A Modified F1 Screening Method for Detecting Resistance Gene Alleles to Bt Cotton in Helicoverpa armigera (Lepidoptera: Noctuidae)

Large-scale cultivation of Bt cotton places high selection pressure on target insects and, consequently, may prompt resistance evolution in pest populations. To better monitor Bt-resistance allele frequency in field populations, a modification of the F1 screen was developed to screen F1 progeny derived from single pair mating between field-collected males and laboratory resistant females (designated as F1 screen). This method was used to survey a field population of Heliocoverpa armigera (Hübner) for resistance alleles at the same loci in the resistant strain. After treatment of the F1 progeny derived from more than 260 single pair mating lines with Bt cotton, there was no clear separation of resistant genotypes from susceptible genotypes based on survival rates which demonstrated a linear distribution within a certain range. We further analyzed larval growth data of the F1 progeny and found a correlation between larval body weight and survival rate. The maximum correlation was obtained when F1 larval body weight reached ≥0.7 mg. To avoid underestimation, body weight ≥0.6 mg was used to initially separate potential positive lines (resistant genotype). After that, a corrected survival rate of >21.3% (the minimal theoretical rate of >25% for F0 males to carry sr and rr genotypes) was used as the criterion to examine F2 progeny derived from single sib-mating of F1 adults and to verify whether the potential positive lines carried resistant alleles.

ABSTRAKKetersediaan induk dalam suatu usaha pembenihan memiliki peran yang sangat penting. Namun demikian, ketersediaan induk yang berasal dari alam sangat terbatas jumlahnya, sehingga perlu dilakukan penyediaan calon induk yang berasal dari hasil budidaya. Ikan kerapu bebek Cromileptes altivelis Valenciennes, 1828 turunan pertama (F-1) dan turunan kedua (F-2) sudah dapat diproduksi dari hasil budidaya, namun benih turunan ketiga (F-3) belum dapat diperoleh. Penelitian ini bertujuan untuk mendapatkan karakteristik morfologis dan pertumbuhan larva ikan kerapu bebek F-3 sebagai calon induk F-3, serta tingkat keberhasilan produksi benihnya. Pemeliharaan larva dilakukan dalam hatchery hingga larva menjadi benih. Parameter yang diamati meliputi panjang total dan panjang duri sirip larva, berat tubuh larva serta sintasan dan jumlah produksi benih. Hasil penelitian menunjukkan bahwa panjang total larva umur 5, 15, 25 dan 35 hari, masing-masing adalah 3,20±0,07; 4,42±1,11; 8,35±1,12 dan 12,51±3,23 mm. Duri sirip mulai terukur pada larva umur 15 hari. Berat larva umur 30 hari adalah 0,11±0,04 g. Pola pertumbuhan panjang total dan berat tubuh larva adalah eksponensial, sedangkan pola pertumbuhan duri siripnya adalah linier. Masa pemeliharaan larva hingga menjadi benih adalah ± 40 hari. Jumlah benih ikan kerapu bebek F-3 yang diproduksi dalam satu kali siklus pemeliharaan berkisar 440 hingga 2.300 ekor dari 50 ekor induk dan 3 kali siklus pemijahan dengan tingkat kelangsungan hidup 1,30% hingga 8,80%. Hasil penelitian ini mengindikasikan bahwa ikan kerapu bebek F-3 dapat diproduksi dari hasil budidaya seperti halnya pada F-1 dan F-2. ABSTRACTBroodstocks are the most important part of humpback grouper culture, but their availability in nature are limited. Therefore, it is necessary to produce broodstock candidates from culture. The first (F-1) and the second (F-2) generation of humpback grouper have already been produced but the third generation (F-3) production is still on the way. This study was conducted to find out morphological characteristic of the third generation (F-3) of humpback grouper larvae as the candidate of the third generation of broodstock, larval growth and the success rate of seed production. Larvae rearing was done in hatchery until larvae metamorphosed to be seeds. Observed variables including larval total length and spine length, larval body weight, survival rate and the juvenile productions. The study result showed the total length of 5, 15, 25 and 35 days old larvae were 3.20±0.07; 4.42±1.11; 8.35±1.12 and 12.51±3.23 mm, respectively. The spine began measured on 15 days old larvae. The body weight of 30 days old larvae was 0.11±0.04 g. The growth pattern of larval total length and body weight were exponential, while the growth of spine was linear. Rearing period from larvae to juveniles was 40 days. Number of F-3 seed production of humpback grouper produced from once rearing cycle range between 440 and 2,300 fish and the survival rate range from 1.30% up to 8.80%. Therefore, this study could indicate that seed of F-3 humpback grouper can be produced as those of F-1 and F-2.

A study was conducted with four genotypes of quail, namely Japanese (J), White (W), Black (Bl) and Brown (Br) quail with the objective to know their hatchability traits, body weight and egg production performances. The parent males and females were maintained in cages for single pair mating through close breeding system for producing each generation. A total of 1866-day-old chicks comprising of 4 types of quail namely J, W, Br, and Bl were hatched in a batch. The egg weight were 10.35 ± 0.3, 11.60 ± 0.42, 10.30 ± 0.42 and 9.78 ± 0.65 for J, W, Br and Bl respectively, which was influenced (p < 0.05) by genotype. White genotype of quail is significantly better for egg weight, chick weight and chick: egg ratio than that of their counterparts. The highest hatchability of incubated eggs was found in Japanese quail genotype (71.01%) and the lowest in Black (31.03%). Significantly higher body weight was found in White and Black followed by Brown and Japanese quail genotype at different period of age. <p>The present study was aimed to identify the effect of non-genetic factors on harvest body weight of <em>Clarias magur</em> (Hamilton, 1822) reared under mono and polyculture systems and to estimate its heritability. Magur fishes from three different geographical regions <em>viz.,</em> Andhra Pradesh, Assam and West Bengal were procured and stocked at Fresh Water Fish Farm (FWFF) of ICAR-Central Institute of Fisheries Education, Balbhadarpuram, Kakinada, Andhra Pradesh, India. A total of 139 pairs of fishes were mated to produce full-sib families of which only 42 families had either 15 or more offsprings or survived till tagging and of these a total of 1373 fishes were PIT (Passive Integrated Transponder) tagged. Tagged fishes were reared in mono and polyculture systems. In polyculture system, magur were stocked along with rohu <em>Lobeo rohita</em> fishes. The average harvest body weight of magur reared in monoculture was 146.27±1.36 g and was significantly higher than the average body weight of fishes reared in polyculture (137.33±2.28 g) (p&lt;0.05). Male fishes were significantly heavier than females at harvest. The heritability for harvest body weight from animal model was 0.55±0.76 and from single pair mating it was 0.63±0.10. The heritability for harvest body weight in mono and polyculture systems differed from one another.</p>


The functional properties of marine invertebrate larvae represent the sum of the physiological activities of the individual, the interdependence among cells making up the whole, and the correct positioning of cells within the larval body. This chapter examines physiological aspects of nutrient acquisition, digestion, assimilation, and distribution within invertebrate larvae from an organismic and comparative perspective. Growth and development of larvae obviously require the acquisition of “food.” Yet the mechanisms where particulate or dissolved organic materials are converted into biomass and promote development of larvae differ and are variably known among groups. Differences in the physiology of the digestive system (secreted enzymes, gut transit time, and assimilation) within and among feeding larvae suggest the possibility of an underappreciated plasticity of digestive physiology. How the ingestion of seawater by and the existence of a circulatory system within larvae contribute to larval growth and development represent important topics for future research.

The body weight of U.S. adults and children has risen markedly over the past three decades. The physical health consequences of obesity are widely documented, and emerging research from the Midlife in the United States study and other large-scale surveys reveals the harmful impact of obesity on adults’ psychosocial and interpersonal well-being. This chapter synthesizes recent research on the psychosocial implications of body weight, with attention to explanatory mechanisms and subgroup differences in these patterns. A brief statistical portrait of body weight is provided, documenting rates and correlates of obesity, with a focus on race, gender, and socioeconomic status disparities. The consequences of body weight for three main outcomes are described: institutional and everyday discrimination, interpersonal relationships, and psychological well-being. The chapter concludes with a discussion of the ways that recent integrative health research on the psychosocial consequences of overweight and obesity inform our understanding of population health.

The influence of temperature on larval survival and development was studied in the edible crab, Cancer pagurus, from a population off the island of Helgoland, North Sea. In rearing experiments conducted at six different temperatures (6°, 10°, 14°, 15°, 18° and 24°C), zoeal development was only completed at 14° and 15°C. Instar duration of the Zoea I was negatively correlated with temperature. A model relating larval body mass to temperature and developmental time suggests that successful larval development is possible within a narrow temperature range (14° ± 3°C) only. This temperature optimum coincides with the highest citrate synthase activity found at 14°C. A comparison for intraspecific variability among freshly hatched zoeae from different females (CW 13–17 cm, N = 8) revealed that both body mass and elemental composition varied significantly. Initial larval dry weight ranged from 12.1 to 17.9 μg/individual, the carbon content from 4.6 to 5.8 μg/individual, nitrogen from 1.1 to 1.3 μg/individual, and the C:N ratio from 4.1 to 4.4. A narrow larval temperature tolerance range of C. pagurus as well as the indication of intraspecific variability in female energy allocation into eggs may indicate a potential vulnerability of this species to climate change. Large-scale studies on the ecological and physiological resilience potential of this commercially fished predator are needed.

Ikan baung (Hemibagrus nemurus) memiliki prospek bagus untuk dikembangkan sebagai komoditas budidaya di Indonesia karena citarasa daging yang enak banyak diminati konsumen. Dewasa ini, peningkatan produksi benih dapat dilakukan melalui pemijahan buatan dengan manipulasi hormonal. Penelitian yang dilakukan bertujuan untuk mengevaluasi produksi induk ikan baung melalui pemijahan buatan dengan dosis penyuntikan GnRH-a berbeda. Penelitian dilakukan di Instalasi Riset Plasma Nutfah Perikanan Air Tawar, Cijeruk, Bogor pada bulan November 2017. Induk betina ikan baung yang digunakan berukuran 465,0 ± 71,8 g; dan induk jantan 426,3 ± 46,8 g. Induksi dilakukan dengan penyuntikan hormon GnRH-a dengan tiga dosis yang berbeda pada induk betina (0,3; 0,5; dan 0,7 mL/kg bobot badan), dan ikan jantan dengan dosis 0,4 mL/kg bobot badan. Masing-masing perlakuan terdiri atas tiga ekor induk betina sebagai ulangan. Hormon disuntikkan secara intramuskular, diberikan dua kali penyuntikan 35% dari dosis total pada penyuntikan pertama, dan 65% diberikan pada penyuntikan kedua, dengan interval waktu penyuntikan delapan jam. Parameter yang diamati yaitu jumlah telur ovulasi, derajat pembuahan, derajat penetasan, dan sintasan larva selama tujuh hari. Hasil penelitian menunjukkan bahwa dosis penyuntikan hormon GnRH-a 0,5 mL/kg pada induk ikan baung saat proses pemijahan buatan menghasilkan derajat penetasan yang lebih tinggi dibandingkan dosis 0,3 dan 0,7 mL/kg; serta sintasan larva yang lebih baik dibandingkan dosis 0,7 mL/kg. Sedangkan nilai jumlah telur yang berhasil ovulasi dan derajat pembuahan yang relatif lebih baik ditemukan pada perlakuan dosis 0,7 mL/kg dibandingkan dengan dosis 0,3 dan 0,5 mL/kg.Asian redtail catfish (Hemibagrus nemurus) has a good prospect to be developed as aquaculture commodity in Indonesia. It was proved by the demand of consumers regarding to its delicious flesh taste. Recently, efforts to increase seedling production could be implemented through artificial spawning with hormonal manipulation. This study was conducted to evaluate the production of broodstocks through artificial spawning with different doses of GnRH-a. Study was conducted at Research Institute for Freshwater Fisheries Germplasm, Cijeruk, Bogor in November, 2017. The broodstock used in this study was 465.0 ± 71.8 g for females, and 426.3 ± 46.8 g for males. Induction was performed by injection of GnRH-a hormone with three different doses on female broodstocks (0.3, 0.5 and 0.7 mL/kg of body weight), and male broodstocks with a dose of 0.4 mL/kg of body weight. Each treatment consisted of three females as replications. The hormone was injected intramuscularly, given twice injection: 35% of the total dose is given at the first injection, and the remaining 65% was given at the second injection, with an injection time interval of eight hours. The results showed the observed parameters of number of the ovulated eggs, fertilization rate, hatching rate, and larval survival for seven days, treatment dosage of 0.5 mL/kg showed better results (P<0.05) than the dosage of 0.3 mL/kg and 0.7 mL/kg. Parameters observed were number of ovulated eggs, fertility rate, hatching rate, and survival rate for seven days. The results showed that the dosage of 0.5 mL/kg GnRH-a injection to the broodstock during artificial spawning resulted in higher hatching rate compared to 0.3 and 0.7 mL/kg and better larval survival rate than the dosage of 0.7 mL/kg. Meanwhile, the number of ovulated eggs and fertility rate was relatively better in the treatment of 0.7 mL/kg compared with the dosage of 0.3 and 0.5 mL/kg.

Efficient, large-scale heterologous production of enzymes is a crucial component of the biomass valorization industry. Whereas cellulose utilization has been successful in applications such as bioethanol, its counterpart lignin remains significantly underutilized despite being an abundant potential source of aromatic commodity chemicals. Fungal lignin-degrading heme peroxidases are thought to be the major agents responsible for lignin depolymerization in nature, but their large-scale production remains inaccessible due to the genetic intractability of basidiomycete fungi and the challenges in the heterologous production of these enzymes. In this study, we employ a strain engineering approach based on functional genomics to identify mutants of the model yeast Saccharomyces cerevisiae with enhanced heterologous production of lignin-degrading heme peroxidases. We show that our screening method coupling an activity-based readout with fluorescence-assisted cell sorting enables identification of two single null mutants of S. cerevisiae, pmt2 and cyt2, with up to 11-fold improved secretion of a versatile peroxidase from the lignin-degrading fungus Pleurotus eryngii. We demonstrate that the double deletion strain pmt2cyt2 displays positive epistasis, improving and even enabling production of members from all three classes of lignin-degrading fungal peroxidases. We anticipate that these mutant strains will be broadly applicable for improved heterologous production of this biotechnologically important class of enzymes.

<b>Objective: </b>Obesity and type 2 diabetes are risk factors for cardiovascular diseases and mortality, and that commonly result in weight variabilities. We aimed to investigate the association between body weight variability and risk of major cardiovascular outcomes and mortality in individuals with type 2 diabetes using large-scale, nationwide cohort data on the Korean population. <div><p><b>Research Design and Methods: </b>We enrolled 624,237 individuals with type 2 diabetes who underwent health examinations provided by the Korean National Health Insurance System between 2009 and 2010, with ≥3 body weight measurements within 5 years since enrollment and followed up until the end of 2017. We assessed body weight variability using four indices, including variability independent of the mean (VIM). Multivariable-adjusted Cox proportional hazards regression analysis was performed.</p> <p><b>Results: </b>During the follow-up, 15,832, 25,038, and 44,716 cases of myocardial infarction (MI), stroke, and all-cause mortality, respectively, were recorded. Body weight variability was associated with increased risks of major cardiovascular outcomes after adjusting for confounding variables. Compared with the hazard ratios (HRs) of the lowest quartile group, the HRs (95% CIs) of the highest quartile group of VIM for body weight were 1.15 (1.10–1.20), 1.22 (1.18–1.26), and 1.58 (1.53–1.62) for MI, stroke, and all-cause mortality, respectively.</p> <p><b>Conclusions: </b>Body weight variability was associated with increased risks of MI, stroke, and all-cause mortality in type 2 diabetes patients and may be a predictor of cardiovascular outcomes in such patients. Appropriate interventions to maintain stable weight could positively influence health outcomes in type 2 diabetes patients.</p> </div> <br>

Целью настоящей работы стало исследование эффектов лактоферрина (Лф) человека у мышей после острого гамма-облучения в сублетальной дозе. Методы. Исследование было проведено на 2-2,5-месячных самцах мышей линии C57Bl/6. Животные из экспериментальных групп были подвергнуты общему воздействию гамма-излучения в дозе 7,5 Гр. Сразу после облучения и повторно через 24 часа после него часть животных получила инъекцию Лф (внутрибрюшинно, 4 мг на животное). Было изучено влияние Лф на выживаемость и среднюю продолжительность жизни мышей. Для оценки общей двигательной и исследовательской активности использовали тест «Открытое поле». Результаты. Введение Лф позволило увеличить выживаемость и среднюю продолжительность жизни облученных мышей в течение эксперимента. Происходила более быстрая нормализация динамики изменения массы тела. Кроме того, Лф оказал компенсаторное действие на исследовательскую активность облученных животных. The aim of this study was to investigate effects of human lactoferrin (Lf) in mice exposed to acute gamma-irradiation at a sublethal dose. Methods. C57Вl/6 2-2.5-month-old male mice were used for the experiments. Animals from experimental groups were exposed to whole-body gamma-radiation at a dose of 7.5 Gy. Some animals received an intraperitoneal injection of Lf (4 mg per animal) immediately and then at 24 hours after the irradiation. The effect of Lf on survival rate and life span was studied. The open field test was used to assess locomotor and research activity. Results. The Lf administration increased the survival rate and life span of irradiated mice during the experiment. The dynamics of body weight normalized faster. In addition, Lf exerted a compensatory effect on the research activity of irradiated animals.
