Indonesian traditional drink such as Sarabba is generally consumed for its taste. However, this drink has certain functional properties which originated from its basic ingredients. To enhance its functional properties, other natural ingredients such as black rice extract can be added to the basic ingredients. Therefore, the objectives of these study were to find out the best treatment for the use of black rice extract in producing of Sarabba and the effect of drying time on proximate value, anthocyanin, and thiamine contents; and solubility level of the product. Black rice is one type of local varieties that contains anthocyanins, a group of flavonoids, which is beneficial to health. This research used black rice as basic ingredient in making instant “Sarabba”. Sarabbais one traditional drink from South Sulawesi which is made from brown sugar, ginger and coconut milk. The aim of this research was to determine the best treatment of the utilization of black rice extract in making “sarraba” instant. Sarabbawas prepared with ratio of black rice and water, 50%: 50%; 30%: 70%; 70%: 30 %. The best treatment was then subjected to drying process at different times, 1 hour, 1.5 hours, 2 hours to determine the effect of drying time on proximate, anthocyanin, thiamine and solubility content. The sensory value and solubility results obtained were treated with quantitative descriptive with three replications. The best result of sensory evaluation based on color, taste, odor and texture was found in treatment 70% rice:30% water. The result showed that best content of water, carbohydrate and solubility level were obtained at two hours drying which were 1.67%, 0.11% and 20 seconds, respectively. The best content of ash, anthocyanin, thiamin and fat content were obtained during one hour drying which were 2.3%, 11.83%, 0.056% and 0.03, respectively. The highest protein content was obtained from of 1.5 hours drying treatment (4.78%). Recommended Sarabbainstant processing was ratio of black rice 70% : water 30% and one hour drying.

Abstract: The objective of this research is to determine the antioxidant activity of the extracts of black rice cultivars Pare Ambo, South Sulawesi. Black rice Pare Ambo cultivars, South Sulawesi extracted with 70% ethanol by maceration. After that, the extract tested antioxidant activity using DPPH with concentration 50, 100, 200, 400, 500 ppm, and methods FRAP for the determination of total content of antioxidants at concentrations 600, 700, 800, 900, 1000 ppm. Research results using DPPH antioxidant test showed a concentration of 50 ppm has an activity that is the lowest free-radical scavengers 22% and 400 ppm had a prophylactic activity of free radicals highest 84,60%. While FRAP method showed a concentration of 600 ppm had the lowest total antioxidant that is 177,64 mmol/g and a concentration of 1000 ppm has the highest total antioxidant that is 272,94 mmol/g. Results of this research concluded that the extract of black rice cultivars Pare Ambo, South Sulawesi has a good potential antioxidant activity.Keywords: black rice, extract, antioxidants, DPPH, FRAPAbstrak: Tujuan penelitian ini adalah untuk menentukan aktivitas antioksidan dari ekstrak beras hitam kultivar Pare Ambo, Sulawesi Selatan. Setelah itu, ekstrak diuji aktivitas antioksidan menggunakan metode DPPH dengan konsentrasi 50, 100, 200, 400, 500 ppm, dan metode FRAP untuk penentuan kandungan total antioksidan dengan konsentrasi 600, 700, 800, 900, 1000 ppm. Hasil penelitian pengujian antioksidan menggunakan metode DPPH menunjukkan konsentrasi 50 ppm memiliki aktivitas penangkal radikal bebas terendah yaitu 22% dan konsentrasi 400 ppm memiliki aktivitas penangkal radikal bebas tertinggi yaitu 84,60%. Sedangkan metode FRAP menunjukkan konsentrasi 600 ppm memiliki total antioksidan paling rendah yaitu 177,64 mmol/g dan konsentrasi 1000 ppm memiliki total antioksidan paling tinggi yaitu 272,94 mmol/g. The present study aimed to assess the drying kinetics of black rice and fit different mathematical models (empirical and diffusive) to the experimental data, and evaluate the effect of drying air temperature on the physical-chemical and bioactive compounds quality of black rice. Drying air temperatures ranged from 40 to 80 ºC and the drying air speed was 1.5 m/s. Physical-chemical characterization of the product was based on the following parameters: moisture, water activity, ashes, total protein content, pH, total acidity, lipids, total carbohydrates, total anthocyanins, flavonoids, total phenolic compounds and antioxidant activity. Among the empirical models, Page showed the lowest mean squared deviations (MSD) and highest coefficients of determination (R2). For the diffusion model, the values of effective mass diffusivity and convective heat transfer coefficient increased with increasing drying air temperature, and the Biot number indicated that the first-type boundary condition would also describe well the drying process. Physical-chemical parameters and bioactive compounds differed between the temperatures used, and the temperature of 60 ºC led to the best relationship between drying time and preservation of product characteristics.

Objective: Based on previous research, black rice (Oryza sativa L.) has been proven to have antioxidant activity which is classified in the very strong category, approximately 41.5 bpj. this The aim of this study was to produce antioxidant functional drink.Methods: Simplicia powder prepared from black rice (O. sativa L.) sample by extracting with 70% ethanol solvent using kinetic maceration, followed by phytochemical screening, determination of extract quality, drying extract with freeze-drying method, then dry extract obtained used to produce functional drinks on which evaluation and antioxidant activity test carried out. The antioxidant activity test used in this research was DPPH-free radical scavenging method.Results: Screening result showed that both simplicia and extract contain flavonoid compounds, saponins, tannins, steroids, and triterpenoids. Result from dried extract quality determination shown to be thick consistency, has blackish-purple color, has specific aroma and bitter, water-soluble extract content of 59.04%; ethanol-soluble extract content of 69.77%; moisture content of 7.65%; loss on drying 9.40%; total ash content of 5.17%; acid-insoluble ash content of 0.84%; water-soluble ash content 4.15%; and residual solvent 0.44%. Pb and Cd metal contamination were 1.1994 mg/kg and 0.0905 mg/kg, respectively. Total plate count microbial contamination TNTC and the number of yeast and mold found to be 0.0387×103 colony/g. Evaluation test of functional drinks with powder type and found to be blackish-purple, and specific aroma, and total plate count microbial contamination was 0.310×103 colony/g, and the number of yeast and mold found was 0.6997×102 colony/g. Result of IC50 antioxidant activity of viscous extract was 37.5587 bpj; dried extract was 231.0101 bpj; dried extract (distilled water) was 44.7412 bpj; and functional drinks were 46.3594 bpj.Conclusion: Thus, results suggest that dried black rice’s extract and functional drink produced found to have very strong antioxidant activity.

Abstract: Rice is the main type of food that is very essential to the people of Asia, especially in Indonesia. Color pigments, which are located on the outer layer of black rice paddy, have been reported to contain a mixture of anthocyanin compounds. Anthocyanin compounds act as antioxidants and free radical catchers, which serve to prevent some degenerative diseases. Black rice contains a nutritional value that is superior to other types of rice. It includes high content of protein, vitamins and minerals although there are distinct varieties depending on the location of production. This research has intended to determine the description of the activity and stability of the antioxidant from black rice extract (Oryza sativa L.) cultivars in Enrekang, South Sulawesi at 100 degrees Celsius heating with the periods of 0, 10, 20 and 30 minutes, and the antioxidant activity has been tested using DPPH and FRAP methods. Based on the results of the research using DPPH method, it was discovered that the activity of radical scavengers increased by the effect of the temperature from extracts of the black rice cultivar in Enrekang heated for 0, 10, 20, and 30 minutes at a temperature of 100 ° C and the antioxidant activity of the extracts of the black rice was relatively stable against heating that was tested using FRAP.Keywords: black rice, antioxidants, DPPH, FRAPAbstrak: Beras merupakan jenis pangan utama yang penting bagi masyarakat di Asia, terutama di Indonesia. Pigmen warna yang terletak pada lapisan luar padi dari beras hitam telah dilaporkan mengandung campuran senyawa antosianin, senyawa antosianin berfungsi sebagai antioksidan dan penangkap radikal bebas, sehingga berperan untuk mencegah terjadinya beberapa penyakit degeneratif. Beras hitam memiliki kandungan nilai gizi yang lebih unggul dari beras jenis lainya. Seperti kandungan protein yang tinggi, vitamin dan mineral. Meskipun terdapat perbedaan varietas tergantung pada lokasi produksinya. Penelitian ini bertujuan untuk mengetahui gambaran aktivitas dan stabilitas antioksidan dari ekstrak beras hitam (Oryza sativa L.) kultivar Enrekang, Sulawesi Selatan pada pemanasan 100oc dengan lama pemanasan 0, 10, 20, dan 30 menit dan diuji aktivitas antioksidan menggunakan metode DPPH dan FRAP. Berdasarkan hasil penelitian menggunakan metode DPPH, didapatkan bahwa aktivitas penangkal radikal bebas meningkat dengan efek pemanasan pada ekstrak beras hitam (Oryza sativa L.) kultivar Enrekang Sulawesi Selatan dengan pemanasan 0, 10, 20, dan 30 menit pada suhu 100oC. Dan aktivitas antioksidan pada ekstrak beras hitam relative stabil terhadap pemanasan yang diuji menggunakan metode FRAP.Kata kunci: beras hitam, antioksidan, DPPH, FRAP

Abstract: Rice is a staple food that is very important in the world, including in Indonesia. The kind of rice that is generally consumed by people in Indonesia is white rice, brown rice, and black rice. Black rice is one type of rice which is popular in the community and consumed as functional foods because of health benefits. The purpose of this study is to describe the nutrient contents of the black rice cultivars ambo Pare, South Sulawesi. This research was conducted by using descriptive method with samples of black rice originally from Toraja. The results of research which conducted at the Laboratory of Integrated Sam Ratulangi University and the Center for Environmental Health Engineering Manado are 85% carbohydrates, 1.9% fat, 1.04% protein, 10.5% water, fiber 0.8%, Abu by 0.4%, which amount of 0.6 mg of Vitamin C, 31.6 mg of Vitamin E, 0.386 mg / ml of Calcium (Ca), 1.95 mg / ml of Magnesium (Mg), 0.886 mg / ml of Potassium (K), 0,391mg / ml of iron (Fe), and 0.021 mg / ml of Zinc (Zn). Beras yang umumnya dikonsumsi oleh masyarakat di Indonesia adalah beras putih, beras merah, dan beras hitam. Beras hitam merupakan salah satu jenis beras yang mulai populer di masyarakat dan dikonsumsi sebagai pangan fungsional karena bermanfaat bagi kesehatan. Tujuan penelitian ini adalah untuk mengetahui gambaran kandungan zat gizi pada beras hitam Kultivar pare ambo, Sulawesi Selatan. Penelitian ini dilakukan dengan menggunakan metode deskriptif dengan sampel beras hitam yang berasal dari Toraja. Berdasarkan hasil penelitian yang dilakukan di Laboratorium Terpadu Universitas Sam Ratulangi dan Balai Teknik Kesehatan Lingkungan Manado diperoleh hasil kandungan zat gizi antara lain Karbohidrat sebesar 85%, Lemak sebesar 1,9%, Protein sebesar 1,04%, Air sebesar 10,5%, Serat sebesar 0,8%, Abu sebesar 0,4%, Vitamin C sebesar 0,6 mg, Vitamin E sebesar 31,6 mg, Kalsium (Ca) sebesar 0,386 mg/ml, Magnesium (Mg) sebesar 1,95 mg/ml, Kalium (K) sebesar 0,886 mg/ml, Besi (Fe) sebesar 0,391mg/ml, dan Zinc (Zn) sebesar 0,021 mg/ml. Teh dan rempah memiliki kandungan antioksidan yang berguna untuk kesehatan. Tujuan dari pembelajaran ini adalah untuk mendapatkan formula minuman fungsional teh dan rempah terbaik. Penelitian ini menggunakan mixture design untuk menentukan formula optimum minuman fungsional terbaik yang terdiri dari gula merah, bubuk kayu manis dan bubuk kapulaga. Berdasarkan input batas atas dan batas bawah dari bahan baku sehingga didapatkan 16 formulasi yang akan diteliti. Untuk batas atas gula merah yaitu 40 dan batas bawah gula merah yaitu 35. Untuk batas atas bubuk kayu manis yaitu 15 dan batas bawah kayu manis yaitu 10. Untuk batas atas bubuk kapulaga yaitu 50 dan batas bawah bubuk kapulaga yaitu 45. Untuk menentukan kualitas minuman fungsional dilakukan uji kimia (uji karbohidrat, gula total dan nilai pH, uji fisik (Uji viskositas), uji organoleptik (uji hedonik untuk warna, aroma, rasa dan kekentalan) dan mutu hedonik yang ditentukan dari kesukaan panelis terhadap warna, aroma, rasa dan kekentalan) dan uji antioksidan. Penelitian ini untuk mendapatkan formula optimum yang terbaik dari hasil pengacakan oleh mixture design. Hasil dari pengolahan mixture design adalah anova, grafik dan duncan. Berdasarkan hasil penelitian formula minuman fungsional yang masih dapat diterima adalah formulasi 11 yang terdiri dari 40% gula merah, 12.67% bubuk kayu manis dan 47.32% bubuk kapulaga. Formula 11 mengandung 0.39% karbohidrat, 15.46% gula total, 5.38% pH 4.98%  viskositas dan 255.41 mg antoksidanSpice tea have antioxidants compounds that are beneficial for health. The purpose of this study was to get the best spice tea formulation as a functional drink. This research used a mixture design to obtain the best optimum formula functional drink that composed by brown sugar, cinnamon powder and cardamom powder. Based on input the upper and lower limits of raw materials then get 16 formulations will be researched. The upper limit of brown sugar, cinnamon powder and cardamom powder is 40, 15 and 50. The lower limit of brown sugar, cinnamon powder and cardamom powder is 35, 10 and 45. The functional drink quality determined by chemical test (carbohydrate level, total sugar and pH value), physical test (viscosity level), organoleptic test and antioxidan level. Data was processed statistically using Design Expert application with one-way analysis (one way ANOVA) at 95% confidence level. Duncan`s follow-up was carried out to find out the differences between treatments if ANOVA had a significant effect. Based on the results of the research, formulation functional drink are still aceptable is formulation 11 of 40% brown sugar, 12.67% cinnamon powder and 47.32% cardamom powder. Formulation 11 contained carbohydrate level 0.39%, total sugar 15.46% and pH value 5.38%, viscosity level 4.98%, and antioxidan level 255.41 mg

Anthocyanins belong to the group of flavonoid compounds broadly distributed in plant species responsible for attractive colors. In black rice (Oryza sativa L.), they are present in the stems, leaves, stigmas, and caryopsis. However, there is still no scientific evidence supporting the existence of compartmentalization and trafficking of anthocyanin inside the cells. In the current study, we took advantage of autofluorescence with anthocyanin’s unique excitation/emission properties to elucidate the subcellular localization of anthocyanin and report on the in planta characterization of anthocyanin prevacuolar vesicles (APV) and anthocyanic vacuolar inclusion (AVI) structure. Protoplasts were isolated from the stigma of black and brown rice and imaging using a confocal microscope. Our result showed the fluorescence displaying magenta color in purple stigma and no fluorescence in white stigma when excitation was provided by a helium–neon 552 nm and emission long pass 610–670 nm laser. The fluorescence was distributed throughout the cell, mainly in the central vacuole. Fluorescent images revealed two pools of anthocyanin inside the cells. The diffuse pools were largely found inside the vacuole lumen, while the body structures could be observed mostly inside the cytoplasm (APV) and slightly inside the vacuole (AVI) with different shapes, sizes, and color intensity. Based on their sizes, AVI could be grouped into small (Ф < 0.5 um), middle (Ф between 0.5 and 1 um), and large size (Ф > 1 um). Together, these results provided evidence about the sequestration and trafficking of anthocyanin from the cytoplasm to the central vacuole and the existence of different transport mechanisms of anthocyanin. Our results suggest that stigma cells are an excellent system for in vivo studying of anthocyanin in rice and provide a good foundation for understanding anthocyanin metabolism in plants, sequestration, and trafficking in black rice.
