutrophil adhesion. These effects could conclude that Cassia occidentalis has an antiasthmatic property.
