Abstract Background Osteomyelitis caused by Staphylococcus aureus (S. aureus) is an important post-operation complication, especially after fracture internal fixation and artificial joint replacement. Animal models play an indispensable role in exploring the pathogenesis of osteomyelitis. Most models use internal fixation, bacterial suspension and vascular sclerosing agent to destroy blood vessels. Vascular sclerosing agents not only damage blood vessels but also lead to local inflammatory immune disorders, which is different from simple vascular disease and osteomyelitis caused by ischemia in clinical practice. Methods The experimental animals were randomly divided into three groups: femoral artery ligation group, vascular sclerosing agent group and non-infection aseptic operation group. In the femoral artery ligation group, the femoral artery was ligated to reduce the blood flow of the affected limb to simulate the clinical ischemic state and increase the susceptibility, then the Kirschner needle with S. aureus biofilm was implanted into the tibia of rats, and the bone defect was sealed with aseptic paraffin. The non-infection aseptic operation group and the infection model group caused by vascular sclerosing agent have been used as the blank and positive control group. After operation, survival rate, body temperature and incision healing were monitored. Four weeks later, radiological and pathological changes of all animals were evaluated, and the secretions from osteomyelitis experienced etiological separation and cultivation. Results The chronic osteomyelitis model was established successfully by ligating femoral artery and implanting Kirschner needle covered with S. aureus biofilm. Signs of chronic osteomyelitis were observed in all rats of femoral artery ligation infection group and positive control group. No signs of infection and chronic osteomyelitis were found in the non-infection aseptic operation control group. Conclusion The method of ligating the femoral artery and implanting Kirschner needle with S. aureus biofilm into the tibia of rats can effectively establish a stable and reproducible chronic osteomyelitis model which is closer to the clinical pathogenesis and natural route of infection. This model could be useful for the study of pathogenesis and therapeutics of chronic osteomyelitis with S. aureus.

The prevalence of infectious diseases in Indonesia is still relatively high, for example Infection caused by Staphylococcus aureus. Indonesia has many traditional plants that can be used for herbal treatments containing tannins, flavonoids, and essential oils which have relatively high antibacterial activity by forming inhibitory zones in the Staphylococcus aureus bacteria which are the cause of skin infections food poisoning to systemic infections. Thereforekencur can be used as an alternative medicine. This study was intented to determine the effect of giving kencur extract (Kaempferia galanga L.) In inhibiting the growth of Staphylococcus aureus bacteria. This study was experimental laboratory used plate diffusion method of “Postest Ony Control Group Design” design. There were 7 groups, 5 treatment groups and 2 control groups. The concentration of treatment group was 100%, 50%, 25%, 12,5%, 6,25% while for positive control using amoxicillin 25μg and aquadest as negative control. The bacterial used were Staphylococcus aureus. The results showed that kencur extract (Kaempferia galanga L.) had an antibacterial effect in inhibiting the growth of Staphylococcus aureus bacteria.With a value of p = 0.001 or p <α. kencur extract (Kaempferia galanga L.) has an antibacterial effect in inhibiting the growth of Staphylococcus aureus bacteria.

Methicillin-resistant Staphylococcus aureus (MRSA) can grow as structured biofilm in different surfaces, including oral mucosa and denture surfaces. Such biofilms can be released into the oral fluids and aspirated, causing systemic infections such as aspiration pneumonia. This study evaluated the efficacy of two disinfectant solutions and microwave irradiation in disinfecting acrylic specimens contaminated with MRSA biofilm. Thirty-six acrylic specimens were made, sterilized and contaminated with MRSA (107 cfu/mL). After incubation (37 °C/48 h), the specimens were divided into 4 groups: not disinfected (positive control); soaking in 1% sodium hypochlorite for 10 min; soaking in 2% chlorhexidine gluconate for 10 min; and irradiating by microwave for 3 min at 650 W. The viability of cells was evaluated by XTT reduction method. All specimens from the positive control group showed biofilm formation after 48 h incubation. The mean absorbance value of the control specimens was 1.58 (OD at 492 nm). No evidence of biofilm formation was observed on specimens after the disinfection methods. Disinfection by soaking in 1% sodium hypochlorite and 2% chlorhexidine gluconate and irradiating by microwaves resulted in 100% reduction of MRSA biofilm metabolism. The use of chemical solutions and microwave irradiation was shown to be effective for eradicating mature MRSA biofilms on acrylic resin specimens.

Background: Resistance is defined as growth inhibition of bacteria with antibiotics. Staphylococcus aureus bacteria is known to be resistant to gentamicin and methicillin. Staphylococcus aureus can cause various infection of the skin, injuries, and system. Methods: This research is true-experimental with Posttest-Only Control Group Design and using Kirby-Bauer method by means of five concentration (20, 40, 60, 80, dan 100%). The negative control uses sterile distilled water, and the positive control uses Vancomycin 30 µg. Result: This research showed the mean of inhibition zone diameter of each concentration consecutively 16 mm, 20 mm, 22 mm, 23 mm, and 23 mm. Oneway ANOVA statistic analysis showed that the value of p is 0,000 that mean there is an influence of encok roots extract to Staphylococcus aureus growth. Conclusion: There is an influence of encok roots extract to Staphylococcus aureus growth. Seen from its ability to inhibit the growth of Staphylococcus aureus.

Abstract Purpose Treatment of chronic osteomyelitis (bone infection) remains a clinical challenge. Our previous study had demonstrated that NEMO-binding domain (NBD) peptide effectively ameliorates the inhibition of osteoblast differentiation by TNF-α in vitro. In this work, NBD peptide was evaluated in vivo for treating chronic osteomyelitis induced by methicillin-resistant Staphylococcus aureus (MRSA) in a rabbit model. Methods Tibial osteomyelitis was induced in 50 New Zealand white rabbits by tibial canal inoculation of MRSA strain. After 3 weeks, 45 rabbits with osteomyelitis were randomly divided into four groups that correspondingly received the following interventions: 1) Control group (9 rabbits, no treatment); 2) Van group (12 rabbits, debridement and parenteral treatment with vancomycin); 3) NBD + Van group (12 rabbits, debridement and local NBD peptide injection, plus parenteral treatment with vancomycin); 4) NBD group (12 rabbits, debridement and local NBD peptide injection). Blood samples were collected weekly for the measurement of leucocyte count, erythrocyte sedimentation rate (ESR), and C-reactive protein (CRP) levels. The rabbits in all four groups were sacrificed 6 weeks after debridement; the anti-infective efficacy was evaluated by radiological, histological, and microbiological examination, and promotion of bone remodeling was quantified by micro-CT using the newly formed bone. Results Except two rabbits in the Control group and one in the NBD group that died from severe infection before the end point, the remaining 42 animals (7, 12, 12, 11 in the Control, Van, NBD + Van, and NBD group respectively) were sacrificed 6 weeks after debridement. In general, there was no significant difference in the leucocyte count, and ESR and CRP levels, although there were fluctuations throughout the follow-up period after debridement. MRSA was still detectable in bone tissue samples of all animals. Interestingly, treatment with NBD peptide plus vancomycin significantly reduced radiological and histological severity scores compared to that in other groups. The best therapeutic efficacy in bone defect repair was observed in the NBD peptide + Van group. Conclusions In a model of osteomyelitis induced by MRSA, despite the failure in demonstrating antibacterial effectiveness of NBD peptide in vivo, the results suggest antibiotics in conjunction with NBD peptide to possibly have promising therapeutic potential in osteomyelitis.

Abstract: Synthetic antibiotics have certain side effects, therefore, it is necessary to find alternative natural antibacterial materials which is easily available and to be cultivated, inter alia Amaranthus hybridus L. Its leaves contain active compounds, so they might have antibacterial potential. This study was aimed to determine the inhibitory effect of Amaranthus hybridus leaf extract on the growth of Staphylococcus aureus. This was a true experimental study using post test only control group design. This study was conducted at the Microbiology Laboratory of Faculty of Medicine and the Natural Phytochemical Laboratory of Faculty of Mathematics at Sam Ratulangi University. The modified Kirby-Bauer method was used with three wells, containing Amaranthus hybridus leaf extract, the positive control, and the negative control; and with 5 repetitions. The results showed that the average diameters of the inhibition zone of Amaranthus hybridus leaf extract and of the negative control were 0 mm meanwhile of erythromycin as the positive control was 38.8 mm. Conclusion: Amaranthus hybridus leaf extract had no inhibitory effect on Staphylococcus aureus. Daun bayam petik memiliki potensi antibakteri karena memiliki kandungan senyawa aktif. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak daun bayam petik terhadap pertumbuhan bakteri Staphylococcus aureus. Jenis penelitian ialah eksperimental murni dengan post test only control group design. Penelitian dilakukan di Laboratorium Mikrobiologi Fakultas Kedokteran dan Labaratorium Fitokimia Fakultas MIPA Unsrat. Metode pengujian yang digunakan ialah modifikasi Kirby-Bauer dengan menggunakan tiga buah sumuran yang diberi ekstrak daun bayam petik, kontrol positif, dan kontrol negatif, sebanyak 5 kali pengulangan. Hasil penelitian menunjukan diameter rerata dari zona hambat yang terbentuk pada sumur dengan ekstrak daun bayam petik dan pada sumur dengan kontrol negatif ialah 0 mm, sedangkan pada sumur yang diberi kontrol positif amoksisilin terjadi resistensi sehingga diganti dengan eritromisin dan didapatkan rerata zona hambat ialah 38,8 mm. Simpulan: Ekstrak daun bayam petik (Amaranthus hybridus L.) tidak memiliki daya hambat terhadap pertumbuhan bakteri Staphylococcus aureus.Kata kunci: daya hambat, Amaranthus hybridus L, Staphylococcus aureus

Staphylococcus aureus is a nosocomial problem that can cause skin infections. Treatment of skin infections can be done by using citronella (Cymbopogon nardus) which is antibacterial because it contains compounds such as flavonoids, steroids, terpenoids, saponins, and essential oils. This study aims to determine the antibacterial power of Cymbopogon nardus leaf starch essence against Staphylococcus aureus inhibition zone. This type of research is a laboratory experiment using a post-test only control group design. The results showed that the extract of Cymbopogon nardus leaves with a concentration of 5%, 10%, 15%, and 20% with positive control of ampicillin had different inhibitory zona against Staphylococcus aureus bacteria. The conclusion is that the concentration of Cymbopogon nardus leaf starch is the most effective and forms an inhibition zone with the largest average (16.28), namely a concentration of 20%.Keywords: Antibacterial, lemongrass, Staphylococcus aureus

Aloe vera (Aloe barbadensis Miller) contains anthraquinone compounds, flavonoids and saponins which are thought to have antibacterial effects so as to inhibit bacterial growth. The purpose of the study was to determine the inhibitory power of Aloe barbadensis Miller on the growth of Staphylococcus aureus bacteria. The pure experimental study was conducted with The Randomized Posttest Only Control Group Design using six (6) treatments, namely aloe vera extract with a concentration of 25%, 50%, 75%, 100%, Chloramphenicol 30 mcg antibiotics as a positive control, and sterile aquadest as a control negative for Staphylococcus aureus bacteria grown on Mueller Hinton media. After being treated, it was incubated for 24 hours and measured its inhibitory power. The growth inhibition data of Staphylococcus aureus bacteria in the form of inhibition zone diameter was analyzed statistically by Kruskal Wallis test, at the level of confidence α = 0.05. The results showed that the average inhibitory effect of Aloe vera extract on the growth of Staphylococcus aureus bacteria in concentrations of 25%, 50%, 75% and 100% were 0 ± 0 mm; 7.125 ± 0.1250 mm; 8.375 ± 0.23936 mm; and 9.0 ± 0.40825 mm. Based on the results of the study concluded that aloe vera extract can inhibit the growth of Staphylococcus aureus bacteria. The higher the concentration of Aloe vera extract the greater the inhibitory effect on the growth of Staphylococcus aureus bacteria.

The rhizome of the bangle contains antibacterial compounds including saponins, flavonoids, essential oils, and tannins. The aim of the study was to determine the inhibitory power of bangle rhizome extract on the growth of Staphylococcus aureus bacteria. True research - experiment with posttest design with control group was carried out by giving 6 treatment groups, namely negative control group (K-) using aquadest, positive control group (K +) using 30 mcg Chloramphenicol antibiotics, 25% ( PE1), 50% (PE2), 75% (PE3), and 100% (PE4) of concentration of rhizome extract treatment. After the treatment is complete, the inhibition is measured. The research data were analyzed statistically by Kruskall Wallis non parametric test and Sperman Rang correlation test. The results showed that the average inhibition of bangle rhizomes in each group in groups K-, K+, PE1, PE2, PE3, and PE4 were 0.000 ± 0.000, 25.000 ± 0.408, 14.250 ± 0.479, 16.500 ± 0.289, 17.750 ± 0.250, 19,500 ± 0.289 differ significantly (p <0.05). There was a very significant positive correlation between the concentration of rhizome bangle extract and the growth inhibition of Staphylococcus aureus bacteria (r count of 0.967 (**)> r table which was 0.503 at α = 0.05 and n = 24 or p <0.01). Conclusion: bangle rhizome extract at concentrations of 25%, 50%, 75%, and 100% has a strong inhibitory power against the growth of Staphylococcus aureus bacteria. The higher the concentration the greater the inhibitory power.

The high prevalence of diabetes mellitus in Indonesia has made antidiabetic treatment efforts increasingly popular. In addition, the many side effects caused by the use of chemical drugs, triggered the development of research on herbal therapy. The purpose of this study was to determine the effectiveness of Dayak onion bulb extract in reducing two-hour postprandial blood glucose, triglyceride levels, and the amount of lipid vacuoles in aorta blood vessels of diabetic rat. The long-term goal of this study is the creation of innovation products in the form of standardized herbs as complementary therapies that can be used daily by the community, so that the risk of morbidity can be reduced. The research design used was randomized control group pretest posttest design. The results of blood glucose analysis showed that there were significant differences between the positive control group with the 400 mg/KgBW and 800 mg/KgBW extract group, with a significance value of 0,000. In the extract group, 400 mg/KgBW with 800 mg/KgBW extract group showed no significant difference with a significance value of 0.390. The results of the analysis of triglyceride levels showed no significant difference between the positive control group with the 400 mg/KgBW extract group, with a significance value of 0.981. However, there were significant differences between the positive control group with the 800 mg/KgBW extract group with a significance value of 0.025. Between the 400 mg/KgBW and 800 mg/KgBW extract groups, there was a difference with a significance value of 0.024. Aorta histopathology results showed that there was a significant difference in the number of lipid vacuoles between the positive control group and the 400 mg/KgBW extract group and 800 mg/KgBW extract group. Based on the research, it can be concluded that the extract dose of 400 mg/KgBW is a dose that is able to influence the decrease in blood glucose levels, triglycerides and the amount of lipid vacuoles in the aorta blood vessels.

Abstract: Mouthwash could inhibit the growth of Staphylococcus aureus bacteria in the oral cavity. Albeit, the most widely used today is mouthwash containing clorhexidine that has side effects in prolonged use. Therefore, it is necessary to find new agents as an alternative antibacterial, especially against Staphylococcus aureus. Squid ink is one of the best known seafood used as alternative medicine which has a wide range of therapeutic applications. This study was aimed to evaluate the inhibitory effect of squid ink extract (Loligo sp) on the growth of S. aureus. This was a true experimental study with a post test only control group design. We used modified Kirby-Bauer method using filter papers. Ciprofloxacin antibacterial was used as the positive control and aquadest as the negative control. Extract of squid ink (Loligo sp) and stock of pure bacteria S. aureus bacteria were prepared. The results showed that mean of zone of inhibition of the squid ink extract (Loligo sp) was 11.22 mm which was less than the zone of inhibition of ciprofloxacin. In conclusion, the squid ink extract (Loligo sp) had a moderate inhibitory effect (Himedia category) on the growth of Staphylococcus aureus.Keywords: extract of squid ink (Loligo sp), Staphylococcus aureus, inhibitory effectAbstrak: Salah satu cara penanganan bakteri Staphylococcus aureus dalam rongga mulut ialah dengan menggunakan obat kumur. Tinta cumi-cumi merupakan salah satu hasil laut yang dikenal dalam dunia pengobatan alternatif serta memiliki jangkauan yang luas pada aplikasi terapeutik. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak tinta cumi-cumi (Loligo sp) terhadap pertumuhan bakteri S. aureus. Jenis penelitian ialah eksperimental murni dengan post test only control group design. Metode yang digunakan yaitu metode modifikasi Kirby-Bauer dengan menggunakan kertas saring. Kontrol positif menggunakan antibakteri siprofloksasin dan kontrol negatif menggunakan akuades. Pada penelitian ini digunakan ekstrak tinta cumi-cumi (Loligo sp) dan stok bakteri murni S. aureus. Hasil penelitian mendapatkan bahwa diameter rerata zona hambat dari ekstrak tinta cumi-cumi (Loligo sp) terhadap pertumbuhan bakteri S. aureus sebesar 11,22 mm namun diameter tersebut lebih kecil daripada diameter zona hambat siprofloksasin. Simpulan penelitian ini ialah ekstrak tinta cumi-cumi (Loligo sp) memiliki daya hambat kategori sedang (Himedia) terhadap pertumbuhan bakteri S. aureus.Kata kunci: tinta cumi-cumi (Loligo sp), bakteri Staphylococcus aureus, daya hambat
