Gastrointestinal Nematodes are one among cause decrease production efificiency in goat and sheep in Indonesia, especially parasite of abomasum namely Haemonchus contortus. This parasite causes disease of Haemonchosis causing anemia, and causes mortality to young goat or sheep. Loss of economics caused by high morbidity and  mortality this parasite so that can improve the expense of prevention and medication of this disease. This research aim was to evaluate ovicidal and larvicidal apus bamboo leaf infusion (Gigantochloa apus) against Haemonchus contortus by in vitro. Examination of apus bamboo leaves infusion to seven concentrations namely : 1.2; 1.0; 0.8,; 0.6; 0.4; 0.2;0.1; 0.05 and 0 mg / ml as negative control. Positive control uses Albendazole with concentration 0,025 mg / ml. The highest concentration 1.2 mg / ml apus bamboo leaves infusion   inhibited 92.35% egg hatching ( LC 0.49 mg / ml ) and have larvicidal activity equal to 90% ( LC50 0,47mg / ml). The concentration 1.2 mg / ml also have similar effect with positive control which use Albendazole. Phytochemical apus  bamboo leaf analysis of the highest concentration revealed the presence  tanin condensed, saponins, and alkaloids.

Medicinal plants with anthelmintic properties are an alternative to the chemical fight against small ruminant’s gastrointestinal nematodes. In order to broaden the spectrum of medicinal plants with anthelmintic properties, ovicidal activity and L3 larvae exsheathment of C. sesamoïdes Endl and S. hermonthica (Deli) Benth, aqueous extracts has been done on H contortus egg and L3 larvae. Three concentrations, 3.12 mg / ml, 6.25 mg / ml, 12.5 mg / ml of each plant extract were used to assess the inhibition of fresh egg hatching and larval paralysis as well as the 'inhibition of L3 larvae exsheathment. A negative control (PBS1x) and a positive control (levamisole, 2.5 mg / ml) were constituted for the fresh egg hatching inhibition assessment while only the negative control (PBS1x) was constituted to evaluate the inhibition of L3 Larvae exsheathment.1 ml of each concentration was contacted with 1 ml of the egg solution and then incubated for 48 hours at 27 ° C in petri dishes (60X15 Cm) for hatching test inhibition.1 ml of each extract concentration was contacted with 1 ml of the embryonated egg solution after 24 hours of incubation at 27 ° C for the larval paralysis test.

The anthelminthic efficacy of the crude extracts of Hypoestes forskaolii (Vahl) R.Br (Acanthaceae) against gastrointestinal nematodes (GIN) in sheep was investigated using the in vitro egg hatch inhibition assay. Faecal samples were collected from sheep with naturally occurring infection of GIN (Trichostrongylus spp., Chabertia ovina, Cooperia spp., Haemonchus contortus and Teladorsagia spp.). Crude leaf extracts of H. forskaolii was obtained using increasing polarity solvents: n-hexane, chloroform, chloroform:methanol 9:1, methanol. Thiabendazole (0.2 µg/mL and 0.5 µg/mL) was used as a positive control and untreated GIN eggs in deionised water served as the negative control. All the extracts exhibited a weak ovicidal activity against GIN (less than 50% of egg hatch). Noteworthy, the n-hexane extract showed a percentage of inhibition of egg hatching greater than other extracts inhibiting the 30.8% at the concentration of 1 mg/mL showing a dose-dependent effect on nematode eggs hatching. Further studies are needed to investigate the effects of extracts used and to evaluate the ovicidal effects of other extracts of H. forskaolii.

AbstractHaemonchus contortus is one of the major gastrointestinal nematodes responsible for significant economic and production losses of sheep. Diseases caused by this species lack effective anthelmintic products, and the search for new compounds to replace synthetic anthelmintics has been extensive. The present investigation assesses the in vitro activity of the essential oil of melaleuca (Melaleuca alternifolia), both free (TTO) and nanostructured (nanoTTO), and terpinen-4-ol (terp-4-ol) on eggs and larvae of H. contortus. Tests of egg hatching (EHT) and inhibition of larval migration (LMIT) were used to assess the in vitro efficacy of TTO, nanoTTO and terp-4-ol. Using EHT, at a concentration of 3.5 mg/ml, 100% inhibition occurred using TTO and terp-4-ol, with LC50 values of 0.43 and 0.63 mg/ml, and LC90 values of 1.75 mg/ml and 3.12 mg/ml, respectively. NanoTTO had lower activity, with 82.6% inhibition at the same concentration. Using LMIT, TTO and nanoTTO had a similar activity with 88.0% and 84.8% inhibition, respectively, at a concentration of 56 mg/ml. Terp-4-ol had a greater effect on larvae, with 85.7% inhibition at a concentration of 56 mg/ml and 82.4% at 3.5 mg/ml, demonstrating high activity at the lowest concentration tested. Therefore, the results indicate that all substances tested showed ovicidal and larvicidal activity against H. contortus. TTO, terp-4-ol and, mainly, nanoTTO may be targeted in in vivo studies, besides being a promising line of research into the control and treatment of veterinary important helminths.

Acacia nillotica and Desmanthus virgatus are two entopharmacologycal plants that thrives throughout the season in East Nusa Tenggara Province, other Provinces in Indonesia or tropical countries. Extraction of young leaves of Acacia nillotica (EDMAN) and Desmanthus virgatus (EDMDV) contains tannin compound. By pharmacodynamic viewpoint, this extraction has potency as an anthelmintic. Objective: to compare the in-vitro effectivity of young leaves extraction of the two plants as a vermicidal power to combat Haemonchus contortus. Materials: young leaves of Acacia nillotica and Desmanthus virgatus and Haemonchus contortus. Method: The study was grouped into four treatments: EDMAN, EDMDV, Positive control (Albendazole 0,055%) and negative control (aquades).  The concentration of the young leaves extracts are 2.5%, 3.5%, 4.5% out of 0.01 g/mL of extraction. Each treatment was applied to 6 female Haemonchus contortus with four replicates allowing immersion time for 1, 3, 5 or 7 hours. Variable measured and tested was mortality of the H. contortus. The vermicidal effectively was descriptively analysed. The results showed that mortality percentage (vermicidal) treatment of 2.5%, 3.5%, 4.5% EDMAN for 7-hour immersion was 16.7%, 45.8%, 12.5%, respectively. That values for EDMDV for similar concentrations and immersion time was 50%, 33.3%, 12.5%, respectively. Conclusion: EDMDV has a more effective vermicidal power between the two etnopharmacological treatments at 2.5% concentration.

The aim of this study was to evaluate the in vitro lethal effect of a methanolic extract (ME) from Caesalpinia coriaria fruits against Haemonchus contortus eggs and infective larvae. The anthelmintic activity was assessed using the egg hatching inhibition assay (EHI) and the mortality test. The ME was assessed using five concentrations as follows: 6.15, 3.12, 1.56, and 0.78 mg/mL to eggs and 150, 100, 75, and 50 mg/mL to larvae, respectively. Ivermectin (5 mg/mL) was used as positive control and 4% methanol and distilled water were used as negative controls. The data of ovicidal and larvicidal effect were analyzed with a completely randomized design through ANOVA analysis using the general linear model (GLM) and lethal concentrations (LC50 and LC90) were estimated through a Probit analysis using the SAS program. A clear ME increased concentration dependence effect was observed in the EHI and mortality tests. The highest activity of the methanolic extract was observed at the highest concentration (P < 0.05) to obtain a similar effect to the positive control (ivermectin), with LC50 = 78.38 and 0.00064 mg/mL and LC90 =235.63 and 0.024 mg/mL, respectively, for larvae and eggs. The results indicate that the C. coriaria fruit ME possesses in vitro ovicidal and larvicidal properties (gallotannins: methyl gallate) against H. contortus that needs to be investigated more in vivo for the control of gastroenteric nematodes in ruminants.

Background and Aim: Hemonchosis has resulted in huge economic losses for sheep farmers worldwide. Secondary metabolite compounds from Indigofera tinctoria L. can be used as anthelmintics. This study aimed to evaluate the in vitro and in vivo effects of I. tinctoria L. aqueous extract (IAE) as an anthelmintic against adult Haemonchus contortus isolated from sheep. Materials and Methods: Ten active adult worms were placed in each Petri dish containing 25 mL of IAE, each having a different concentration of IAE (at concentrations 100, 120, 140, 160, 180, 200, and 220 mg/mL). Each experiment was repeated. The positive control used 1% albendazole, and the negative control used 0.62% saline water. The number of immobile worms and the time of mortality were recorded after 2, 4, 6, and 8 h. The dead worms were subsequently tested using scanning electron microscopy (SEM) and sodium dodecyl sulfate-polyacrylamide gel electrophoresis. In the in vivo study, 15 sheep with an average fecal egg count (FEC) of 1000 eggs per gram were treated with IAE for 9 days: Group A (negative control, saline water 0.62%), Group B (21 mg/mL), Group C (41 mg/mL), Group D (62 mg/mL), and Group E (positive control, albendazole 100 mg/mL). Measurements of the body weight, FEC reduction test, and hematology testing were performed on days 0, 7, and 14. SEM was performed using worms found from the abomasum of slaughtered sheep. Results: The leaves of I. tinctoria L. contained a number of secondary metabolites, including total tannins, saponins, flavonoids, and alkaloids. The most effective concentration that killed the adult H. contortus worms was 220 mg/mL (93.33% mortality) after 8 h of treatment. The electrophoresis results showed that the protein band at a dose of 22% was less than that of the control. The highest FECR value of the treatment group on the 14th day after treatment was at a dose of 62 mg/mL. The highest weight gain as well as the highest increased hemoglobin (Hb), packed cell volume (PCV), and total erythrocyte count (TEC) values on the 14th day after treatment were at a dose of 41 mg/mL. The SEM results showed that IAE treatment caused the worms' anterior parts to become wrinkled with thick creases and cuticle abrasion (in vitro) and the anterior part to shrink along with the presence of aggregates in the worm cuticle (in vivo). Conclusion: The aqueous extract of I. tinctoria contains tannins, saponins, flavonoids, and alkaloids and has an anthelmintic effect with decreased FEC, increased weight gain, Hb, PCV, and TEC, causing damage to the worms' body and reducing the protein profile of adult H. contortus worms.

The anthelmintic resistance problem is widely recognized in sheep production. Therefore, new methods of control against gastrointestinal nematodes (GIN) need to be integrated. The aim of this work was to assess the toxicity of A. cina 30 CH as a homeopathic product against Haemonchus contortus in in vitro and in vivo assays. A. cina 30 CH was obtained from a commercial laboratory, and confirmation of artemisinin as a key ingredient was performed with mass spectrophotometry. The A. cina 30 CH and the artemisinin pure reagent were used for the inhibition of egg hatching (IEH) and for the inhibition of larval migration of H. contortus L3 (ILM). In addition, three groups of 10 naturally infected lambs with GIN were treated with A. cina 30 CH and albendazole, and 10 were used as control. The parasitic infection was monitored at 0, 7, 14 and 28 d postreatment (PT) to determine the number of eggs per gram  (epg) and  FAMACHA index.  The in vitro data  showed 100 %  IEH and 64.7 % ILM by A. cina 30 CH, and nonlethal activity was observed with the artemisinin pure reagent. The toxicity of A. cina 30 CH against H. contortus in infected lambs was observed after 7 d of infection. Administration of the A. cina 30 CH yielded a 69 % reduction in the epg at 28 d PT, similar to the albendazole (P<0.05). In conclusion, A. cina 30 CH had the ability to IEH and ILM of H. contortus in in vitro assays and reduced the number of eggs of H. contortus, which is the primary parasitic nematode in grazing lambs, thereby reducing infection.

Candidiasis is an important nosocomial infection with high morbidity value, high mortality rate, and expensive clinical cost. However, public access to well-developed treatment is not acquired. Current medicines mostly used like -azol drugs had been showing the resistant effects because of the long period use of the same medicines. Alternative medicines like herb medicine are discussed to reduce multi-resist infections, such as lemongrass Cymbopogon citratus essential oil. This study aims to know the effect of lemongrass essential oil on the growth of Candida albicans in vitro. Laboratory experimental (in vitro) was conducted in this research. Candida albicans strains were being used as the objects, which were picked by random sampling. Candida albicans were divided into 12 groups of treatment, the group I was treated with ethanol 96% as the negative control, group II with fluconazole 25μg as the positive control, and group III-XII with Cymbopogon citratus essential oil with 10%, 20% to 100% concentrated. The diameter of inhibition zones was measured after 2x24 hours incubation. The data was analyzed by post-hoc Mann Whitney test with SPSS 18.0 (p<0.05 considered as significant). Cymbopogon citratus essential oil showed antifungal activity to the Candida albicans began in the 10% to 100% concentration (p<0.05). Inhibition zones with the 50% to 100% concentration had similar results to the positive control (p>0.05). The Cymbopogon citratus essential oil has an antifungi effect toward Candida albicans in vitro significantly by the negative control.

The anthelmintic resistance has limited the control of gastrointestinal nematodes of small ruminants and thus has awakened interest in the study of plants extract as a source of anthelmintics. These experiments were carried out to evaluate the in vitro efficacy of Jatrophacurcas latex extract against Haemonchuscontortus larval motility. To evaluate the larvicidal activity, H.contortus L3 were incubated with the extracts with varying concentration of 5 mg/mL, 10 mg/mL, 15 mg/mL and 20 mg/mL at 27°C for 48, 72 and 96 hrs. The results were subjected to the Kruskal-Wallis test (P less than 0.05). The extracts showed dose-dependent larvicidal effects. These results suggest that J.curcas can be used to control gastrointestinal nematodes of small ruminants.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.
