The aim of this study was to compare canine cortisol results obtained by the Vcheck method with those obtained by the IMMULITE 2000 immunoassay, which had previously been validated for canine serum. The concentration of cortisol in 44 canine serum samples was determined concurrently by the Vcheck and IMMULITE methods, the latter as the reference method. Cortisol values were compared using Pearson’s correlation analysis and simple regression analysis. Agreement between the two methods was calculated with a Bland-Altman plot. According to regression analysis and the Bland-Altman plot, the two methods gave comparable results. The results of the Vcheck method were comparable to those of the IMMULITE 2000 reference method, so the Vcheck may be used as an alternative assay to evaluate serum cortisol concentration in dogs for the diagnosis of adrenal disease.

Radioimmunoassay (RIA) is the “gold standard” method for evaluation of serum cortisol concentration. The VIDAS cortisol test is an enzyme-linked fluorescent assay designed for the MiniVidas system. The aim of this study was to compare the VIDAS method with RIA for measurement of bovine serum cortisol concentration. Cortisol concentrations were evaluated in 40 cows using both VIDAS and RIA methods, the latter as the reference method. A paired Student’s -test, Pearson’s correlation analysis, Bland-Altman plot, and Deming regression analysis were used to compare the two methods. There was no statistically significant difference between mean serum cortisol concentrations measured by VIDAS or RIA methods (). Both methods were able to detect significant differences in mean low and high cortisol concentrations ( RIA and VIDAS). The correlation coefficient was low, but a Bland-Altman plot and Deming regression analysis show neither constant nor proportional error. The VIDAS method produced slightly higher values than RIA, but the difference was small and in no case did the mean value move the normal range. Results suggest that VIDAS method is suitable for the determination of bovine serum cortisol concentration in studies of large numbers of animals.

Abstract Introduction Simpson's biplane (SBP) ignores the apical long-axis plane. It is estimated that 40% of myocardial infarction (MI)-related segmental abnormalities are located in this plane. The real-time triplane (RT3P) overcomes this issue, allowing to obtain all apical views simultaneously, at the same cardiac cycle. This method could be a more precise and reliable alternative to SBP in ejection fraction (EF) quantification in this setting. Purpose The authors aimed to compare EF assessed by two quantification methods, SBP and RT3P, using quantitative three-dimensional echocardiography (3DE) as the reference method, in patients with MI. Methods We have prospectively gathered data from 52 adult patients, which had recent or past history of MI. Exclusion criteria were the presence of atrial fibrillation, unknown coronary anatomy, significant valvular disease, left branch block, ventricular pacing and poor definition of endocardial borders. Estimation of EF by SBP, RT3P and 3DE was performed in all patients and gathered by two experienced operators. The two operators were blinded for the coronary angiography results before volume quantification. They were also blinded for their own measurements, as well as for the other operator's measurements. Spearman's correlation and linear regression were performed for correlation analysis. Bland-Altman plot was used for agreement assessment among the different methods. Interobserver agreement was assessed by Cohen's kappa. Results Patient characteristics are shown in table 1. EF calculation was feasible in all patients. There were excellent correlations between EF measured by SBP vs 3DE and RT3P vs 3DE (r=0.813 and r=0.9 respectively). Test of equality between two correlation coefficients confirmed that EF by RT3P method is significantly more correlated with the reference method, compared with EF by SBP (p=0.004). We have also performed a further analysis to study this results in special subsets. In anterior MI subset, EF by RT3P correlated by 0.909 with 3DE, versus 0.826 from EF by SBP (p=0.019). In inferior MI subset, EF by RT3P correlated by 0.779 with the reference method, versus 0.706 from EF by SBP (p=0.246). Fig 1 Correlation and Bland-Altman plot Conclusions Estimation of EF using SBP and RT3P methods by experienced operators strongly correlate with EF determined by 3DE. The RT3P method showed the strongest correlation between the two methods, which may point to its usefulness in the evaluation of EF in patients with anterior wall motion abnormalities after myocardial infarction.

We report the results of validation of canine serum cortisol determination with the Immulite 2000 Xpi cortisol immunoassay (Siemens), with characterization of precision (CV), accuracy (spiking-recovery [SR] bias), and observed total error (TEo = bias + 2CV) across the reportable range, specifically at the most common interpretation thresholds for dynamic testing. Imprecision increased at increasing rate with decreasing serum cortisol concentration and bias was low, resulting in increasing TEo with decreasing serum cortisol concentration. Inter-laboratory comparison study allowed for determination of range-based bias (RB) and average bias (AB). At 38.6 and 552 nmol/L (1.4 and 20 μg/dL), between-run CV was 10% and 7.5%, respectively, and TEo ~30% and ~20%, respectively (TEo remained similar regardless of the considered bias: SR, RB, or AB). These analytical performance parameters should be considered in the interpretation of results and for future expert consensus discussions to determine recommendations for allowable total error (TEa). Importantly, the commonly used thresholds for interpretation of results were determined ~40 y ago with different methods of measurements and computation, hence updating is desirable. Quality control material (QCM) had between-run imprecision of 4% for QCM1 and 7% for QCM2; the bias was minimal for both levels. Acceptable QC rules are heavily dependent on the desired TEa for the QCM system (TEaQCM), itself limited by the desired clinical TEa. At low TEaQCM (20–33%), almost no rules were acceptable, whereas at high TEaQCM (50%), almost all rules were acceptable; further investigation is needed to determine which TEaQCM can be guaranteed by simple QC rules.

AbstractBackgroundAlthough majority of the previous studies have shown a good correlation between enzyme linked immuno sorbent assay (ELISA) and flow cytometry in terms of cytokines, two laboratory methods usually were compared with the regression analysis and correlation in the literature. This study aimed at comparing the ELISA and flow cytometry assay for measuring cytokines by using two different statistical methods, regression analysis and Bland-Altman plot.Materials and methodsFifty patients, diagnosed with hypercholesterolemia and expecting high level serum cytokines, and 30 healthy volunteers, expecting normal level serum cytokines, were enrolled in the study. The interleukin-1 beta (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) were measured using ELISA, and compared with obtained levels using flow cytometric method.ResultsAlthough regression analysis showed that the two methods are compatible with measurements of IL-1β, IL-6 and TNF-α, they tended to show dissimilarity with measurements of IL-1β and TNF-α based on Bland-Altman graphs.ConclusionAccording to Bland-Altman plot, our results providing evidence of ELISA and flow cytometry assays were compatible with each other for IL-1β and IL-6 measurements compared to TNF-α measurement. However, our study has a small number of participants, hence this study need to be confirmed by investigations involving more participants.

Introduction: Effective diagnosis of cardiovascular diseases requires the right tools to be used enabling selective and sensitive analysis of their biomarkers. One of them is homocysteine (Hcy), nowadays determined by immunoassays and chromatographic methods. This study aims to compare the results obtained by direct chemiluminescence immunoassay (CLIA) and high performance liquid chromatography with fluorescent detection (HPLC-FD) using commercial kits. Materials and methods: Homocysteine concentration was determined in serum samples obtained from 101 individuals, using Atellica IM HCY (Siemens Healthineers, Erlangen, Germany) and HCY in plasma/serum – HPLC-FD (Chromsystems Instruments & Chemicals GmbH, Gräfelfing, Germany) tests validated for routine analysis. The latter was applied as a reference method. The comparability and agreement between the tested methods were evaluated using the Passing-Bablok (PB) regression analysis and the Bland-Altman (BA) method of the differences analysis. Results: Studies showed that CLIA gives higher Hcy concentrations (15.7 ± 4.14 μmol/L). Passing-Bablok regression analysis of the results obtained with CLIA (y) compared with HPLC-FD (x) yielded an intercept of 0.22 (95%CI: - 2.16 to 2.46) and slope of 1.58 (95%CI: 1.33 to 1.87). Bland-Altman analysis demonstrated a systematic positive bias for CLIA of 5.85 ± 2.77 μmol/L. Conclusions: Methods disagreement precludes their interchangeability. Lower Hcy values by HPLC-FD result from its greater selectivity. High performance liquid chromatography with fluorescent detection should be considered as preferential method for analysing Hcy in blood serum as well as the recommended reference method for routine clinical analysis. This fact, however, imposes the need to establish new reference ranges.

AbstractGenerally, there is a need for short questionnaires to estimate diet quality in the Netherlands. We developed a thirty-four-item FFQ – the Dutch Healthy Diet FFQ (DHD-FFQ) – to estimate adherence to the most recent Dutch guidelines for a healthy diet of 2006 using the DHD-index. The objectives of the present study were to evaluate the DHD-index derived from the DHD-FFQ by comparing it with the index based on a reference method and to examine associations with participant characteristics, nutrient intakes and levels of cardiometabolic risk factors. Data of 1235 Dutch men and women, aged between 20 and 70 years, participating in the Nutrition Questionnaires plus study were used. The DHD-index was calculated from the DHD-FFQ and from a reference method consisting of a 180-item FFQ combined with a 24-h urinary Na excretion value. Ranking was studied using Spearman’s correlations, and absolute agreement was studied using a Bland–Altman plot. Nutrient intakes derived from the 180-item FFQ were studied according to quintiles of the DHD-index using DHD-FFQ data. The correlation between the DHD-index derived from the DHD-FFQ and the reference method was 0·56 (95 % CI 0·52, 0·60). The Bland–Altman plot showed a small mean overestimation of the DHD-index derived from the DHD-FFQ compared with the reference method. The DHD-index score was in the favourable direction associated with most macronutrient and micronutrient intakes when adjusted for energy intake. No associations between the DHD-index score and cardiometabolic risk factors were observed. In conclusion, the DHD-index derived from the DHD-FFQ was considered acceptable in ranking but relatively poor in individual assessment of diet quality.

Bioelectrical impedance analysis (BIA) is used to estimate body composition characteristics. The values of body fat and fat free mass (FFM) are obtained as per algorithms of the device that are often unknown to the researchers. Some models of the analyzer provide resistance and reactance values that may be useful to estimate FFM. Objective of the present study was to test the agreement and proportional bias in the estimation of FFM obtained through BIA and that derived from the resistance and reactance values using a formula for Mexican adults. A cross-sectional study was carried out in 2019 selected 60 university male students aged 21 to 23 years from Merida, Yucatan. A multifrequency whole body bioelectrical impedance analyzer Tanita MC 180 MA (Tanita Corporation, Tokyo-Japan) was used to evaluate body composition characteristics. The device gives estimates of FFM in kg (based on algorithm) and the resistance and reactance values (ohms). There is an existing formula for Mexican adults to estimate FFM (FFM_FOR) from the resistance and reactance values obtained through BIA. An agreement between the two estimates of FFM has been tested using Bland-Altman plot and linear regression analysis. Mean value of age of the participants was 21.88 years. FFM estimated by BIA (FFM_BIA = 41.44 kg) and that derived from FFM_FOR (41.36 kg) had signifi cant intraclass correlation coeffi cient (ICC) (Cronbach’s alpha = 0.99, p<0.0001). One sample t-test estimating the diff erence of mean values between FFM_BIA and FFM_FOR was not signifi cant (t = 1.37, mean diff erence -0.02, p = 0.18). The Bland-Altman plot shows almost all data points lie within 95% confi dence interval limits. A linear regression analysis using the diff erence of FFM values as dependent variable and the average of the measurements as the independent variable showed no signifi cant interrelationships. In conclusion, the formula to estimate FFM using the resistance and reactance values of BIA has been found to be useful in the present study.

Today, the sharing economy is a widely discussed topic where people ask themselves what it is, how it works, and what it brings to the society. It occurs in many social spheres; this research paper focuses on the sharing economy in the field of transport. The sharing economy is a phenomenon of the 21st century; the fact that it is a very current topic is also evidenced by the amount of related research published in the databases. This paper aims to apply the BlandAltman model, commonly used in medicine, to the sharing economy. Its aim is to point out the functionality and possibilities of using non-traditional approaches in various areas of study. The introduction describes the sharing economy and the latest publications dealing with it in the field of transport, with a focus on Uber. The methodology section describes the Bland-Altman model and the authors who discuss it. In the results section, we offer the results of the application of the Bland-Altman model on the data obtained from 110 Uber rides in Prague. In addition to this method, regression analysis was also applied. The discussion and conclusion section summarizes the results and mentions essential publications in the field.

SummaryTen patients for elective cholecystectomy were studied pre-, per- and postoperatively. All had neurolept anesthesia. Plasma concentrations of β-TG, TXB2 and 5-HT and intraplatelet 5-HT were measured. Aggregation to ADP was recorded.Serum cortisol concentration was used as index of the stress response, showing peroperative increase and postoperative decrease. Closely related to this we observed a significant increase in P-β-TG and P-TXB2 with postoperative normalization in 6 patients without complications. P-5-HT had a peak peropera-tively and remained elevated postoperatively. A negative correlation between P--5-HT and decreasing intraplatelet 5-HT postoperatively was observed.High postoperative levels of P--5-HT seem to be related to low arterial Po2 and pulmonary dysfunction. In 3 patients with complications a second increase in P-β-TG, P-TXB2 and partly in P--5-HT was found. Platelets were temporarily refractory to ADP immediately following surgery and showed increased aggregabil-ity postoperatively. We conclude that platelets are activated in surgical stress.

Abstract This study was conducted to determine if feeding calves NaturSafe would reduce the acute phase response (APR) to lipopolysaccharide (LPS) challenge. Crossbred steers (n=32; 274±2 kg) were randomly allotted to two treatment diets: 1) Control, fed a standard receiving ration, and 2) NaturSafe, fed the Control ration supplemented with NaturSafe at 12 g/hd/d (NaturSafe®, Diamond V). On d22, steers were fitted with indwelling jugular catheters and rectal temperature monitoring devices and placed in individual stalls. On d23, steers were challenged i.v. with 0.25 µg/kg BW LPS. Serum samples were collected and sickness behavior scores (SBS) recorded at 0.5-h intervals from -2 to 8h and at 24h relative to LPS challenge. Rectal temperatures were greater (P=0.01) in NaturSafe compared to Control steers for the following time intervals following LPS challenge: 6 to 11h, 13h, 15 to 20h, and 22 to 24h. Additionally, SBS were reduced (P&lt; 0.01) in NaturSafe compared to Control steers. White blood cell concentrations were greater (P=0.05) in NaturSafe compared to Control steers prior to the LPS challenge, yet the response to LPS did not differ between treatments (P &gt;0.05). A treatment × time interaction for serum cortisol concentrations (P&lt; 0.01) showed an increase at 0.5 and 2h post-challenge but a reduction at 3h in NaturSafe compared to Control steers. Additionally, fibrinogen was greater (P&lt; 0.01) in NaturSafe compared to Control steers. There was a treatment × time interaction (P&lt; 0.01) for TNF-α where concentrations were reduced from 1 to 2h post-challenge in NaturSafe compared to Control steers. Serum IL-6 tended (P=0.09) to show a reduction in serum concentrations in NaturSafe compared to Control steers. There was a tendency (P=0.07) for a treatment × time interaction for IFN-γ. Overall these data suggest a priming effect of NaturSafe on the innate immune system of steers, resulting in an attenuated APR to the LPS challenge.
