Objective: The study was aimed to extract, formulate and characterize collagen extract of Stichopus horrens into serum preparations and decide antioxidant activity in powders and serum preparations. Methods: The sea cucumber meat was extracted collagen in three stages, namely the pretreatment stage using 0.3 M NaOH solution 1:10 (w/v) for 48 h, the hydrolysis step in the 0.3 M 1:10 acetic acid solution (w/v) for 48 h, and the extraction stage with distilled water 1: 2 (w/v) for 2 h at 45 °C. The collagen extract was freeze-dried to obtain collagen powder. Collagen powder was characterized by HPLC and its antioxidant activity was determined using the DPPH method. Collagen powder formulated with extract variation of 0, 0.5 and 1%. Evaluation of serum included organoleptic, homogeneity, stability of pH and viscosity as well as antioxidant activity. Results: The results showed that collagen powder had a % yield of 0.24%, which consisted of the amino acids glycine, proline, alanine, and glutamic acid as the dominant amino acids. The % Free radical inhibition of collagen powder at concentration of 5000 ppm was 63.23%. IC50 values were obtained at 4045.37 ppm. The stability test resulted in stable serum preparations without significant changes at 4 °C and 27 °C±2 °C storage temperatures. Conclusion: The measurement of DPPH Radical reduction activity in the highest serum preparation was 1% extract with a value of 2.4%.

Golden sea cucumber or locally known as “teripang emas” is one of Indonesia’s most popular sea cucumber and widely processed as functional food or supplement due to its bioactivities. The sea cucumber is often misidentified due to its morphological similarities with other Stichopus spp. This study aimed to identify the golden sea cucumber obtained from West Nusa Tenggara, Indonesia, by a molecular method and study the antioxidant activities of its collagen hydrolysates. The hydrolysates were produced by hydrolyzing acid collagen extract using neutrase for 30, 60, 120, 180, and 240 mins. The products were then analyzed for their degree of hydrolysis, peptide content, molecular weight distribution and radical scavenging activity by the 2,2-Diphenyl-1-picrylhydrazyl (DPPH) method. Results showed that hydrolysis for 180 mins was optimal in producing the highest peptide content, 12.79 ± 0.44 mg/mL, with a degree of hydrolysis (DH) of 55.2 ± 1.50%. However, the highest antioxidant activity (IC50 of 5.25 ± 0.15 mg/mL) was demonstrated after 60 mins hydrolysis with molecular weight (MW) ranged from less than 14.4 kDa to approximately 25 kDa. The hydrolysate might be categorized as a weak to moderate antioxidant. Based on the molecular identification, the golden sea cucumber had 99% similarities with Stichopus horrens and S. monotuberculatus.

Gold sea cucumber extract (Stichopus hermanni) is rich in collagen and unsatturated fatty acid also known omega 3 such as docosahexaenoic acid and eicosapentaenoic acid that highly effective as an antioxidant. Oligomeric proanthocyanidin is the main content in grape seed oil and the most powerful antioxidant agent that can protect collagen. The aim of research was to determine the effect of gold sea cucumber extract concetration on antioxidant activity of grape seed oil nanoemulsion. The three formulas with results test of antioxidant activity with DPPH method using 15% Brij CS12® and 30% propylene glycol has done which the oil phase was dispersed in the aqueous phase.Variation that have been done is to use the antioxidant activity (IC50) of gold sea cucumber extract  for 2, 4, and 6 times. The resulting preparations was conformed of organoleptic, nanoemulsion type, clarity, globule size, viscosity and rheology, pH and freez thaw test. Antioxidant activity of gold sea cucumber extract and grape seed oil were  (IC50= 53.79 and 17.41 μg/mL respectively). The nanoemulsion was analysed using two-way ANOVA method. Antioxidant  activity at storage for 0; 2 and 4-weeks of formula I, II, III were (IC50=  49.15; 49.45, 51.15 µg/mL), (IC50= 44.12, 47.48, 48.14 µg/mL), and (IC50= 42.14, 43.45, 45.33 µg/mL) respectively. Can be concluded that the higher concentration of gold sea cucumber extract, the antioxidant activity of nanoemulsion is higher, but it decreased during the storage.

The goal of this study was to investigate the efficacy and the safety of grape pomace (Vitis labrusca L.) obtained from the winemaking process as an antioxidant raw material for cosmetic formulations. Grape pomace was dried and submitted to extraction with solvents. The extracts were filtered and freeze-dried. The extraction carried out with 75% acetone-water showed a better yield (1.9 g·g−1). High performance liquid chromatography (HPLC) analysis confirmed the presence of ellagic acid as an active biocompound in the extracts. The Total phenolic content of freeze-dried extracts reached 69.83 ± 1.02 mg for 75% acetone-water system. Antioxidant activity determined by the 2,2-Diphenyl-1-picrylhydrazyl (DPPH) method revealed there was no statistical difference between the extract (EC50 6.9 ± 0.21) and butylated hydroxytoluene (BHT) (EC50 7.6 ± 0.71). Results of cytotoxicity showed that extracts from grape pomace were safe, even at the highest concentration tested (200 mg·mL−1) against the 3T3 cell line. Scanning Electron Microscopy (SEM) analysis revealed that there were no morphological cell changes of cells treated. Cytoprotection of cells towards the oxidation promoted by the peroxide solution was observed in the lowest concentration tested (0.73 mg·mL−1). Grape pomace from the winemaking process revealed a significant antioxidant activity attributed to its biocompounds. The safety and efficacy of extracts of grape pomace as raw material for cosmetic formulations was proposed through cell culture assays.

Teripang merupakan hewan jenis Echinodermata. Beberapa species teripang, misalnya H. scabra, H. fuscogilva dan T. ananas memiliki nilai jual yang tinggi. Meskipun tidak semua spesies memiliki nilai jual yang tinggi, teripang berpotensi untuk dikembangkan dalam bidang kesehatan, terutama dalam kemampuannya untuk meredam radikal bebas dan mencegah berbagai penyakit degeneratif. Tujuan dari penelitian ini yaitu untuk mengidentifikasi, menentukan aktivitas antioksidan, kandungan total fenol, serta kadar karotenoid pada ekstrak beberapa teripang yang berasal dari Perairan Karimunjawa Jepara. Hasil identifikasi dari ke empat spesies adalah Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, dan Holothuria atra. Ekstraksi dilakukan dengan teknik maserasi dinding tubuh teripang menggunakan pelarut metanol. Uji aktivitas antioksidan dilakukan dengan menggunakan metode DPPH (1,1-difenil-pikrilhidrazil) sebagai agen radikal bebas. Penentuan total fenolik dilakukan menggunakan metode folin-ciocalteu, dan penentuan kadar karotenoid dilakukan dengan menggunakan spektrofotometer. Hasil menunjukkan bahwa aktivitas antioksidan Bohadschia vitiensis, Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, dan Holothuria atra tergolong sangat lemah dengan nilai IC50 secara berturut-turut adalah sebesar 454,28 ± 14,4; 713,51 ± 9,1; 801,57 ± 8,4 dan 1535,47 ppm. Kandungan total fenol sebesar 21,08 ± 0,49; 14,325 ± 0,21; 14,033 ± 0,33 dan 10,67 ± 0,12 mg GAE/g sampel, dan kadar karotenoidnya sebesar 23,28 ± 0,07; 25,78 ± 0,56; 11,85 ± 0,04; dan 41,44 ± 0,008 µmol/g sampel.Kata kunci: Teripang, Antioksidan, DPPH, Fenolik, Karotenoid Sea cucumbers are sea benthos from phylum Echinoderms phylum. Some sea cucumbers such as H. scabra, H. fuscogilva, T. Ananas have a high economical price. Eventhough, some sea cucumbers have the potency of being a healthy food. It it scientifically proven that some sea cucumbers have the ability to reduce free radicals and prevent various degenerative diseases caused by the excessive free radicals. The aims of this study were to identified, determine antioxidant activity, total phenol content, and carotenoid levels in sea cucumber extracts originally from Karimunjawa waters. Some of the sea cucumbers obtained are Stichopus cf. quadrifasciatus, Pearsonothuria graeffei, Bohadschia vitiensis, and Holothuria atra. The extraction was done by macerating the body walls with methanol. The antioxidant activity test were tested using the DPPH method as free radicals, the total phenolic content tested using the folin-ciocalteu method, and determined the carotenoid levels spectrophotometrically. Based on the results, antioxidant activity of  B. vitiensis, S. cf. quadrifasciatus, P. graeffei, and H. atra were  classified as very weak with IC50 454.28 ± 14.4; 713.51 ± 9.1; 801.57 ± 8.4 dan 1535.47 ppm. Total phenolic content were 21.08 ± 0.49; 14.325 ± 0.21; 14.033 ± 0.33 dan 10.67 ± 0.12 mg GAE / g samples, while carotenoid levels were 23.28 ± 0.07; 25.78 ± 0.56; 11.85 ± 0.04; dan 41.44 ± 0.008 µmol / g sample.Keyword : Sea cucumber, Antioxidant, DPPH, Phenolic, Carotenoid

Pseudocydonia sinensis (Thouin) C. K. Schneid. less known plant species in the Ukraine conditions, but the fruits were widely used in traditional Chinese medicine for the treatment of asthma, colds, sore throat, mastitis, rheumatoid arthritis, and tuberculosis. The content of protein, ash, and lipids in the seeds was found to be greater than in the pulp and peel. Monosaccharide analysis of neutral carbohydrate part showed the presence of two main sugars fructose and sucrose in the seeds, pulp, and peel. There is a higher beta-carotene content in the rind of the fruit than in the seeds and pulp. The total amount of fatty acids varied from sample to sample and contained mainly oleic acid, palmitic acid, linolenic acid, and linoleic acid. Linoleic acid in the seeds was 48.02% of total fatty acids, slightly less in the rind 42.70%. Palmitic acid, oleic acid, and linoleic acid in the pulp samples were 45.38, 21.32, and 14.93%, respectively. The total amount of amino acids found in the seeds was 105.0 g.kg-1 DM, including total essential amino acids (32.70 g.kg-1 DM). Glutamic acid was found in seeds to be the dominant free amino acid followed by aspartic acid and arginine in the seed. In our study, the antioxidant activity carried out by the DPPH method and measured by molybdenum reducing antioxidant power of peel, pulp and seeds were 9.41, 7.08, 6.21, and 158.81, 92.83, 78.58 mg TEAC.g-1 DM, respectively. Micro and macronutrients and amino acids predominated in the seeds, total fatty acids predominated in the pulp. The highest content of bioactive compounds (total polyphenols, flavonoid, and phenolic acid) and antioxidant activity was found in the peel. P. sinensis can be considered as a nourishing fruit with a copious potential with health-promoting roles and medicinal properties.

Background: This study aimed to determine the potential anti-aging effects of sea grapes and tempe (Fermented soybeans) collagen particle size, by measuring the activities of anti-glycation, antioxidant and tyrosinase inhibitors. Methods: Collagen was isolated from sea grapes and tempe freeze dried powder and treated with different NaOH concentrations (0.10 M; 0.20 M; 0.30 M), and CH3COOH 1 M solution, separately. The collagen particle size was adjusted by stirring at 1000 rpm for 5 and 10 hours. 2,2-diphenyl-1-picrylhydrazyl (DPPH) was used to measure the antioxidant activity, and L-tyrosine and L-DOPA (l-3,4-dihydroxyphenylalanine) were used as a marker of tyrosine inhibition.  Results:  The collagen treated with 0.10 M NaOH produced the highest collagen yield (11.65%), and largest particle size (2455 nm). Additionally, this collagen, when treated for 5 hours, exhibited 24.70% antioxidant activity, 62.60% anti-glycation, 8.97% L-tyrosine, and 26.77% L-Dopa inhibition activities. Meanwhile, the collagen treated for 10 hours had a 9.98% antioxidant activity, 41.48% anti-glycation, 7.89% L-tyrosine, and 2.67% L-Dopa inhibition activity.  Conclusion: Sea grapes and tempe collagen powder treated with 0.10 M NaOH and stirred for 5 hours, as functional foods have anti-aging properties.

The aim of the study was to determine the effect of controlled micro-oxygenation on the fermentation process of two types of white wine (ʻPinot Blancʼ and ʻPinot Grisʼ). The wine was micro-oxygenated during fermentation and maturation. During fermentation, the antioxidant activity was monitored by the DPPH method and the ammonia nitrogen and nitrogen content of the amino acids and the number of cells in 1 mL of fermenting must were also monitored. In micro-oxygenated wine, the growth of cell mass in the first third of the fermentation was recorded and showed an increase in ammonia nitrogen consumption compared to the control experiment. During fermentation, a slight increase in antioxidant activity was observed in the micro-oxygenated wine. For the aromatic profile measured by GC analysis, a higher content of aromatics, mainly esters, was observed in the micro-oxygenated wine. This was also present in the sensory evaluation.

The aim of this study was to investigate the ability of NLC in increasing photostability of tomato extract in term of antioxidant activity. Photostability testing on antioxidant activity of samples were conducted by accelerating method using UVB radiation 32.400 joule for 21 hours radiation. Antioxidant activity was measured by DPPH method. NLC was made by High Shear Homogenization (HPH) method at 24000 rpm for 4 cycles, while conventional creame was made by low speed at 400 rpm. The product were characterized include: pH, viscosity, and particle size. There were had difference characters and physical stability. NLC had smaller size, more homogenous and more stable than conventional creame. It was known that stability of antioxidant activity of tomato extract in NLC system higher than in conventional creame. That was showed with k value, as constanta of rate scavenging activity decreasing in antioxidant power between time (Sigma 2-tail less than 0.005) of NLC and conventional creame were: 2.03x10-2 %/hour ±0.08 (3.94) and 4.71x 10-2 %/ hour ±0.23 (4.88) respectively.

Background: P. mucronata (Pm) comes from South America, Brazil and is characterized as “Maracujá de Restinga”. It is used in folk medicine for its soothing properties and in treating insomnia. Objective: The present study for the first time analyzed the antioxidant and cytotoxicity of the hydroalcoholic leaves extract and fractions from Pm. Method: The cytotoxicity test will be evaluated by different assays (MTT and CV) against human prostate cancer (PC3) and mouse malignant melanoma (B16F10) cell lines, and the antioxidant test by DPPH method. Results: β-Amyrin, oleanolic acid, β-sitosterol and stigmasterol were isolated of the most active, hexane fraction. These substances were tested against the tumor cell lines: β-sitosterol and stigmasterol showed the most relevant activity to PC3 in CV assay and, oleanolic acid to B16F10 by the MTT assay. In addition, it was possible to indicate that the mode of cell death for stigmasterol, presumably is apoptosis. In terms of antioxidant activity, the hydroalcoholic leaves extract presented higher activity (EC50 133.3 µg/mL) compared to the flower (EC50 152.3 µg/mL) and fruit (EC50 207.9 µg/mL) extracts. By the HPLC-MS, it was possible to identify the presence of flavones in the leaf extract (isoschaftoside, schaftoside, isovitexin, vitexin, isoorientin, orientin). Conclusions: P. mucronata hexane fraction showed promising cytotoxic effect against cancer cell lines, and stigmasterol contributes to this activity, inducing apoptosis of these cells. Furthermore, as other Passiflora species, Pm extract showed antioxidant activity and flavones are its major phenolic compounds.

Pineapple peel still contains an important amount of phenolic compounds and vitamins with valuable antioxidant activity. In this way, the aim of this study was the recovery of the bioactive compounds from pineapple peel using environmentally friendly and low-cost techniques, envisaging their application in food products. From the solid-liquid extraction conditions tested, the one delivering an extract with higher total phenolic content and antioxidant capacity was a single extraction step with a solvent-pineapple peel ratio of 1:1 (w/w) for 25 min at ambient temperature, using ethanol-water (80–20%) as a solvent. The resulting extract revealed a total phenolic content value of 11.10 ± 0.01 mg gallic acid equivalent (GAE)/g dry extract, antioxidant activity of 91.79 ± 1.98 µmol Trolox/g dry extract by the DPPH method, and 174.50 ± 9.98 µmol Trolox/g dry extract by the FRAP method. The antioxidant rich extract was subjected to stabilization by the spray drying process at 150 °C of inlet air temperature using maltodextrin (5% w/w) as an encapsulating agent. The results showed that the antioxidant capacity of the encapsulated compounds was maintained after encapsulation. The loaded microparticles obtained, which consist of a bioactive powder, present a great potential to be incorporated in food products or to produce bioactive packaging systems.
