The experiment was conducted to compare estrous performance between and estradiol level kacang and Nubian goats estrous induction by prostaglandin F2 alpha (PGF2α). Eight female goats were divided into two groups, groups 1 (K1) consisted five Nubian goats and group 2 (K2) included three kacang goats with criterias as follows: age 1.5-3.0 years old, not pregnant, at least two months postpartum, and healthy clinically. All goats were estrous synchronized by double injection 7,5 mg PGF2α intramuscularly with 10 days interval. Goats that appeared estrus symptoms were mated naturally to a male goat. Determination of estrous intensity was performed using scoring method. Blood samples were taken at the first time the goats were mated (day 0). The measurement of estradiol concentration was done by enzyme linked immunosorbent assay (ELISA) method. Data of intensity, onset, duration, and estradiol level were analyzed using t test. The results showed that the rate of intensity, onset, duration, and estradiol level of estrous at KI vs KII were 2.08±0.52 vs 1.85±0.67, 56.00±34.11 vs 40.80±18.20 hours, and 24.00±26.15 vs 33.80±9.10 hours, and 103.51±42.49 vs 67.95±38.44 pg/ml (P>0,05). It can be concluded there was no difference of estrous performance and estradiol level between kacang and Nubian which estrous induction with PGF2α. The relationship between the intensity of estrus estradiol level in the Nubian goats was 0.995 while the kacang goats was 0.890.

This study aims to determine the relationship between the intensity of estrus and estradiol concentrations during insemination on local cattle. Ten cows were divided into two groups with each group consist of five cows with category: aged 5-8 years, weight 150-250 kg, clinically healthy, and not pregnant. Plasma samples for examination of the hormone estradiol concentrations were collected during insemination time from jugular vein. Prior to the collection of plasma samples, whole cow were estrus synchronized using prostaglandin F2 alpha (PGF2α) and Ovsynch protocol. The PGF2α were injected twice at intervals of 11 days. Examination of estradiol hormone concentration was performed with enzyme-linked immunosorbent assay (ELISA) using estradiol commercial kits. The results of this study indicate that statistically there is no relationship between estrous intensity and estradiol concentration.

Os ensaios foram realizados no Laboratório de Hidráulica do Departamento de Engenharia da Universidade Estadual do Oeste do Paraná – UNIOESTE. Na  avaliação dos microaspersores  estudou-se, a variação decorrente do processo de fabricação e a determinação da equação característica da relação vazão–pressão. Os microaspersores do Grupo modular com diâmetros de bocais 0,94; 1,16;1,41;1,92 e 2,34 mm,  foram submetidos às pressões de 100, 150, 200, 250, 300 e 350 kPa. As equações características determinadas indicaram que o microaspersor testado não é auto-compensante , tolerante a sensibilidade de variações de pressões e que a equação potencial se ajusta bem aos dados. Os coeficientes de variação de fabricação obtidos foram menores que 5%, classificando-se, de acordo com a Norma ISO, como de categoria A. UNITERMOS: Microaspersão, coeficiente de variação, modelo potencial.  VILAS BÔAS, M. The tests was accomplished in the Laboratory of Hydraulics of the Department of Engineering of the State University of the West of Paraná - UNIOESTE. In the evaluation of the microasprinklers it was studied such characteristics as, the variation due to the production process and the determination of the characteristic equation of the relationship vazão-pressure. The microsprinklers of the Group to modulate with diameters of nozzle 0,94; 1,16;1,41;1,92 and 2,34 mm, the pressures were submitted 100, 150, 200, 250, 300 and 350 kPa. The certain characteristic equations showed that the tested microsprinklers is not solemnity-compensante and that the potential equation was fit well to the data. The obtained coefficients of production variation were everybody below 5% being able to not this way to classify them in agreement with for ISO  category as A. KEYWORDS: Microsprinkler, coefficient variation, power function.

The black mould Stachybotrys chartarum and its mycotoxins have been linked to damp building-associated illnesses. The objective of this study was to determine the effects of water availability (water activity, aw) and temperature on growth and production of satratoxin G (SG) by a macrocyclic trichothecene-producing strain (IBT 7711) and non-producing strain (IBT 1495) of S. chartarum. Growth studies were carried out on potato dextrose agar modified with glycerol to 0.995-0.92 aw at 10-37 °C. Growth extension was measured and the cultures were extracted after 10 days and a competitive enzyme-linked immunosorbent assay (ELISA) method used to quantify the SG content. Growth was optimal at 25 to 30 °C at 0.995 aw, but this was modified to 0.98 aw at 30 °C for both strains (1.4-1.6 mm/day, respectively). The ELISA method revealed that, in contrast to growth, SG production was maximal at 20 °C with highest production at 0.98 aw (approximately 250 μg/g mycelia). When water was freely available (0.995 aw), SG was maximally produced at 15 °C and decreased as temperature was increased. Interestingly, the strain classified as a non-toxigenic produced very low amounts of SG (<1.6 μg/g mycelia) that were maximal at 25 °C and 0.98 aw. Contour maps for growth and SG production were developed from these data sets. These data have shown, for the first time, that growth and SG production profiles are very different in relation to key environmental conditions in the indoor environment. This will be very useful in practically determining the risk from exposure to S. chartarum and its toxins in the built environment.

     The aim of the present study is to establish the relationship between gestational age and placentome measurement via real-time ultrasound. This study is conducted in the college of Veterinary Medicine at Diyala University. Seventeen multiparous does were synchronized for estrus and naturally served to obtain pregnancy and subsequently scanned using both transducers. Ultrasonic (U.S) examination was performed transabdominally (TA) with sector-probe (5MHz) and transrectally (TR) with linear-probe (7.5MHz), in 10 day interval done started from day 35-135 post mating. The placentome was observed for the first time on day 35 by using TR ultrasonic examination (7.5MHz) linear transducer, as echogenic densities on the surface of endometrium. The results showed a significant increased (P≤0.05) in placentomes growth with gestation age. The placentome diameter (PD) reached maximized size around days 126 (39.6±2.37mm). Also, the results indicated significant difference between placentome diameter (PD) size in single and twin-pregnant does at (P≤0.05). The average of PD size in the single and twin-pregnant does was, respectively, 7.5±0.41mm and 9.2±0.74 mm at the day 35 and 45 of gestation. The maximal size of PD was 39.2±2.50mm in singleton-bearing does during 116-125 day and 41.0±2.19 mm in twin-bearing does during 126-135 day of gestation. In conclusion, the determination of gestation age according to placentome measurement was not reliable after day 90. Also, the use of 7.5 MHz linear array transducer for TR examination was found to be efficient, for early recognition of placentomes starting point from 35 days of gestation, as well as, the larger placentomes are expected in twins-bearing does.

Estradiol is one factor that can alter the outcome of the treatment of infertile couples following the application of in vitro fertilization techniques. Currently, the estradiol level is measured by two diagnostic methods Enzyme-Linked Immunosorbent Assay (ELISA) and Electrochemiluminescence (ECL). Accordingly, this study determines ELISA and ECL's sensitivity and consistency to measure different levels of estradiol and determine its reliable range and provide this information to laboratories and gynecologists. This study is performed on 250 patients of the Avicenna Fertility Center. The data of the study are analyzed in SPSS18 and MiniTab. Consent was obtained for experimentation with human subjects. Pearson correlation coefficient was used to investigate the relationship between these two methods. The results indicated a strong correlation between the two variables ECL and ELISA ( r= 0.735, P-value<0.001). High numbers indicate that the decrease and increase of one variable are proportional to the other variable's fluctuation. This study shows that the results of estradiol obtained from both ECL and ELISA are similar. In the ELISA method, due to the linear values' limitation, samples with estradiol concentration above the highest standard level should be diluted and the dilution coefficient should then be applied.

Abstract Dinophysistoxin-2 (DTX-2), an isomer of okadaic acid (OA), recently has been found in Irish waters. DTX-2 was the predominant toxin during prolonged infestations in cultivated mussels along the southwest coast of Ireland. Substantial variations in toxin levels may exist both horizontally and vertically in the water column. The need to take multiple samples and the ethical concern about the use of mammals for routine quality control of shellfish prompted examination of 2 commercially available enzyme-linked immunosorbent assay (ELISA) methods, designed to detect OA, for determination of both OA and DTX-2. One ELISA method (DSPCheck, Sceti Co. Ltd., (Tokyo, Japan) showed good cross-reactivity (40 ± 5%) with standard DTX-2. This study showed that both ELISA methods show good correlation with the liquid chromatographic analysis of 9-anthryldiazomethane derivatives when OA is the predominant toxin present. The sensitivity was also good for OA determination using both methods, which allowed toxin measurement at 10 ng/mL (0.5 ng/well). This level is equivalent to 0.03 μg/g mussel meat. Blank mussel samples spiked with DTX-2 standards gave a good linear correlation (r = 0.997) with this ELISA method when toxin levels were 0.03-0.3 μg/g mussel meat. This range is appropriate for regulatory control of diarrhetic shellfish poisoning.

We measured folding of elastin lamellae and collagen fibers in human carotid arteries and correlated the results with the elastic properties of the vessels. Specimens cut into circumferential strips were stretched to various degrees, fixed, and stained for elastin and collagen. Folding was measured on photographic projections. Elastin lamellae unfolded quickly with initial stretch. In old vessels they did not straighten as much as in the young suggesting that the content of the interlamellar space may interfere with the unfolding. Collagen fibers straightened more during stretch in stiffer older vessels than in the young, already at low degrees of stretch. Young's modulus at extension of 5% appeared to correlate with unfolding of elastin lamellae in young extensible vessels. The modulus correlated significantly with unfolding of collagen fibers at extensions of 15% and greater, and the slope of the regression of the modulus on folding increased with stretch. Our findings provide for the first time morphologic evidence for the role of elastin lamellae in the determination of the elastic properties at low extensions, for the importance of collagen fibers in increasing stiffness with further stretch, and for the relationship between increased stiffness with age and the earlier recruitment of the collagen fibers.

The objective of the study was to determine ochratoxin A (OTA) concentrations in serum and urine of pigs during 30-day OTA treatment. OTA was administered orally to the experimental group (n=5) at a dose of 0.78 mg per animal per day, whereas control animals (n=5) were left untreated. OTA concentrations were determined using a validated enzyme-linked immunosorbent assay (ELISA). Method validation resulted in mean recoveries of 93-101% for serum and 98-106% for urine, with acceptable mean inter- and intraday relative standard deviations (<8% for urine and <7% for serum). The ELISA method can be effectively used as a simple screening method to determine OTA exposure in pigs during fattening. The maximum mean OTA concentration in serum was recorded on day 22 (8.75±2.93 ng/ml) and in urine on day 20 (43.56±35.76 ng/ml), indicating significant differences in OTA concentrations between these two matrices.

AbstractObjectivesFaced with the COVID-19 pandemic and its impact on the availability and quality of both therapeutic and diagnostic methods, the Belgian authorities have decided to launch a procedure for additional evaluation of the performance of serological tests offered for sale on the national territory. This has been proposed with a double aim: (1) an in-depth verification of the analytical and clinical performances presented by the manufacturer and (2) an economy of scale in terms of centralized validation for all the laboratories using the tests subject to evaluation.MethodsA retrospective validation study was conducted including the serum of 125 patients in order to determine the analytical and clinical performances of the LIAISON®SARS-CoV-2 from DiaSorin® detecting anti-SARS-CoV-2 IgG and to compare its clinical performance with the enzyme-linked immunosorbent assay (ELISA) test from Euroimmun®, one of the first commercially available tests allowing the detection of anti-SARS-CoV-2 IgA and IgG.ResultsThe performances of the LIAISON®SARS-CoV-2 satisfied all the acceptance criteria and provided “real world” analytical and clinical performances very close to the ones reported by the manufacturer in its insert kit. Comparison between the LIAISON®SARS-CoV-2 and the ELISA method did not reveal any difference between the two techniques in terms of sensitivities and specificities regarding the determination of the IgG.ConclusionsThis study reports the validation of the LIAISON®SARS-CoV-2 allowing to detect IgG antibodies specifically directed against SARS-CoV-2. The analytical and clinical performances are excellent, and the automation of the test offers important rates, ideal for absorbing an extension of testing.
