Gladiolus is one of the most popular flower crops grown in Romania. The breeding program at the University of Agricultural Sciences and Veterinary Medicine Cluj-Napoca of this species has been especially focused on the improvement of main characteristics, such as color, number and form of florets, plant height, multiplication capacity, diseases resistance, etc. A program for genetic breeding of gladiolus varieties using different genitors (Romanian and foreign cultivars) was initiated. An intraspecific crossing between cultivars has been made followed by clonal selection and vegetative multiplication of the selections. In this paper, we show 15 new gladiolus selections, which were observed and analyzed in 2004–05. The selections obtained have a great uniformity of their morphological characteristics. These selections are more vigorous, producing greater number of florets with superior quality, have new colors of flower, and are distinguished by a long blooming time. The intraspecific variability of the above-mentioned characteristics was more than low, and rarely medium, high, or very high. These data suggest fair possibilities to choose the best selections that will be proposed for testing and homologation as new cultivars. The hybrids represent a step forward in combining high qualities in gladiolus. They will contribute to improving the assortment of gladiolus for cut flowers, landscape, or as genetic material, which can be used for new crossing in order to obtain new cultivars. Scientifically, a series of findings appeared considering the combining capacity of genitors, transmission of some useful qualities, and other aspects that contribute to improve of the new varieties.

Oreochromis mossambicus is currently receiving much attention as a candidater species for aquaculture programs within Southern Africa. This has stimulated interest in its breeding cycle as well as the morphological characteristics of the gonads. Limited information is available on SEM and TEM observations of the male gonads. It is known that the testis of O. mossambicus is a paired, intra-abdominal structure of the lobular type, although further details of its characteristics are not known. Current investigations have shown that spermatids reach full maturity some two months after the female becomes gravid. Throughout the year, the testes contain spermatids at various stages of development although spermiogenesis appears to be maximal during November when spawning occurs. This paper describes the morphological and ultrastructural characteristics of the testes and spermatids.Specimens of this fish were collected at Syferkuil Dam, 8 km north- west of the University of the North over a twelve month period, sacrificed and the testes excised.

AbstractThe Anemone genus is a tuberous geophyte which undergoes a dormancy period during unfavorable environmental conditions for growth. Five species of the Anemone genus naturally grow in several regions of Iran. The diverse uses of Anemone in gardens for landscaping, cut flowers, and potted plants indicate its high ornamental potential. Its dormancy and flowering are influenced by various factors. The present paper was conducted to explore the flowering behavior of Anemone accessions in response to different pre-treatments. For this purpose, tubers of 18 Anemone accessions (A. coronaria and A. biflora) were collected from natural regions of six provinces in Iran. These tubers were subjected to different conditions of non-chilling (20 °C, 90 days), chilling (4 °C, 90 days), GA3 (150 mgL-1; 24 h), and 5-azaCitidine (5-azaC; 40 µM; 24 h) prior to the cultivation. Most of the accessions were able to enter the flowering stage without chilling. The shortest period for the sprouting of tubers (16.89 ± 7.83 days) belonged to 5-azaC pre-treatment. In addition, this treatment accelerated the flowering time (about 30 days earlier) and diameter of the stem, bud, and flower. Morphological characteristics, such as stem height, number of leaves, bud, and petal and the longevity of flowers on the plant were significantly affected by GA3 pre-treatment. Our results indicated a positive correlation between flower length, stem height, and stem diameter with flower longevity under different pre-treatment conditions. The present study demonstrated that accessions Anm3, Anm12, and Anm18 had ornamental values higher than the population mean across four conditions.

In forty years of genetic breeding of wheat, Embrapa (Brazilian Agricultural Research Corporation) has developed over a hundred new cultivars for different regions of Brazil. Information regarding identification of these cultivars is often requested from Embrapa breeders. Data on year of release, name of pre-commercial line, the cross made, and the company unit responsible for indication of the cultivar are not always easily accessible and are often scattered throughout different documents. The aim of this study was to conduct a historical survey of all the wheat cultivars released by Embrapa, aggregating the information in a single document. Since 1974, Embrapa has released 112 wheat cultivars, including 12 by Embrapa Soybean - CNPSo (Londrina, PR), 14 by Embrapa Cerrado - CPAC (Brasília, DF), 9 by Embrapa Agropecuária Oeste - CPAO (Dourados, MS), and 77 by Embrapa Wheat - CNPT (Passo Fundo, RS).

Abstract Background Chinese indigenous pigs exhibit considerable phenotypic diversity, but their population structure and the genetic basis of agriculturally important traits have not been explored. Results Here, we sequenced the whole genomes of 24 individual pigs representing 22 breeds distributed throughout China. For comparison with European and commercial breeds (one pig per breed), we integrated seven published pig genomes with our new genomes. Our results showed that pig domestication occurred at three places in Southeastern Asia, namely the Mekong region, the middle to downstream regions of the Yangtze River, and Tibetan highlands. Moreover, we demonstrated that classic morphological characteristics such as coat color are not consistent with genetic data. We found that genetic material from European pigs likely introgressed into five Chinese breeds. Two new subpopulations of domestic pigs have been identified in South and North China that encompass morphology-based criteria. The Southern Chinese subpopulation comprises the classical Southern China Type and part of the Central China Type, whereas the Northern Chinese subpopulation comprises the North China Type, the Lower Yangtze River Basin Type, the Southwest Type, the Plateau Type, and the remainder of the Central China Type. Eight haplotypes and two recombination sites were identified within a conserved 40.09 Mb linkage-disequilibrium block on the X chromosome. Potential selection and domestication signatures were identified, mainly influencing body size, along with adaptations to cold and hot temperature environments. Conclusions Our findings provide insights into the phylogeny of Chinese indigenous pig breeds, and will be of enormous benefit in identifying beneficial genes to develop superior pig breeds.

ABSTRACT The genetic variability within the crop species Ipomoea batatas is broad, hence, in order to support future breeding programs it is of the utmost importance that germplasm banks be created, conserved, and characterized. Therefore, the objective of this work was to rescue and evaluate the genetic divergence in sweet potato accessions collected in traditional communities of Vale do Ribeira Paulista. Sweet potato samples were collected from quilombos, indigenous villages, caiçaras communities, and small farms. The study was conducted between February 2013 and August 2014 in a randomized block design with three replications. Genetic material included 95 collected accessions and two commercial cultivars. Morphological characteristics of the accessions were evaluated and distances in the genetic distance matrix were estimated by means of multi-category variables, the data being subsequently clustered by the Tocher method. Analysis of the relative contribution of each characteristic and phenotypic correlation of descriptors was also performed. Results evidenced wide genetic diversity among the sweet potato accessions collected in Vale do Ribeira, which were not grouped according to the collection point. The descriptors that contributed more than 60% of genetic diversity included: leaf size, general leaf profile, immature leaf color, petiole pigmentation, predominant branch color, branch secondary color, stem length, cortical thickness, predominant periderm color, and periderm color intensity. Correlations between morphological descriptors was observed in 22.26% of the paired traits.

Rabbit can serve as an excellent model for human reproduction and relevant disease study. The objective of the study was to develop an effective procedure to preserve the rabbit genome by vitrifying zygotes. Sexually matured New Zealand White rabbits maintained under a 16 h light:8 h dark cycle were superovulated with a regime of two 0.3-mg, two 0.4-mg, and two 0.7-mg injections of FSH with an interval of 12 h between, followed by an injection of 200 IU hCG and mating. Presumptive zygotes were flushed from the oviducts and collected by midventral lapartomy 18 h after hCG injection. One-cell-stage embryos were assigned to pre-equilibration at 38.5�C in HEPES-buffered TCM199 supplemented with 20% FBS + 7.5% ethylene glycol (EG) (v/v) + 7.5% dimethylsulphoxide (DMSO) (v/v) (dehydration medium) for 3 min, and subsequently to 1.0 ml of vitrification medium (TCM199 supplemented with 20% FBS + 20% EG and 20% DMSO), as described by Vajta et al. (1998 Mol. Reprod. Dev. 51, 53–58), at room temperature for 3 min. Five to six embryos per group were then vitrified in a micro-droplet (approximately 1–2 µL) by directly dropping them into a thin layer of liquid nitrogen on the solid surface that generated a super cold surface for vitrification. Vitrified embryos were sequentially warmed, rehydrated in 20% FBS M199 with different concentrations of sucrose, and washed in 20% FBS M199 for 5 min. Warmed embryos were assigned either for further in vitro culture or for embryo transfer to test corresponding developmental potentials. Dutch rabbits were used as recipients in the procedure of asynchronous ovulation induction (22-h delay to zygote donors) by an intramuscular injection of 15 µg of GnRH per doe. Three or four warmed zygotes were surgically transferred into the recipients on the same day of thawing. Pregnancy was monitored by palpation and/or ultrasound on Days 14–16 post-embryo transfer (ET); C-sections were performed on Day 31 to retrieve full-term developed newborns. Preliminary results showed that a 96% (n = 35) post-warming survival rate was achieved with vitrified rabbit zygotes; subsequent cleavage and blastocyst development were 41.6 and 33.3%, respectively. Transfer of a total of 23 warmed embryos into 6 recipients resulted in 5 pregnancies (83.3%, 5/6). Five live kits (21.7%, 5/23) were delivered. Our study suggests that vitrification of rabbit zygotes is a feasible approach for preserving rabbit genetic material. The establishment of suitable conditions for the vitrification of rabbit zygotes could be useful as a model for vitrification of human 1-celled embryos. Cryopreservation of embryos is an important technique in the whole MOET program, which could help improve the transportation of genetic material across South Africa and globally. This trial evaluated the survival rate of goat embryos following transfer with cryopreserved Boer goat embryos. Twenty-seven multiparous Boer goat recipients were synchronized with CIDR for 16 days and injected with 300 IU of eCG at CIDR withdrawal. The recipients were allocated into 3 groups (n = 9). Group 1 received fresh embryos; Group 2 received slow frozen embryos; and Group 3 received vitrified embryos. Expanded blastocysts used were surgically collected from donors superovulated with pFSH on 7 following AI. Two blastocysts were transferred laparoscopically to the uterine horn ipsilateral to functional CL. A pregnancy rate of 85.7% (6) was obtained following the transfer of fresh embryos and tended to be better than in the does receiving slow frozen and vitrified embryos, (n = 4; 50.0% and n = 3; 37.5% does pregnant, respectively) with no significant differences. The kidding rate of the recipient does declined to 57.0% (4) and 25.0% (2) for fresh and slow frozen groups, respectively. The embryo survival rate of 35.7% (n = 5) for fresh, 25.0% (n = 4) for conventional slow freezing and 31.3% (n = 5) for vitrification was obtained and was not affected by the number of CL present on the respective ovaries at the time of embryo transfer. Although the pregnancy rate following the transfer of fresh embryos was satisfactory, the embryo survival rate following the transfer of fresh or cryopreserved embryos tended to be lower. Abiotic stress represents a main constraint for agriculture, affecting plant growth and productivity. Drought and soil salinity, especially, are major causes of reduction of crop yields and food production worldwide. It is not unexpected, therefore, that the study of plant responses to abiotic stress and stress tolerance mechanisms is one of the most active research fields in plant biology. This Special Issue compiles 22 research papers and 4 reviews covering different aspects of these responses and mechanisms, addressing environmental stress factors such as drought, salinity, flooding, heat and cold stress, deficiency or toxicity of compounds in the soil (e.g., macro and micronutrients), and combination of different stresses. The approaches used are also diverse, including, among others, the analysis of agronomic traits based on morphological characteristics, physiological and biochemical studies, and transcriptomics or transgenics. Despite its complexity, we believe that this Special Issue provides a useful overview of the topic, including basic information on the mechanisms of abiotic stress tolerance as well as practical aspects such as the alleviation of the deleterious effects of stress by different means, or the use of local landraces as a source of genetic material adapted to combined stresses. This knowledge should help to develop the agriculture of the (near) future, sustainable and better adapted to the conditions ahead, in a scenario of global warming and environmental pollution.

Abstract Dinoflagellates in the genus Gambierdiscus are known to produce potent neurotoxins known as ciguatoxins. These toxins vector through the food web and can reach sufficiently high levels to cause ciguatera fish poisoning (CFP). In many regions of the Caribbean, little is known about the occurrence of CFP-causing species or the incidence of CFP, despite its apparent regional increase over the last few decades. Given the intraspecific variability in content and potencies of these toxins among Gambierdiscus species, the precise identification of local species of this genus is crucial. In this study we confirm the presence of the epibenthic dinoflagellate Gambierdiscus caribaeus in the Colombian continental Caribbean. Cells were collected in seagrass beds of Thalassia testudinum in Bonito Gordo, a site located on the western side of Bahía Concha, Tayrona National Natural Park, near the city of Santa Marta, Colombia. Subsequently, a single strain of Gambierdiscus could be established for further identification. Morphological (light and scanning electron microscopy), and molecular results (partial LSU and ITS rDNA sequencing) were consistent with the original description of G. caribaeus, as well as with the morphological characteristics shown by other authors in organisms obtained near the sampling area.

Abstract Conservation of fruit tree biodiversity is important for the mankind according to the Convention on Biological Diversity. In Romania, due to favorable environmental conditions, numerous genetic resources of plum, apple, walnut, hazelnut, sweet chestnut and other fruit crops are present. Identification, evaluation and conservation of fruit genetic resources activities were launched in 1970’ in order to limit the loss of the biodiversity due to erosion and genetic vulnerability. Fruit Growing Research & Extension Station (SCDP) of Valcea, which is belonging now to the University of Craiova, was assigned to deal with conservation of genetic resources for the Prunus, Juglans, Corylus and Castanea genera. As result, national hazelnut collection, the sweet chestnut collection and a part of the plum and walnut national collections are located here. Genetic resources of Malus, Pyrus, Sambucus, Carya and Salix are hold in the active collections too. The ex situ collections include 1160 accessions, out of those 48 species, 533 cultivars and 565 other types like hybrids, biotypes, mutants, etc. Autochthonous or ‘original’ accessions include 173 local cultivars and 565 other genotypes. Breeding activity based on valuable germplasm conducted to releasing of a total number of 31 cultivars and 8 rootstocks registered in the Romanian Official Catalogue for Varieties, 15 varieties have been patented in Romania and for one by CPVO. Identification, in situ evaluation, collection, ex situ evaluation, propagation and regeneration activities regarding fruit tree genetic resources have to be continued in order to conserve the local fruit tree biodiversity and to value it through breeding and use of the new varieties in the orchards.
