Endosulfan an organochlorine insecticide that is commonly used even though it has been banned due to its toxic and teratogenic effect. This study aims to determine the effect of orally endosulfan exposure to pregnant rat (Rattus norvegicus) at day 6-15th of gestation period to the foetus skeletal malformation. A Completely Randomized Design (CRD) by endosulfan dosages: 0 (control); 0.083; 0.190; 0.440; 1.000 mg/kg BW/day with 5 replicates was applied. At 20th of the gestational period, the rats were sacrificed, their reproductive organs and the fetal skeletal malformation were observed using Alizarin red S method. The results of one-way ANOVA test showed that endosulfan exposure did not significantly affect the pregnancy outcomes ie. female rats weight gain, the numbers of implanted foetus, foetal body weight and length, respectively. Malformation of foetus implantation, foetus size and foetus skeletal occured in the endosulfan exposure groups. Fisher’s exact test results showed a significant difference between the control group and the endosulfan exposure group on the number of foetuses that have abnormalities in the number of skeletons of the sternum, fore and hind paws. The exposure of endosulfan at 0.083-1.000 mg / kg BW / day in pregnant rats caused skeletal malformations of the foetus ie. decreased in the number on sternum, fore- and hind paw bones. ABSTRAK Endosulfan merupakan insektisida golongan organoklorin yang masih digunakan hingga kini meski telah dilarang karena bersifat toksik dan teratogenik. Penelitian ini bertujuan untuk mengetahui efek pemajanan endosulfan secara oral terhadap malformasi rangka fetus tikus (Rattus norvegicus Berkenhout, 1769) selama umur kebuntingan 6-15 hari. Penelitian dilakukan secara eksperimental di laboratorium menggunakan Rancangan Acak Lengkap (RAL) dengan pemajanan endosulfan dosis: 0 (kontrol); 0,083; 0,19; 0,44; dan 1 mg/kg BB/hari dan masing-masing diulang sebanyak lima kali. Induk betina dengan umur kebuntingan 20 hari, dikorbankan nyawanya lalu dibedah dan diamati tampilan reproduksi induk serta malformasi rangka pada fetus dengan metode pewarnaan Alizarin red S. Hasil uji sidik ragam satu arah menunjukkan pemajanan endosulfan tidak berpengaruh secara signifikan terhadap rataan pertambahan bobot badan induk, jumlah fetus terimplantasi, bobot badan dan panjang fetus. Malformasi implantasi fetus, ukuran fetus dan rangka fetus terjadi pada kelompok pemajanan endosulfan. Hasil uji eksak Fisher menunjukkan adanya perbedaan yang signifikan antara kelompok kontrol dengan kelompok pemajanan endosulfan terhadap jumlah fetus yang mengalami kelainan jumlah rangka penyusun sternum, cakar depan dan cakar belakang. Simpulan dari penelitian ini adalah pemajanan endosulfan dosis 0,083- 1,000 mg/kg BB/hari pada tikus bunting menimbulkan malformasi rangka pada fetus yaitu berkurangnya jumlah tulang penyusun sternum, cakar depan, dan cakar belakang.

Objectives: Drugs can cause undesired effects on the fetus during pregnancy, especially embryonic/organogenesis which could lead to defects in the fetus because some types of drugs can penetrate the placenta and will undergo biotransformation into a highly reactive compound that has the potential to become a teratogenic compound. The aim of this research was to examine the teratogenic effect of bintangur leaves (Calophyllum soulattri Burm. F) ethanol extract to Sprague Dawley strain white rats. Methods: The white rats are divided into four treatment groups: Control group was given carboxymethyl cellulose Na 1%, comparison group was given trimethoprim 360 mg/kg BW, C. soulattri leaves ethanol extract (CLE) 100 mg/kg BW, and CLE 500 mg/kg BW. The treatment was administrated since organogenesis period. Cesarian section was performed to pregnant rat at the 20th day to separate the fetuses. Observation covered body weight of pregnant rats, fetal biometric, morphological malformation, and skeletal formation. Results: CLE 100 mg/kg BW and 500 mg/kg BW did not cause any change in the number of a living fetus, body weight, and length of fetuses like the comparison group. Both doses of CLE shown have a normal skeletal formation. Resorption was found in the comparison group and CLE 100 mg/kg BW with the percentage was 65.21% and 6.67%. It was found that there is no significant difference (p<0.05) between both doses of CLE compared to control group. Conclusion: From the results, it is concluded that CLE did not have the teratogenic effect.

Then blood samples were taken from the heart for measurement of serum estradiol 17b levels by ELISA and uterine organs were taken for measurement of Malondialdehyde (MDA) with spectro-photometry method.Results: The results of the measurement of serum estradiol 17b and uterus malondialdehyde (MDA) levels of female Wistar strain rats (Rattus norvegicus) showed an opposite pattern, where there was a decline in serum estradiol 17b levels and an increase in uterus malondialdehyde (MDA) level. There was a significant difference (p=0.000<alpha) in 17b estradiol serum and uterus Malondialdehyde (MDA) levels of female rats between control group and group exposed to sipermetrin treatment for 28 days.Conclusions: Oral sipermetrin exposure can decrease serum levels of estradiol 17b and increase uterine levels of malondi-aldehyde (MDA) of female Wistar strain rats (Rattus norvegicus).

Based on the National Population and Family Planning Agency (BKKBN) data in 2013, there were 8,500,247 couples of childbearing age who were new KB (family planning) participants with the total number using injection was 4,128,115 (48.56%). Research Objectives to find out The Effect of The Administration OF Medroxy Progesterone Acetate Hormone to Endometrial Thickness of The Female Rattus Norvegicus Wistar Strain. The design of this research is the post test only control group design by having  white rats species Rattus Norvegicus Wistar Strain as the poplulation, the samples are taken randomly from all populations that met the following sample criteria: female rats, Aged 12-14 weeks, having body weight of 150 - 200 grams. Based on the calculation of the sample size, the sample size / number of animals r = 10 rats was obtained, with a total sample size for both groups of treatment and control were 20 female rats which included: group 1 amounted to 10 rats that were not given Medroxy Progesterone Acetat hormones as a control group where group 2 amounted to 10 rats which given Medroxy Progesterone Acetat as a treatment group. The results showed the average endometrial thickness in the (P1) Medroxy Progesterone Acetate Hormone group was 493.17 µm ± 114.96, this result was lower than the control group 1192.56 µm ± 428.94. From the ANOVA test table, the P value <0.05 means that there is a very significant difference in the average endometrial thickness between the control group and the treatment group. Based on the results of the Post Hoc Bonfferoni Test, it can be seen that the average endometrial thickness between Control and P1 showed a significant difference (P <0.05). The analysis of the result and discussion of the research can be concluded that: the Medroxy Progesterone Acetate hormone affects the endometrial thickness, and there is a difference in the histological picture of endometrial thickness due to the administration of the Medroxy Progesterone Acetate hormone and without the administration of to the administration of the Medroxy Progesterone Acetate hormone.

Hyperbaric Oxygen Therapy (HBOT) can increase oxygen delivery to tissues and stimulate the formation of H2O2 as a secondary messenger for phosphorylation of nuclear factor kappa beta (NF-kB) which plays an important role in the transcription of the anti apoptotic gene. This study aimed to determine the effects of Hyperbaric Oxygen Therapy (HBOT) in enhancing the expressions of Bcl-2 in the myometrium of pregnant rats infected by Toxoplasma gondii. This study was an experimental study with a randomized control group of post-test only and designed by 37 pregnant Rattus norvegicus Sprague Dawley. Randomly, the rats were divided into four groups. Group A is infected pregnant rats that exposed by 10 sessions of HBOT 2.4 ATA in 3x30 minutes. Group B is non-infected pregnant rats and exposed by 10 sessions of HBOT 2.4 ATA in 3x30 minutes. Group C is infected pregnant rats without any exposure. Group D is non-infected pregnant rats without any exposure. Each infected pregnant rat was given a 103 tachyzoite of T.gondii by intraperitoneal injection. Bcl-2 expressions were measured through immunohistochemistry. All data were analyzed using ANOVA test through SPSS 21 program application. There was a significant difference in Bcl-2 expression between Group A and Group C because p<α (p<0.017). HBOT can increase the expression of Bcl-2 from infected and not infected rat myometrium, in the provision of HBOT 2.4 ATA for 3x30 minutes, twice a day for 5 days.

Background: Gelatin has been used as a hemostatic agent to stop bleeding and it can be absorbed by human body within 6 hours. The patin fish (Pangasius djambal) gelatin has a high amount of amino acid, especially glutamine which has an important role in increasing the proliferation of fibroblast in wounds. The purpose of this study was to determine the effect of patin fish gelatin to the number of fibroblast in wound after tooth extraction.Method: Rat’s tooth extraction is done on the lower left incisive tooth. Thirty white rats were divided into two groups randomly with 3 time series. Control group was the rats without given of patin fish gelatin and treatment group was the rats given 1cc of patin fish gelatin with pipette. Socket taken for histologically processed by Hematoxylin Eosin staining.Result: It showed there were significant differences in the number of fibroblasts between groups. The Post Hoc Tukey test result showed there was a significant difference between the control group and the treatment group.Conclusion: Patin fish gelatin has effect increasing the number of fibroblast inwound after white rat’s (Rattus norvegicus) tooth extraction

Abstract Background: Premature ovarian failure is among the most important side effects of chemotherapy during reproductive period. Preserving ovarian function is gradually gaining importance during oncologic treatment. The present study aims to investigate the potential of melatonin to protect from cisplatin-induced ovarian toxicity in rats. Twenty nine female rats were divided to three groups: Saline control group (Group 1), cisplatin group (Group 2), and cisplatin+melatonin group (Group 3). While the rats in Groups 2 and 3 were administered 5 mg/kg single dose of cisplatin via intra-peritoneal (IP) route, the rats in Group 3 were started on melatonin (20 mg/kg IP) before cisplatin administration and continued during 3 consecutive days. Ovaries were removed one week after cisplatin administration in all groups. Blood samples were obtained before the rats were decapited. Histological evaluation, follicle count, and classification were performed. TAp63 mRNA expression was evaluated using mRNA extraction and real-time polymerase chain reaction (PCR) method. Serum estradiol (E2) and anti-mullerian hormone (AMH) values were measured with enzyme immune-assay technology. Results: While primordial follicles were seen to decrease in Group 2 as compared to Group 1 (p:0.023), primordial follicle count was observed to be preserved significantly in melatonin group as compared to Group 2 (p:0.047). Moreover, cisplatin-induced histo-pathological morphology was preserved in favor of normal histology in melatonin group. A significant difference was not observed between groups with regard to mean serum AMH and E2 values (p:0.102 and p:0.411, respectively). While TAp63 gene expression significantly increased in Group 2 as compared to control group (p:0.001), we did not detect a statistically significant difference in cisplatin+melatonin group, although gene expression decreased (p:0.34). Conclusion: We conclude that concurrent administration of melatonin and cisplatin may protect from ovarian damage.

The effects of a new experimental drug, KF-868, were investigated after administration to pregnant Sprague-Dawley rats at 0(vehicle), 0.1, 2.0, and 40.0 mg/kg per day during Days 7 through 17 of gestation by examination of term fetuses and naturally delivered offspring. Pregnant rats administered 0.1, 2.0, and 40.0 mg/kg per day gained significantly more weight during the dosage period than did the vehicle control group. Treatment-related physical signs, bloody crust on nose and stains on fur, were observed in the high dosage group. Fetal viability was significantly increased, and resorptions were significantly decreased for the mid and high dosage groups, when compared with the control group. Average fetal body weights for cesarean-delivered fetuses were less for the 40.0 mg/kg per day dosage groups than for the vehicle control group. Visceral and skeletal evaluations of fetuses revealed no difference between the control and test groups. Percent survival of pups was significantly less for the high dosage group than for the control group. Average rat body weights prior to mating for the high dosage group were generally less than for the control group. All physical and functional developmental values were comparable among the control and test groups. Evaluation of postweaning parameters of pups revealed no significant difference in sex maturation, behavior (open-field and water maze), and reproductive capacity. Average body weight gains during the 9-week growth period before mating were significantly less for the 40.0 mg/kg per day dosage group F1 generation female rats. Toxicity in fetuses and offspring was observed only at the highest dosage level. Dosage-dependent, significant increases in maternal body weight gain, as compared with control values, occurred for doses in the 3 KF-868-administered groups. These results indicate that 0.1 and 2.0 mg/kg per day dosages of KF-868 were not lethal and did not produce any adverse effects on the morphological or functional development of offspring. Toxicity was evident in offspring and fetuses of dams administered 40.0 mg/kg per day KF-868, 40,000 times as high as the daily therapeutic dose.

Juveniles of pejerrey, Odontesthes bonariensis, were exposed to 0.1% Alizarin Red S (ARS) alone or with a previous immersion in 2.2% saline solution (Osmotic Induction, OI) to enhance the ARS marking method. Fish were marked in the field and immediately released in 1 m3 cages in "La Salada de Monasterio" lagoon, Chascomús, Buenos Aires , Argentina. After 73 days, clear marks were observed in the otoliths, caudal fin rays and scales with both treatments, being the intensity of the signal in the scales of OI+ARS treated fish higher. On the other hand, no marks were observed in the control group on the same structures. Approximately one year post-treatment (385 days), only marks in caudal fin rays were found clearly in OI+ARS treated fish. After this period, no significant differences in total length or weight between marked or control fish were observed and the mortality ranged between 30-40 % in all cages. These results provide strong evidence for the potential applicability of this cost-effective marking technique in differentiation of wild and hatchery-produced pejerrey. The success in the caudal fin rays marking is also important because it is easy to do and does not require the sacrifice of fish.

Objective: To analyze the effect of curcumin in VEGF expression on Rattus norvegicus cervical cancer cell xenograft model.Methods: An experimental study with randomized post test only control group design. The subjects were Rattus norvegicus (Sprague Dawley), inoculated with He-la cervical cancer cells from American Type Culture Collection (ATCC) processed in stem cell laboratory Institute of Tropical Disease (ITD) Airlangga University. 5x106 of He-La cells were injected subcutaneously in dorsal flank area of Rattus norvegicus. After 30 days of observation we performed histopathological examination of xenograft tissue and randomized into 2 groups which were given curcumin orally 1000 mg/kg (curcumin group) vs. no therapy (control group). After another 30 days the xenograft tissue was dissected and underwent immunochemistry examination for VEGF expression.Results: 32 samples of Rattus norvegicus were divided into 2 groups, In curcumin group the VEGF median expression was 2,2 (0,3-7,6) and in control group the VEGF median expression was 6,6 (1,2-12). There was a statistically significant difference with p value =0,009 with Mann Whitney test (p<0,05).Conclusion: VEGF expression in Rattus norvegicus xenograft model of cervical cancer was suppressed by giving Curcumin 1000 mg/kgBB orally.
