Arecanut is being grown in an area of 2.71 lakh ha in Karnataka and contributes 65 per cent area to the country. Arecanut sheds the leaf sheath and they are widely used for consumption and packing of food in various occasions. The survey was conducted to know the microbial contaminants associated with leaf sheath plates and during the survey visited to 30 plates manufacturing units situated in Shivamogga and Davanagere districts in Karnataka. Randomly collected areca plates were used for laboratory study to know the contaminants. From these, fungal and bacterial contaminants were isolated and identified by cultural, morphological and biochemical studies. The result revealed that Rhizopus stolonifer, Penicillium citrinum, Aspergillus niger, Aspergillus flavus and Trichoderma harzianum and prevalence of Pseudomonas sp. (32×10-6 CFU/mL), Bacillus sp. (19×10-6 CFU/mL), Micrococcus sp. (12×10-6 CFU/mL) and E. coli (4×10-6 CFU/mL) were observed in contaminated plates under laboratory studies. The study concluded that the prevalence of fungi and bacteria may cause deleterious effects on human health is a chance factor. In future, it is required to concentrate on the sterilizing process which may prevent the health hazardous on human beings.

Microorganisms like fungi, actinomycetes and bacteria are considered nanofactories and are helpful in the production of nanoparticles useful in the welfare of human beings. In the present study, we investigated the production of silver nanoparticles from Streptomyces species JF714876. Extracellular synthesis of silver nanoparticles by Streptomyces species was carried out using two different media. Silver nanoparticles were examined using UV-visible, IR and atomic force microscopy. The size of silver nanoparticles was in the range of 80-100 nm. Antimicrobial activity of silver nanoparticle against bacteria such as E. coli, S. aureus, and dermatophytes like T. rubrum and T. tonsurans was determined. Thus, this study suggests that the Streptomyces sp. JF741876 can produce silver ions that can be used as an antimicrobial substance.

Enterobactérias foram identificadas em adultos de Musca domestica (Linnaeus, 1758) (Diptera: Muscidae) e Chrysomya megacephala (Fabricius, 1754) (Diptera: Calliphoridae). Ambas as espécies foram capturadas no Jardim Zoológico da cidade do Rio de Janeiro e tiveram a superfície externa do corpo lavada e o sistema digestivo dissecado, para análise bacteriológica. Identificaram-se Escherichia coli, Citrobacter sp., Proteus mirabilis, Morganella sp., Klebsiella sp., Pseudomonas sp., Enterobacter sp. e Salmonella Agona. P. mirabilis foi o isolado bacteriano mais freqüente. Em duas amostragens (8%) de C. megacephala, isolou-se Salmonella Agona. As amostras de E. coli não foram enteropatogênicas. M. domestica e C. megacephala são potenciais veiculadoras de bactérias causadoras de enterites em humanos e animais.

ABSTRACTSubtilase cytotoxin (SubAB) from verotoxin (VT)-producingEscherichia coli(VTEC) strains was first described in the 98NK2 strain and has been associated with human disease. However, SubAB has recently been found in two VT-negativeE. colistrains (ED 591 and ED 32). SubAB is encoded by two closely linked, cotranscribed genes (subAandsubB). In this study, we investigated the presence ofsubABgenes in 52 VTEC strains isolated from cattle and 209 strains from small ruminants, using PCR. Most (91.9%) VTEC strains from sheep and goats and 25% of the strains from healthy cattle possessedsubABgenes. The presence ofsubABin a high percentage of the VTEC strains from small ruminants might increase the pathogenicity of these strains for human beings. Some differences in the results of PCRs and in the association with some virulence genes suggested the existence of different variants ofsubAB. We therefore sequenced thesubAgene in 12 strains and showed that thesubAgene in most of thesubAB-positive VTEC strains from cattle was almost identical (about 99%) to that in the 98NK2 strain, while thesubAgene in most of thesubAB-positive VTEC strains from small ruminants was almost identical to that in the ED 591 strain. We propose the termssubAB1to describe the SubAB-coding genes resembling that in the 98NK2 strain andsubAB2to describe those resembling that in the ED 591 strain.

AbstractEmergence and spread of antimicrobial-resistant bacteria, including extended-spectrum β-lactamase (ESBL)-producing Escherichia coli, have become serious problems worldwide. Recent studies conducted in Vietnam revealed that ESBL-producing E. coli are widely distributed in food animals and people. CTX-M-9 and CTX-M-1 are the most prevalent β-lactamases among the identified ESBLs. Furthermore, most of the ESBL-producing E. coli isolates were multi-drug resistant. Residual antimicrobials such as sulfamethoxazole, trimethoprim, sulfadimidine, cephalexin, and sulfadiazine were also detected at a high level in both animal meats and environmental water collected from several cities, including Ho Chi Minh city and Can Tho city. These recent studies indicated that improper use of antimicrobials in animal-originated food production might contribute to the emergence and high prevalence of ESBL-producing E. coli in Vietnam. Although clonal ESBL-producing E. coli was not identified, CTX-M-55 gene-carrying plasmids with similar sizes (105–139 kb) have been commonly detected in the ESBL-producing E. coli strains isolated from various food animals and human beings. This finding strongly suggests that horizontal transfer of the CTX-M plasmid among various E. coli strains played a critical role in the emergence and high prevalence of ESBL-producing E. coli in Vietnam.

The article describes the results of laboratory tests of the effectiveness of the disinfectant Monochloride (Iodine chloride) 2%. Laboratory studies were carried out on test objects and test surfaces contaminated with test cultures of microorganisms, which included museum cultures of E. coli (E. coli 1257), S. aureus 209-P, mycobacteria (pcs. B5), and spores (B. cereus pieces. 96). Disinfection of test objects was carried out by the method of irrigation at a consumption rate of 0,25...0,3 l/ m2 with disinfection of smooth surfaces and 0,5 l/m2, with disinfection of rough surfaces. The treatment was performed twice with an interval of 60 minutes. Our work established that the Monochloride (Iodine chloride) 2% has a high disinfectant activity against Escherichia coli, Staphylococcus aureus, mycobacteria and spores. On the basis of the laboratory work, this tool can be recommended for production tests at veterinary surveillance facilities.

Abstract Background: Worldwide, food borne illness is often associated with consumption of meat and poultry products sold at retail markets. A study on the bacteriological status of chicken carcass in Osogbo, Nigeria, was carried out to determine the prevalence of Arcobacter species, Escherichia coli and Staphylococcus aureus in chicken carcasses. Methodology: A total of 100 samples of chicken carcasses were collected from two major processing points in Osogbo, Nigeria. The samples were analysed for the presence of bacterial contaminants using standard microbiological isolation and identification procedures, with antimicrobial susceptibility test performed using the disk diffusion method. Result: Of hundred chicken carcasses sampled, 38% were positive for Arcobacter species and E. coli while 60% accounted for S. aureus isolates. Ninety percent of Arcobacter spp isolates were susceptible to ciprofloxacin, 85% to gentamicin, and pefloxacin, 70% to chloramphenicol and 90% were resistant to amoxicillin, 85% to augumentin and 80% to streptomycin. Hundred percent of E. coli isolates were susceptible to ciprofloxacin, pefloxacin, 95% to gentamicin and 100% were resistant to streptomycin, 85 % resistant to amoxicillin, augumentin, while 100% of S. aureus isolates were susceptible to trimethoprim sulphamethoxazole, 90% susceptible to gentamicin, 80% to streptomycin and 100% of the S.aureus isolates were resistant to ampliclox. Conclusion: The bacteriological status of chicken carcass revealed high contamination with Arcobacter, E coli and S. aureus with varying degree of antibiotic resistance therefore, improvement in meat processing procedures and strict hygiene measures towards reduction of these pathogens in food products should be encouraged.

Mortality in broilers (6.56%) and growing ducks (11.0%) caused by Escherichia coli was recorded in the experimental flocks study during the period from May to August 2003. E. coli organisms isolated from broiler birds affected with characteristic lesions of omphalitis and yolk sac infection, fibrinous pericarditis and peri-hepatitis , hemorrhagic enteritis, and accumulation of excessive pericardial and peritoneal fluid, whereas from ducks with lesions of hemorrhagic enteritis and extensive epicardial hemorrhages. Each of the 21 isolates collected from broilers and 11 isolates from ducks was characterized by cultural and biochemical studies, of which 8 isolates from broilers and 5 isolates from ducks were tested for antibiotic sensitivity with 9 different antibiotics. The antibiotic sensitivity pattern showed that the isolates were highly sensitive to ciprofloxacin but an increasing trend of resistance was recorded in broilers (7 / 9) than duck (4 / 9) isolates. It may be concluded from the results of this study that the high rate of E. coli infection in broilers and ducks along with the high resistance of isolates to antibiotics constitute a threat to the poultry industry in Bangladesh. The mould genus, Penicillium, is known to be a significant source of environmental aero-allergens. One important allergen from Penicillium citrinum, Pen c 2, has been identified by means of two-dimensional immunoblotting using IgE-containing patients' sera. This novel allergen was cloned, sequenced and expressed in Escherichia coli. The cloned cDNA encodes a large 457-amino acid protein precursor containing a 16-amino acid signal peptide, a 120-amino acid propeptide and the 321-amino acid mature protein. Comparison of the Pen c 2 sequence with known protein sequences revealed shared high sequence similarities with two vacuolar serine proteases from Aspergillus niger and Saccharomyces cerevisiae. Asp-46, His-78 and Ser-244 were found to constitute the catalytic triad of the 39-kDa Pen c 2. The DNA coding for Pen c 2 was cloned into vector PQE-30 and expressed in E. coli as a His-tag fusion protein that bound serum IgE from Penicillium-allergic patients on immunoblots. Recombinant Pen c 2 could therefore be used effectively for diagnosis and also potentially for the treatment of mould-derived allergic disorders.

There is strong correlation between changes in abundance of specific bacterial species and several diseases including schistosomiasis. Several studies have described therapeutic effects of curcumin (CUR) which may arise from its regulative effects on intestinal microbiota. Thus, we examined the impact of CUR on the diversity of intestinal microbiota with/without infection by Schistosoma mansoni cercariae for 56 days. Enterobacteriaceae was dominating in a naive and S. mansoni infected mice group without CUR treatment, the most predominant species was Escherichia coli with relative density (R.D%) = 80.66% and the least one was Pseudomonas sp. (0.52%). The influence of CUR on murine microbiota composition was examined one week after oral administration of high (40) and low (20 mg/kg b.w.) CUR doses were administered three times, with two day intervals. CUR induced high variation in the Enterobacteriaceae family, characterized by a significant (p < 0.001) reduction in E. coli and asignificant (p < 0.001) increase in Pseudomonas sp. in both naïve and S. mansoni-infected mice, compared to untreated mice, in a dose-dependent manner. Additionally, our study showed the effects of high CUR doses on S. mansoni infection immunological and parasitological parameters. These data support CUR’s ability to promote Pseudomonas sp. known to produce schistosomicidal toxins and offset the sequelae of murine schistosomiasis.
