Objective: The objective of the study was to evaluate the anti-oxytocic activity of ethanolic extract of Asparagus racemosus root. Methods: Ethanolic extract of A. racemosus root was taken by Soxhlet extraction method. Wistar strain albino non-pregnant female rats of weight 200–300 g were pre-treated with estrogen and were sacrificed to take their uterus. The tissue was then mounted in an organ bath containing de Jalon’s solution. The response of the tissue to various doses of oxytocin alone and then on adding increasing doses of the alcoholic extracts of A. racemosus along with the dose of oxytocin which produces sub-maximal contraction were recorded on a smoked drum. Results: Extract in doses up to 40 mg when given along with oxytocin 0.1 unit produced graded increase in contractions in rat uterus. From 80 mg onward graded blockade of contractions occurred with complete blockade at 200 mg. On complete removal of the extract by thorough washing, it was seen that oxytocin was again able to produce contractions of the rat uterus tissue. Conclusion: The ethanolic extract of A. racemosus root had demonstrated good anti-oxytocic property.

Invitro membrane-stabilizing potentials of fractionates of ethanolic extract of Carica Papaya leaf was investigated in this study. The soxhlet extraction method was used to extract the plant, fractionated with 6 different solvents to give 6 different fractions (hexane, ethyl acetate, chloroform, butanol, methanol benzene). Hbss red blood cells samples were obtained from non-crises state sickle cell patients from Eku Baptist hospital Abraka Delta State, Nigeria. These tests involved the use of positive (p-hydroxy benzoic acid 5ug/ml) and negative controls (normal saline) for membrane stability experiments. Hbss blood was treated with 2mg/ml to 10mg/ml in seven groups with leaf fractionates. Data was analyzed using ANOVA test. The results shows that osmotic fragility was reduced by the introduction of the leaf fractionate, with the highest rate of reduction noticed in the hexane 1 fractionate. PHBA reversal rate and osmotic fragility effect was normal at low doses, but as concentration increases, reversal rate and percentage reduction of sickling decreases. It was concluded that Carica papaya leaf extract fractions, just as its crude extracts, have as much osmotic fragility activities, and this is dose-dependent and has no negative effect on tested blood samples as compared with the treatments with PHBA.

ABSTRAK             Kulit buah Mundar (G. forbesii King.) merupakan tanaman endemik yang berasal dari Kalimantan Selatan yang berpotensi memiliki aktivitas antioksidan. Penelitian ini dilakukan untuk menguji aktivitas antioksidan ekstrak etanol 30% dan 70% kulit buah Mundar (G. forbesii King.) yang diperoleh dengan metode ekstraksi sokhletasi. Pengujian aktivitas antioksidan dilakukan dengan metode DPPH (2,2-Diphenyl-1-Picrylhydrazil) secara kualititatif dan kuantitatif dengan kuersetin sebagai kontrol positif. Hasil uji aktivitas antioksidan ekstrak etanol 30% dan 70% kulit buah Mundar (G. forbesii King.) secara kualitatif menunjukkan adanya aktivitas antioksidan yang ditandai dengan noda kuning pada plat kromatografi lapis tipis (KLT) setelah disemprot dengan larutan DPPH 0,1mM. Hasil uji aktivitas antioksidan secara kuantitatif menghasilkan nilai IC50 (Inhibitory Concentration) secara berturut-turut dari ekstrak etanol 30% kulit buah Mundar (G. forbesii King.); ekstrak etanol 70% kulit buah Mundar (G. forbesii King.); dan kuersetin adalah 717,01 ppm; 534,69 ppm; dan 2,04 ppm.  Ekstrak etanol 70% kulit buah Mundar (G. forbesii King.) memiliki aktivitas antioksidan yang lebih baik dibandingkan ekstrak etanol 30% kulit buah Mundar (G. forbesii King.).  Kata kunci: Kulit buah Mundar (G. forbesii King.), antioksidan, DPPH, kromatografi lapis tipis (KLT), IC50  ABSTRACT Antioxidants are compounds that can counteract free radicals. Mundar rind (Garcinia forbesii King.) is endemic plants from South Kalimantan that has the potential antioxidant activity. This research was conducted antioxidant activity test of ethanolic extract 30% and 70% of Mundar rind (G. forbesii King.) obtained by soxhlet extraction method. Antioxidant activity test of Mundar rind (G. forbesii King.) used the DPPH (2,2-Diphenyl-1-Picrylhydrazil) method qualitatively and quantitatively with quercetin as a positive control. The results of the antioxidant activity test qualitatively of ethanolic extract 30% and 70% of Mundar rind (G. forbesii King.) showed antioxidant activity marked with a yellow spot with purple background on kromatogram after sprayed with DPPH solution 0,1mM. The results of the antioxidant activity test quantitatively obtained IC50(Inhibitory Concentration)  value respectively of ethanolic extract 30%; ethanolic extract 70%; and quercetin were 717,01 ppm; 534,69 ppm; and 2,04 ppm. The ethanolic extract 70% of Mundar rind (G. forbesii King.) had better antioxidant activity than the ethanolic extract 30% of Mundar rind (G. forbesii King.) Keywords: Mundar rind (G. In this paper, ultrasonic cellulase extraction (UCE) was applied to extract polyphenols from passion fruit. The extraction conditions for total phenol content (TPC) and antioxidant activity were optimized using response surface methodology (RSM) coupled with a Box-Behnken design (BBD). The results showed that the liquid-to-solid ratio (X2) was the most significant single factor and had a positive effect on all responses. The ANOVA analysis indicated quadratic models fitted well as TPC with R2 = 0.903, DPPH scavenging activity with R2 = 0.979, and ABTS scavenging activity with R2 = 0.981. The optimal extraction parameters of passion fruit were as follows: pH value of 5 at 30 °C for extraction temperature, 50:1 (w/v) liquid-to-solid ratio with extraction time for 47 min, the experimental values were found matched with those predicted. Infrared spectroscopy suggested that the extract contained the structure of polyphenols. Furthermore, three main polyphenols were identified and quantified by HPLC. The results showed the content of phenolic compounds and antioxidant activity of the optimized UCE were 1.5~2 times higher than that determined by the single extraction method and the Soxhlet extraction method, which indicates UCE is a competitive and effective extraction technique for natural passion fruit polyphenols.

ABSTRACT In vitro experiments carried out with uterus preparations of ovariectomized adult rats indicate the presence in this tissue of a 20β-hydroxysteroid-oxidoreductase which catalyzes the conversion of 20β-hydroxy-4-pregnen-3-one to progesterone. Since a hepatic 20β-hydroxysteroid-oxidoreductase is absent in adult female rats, the myometrial enzyme can be responsible for the biological activity of 20β-hydroxy-4-pregnen-3-one in these animals. Besides progesterone five metabolites were isolated and identified after incubation of [4-14C]20β-hydroxy-4-pregnen-3-one with uterine tissue: 20β-hydroxy-5α-pregnan-3-one, 20β-hydroxy-5β-pregnan-3-one, 5α-pregnane-3α,20β-diol, 4-pregnene-3α,20β-diol and 4-pregnene-3β,20β-diol. The conversion of 20β-hydroxy-4-pregnen-3-one to progesterone permits us to regard all five steroids isolated as progesterone metabolites in the rat uterus. 20β-hydroxy-5β-pregnan-3-one is the first C21-metabolite with a 5β(H)-configuration isolated in the rat uterus, which indicates the presence of 5β-reductase in this tissue.

Objectives: The purpose of the present study was aimed at evaluating the antipyretic and antiulcer activity on leaves of Trema orientalis L. (family Cannabaceae) on albino Wistar rats. The antipyretic and antiulcer activity of the ethanolic extract of the T. orientalis L. leaves in three different concentrations (100, 200, and 300 mg/kg) was compared with standard paracetamol and pantoprazole, which was evaluated by employing Brewer’s yeast-induced pyrexia and ethanol-induced ulcer model. The biochemical parameters such as the volume of gastric juice secretion, pH, total acidity, ulcer index, percentage protection, and lipid peroxidation were also studied. Methods: T. orientalis leaves were extracted with ethanol by the Soxhlet extraction method. The dried extract was used for further phytochemical and pharmacological analysis. The antipyretic effect was studied using Brewer’s yeast-induced pyrexia. The ethanol-induced ulcer model was used to study the antiulcer effect. Results: The percentage yield of ethanolic extract of T. orientalis leaves was found to be 6.71% w/w, respectively. The extract showed significant antipyretic and antiulcer effect when compared with standard paracetamol and pantoprazole. Conclusion: The ethanolic extract of T. orientalis has significant antipyretic and antiulcer action.

The aim of this research is to know the extraction method of caterpillar pupa without detrimental to chemical composition to know the amount of pupa production of German caterpillar with two extraction methods per 100 grams of pupa To know the content of dry matter, organic matter, crude protein, crude fat, crude fiber and amino acid lysin, methionine, and vitamin E pupa oil of German caterpillar. The study was conducted from February to April 2017, at the Lab. Central University Tribhuwana Tunggadewi. Oil extraction analysis was conducted in the chemistry laboratory of Tribhuwana Tunggadewi University and chemistry laboratory of Poltek Malang. German caterpillar research material, German caterpillar pupa, pupa caterpillar germany extracted by maceration and sokletasi. German caterpillar pupa is obtained from German caterpillar breeders in Amadanom, Dampit, Malang Regency. This research uses two different extraction methods, namely cold extraction method (maseration method) and using heat method (soxhlet method). The conclusions of both research methods are known that the quantity of German caterpillar yield using soxhlet extraction method is more than the maceration extraction method. However, for the quality of the two methods it is still necessary to do further analysis to know the exact comparison of the two methods.

The menopause women have the low level of estrogen in the body. The lack of estrogen changes physiological function in women’s body that affects in health condition. Carica papaya L. leaf contains flavonoid quercetin which exhibits estrogenic effect. The aim of this study is to determine the estrogenic effect of papaya leaves extract (PLE) in vivo, and in silico. Papaya leaves were extracted by ethanol 70% maceration. The in silico study were done by molecular docking between quersetin and Estrogen Receptor (ERα and ERβ) to obtain the docking score. Based on this study, docking score of quercetin was almost similar to the native ligand of ER. The in vivo study was done as follow: 36 female rats Sprague Dawley divided into six groups.  The groups are shame-ovariectomized (S-OVX), control ovariectomized (OVX), CMC-Na control (OVX+CMC-Na), positive control (OVX+Estradiol), and the PLE treatment groups dose 750 mg/kgBW (OVX+750mg/kgBW) and dose 1000 mg/kgBW (OVX+1000 mg/kgBW). Administrations of PLE were done in three weeks orally, while estradiol was administrated intraperitonially. The mammae and uterine were sliced for analysis. Based on the study, the treatment of PLE increased the number of mammae lobules and uterine weight as well as estrogen does.  In summary, PLE can be developed as a source of phytoestrogens.Keywords: Carica papaya L., phytoestrogen, estrogen receptor, mammae lobule, uterine

Abstract In popular medicine Silybum marianum is used as a hepatoprotective agent. Silymarin is the major constituent. The present work deals with the formulation and evaluation of S. marianum tablets from ethanolic extract by direct compression. The ethanolic extract was obtained from seeds by soxhlet extraction. Two pharmaceutical formulations were prepared using fluid extract as an active principle, and Aeroperl® 300 Pharma as a carrier. In order to improve flow ability and compressibility, co-processed excipients MicroceLac® 100 and FlowLac® 90 were employed. Pre-compression and post-compression parameters were evaluated according to USP 34-NF 29. Besides, silymarin was determined by NMR spectral data. Both formulations showed excellent rheological properties and the best biopharmaceutical parameters were observed in F2 (S. marianum ethanolic extract, aeroperl® 300 Pharma, flowLac® 90, glycolate starch and magnesium stearate) in terms of the friability (0.82 %) and the disintegration time (8.05 min).
