Background. The prevalence of endodontic infection after root canal treatment caused by the Enterococcus faecalis bacteria ranged between 24-77%. It is caused by resilience and virulence from Enterococcus faecalis. An alternative  solution have to be done toward irrigation of root canal wall which is effective to kill bacteria. Kedondong Bangkok (Spondias dulcis Forst.) is one of the plants or natural substance potentially as an antibacteria. The antibacterial potencies of Kedondong Bangkok leaves extract (Spondias dulcis Forst.) against Enterococcus faecalis bacteria could be identified by determining Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC). Purpose. This study is aimed to prove antibacterial potencies by identifying Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC) of Kedondong Bangkok leaves extract (Spondias dulcis Forst.) against Enterococcus faecalis bacteria. Method. This study is an experimental laboratories through research design of The Post Test Only Control Group Design. Value of MIC and MBC were known by counting the growth of Enterococcus faecalis bacteria by treating the leaves extract of Kedondong Bangkok  with concentration respectively 25%, 22,5%, 20%,17,5%, 15%, and 12,5% on nutrient agar media in CFU/ml. Result. In the concentration 12,5% there are 8.8% bacterial growth and in the concentration 15% there are no bacterial growth. Conclusion. Kedondong Bangkok leaf extract (Spondias dulcis Forst.) has an antibacterial potency against Enterococcus faecalis bacteria. The MIC shows in concentration of 12,5% and the MBC shows in concentration of 15%.

Abstract: Endodontics is the a type of treatment that aims to keep the teeth in function. Disinfection of the root canal is very important in endodontic treatment. Root canal disinfection can be done by giving the material a root canal medication. One of the bacteria that causes the failure of root canal treatment is the bacterium Enterococcus faecalis. The lemongrass essential oil contains geranial, neral, and mirsen which have antimicrobial activity against Gram-positive and Gram-negative. This study aimed to determine whether the lemongrass essential oil was effective against bacteria Enterococcus faecalis. This was an experimental study using post test only control group design with agar plate diffusion method. Samples consisted of a group of lemongrass essential oil with several concentrations: 25%, 50%, 75%, 100%, each consisted of 4 samples. The 16 control groups consisted of positive and negative groups. Diameter of inhibition was determined by the ability to inhibit Enterococcus faecalis cultured on MHA agar. The results showed that the average inhibitory diameter of 25% lemongrass essential oil was 2.60 mm; of 50% was 4.73 mm; of 75% was 4.50 mm; and of 100% was 5.34 mm. Conclusion: Lemongrass essential oil showed an antibacterial effect to inhibit the growth of bacteria Enterococcus faecalis.Keywords: enterococcus faecalis, lemongrass oil, antibacerialAbstrak: Endodontik merupakan salah satu jenis perawatan yang bertujuan mempertahankan gigi agar tetap dapat berfungsi. Disinfeksi saluran akar sangat penting dalam perawatan endodontik. Disinfeksi saluran akar dapat dilakukan dengan memberi bahan medikamen saluran akar. Salah satu bakteri yang menyebabkan kegagalan perawatan saluran akar ialah bakteri Enterococcus faecalis. Minyak atsiri sereh dapur mengandung geranial, neral dan mirsen yang memiliki aktifitas antimikrobapada gram positif dan gram negatif. Penelitian ini bertujuan untuk mengetahui apakah minyak atsiri sereh dapur memiliki efek terhadap bakteri Enterococcus faecalis. Penelitian ini menggunakan metode eksperimental dengan desain post test only control group design dan metode difusi lempeng agar. Sampel penelitian terdiri dari kelompok minyak atsiri sereh dapur dengan konsentrasi 25%, 50%, 75%, dan 100% yang masing-masing terdiri dari 4 sampel; 16 kelompok kontrol terdiri atas kelompok positif dan negatif. Diameter hambat ditentukan berdasarkan kemampuan menghambat Enterococcus faecalis yang dibiakkan pada media agar MHA. Hasil penelitian memperlihatkan rata-rata diameter hambat minyak atsiri sereh dapur dengan konsentrasi 25% sebesar 2,60 mm, 50% sebesar 4,73 mm, 75% sebesar 4,50 mm, dan 100% sebesar 5,34 mm. Simpulan: Minyak atsiri sereh dapur memiliki efek antibakteri untuk menghambat pertumbuhan bakteri Enterococcus faecalis.Kata kunci: enterococcus faecalis, sereh dapur, antibakteri

Background: Enterococcus faecalis is generally found on the failure of root canal treatments. Zinc oxide propolis is believed to have an antibacterial effect on that bacteria. This research aimed to compare bacteriostatic effect of zinc oxide eugenol (ZOE) and zinc oxide propolis (ZOP) as the sealer materials of root canal.Method: This was an experimental research with post-test only control group design with two different groups (ZOE and ZOP). Culture of Enterococcus faecalis bacteria was smeared on Blood Agar Plate media with six times replication per group and kept inside incubator for 24 hours. The result was obtained from the inhibition zone formed around the pasta.Result: The average result of ZOE and ZOP was 27.7 mm and 13.45 mm respectively. Normality test using Shapiro-Wilks showed that data was normal (p>0.05). Then, the data was analysed using Independent Samples T-test. The result showed that there was different inhibition zone between ZOE group and ZOP group (p<0.05).Conclusion: Based on the result, it can be concluded that ZOP has lower antibacterial effectiveness of the Enterococcus faecalis than ZOE.

Background. Failure of endodontic treatment is commonly caused by the persistent microorganisms remaining in the root canal such as Enterococcus faecalis. Enterococcus faecalis can form a biofilm in tough environmental conditions within the root canals and caused biofilm-mediated infections which needs more complicated treatment due to the increasing of antimicrobial resistance. The biofilm formation initial and most important step is bacteria adherence to the solid surface that is mediated by glucan. NaOCl 2.5% is a commonly used root canal medicaments but can cause injury of periapical tissue. Mangosteen pericarp extract contains flavonoid, tannin, and xanthone have mechanism for inhibiting adherence of bacterial biofilm. Difference of antibacterial activity between NaOCl 2.5% and mangosteen pericarp extract 0.09% can be determined by experimental laboratory to determine the adherence of bacteria in each treatment. Purpose. The aim of this study was to assess the difference of antiglucan activity between NaOCl 2.5% and mangosteen pericarp extract 0.09% on Enterococcus faecalis. Method. This study was designed as an experimental laboratory study with post  test only control group design using Enterococcus faecalis ATCC 29212. Mangosteen pericarp was extracted using maceration method. Adherence analysis was observed after 24 hours by examining the viable cells in suspension. These viable cells are measured by UV-Vis spectrophotometer to compare the suspensions’ turbidity. Using the Independent T-Test, significantly less bacteria were found adhering to the mangosteen pericarp extract. Results. Absorbancy difference level by mangosteen pericarp extract 0.09% is significantly greater than the NaOCl 2.5% (p<0.05). Conclusion. Antiglucan activity that generated by mangosteen pericarp extract 0.09% is greater than NaOCl 2.5%

Pendahuluan: Bahan irigasi yang biasa digunakan berasal dari bahan kimia dapat memberikan efek samping yang lebih besar dibandingkan dengan obat tradisional. Penggunaan bahan yang berasal dari alam dapat dijadikan pilihan sebagai alternatif bahan irigasi saluran akar karena beberapa dari bahan tersebut dapat bersifat menghambat pertumbuhan (bakteriostatik) maupun membunuh bakteri (bakterisid). Tujuan Penelitian: Untuk mengetahui hubungan Ekstrak Sarang Semut (Myrmecodia pendens) sebagai bahan irigasi saluran akar dengan daya hambat bakteri Enterococcus faecalis. Bahan dan Metode: Menggunakan metode eksperimental laboratorium. Bentuk penelitian berupa Post test only control group design dan pengambilan sampel dengan Purposive Sampling menggunakan 4 perlakuan dan 5 kali pengulangan. Uji statistik menggunakan One Way Anova. Hasil: Hasil penelitian ini menunjukkan diameter zona inhibisi dalam menghambat pertumbuhan bakteri Enterococcus faecalis pada ekstrak tanaman sarang semut (Myrmecodia pendens) konsentrasi 25% sebesar 21,10 ± 0,18. Hasil pada ekstrak tanaman sarang semut (Myrmecodia pendens) konsentrasi 50% sebesar 23,47 ± 0,24. Uji statistik memperoleh nilai signifikan p = 0,000 <? = 0,01. Kesimpulan: Hipotesis alternatif penelitian ini diterima dan hasil penelitian ini menunjukkan bahwa ada hubungan ekstrak tanaman sarang semut konsentrasi 25% dan konsentrasi 50% sebagai bahan irigasi saluran akar yang dapat menghambat pertumbuhan bakteri Enterococcus faecalis.

Abstract: Inadequate oral and dental hygiene can cause plaques containing various kinds of bacteria; one of them is Streptococcus mutans which is the main cause of dental caries. Binahong leaf (Anredera cordifolia Steenis) is a medicinal herb that contains antibacterial compounds namely flavonoids, alkonoid, terpanoid, and saponins. This study aimed to determine the minimal inhibitory concentration (MIC) of binahong leaf extract to the growth of Streptococcus mutans. This was a true experimental study with a randomized pretest-posttest control group design. The method used in this study was serial dilution method with turbidimetry and spectrophotometry as the test methods. Binahong leaves were taken from Tempok village and were extracted with maceration method using ethanol 96%. Streptococcus mutans bacteria were obtained from a pure bacterial stock in Microbiology Laboratory of Pharmacy Study Program, Faculty of Mathematics and Natural Science University of Sam Ratulangi Manado. Penelitian ini menggunakan metode serial dilusi dengan metode pengujian turbidimetri dan spektrofotometri. Daun binahong diperoleh di desa Tempok, dan diekstraksi dengan metode maserasi menggunakan etanol 96%. Bakteri Streptococcus mutans diambil dari stok bakteri murni Laboratorium Mikrobiologi Program Studi Farmasi Fakultas MIPA Universitas Sam Ratulangi Manado. The authors compared the activity of daptomycin with that of ampicillin, penicillin, teicoplanin and vancomycin against 304 strains ofEnterococcusspecies isolated from blood and urine. Daptomycin was as active as penicillin againstEnterococcus faecalis: 90% of strains were inhibited by 2 mg/L. Daptomycin was more active than vancomycin (90% minimal inhibitory concentration [MIC90] 2 mg/L; 90% minimal bactericidal concentration [MBC90] 8 mg/L) but was less active than teicoplanin (MIC500.25; MBC908 mg/L) or ampicillin (MIC901 mg/L; MBC902 mg/L) againstE faecalis. In time-kill studies daptomycin was not more rapidly bactericidal than ampicillin or penicillin but was significantly more rapidly bactericidal than either teicoplanin or vancomycin. In combination with gentamicin, daptomycin has activity similar to that of penicillin, vancomycin and teicoplanin. Daptomycin may be a suitable alternative to penicillin in patients allergic to penicillins or for the treatment of enterococcal infections caused by beta-lactamase-producing enterococci.

Pendahuluan: Prevotella intermedia merupakan salah satu bakteri utama pada penyakit periimplantitis. Periimplantitis merupakan inflamasi jaringan lunak dan keras disekitar implan yang dapat dicegah menggunakan ekstrak tanaman antibakteri. Salah satunya yaitu kulit jeruk nipis, yang dapat menghambat proses inflamasi karena mengandung alkaloid, steroid, saponin, flavonoid, tanin sebagai senyawa antibakteri. Penelitian ini bertujuan untuk mengetahui peranan antibakteri kulit jeruk nipis dengan mengukur Konsentrasi Hambat Minimal (KHM) dan Konsentrasi Bunuh Minimal (KBM) ekstrak kulit jeruk nipis terhadap pertumbuhan bakteri Prevotella intermedia. Metode: Eksperimental laboratorium dengan rancangan penelitian post test only control group design. Pengujian KHM dan KBM dilakukan dengan metode dilusi, kulit jeruk nipis dimaserasi menggunakan pelarut etanol 70% sehingga didapatkan ekstrak kulit jeruk nipis dengan konsentrasi 0,78, 1,56, 3,125, 6,25, 12,5, 25, 50, dan 100% dengan chlorhexidine sebagai kontrol positif dan akuades sebagai kontrol negatif. Media kultur bakteri menggunakan Tripton Soya Agar. Hasil: Berdasarkan analisis statistik menggunakan Kruskal Wallis menunjukkan perbedaan penurunan jumlah koloni bakteri yang signifikan (p=0,0001) pada KBM dan KHM dari berbagai konsentrasi ekstrak kulit jeruk nipis terhadap pertumbuhan bakteri Prevotella intermedia, uji lanjutan Mann Whitney  menunjukkan perbedaan penurunan jumlah koloni bakteri yang signifikan (p=0,021) antar masing-masing konsentrasi dan kelompok kontrol. Methods: Experimental laboratory with post-test only control group design. The MIC and MBC tests were performed by the dilution method. The lime peel was macerated using 70% ethanol solvent to obtain lime peel extract with concentrations of 0.78, 1.56, 3.125, 6.25, 12.5, 25, 50, and 100% with chlorhexidine as a positive control and aquadest as a negative control. Bacterial culture media using Tripton Soy Agar. Results: Based on statistical analysis using Kruskal Wallis showed a significant difference in the decrease of the number of bacterial colonies (p=0.0001) in MBC and MIC from various concentrations of lime peel extract on the growth of Prevotella intermedia bacteria. Furthermore, the Mann Whitney follow-up test showed differences in the decrease of the number of bacterial colonies, which was significant (p=0.021) between each concentration and control group. Conclusions: The minimum inhibitory concentration of lime peel extract towards the growth of Prevotella intermedia was 12.5%, with the minimum bactericidal concentration of 25%. Keywords: antibacterial agent; citrus extracts; Prevotella intermedia

Background: Fusobacterium nucleatum is a common bacterial in root canal with pulp necrosis and periradicular lesion. A way to eliminate these bacteria from root canal is by root canal irrigation. Root canal irrigation materials that are widely used nowadays has many shortcomings. The pericarp of mangosteen (Garcinia mangostana L) has antibacterial potency. Therefore mangosteen pericarp can be an alternative material which could inhibit and bactericidal function to Fusobacterium nucleatum. Purpose: The aim of this study was to determine the antibacterial potency of mangosteen pericarp extract (Garcinia Mangostana L.) against Fusobacterium nucleatum. Methods: This study was laboratory experimental with pos-test only control group design. A microdilution method was used to determine minimum inhibitory concentration and minimum bactericidal concentration by colony counting bacteriae in Tryptone Soya Agar (TSA) media with drop plate technique. Growth of bacterial colonies in TSA is calculated manually in colony forming unit (CFU/ml). Results: Bacterial colonies growth at concentration 0.78% was 90% less than positive control group and there were no bacterial colonies growth at concentration 0.975%. Conclusion: The Minimum Inhibitory Concentration (MIC) of mangosteen pericarp against Fusobacterium nucleatum was at 0,78% concentration and the Minimum Bactericidal Concentration (MBC) was at 0.975% concentration.

Background: Root canal infections is caused by the colonization of microorganisms. Microorganisms that commonly found in root canal failure is Enterococcus faecalis. These bacteria can be found in 80-90% of root canal infections. Currently, many plants are used as antibacterial drugs. Pineapple is one plant that can be used as antibacterial drugs. Pineapple peel has a variety of antibacterial chemical compounds, such as, bromelain, saponins, tannins and flavonoids that can inhibit the growth of Enterococcus faecalis bacteria. Aim: To obtain concentration of antibacterial from pineapple peel extract on Enterococcus faecalis growth. Methods: This study was an experimental laboratory with Post Test Only control group design using Enterococcus faecalis ATCC 29212 bacteria that were diluted based on Mc. Farland standard 1.5 x 108 CFU / ml with pineapple peel extract treatment concentration of 100%, 50%, 25%, 12.5%, 6.25%, 3.125%, 1.56%, 0, 78% and then planted in nutrient agar media surface evenly. Results: At concentration of 3.125% pineapple peel extract showed that the growth of colonies was less than 90% of positive control and concentration of 6.25% pineapple peel extract had no visible Enterococcus faecalis bacteria growth as much as 99.9%. Conclusion: The pineapple peel extract (Ananas comosus) have antibacterial power on the growth of Enterococcus faecalis bacterial colonies with Minimal Inhibitory Concentration (MIC) of 3.125% and the Minimum Bactericidal Concentration (MBC) of 6.25%.

Abstract: Generally, failures in root canal treatment are caused by the bacterium Enterococcus faecalis. Several studies using different types of herbs showed their inhibition effect on the growth of bacteria in the oral cavity. Coconut oil can be processed into pure coconut oil (virgin coconut oil, VCO) that contains lauric acid with its antibacterial effect. This study was aimed to determine the inhibitory effect of coconut oil on the growth of the bacteria Enterococcus faecalis. This was an experimental laboratory study with the post-test only control group design. We used a modified method of the Kirby-Bauer disk paper. Sample of VCO was made by using heating process. Enterococcus faecalis bacteria obtained directly from the patients’ necrotic pulps were identified at the Pharmaceutical Microbiology Laboratory, University of Sam Ratulangi Manado. The results showed that Enterococcus faecalis bacteria were identified in the samples. The inhibition zone of VCO to Enterococcus faecalis was 10 mm. Conclusion: Virgin coconut oil could inhibit the growth of Enterococcus faecalis.Keywords: virgin coconut oil (Virgin coconut oil), Enterococcus faecalis, inhibition zoneAbstrak: Pada perawatan saluran akar dapat ditemukan kegagalan perawatan yang disebabkan oleh bakteri Enterococcus faecalis. Terdapat beberapa penelitian mengenai berbagai jenis tumbuhan herbal yang telah dilakukan untuk menghambat pertumbuhan bakteri dalam rongga mulut, salah satunya yaitu tanaman kelapa yang dapat diolah menjadi minyak kelapa murni (virgin coconut oil, VCO) yang mengandung senyawa aktif asam laurat dengan efek antibakteri. Penelitian ini bertujuan untuk mengetahui daya hambat VCO terhadap pertumbuhan bakteri Enterococcus faecalis. Jenis penelitian ialah eksperimental laboratorium dengan post-test only control group design. Metode pengujian menggunakan metode modifikasi Kirby-Bauer dengan paper disk. Sampel minyak kelapa murni dibuat sendiri dengan proses pemanasan. Bakteri Enterococcus faecalis diambil langsung dari pasien nekrosis pulpa lalu diidentifikasi di Laboratorium Mikrobiologi Farmasi FMIPA Universitas Sam Ratulangi Manado. Hasil penelitian menunjukkan adanya bakteri Enterococcus faecalis pada sampel hasil identifikasi. Dari hasil uji daya hambat didapatkan minyak kelapa murni (Virgin coconut oil) memiliki efek antibakteri dalam menghambat pertumbuhan Enterococcus faecalis dengan zona hambat sebesar 10 mm. Simpulan: Virgin coconut oil dapat menghambat pertumbuhan bakteri Enterococcus faecalis.Kata kunci: VCO, Enterococcus faecalis, zona hambat
