Background: Phosalone is an organophosphate insecticide, applied to control of plant pests. This compound has various side effects because it acts as an acetyl cholinesterase enzyme inhibitor. Objective: To investigate the effects of phosalone on the sperm parameters of and levels of sex hormones in adult male rats. Materials and Methods: In this experimental study, 16 adult (8-12 wk) male Wister rates (weighing 220-280 gr) were randomly assigned into 4 groups (n = 4/each). Group 1 (control) received only routine adequate water and food; Group 2, 3, and 4 received different low doses of phosalone (60, 90, and 120 mg/kg respectively). The rats were weighed and anesthetized after 48 days. Sperm parameters including number, motility, and viability as well as sex hormones (such as Luteinizing Hormone, Follicle Stimulating Hormone, and testosterone) were evaluated and compared after removing the epididymis tail. Results: Our results showed that phosalone decreased sperm motility, viability, and number in a dose-dependent manner. The level of FSH and LH was increased, and testosterone was decreased. Also, depending on the dose, phosalone decrease sperm motility and viability (p ≤ 0.001), while the level of FSH and LH was increased and testosterone was decreased (p = 0.861). Conclusion: Phosalone has negative effects on reproductive indices in male rats and can cause serious damage and decrease the number and sperms motility. It can also cause infertility due to changing the concentration of hormones. Key words: Organophosphate, Pesticides, Phosalone, Sperm, Sex hormones.

Background and Aim: Tramadol is a potent analgesic effective in the treatment of mild to severe pains. However, the use of the drug can pose a threat to other organs and systems. Therefore, this study evaluated the effect of graded doses of tramadol on sperm profile of male albino rats. Materials and Methods: Eighteen male rats were divided into three groups (A, B and C) using completely randomized design (CRD) with six rats in each group. Rats in group A served as the control group and were given just food and water while groups B and C were given tramadol at 50 and 100 mg/kg body weight (BW) respectively, daily for the period of 65 days. The treatment was administered via oral gavage and at the end of the treatments, the rats were sacrificed. Immediately after sacrifice, a puncture was made in the epididymis with a sterile pin and examined for semen pH. The epididymes were processed for epididymal sperm motility, viability, count and sperm head abnormality. Results: There was no significant difference in the weight of testes and semen pH. Sperm viability, sperm motility, sperm count and weight of epididymes significantly reduced (p<0.05) in tramadol treated animals when compared with the control. Results also indicated statistically significant (p<0.05) increase in sperm head abnormalities in rats treated with tramadol when compared with the control. Conclusion: The results obtained from this study reveal that tramadol has negative effects on weight of epididymes, sperm count, sperm viability, sperm motility and sperm head abnormalities in male albino rat as mammalian models in a dose dependent manner.

Background: Formalin is commonly applied as an antiseptic and tissue fixative. It has reactive molecules that lead to its cytotoxic effects. According to recent studies, formalin causes a change in the testicular and sperm structure and L-carnitine (LC) acts as an antioxidant to counteract its effects. Objectives: This study aimed to investigate the protective effects of LC on the parameters, chromatin condensation and apoptosis of mice sperm exposed to formalin. Materials and Methods: In this experimental study, 24 balb/c mice (25-40 gr ,10-12 wk) were divided into three groups (n = 8/each): group I without any injections or gavage; group II, received 10 mg/ kg formalin intraperitoneally (I.P); and group III was exposed to formalin and LC, where a dose of 10 mg/kg formalin was injected I.P daily and LC the dose of 100 mg/kg was kept in a solvent solution. After 31 days, the sperm examination was performed as follows: to evaluate chromatin and DNA quality of the sperm, we applied aniline blue (AB), toluidine blue (TB), chromomycin A3 (CMA3), and terminal transferase-mediated deoxy uridine triphosphate biotin end labeling (TUNEL) tests. Results: Sperm parameters such as count, motility, morphology, and viability displayed a significant decrease in the formalin group. While the data exhibited a considerable augment in sperm parameters in the formalin + LC than the formalin and control groups (p < 0.001), significant differences were detected between groups with respect to TB staining, TUNEL test, CMA3 test and AB staining in the formalin and formalin + LC groups. Conclusion: LC can reduce the negative effects of formalin on sperm parameters, chromatin stability, and percentage of apoptosis in an animal model. Twenty-fourSprague-Dawleymale rats (7–9 weeks old, 200–250 g) were divided into Nicotine (N) (0.5 mg/100 g body weight (BW), Nicotine Control (NC) (saline, 0.1 mL/100 g BW),Habbatus saudaoil (HS) (6.0 μL/100 g BW), andHabbatus saudaControl (HSC) (corn oil, 0.1 mL/100 g BW) groups and treated for 100 days. Sperm parameters and seminiferous tubules measurements were evaluated. The N showed a significantly lower sperm motility (1.03±0.05×106 sperm/mL) and percentage of normal (82.61±0.03%) and live (93.88±0.01%) sperm, higher value for the seminiferous tubule (253.36±1.83 μm) and lumen (100.15±2.38 μm) diameters and spermatogonia (19.85±0.39 μm) and spermatocytes (33.37±0.59 μm) layers, and thinner spermatid-sperm layer (22.14±0.71 μm) than the NC (P<0.05). The HS had significantly higher sperm motility (1.49±0.04×106 sperm/mL) and percentage of normal (90.61±0.01%) and live (96.98±0.01%) sperm, smaller lumen diameter (67.53±2.34 μm) and thinner spermatogonia (17.67±0.32 μm) and wider spermatid-sperm (36.95±0.79 μm) layers than the HSC (P<0.05). This research confirmed that nicotine reduced sperm motility and morphology of normal and live sperms and also affected the testis histology, whileHabbatus saudaoil increased sperm quality and gave better testis histological features.

Infertility is a major problem for couples and half of the total number of infertilities belong to men, diagnosable in 40% and pathologically undiagnosable in 60% of the cases. Complications of exposure to pesticides is a concern, with a considerable share in male infertility. Phosalone is an organophosphate insecticide usually used to control various types of pests in agriculture. Considering the extensive use of this compound worldwide, the present study examined the effects of environmental agents related to organophosphate on male infertility. To perform the experiments, 16 adult male rats were randomly assigned to four groups with four each. Group 1 received routine rat feed and water as the first control group; Group 2 received low doses (60 mg/kg); Group 3 medium doses (90 mg/kg); and Group 4 high doses of Phosalone (120 mg/kg). After 48 days (end of the treatment), the rats were weighed and anesthetized using ether. Then, their tail of epididymis was removed, and sperm parameters, including number, motility, and viability, were evaluated and compared. Moreover, their reproductive organs were removed and the sexual tissue was processed and underwent histopathological examinations. In addition, in this study, important biochemical parameters in infertility such as the level of sex hormones (LH, FSH, and testosterone) were evaluated using the ELISA test. Results showed that Phosalone decreased sperm motility, viability, and number in a dose-dependent manner. The level of FSH and LH was increased, and that of testosterone was reduced. This increase in FSH and LH must have occurred as the result of reduced testosterone, and all these changes were dependent on the dose of Phosalone. Results of histological studies also revealed that the trend of degeneration in the tissues of testes, epididymis, and seminal vesicle was completely dose-dependent. Phosalone has considerable effects on reproductive indices and can cause serious damage and impose undesirable effects on the number and motility of sperms, causing infertility by altering the concentration of hormones

The effects of chronic dietary Roundup (RU) exposure on rooster sperm parameters, fertility, and offspring are unknown. We investigated the effects of chronic RU dietary exposure (46.8 mg kg−1 day−1 glyphosate) for 5 weeks in 32-week-old roosters (n = 5 RU-exposed and n = 5 control (CT)). Although the concentrations of glyphosate and its main metabolite AMPA (aminomethylphosphonic acid) increased in blood plasma and seminal fluid during exposure, no significant differences in testis weight and sperm concentrations were observed between RU and CT roosters. However, sperm motility was significantly reduced, associated with decreased calcium and ATP concentrations in RU spermatozoa. Plasma testosterone and oestradiol concentrations increased in RU roosters. These negative effects ceased 14 days after RU removal from the diet. Epigenetic analysis showed a global DNA hypomethylation in RU roosters. After artificial insemination of hens (n = 40) with sperm from CT or RU roosters, eggs were collected and artificially incubated. Embryo viability did not differ, but chicks from RU roosters (n = 118) had a higher food consumption, body weight and subcutaneous adipose tissue content. Chronic dietary RU exposure in roosters reduces sperm motility and increases plasma testosterone levels, growth performance, and fattening in offspring.

Abstract Green tea is a popularly consumed beverage worldwide and contains polyphenols, whose antioxidant activities could improve sperm parameters and fertility thereof. We investigated the effect of green tea on the male rat reproductive system as well as its safety. Male Wistar rats were administered 2 and 5% aqueous extract of green tea for 52 days’ ad libitum, while the control group received tap water. Total polyphenol, flavanol, flavonol and soluble solids significantly increased in a concentration-dependent manner in vitro (P < 0.01). Weights of body, testis, epididymis, prostate gland, seminal vesicles, and liver, serum levels of testosterone, ferric reducing antioxidant power, creatinine, and sperm motility, remained unchanged (P > 0.05). Kidney weight, sperm concentration and vitality, spontaneous acrosome reaction increased (P < 0.05), while alanine transaminase and aspartate transaminase levels decreased (P < 0.05). Catalase, superoxide dismutase, glutathione and lipid peroxidation remained unchanged in the testes, liver and kidney (P > 0.05). Histological sections of testis, epididymis, kidney and liver showed no conspicuous alteration. Diameter and epithelial height of seminiferous tubule decreased, while caudal epididymis epithelial height increased (P < 0.01). Consumption of green tea in the conditions used in the present study seems to be safe and improved sperm parameters. However, subtle structural changes observed in the decreased diameter and epithelial height of the seminiferous tubule and increased acrosome reaction needs further investigation.

Opioids are the most potent and effective analgesics available and have become accepted as appropriate treatment for acute, cancer and non-cancer. Morphine, which is commonly used for the treatment of severe pain, gastrointestinal tract and kidneys. Curcumin petals consist of, glycosides, flavonoids, and anthocyanin. The study aims at evaluating curcumin effect and morphine on sperm parameters, testis tissue and serum testosterone level in rat. In this experimental study, 48 male rats with 28 weeks of age and limited weight of 270 to 300g were selected. They were divided into eight groups of 6, untreated control group; morphine – treated group (20 mg/kg/day); curcumin -treated groups (10, 30, 60 mg/kg/day); and morphine and curcumin treated group intraperitoneal administration for successive 28 days. After 24hours animals were killed. Sperm motility was measured using WHO protocols. The sperm parameter such as motility, sperm count, morphology, seminiferous tubules diameter, weight testis, and serum testosterone level were analyzed (oneway ANOVA). Curcumin (10, 30 and 60 mg/kg) significantly increased mean percentage of sperm motility, count, testis weight, and serum testosterone level compared to control group (p<0.05). Testosterone level decreased significantly in rats treated with morphine. Co-administration of curcumin to morphine-treated rats improved the histopathological alterations induced by morphine in testis and increased the sperm count. Curcumin has a very strong antioxidant effects at applied doses and it can probably be used as an antioxidant and food supplement in reproductive disorders.Journal of Medical and Biomedical Sciences (2017) 6(2), 1-10


Contrast- induced nephropathy (CIN) is an elevation of serum creatinine of ≥ 0.5 mg/dL from baseline after two to three days of exposure to contrast substance if there is no other cause for acute kidney injury. Atorvastatin may protect normal kidney physiology from contrast- induced kidney injury by effects unrelated to hypolipidemia termed pleiotropic effect by decline of endothelin production, angiotensin system down regulation, and under expression of endothelial adhesion molecules. This study was conducted to assess the strategy by which atorvastatin can achieve protective effect for kidneys after exposure to contrast media in an animal model. A 40 male rats were distributed randomly into 4 groups; ten rats for each: group (1): given normal saline; group (2): CIN group given iopromide as contrast media; group (3): given atorvastatin (20mg/kg) and iopromide; and group (4): given atorvastatin (40mg/kg) and iopromide. Blood collected by cardiac puncture for detection of serum glutathione, malondialdehyde, matrix metalloproteinase-9, and interleukin-18. The results have shown a significant increase in inflammatory and oxidative stress markers in contrast media group, and significant reduction in these markers in atorvastatin treated groups, in a dose-dependent manner. As conclusion, atorvastatin mechanism for protection against CIN in a dose-dependent manner can mediate by anti-inflammatory and antioxidant effects.
