<p style="text-align: justify;">Wine contains natural plant phenolic antioxidants that may protect circulating lipoproteins from oxidative damage. By inhibition of the copper-catalyzed oxidation of LDL we determined the activity of thirteen Rhône coast wines exclusively from Syrah and Grenache varieties. About 50 p. cent of the wines were made with a long maceration process. Major monomeric phenolic compounds and procyanidin dimers were analyzed in each sample by HPLC and correlated with relative LDL antioxidant activity. Correlations obtained can be grouped in 3 classes: catechin (r = 0.75), procyanidins, BI, B2, B3 (r = 0.43-0.55), malvidin-3-glucoside and cyanidin (r = 0.43), gallic acid, myreeitin, rutin (r = 0.2-0.4). On the same basis total phenol contents of wines gave a correlation with LDL antioxidant activity of r = 0.72. Comparison, at the same total phenol concentration, with different red California wines shows that antioxidant activity of French Syrah and Grenache range between that of Merlot (56-65 p. cent) and Cabernet Sauvignon (37-45 p.cent). In contrast Syrah wines made with a short extraction process gave lower inhibition of LDL oxidation of 16 p. cent which is less than white Califomia wines averaging 36 p. cent. Activity of each wine phenolic compound can play a role in protecting LDL against oxidation.</p>

Biofarmasi 14: 1-9. Curcuma (Curcuma xanthorrhiza Roxb.) is a type of drug plant that has high enough capacity of production in Indonesia. Generally, commerced in the form of fresh curcuma or processed product as simple as simplicia and curcuma powder. Processed products that could be developed is curcuma oleoresin. It is a mixture of essential oils and resins obtained from extraction process of curcuma powder using an organic solvent. Oleoresin has the same flavor and aroma to the extracted material. Due to these characteristics, it is used as a flavor and food coloring, other than as a raw material in pharmaceutical industry. In addition, it also contains active compounds which can support the utilization of drug and food industries. This study aims to determine whether the size of the powder, powdered curcuma immersion time, and interactions between them that can be influenced the content of curcuminoids, total phenol and antioxidant activity of curcuma oleoresin. Selection of solar dryers in the drying process is based on previously studied that compare the natural drying technique with a solar dryer, and the best results of those studies are shown in the solar dryer. This research using completely randomized design with two factors: the size variation of curcuma powder (60, 80 and 100 mesh) and immersion time variation (extraction) of curcuma powder (12, 24 and 36 hours). The results showed that the powder size of curcuma and immersion time has an effect on curcuminoid content, total phenol and antioxidant activity of curcuma oleoresin. However, there are no interaction between both factors. That is, the size and the immersion time of curcuma powder do not affect each other on the content of curcuminoid, total phenol and activity of antioxidant.

Brewers’ spent grain (BSG) is the most abundant by-product of the brewing process. BSG is currently disposed of or used as a supplement for animal feed, although it contains significant amounts of bioactive compounds of great interest to the pharmaceutical, cosmetic and food sectors. In this study we investigate the feasibility of using a simple solvent extraction procedure to recover phenolic antioxidants from BSG. Acetone–water and ethanol–water mixtures were used as extraction solvents. Phenolic extracts obtained by treatment of BSG with the two solvent systems were characterized in terms of total phenolics and antioxidant activity. For both systems, the extraction yield was maximum at 60% (v/v) organic solvent concentration. At all solvent compositions, mixtures containing acetone provided higher extraction yields. As suggested by the strong correlation between the antioxidant activity of BSG extracts and their phenolic content, the antioxidant capacity of the extracts can be mainly attributed to polyphenols. Overall, the obtained results strongly support the exploitation of BSG as a source of phenolic antioxidants and the possibility of recovering them by a mild and green extraction process.

Background: Microalgae are the source of various compounds with high potentials for being used in different industries. The production of such compounds can be raised under extreme conditions. In the present study, four cyanobacteria and one coccoid green alga were examined which were isolated from hot springs in high background radiation areas in Ramsar, a city in the north of Iran. Methods: Cadmium adsorption from aqueous solution, response towards cadmium stress, antioxidant activity, total phenolic compound and drought tolerance were investigated in these microalgae. Results: The results showed that these extremophile microalgae contain valuable biological compounds which can be useful in remediation of heavy metals from contaminated water and soils and pharmaceutical applications. The unicellular cyanobacterium, Chroococidiopsis thermalis IBRC-M50002, was the best strain with the highest biological activity in various testes such as cadmium adsorption (225 mg g-1), cadmium tolerance stress (100 mg ml-1), antioxidant activity (IC50= 18 μg mg-1) and total phenol content (100 μg ml-1). The coccoid green algae Grasiella emersonii IBRC-M50001, also exhibited significant antioxidant activity (IC50=10 μg mg-1) and total phenol compound (116 μg ml-1), but its cadmium adsorption, tolerance at cadmium stress and desiccation were lower than Chroococidiopsis thermalis. Conclusion: HBRAs microalgae, isolated from extreme conditions, are useful microorganisms for the production of bioactive substances and natural antioxidants. In other words, they exhibited high capacity to be used in pharmaceutical, industrial and commercial applications.

AbstractThe aim of this work is the optimization of phenolic compound extraction from three by-products of banana crops (rachis, discarded banana, and banana’s pseudostem pulp), as a way to valorize them through a green extraction process. The influence of the temperature and aqueous ethanol concentration (Et-OH) on extract properties (total phenol content (TPC) and antioxidant activity) was firstly analyzed. 78 ℃ and ethanol concentrations close to 50% yielded the best results for the three materials. The equations obtained by the response surface methodology gave a satisfactory description of the experimental data, allowing optimizing the extraction conditions. Under optimized conditions, time influence was then assessed, although this parameter seemed not influence results. Among the three by-products, rachis extract (60% Et-OH, 78 ℃, and 30 min) presented the highest TPC (796 mg gallic acid/100 g of dried material) and antioxidant activity (6.51 mg Trolox equivalents/g of dried material), followed by discarded banana, and pseudostem pulp. Under the optimal conditions, experiments were performed at a larger scale, allowing to determine the extraction yields (EY) and to characterize the extracts. The highest EY was obtained for the rachis (26%), but the extract with the highest activity was obtained for discarded banana (50% Et-OH, 78 ℃, and 60 min), which presented a TPC of 27.26 mg/g extract corresponding to 54.59 mg Trolox equivalents/g extract. This study contributes to the valorization of banana crops residues as a source of polyphenolic compounds with bioactive functions that can be extracted under economic extraction conditions. This research was conducted to identify the phytochemical compounds and antioxidant activity of pepe leaves (Gymnema reticulatum Br.) crude extract in various solvent types. The experimental design used in this research was a completely randomized design with solvent type treatments consisting of 5 types : aquadest, ethyl acetate 96%, acetone 70%, ethanol 70%, and methanol 95%. The treatment was repeated three times, so that 15 experimental units were obtained. Qualitative data obtained in this research were displayed with tables, while quantitative data were analyzed by variance and if the treatment had a significant effect, then followed by Duncan's test. The results showed that the treatment of solvent types affected phytochemical compounds that was extracted from pepe leaves and had a significant effect (P<0.01) on yield, total phenol, total flavonoids, and antioxidant activity. The best results showed that 70% acetone solvents were able to extract phytochemical compounds from alkaloids, saponins, phenols, flavonoids, and steroids in the form of sterols and had the highest antioxidant activity based on 62.06% free radical inhibition percentage with IC50 values of 200.775 mg/L, yield 32.35%, total phenol 36.14 mgGAE/g extract, and total flavonoids 50.37 mgQE/g extract.

Chlorogenic acid has been proved to have cardiovascular protection, antibacterial, antiviral, hemostatic, and hypolipidemia effects. Modern scientific research on the bioactivity of chlorogenic acid has been extended to the fields of food, medicine, health care and daily-use chemical industry. The aim of this research was to optimize the extraction conditions for chlorogenic acid from Eucommia ulmoides (Eucommiaceae) leaves. The significant variables were screened and optimized by a combination of Plackett-Burman test and Box-Behnken design. Optimum extraction parameters with ethanol concentration of 50%, solvent pH value of 3, and particle size of 60 mesh were determined according to variance analysis and contour plots. Under these conditions, the yield of chlorogenic acid was up to 4.36 mg/g, which was basically consistent with the theoretical prediction value of 4.50 mg/g. This study also proved the potential antioxidant activity of E. ulmoides leaves. The optimal extract of E. ulmoides leaves rich in chlorogenic acid showed the highest antioxidant activity in the FRAP method, which was 219.8 μM Trolox equivalents (TE) per g extract weight (EW) (μM TE/g EW). The DPPH method gave a similar value (168 μM TE/g EW) to the ABTS method (152 μM TE/g EW). The established extraction process was efficient in the recovery of chlorogenic acid from E. ulmoides leaves, encouraging its valorization as a cheap and sustainable alternative for the isolation of chlorogenic acid.

ABSTRACTNanamuha (Bridelia monoica Merr.) is a plant that was used as a cancer and tumor drug by the Sangihe Islands community. This plant contains phenol compounds, flavonoids, alkaloids and tannins. The purpose of this study was to determine the potential of nanamuha leaves that grow in the Sangihe Island to have antioxidant activity and determine the total phenolic ethanol extracts of nanamuha leaf. The extraction method used is maceration with ethanol solvent p.a. The extracts obtained was determined total phenolic and antioxidant activity test using DPPH method. Based on the results obtained, the ethanol extracts of nanamuha leaves has antioxidant activity with an IC50 value of 75,03 µg / mL and the result showed that the total phenol content the ethanol extracts of nanamuha leaves of  53,34 mg / L.  Keywords : Nanamuha leaf, total phenol, antioxidant activity, DPPH  ABSTRAKNanamuha (Bridelia monoica Merr.) merupakan tanaman yang dimanfaatkan sebagai obat kanker dan tumor oleh masyarakat Kepulauan Sangihe. Metode ekstraksi yang digunakan adalah maserasi dengan pelarut etanol p.a. Ekstrak yang diperoleh ditentukan total fenolik dan uji aktivitas antioksidan dengan metode DPPH. Berdasarkan hasil yang didapatkan ekstrak etanol daun nanamuha memiliki aktivitas antioksidan dengan nilai IC50 75,03 µg/mL dan hasil penelitian menunjukan total kandungan fenolik ekstrak etanol daun nanamuha sebesar 53,34 mg/L. Kata kunci : Daun Nanamuha, Total fenol, Aktivitas antioksidan, DPPH

In Peru, Caulerpa filiformis is a marine algae listed as an invasive species. For years, its distribution has been considered to be in the north coast (Isla Lobos de Afuera and Piura) until a recent report of its distribution in the central coast (Ancash, Lima, and Ica). The present investigation aims to determine the main groups of secondary metabolites, total phenol content, and antioxidant activity of the methanolic extract of C. filiformis from Sechura Bay (Piura) and Paracas Bay (Ica). The main chemical groups were determined through phytochemical screening, the content of phenols by the Folin–Ciocalteu method, and antioxidant activity by the ABTS method (2,2-azinobis-[3-ethylbenzothiazoline-6-sulfonic acid]) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH). The phytochemical screening of the methanolic extract of C. filiformis from Sechura Bay and Paracas Bay revealed the presence of carbohydrates, polyphenols, tannins, flavonoids, lipids, alkaloids, steroids, and triterpenes for both extracts. The total phenol content of the extract of C. filiformis from Sechura Bay (39.31 ± 0.39 mg of AGE/g extract) was significantly higher (p < 0.05) than that from Paracas Bay (18.78 ± 0.31 mg of AGE/g extract). In the ABTS and DPPH assays, the antioxidant capacity of the Sechura C. filiformis extract (IC50 = 3.49 ± 0.01 and 2.18 ± 0.02 mg/mL) was significantly higher (p < 0.05) than that of the Paracas C. filiformis extract (IC50 = 6.41 ± 0.02 and 2.42 ± 0.04 mg /mL). These findings suggest that the methanolic extract of C. filiformis is a source of secondary metabolites with an antioxidant potential.
