To assess the effects of Curcuma longa aqueous extract (CE) on fertility in male rats exposed to Aluminum chloride (AlCl3), Twenty four male rats, 3 months old, divided into 4 groups (n=6) were used. Animal from the first group received, by the oral route, water-containing AlCl3 at the dose of 34 mg/kg body weight (BW). Rats from the second group received both AlCl3 at the same dose in combination with oral treatment of CE at a dose of 200 mg/kg BW. The third group received only an oral administration of CE with the same dose. Rats from the fourth group, without any treatment, served as control. After 4 weeks of experimentation, AlCl3 exposure showed a significant decrease in sperm concentration (4.58±0.65 × 106 cells /ml), and percentage of viability (61.53±23.63 %), and an increase of morphological abnormalities (26.11±17.84 %). A significant decreased in serum testosterone levels (0.31±0.26 ng/ml) and an increase of testicular malondialdehyde (MDA) level (0.16±0.015 μM/g) were also observed. Histological examination of the testes showed degeneration of the seminiferous tubules, germ line cells, and interstitial cells. However, CE treatment concomitant to AlCl3 showed that the rate of morphological abnormalities (19±2.65 %) is significantly decreased compared to AlCl3 group, with a significant increase in serum testosterone (1.17±0.24 ng/ml) and a significant decrease in MDA (0.11±0.003μM/g) level. Microscopic examination revealed a significant regeneration of seminiferous tubules and interstitial cells. This study demonstrated an ameliorative effect of Curcuma longa aqueous extract in testicular tissue and sperm quality.      The present study was undertaken to evaluate the effect of aqueous extract of Date Palm Pollen DPP on the testicular function and serum testosterone, FSH and LH hormones value. Thirty five male rats were divided randomly into five equal groups. Group 1: received 0.5 ml of distilled water (control group), group 2: was treated orally 0.250 p.p.m of sodium florid (NaF) (with volume of 0.5 ml / rat), Group 3: was treated with 0.250 p.p.m of NaF and 50 mg/kg. B.W. of DPP extract (0.5ml D.W \rat), Group 4: was treated with 0.250 p.p.m of NaF and 100 mg/kg. B.W. of DPP extract and Group 5: was treated with 0.250 p.p.m of NaF and 150 mg/kg. B.W. of DPP extract. The results showed  significant (P< 0.05) decrease in sperm concentration, motility and significant (P< 0.05) increases in dead and abnormal sperm in the group 2 in comparison to control, while all groups of  DPP extract showed significant (P< 0.05) increase in  sperm concentration, motility and decrease in dead and abnormal sperm. Maximum effect was observed in animals treated with a dose of 150 mg/kg of DPP extract, also the results revealed significant (P< 0.05) increase in testosterone, FSH and LH hormones in groups treated with DDP in comparison to G1andG2. Male rats received DPP for 50 days showed significant (P< 0.05) increases in body and testes weight as compared to G1andG2. In conclusion the results revealed that the aqueous extract of DPP pollen can be used as a sex enhancer and seems to cure male infertility.

Diabetes mellitus is a metabolic disorder that often affected fertility health, specifically in men. Gynura procumbens (G. procumbens) has been used as traditional medicine due to its ability in treating various types of illness. Hence, this study was carried out to determine the potential beneficial effect of G. procumbens as anti-hyperglycaemia, pro-fertility and libido agent towards diabetes-induced male rats. A total of 42 male Sprague Dawley rats were randomly assigned into six groups; normal, negative, and positive control, and three treated groups of different dosages of G. procumbens aqueous extract (GPAE); 150 mg/kg, 300 mg/kg and 450 mg/kg. Each group was given treatment via oral gavage for seven consecutive days. The rats were sacrificed on day eighth for further analysis. The fasting blood glucose (FBG) level in all GPAE treated group showed significant decrease, and 450 mg/kg treated group showed significant reduction to a normal blood glucose level compared to all groups. There were significant increases in the sperm quality of GPAE treated groups after seven days of treatment compared to the negative and positive control group, specifically in 450 mg/kg treated group. Testis histology showed that GPAE treated groups produced a significant result whereby the seminiferous tubules were seen packed with sperm and successive stage of spermatogenesis were shown compared to control groups. Hormone analysis suggested that the luteneizing hormone (LH), follicle stimulating hormone (FSH) and testosterone hormone of the treated groups were elevated after seven days of treatment compared to the negative and positive control groups. Libido analysis of seven days treatment showed that GPAE treated groups, significantly improved in 450 mg/kg dose compared to other groups, with the highest number of mounting frequency and shortest mounting latency. Fertility test revealed a significant increase in number of implantation sites produced in GPAE treated groups compared to positive and negative control groups. In conclusion, GPAE exhibited a potential beneficial effect as an anti-diabetic and also as fertility agent in diabetic-induced male rats.

During this period, the greatest amount of obtained nanomaterials based on transition of rare earth metals, some of which have significant biological activity. In particular, vanadium compounds have expressive insulin-mimetic action and gadolinium orthovanadate (GdVO4) in nanoform capable in aging laboratory animals inhibit the development of age-related changes, namely a gradual increase (in the physiological range) age glucose level, body weight, reduction of the nervous system reactions and increases the sperm concentration. Given the urgency of anti-aging drug development focus, found a positive effect on spermatogenesis and the lack of information on the effect of this compound on the functional state of the gonads, studied morphofunctional status and endocrine function of the testes in older (18–18.5 months) male rats in long-term (70 days) using nanoparticles (NP) based on GdVO4 dose of 0.33 mg/kg body weight. It was establish the positive effect on bass GdVO4 testicular hormonal function, as evidenced by an increase in serum testosterone (56.3 % at 60 days compared to the injection on the 20th day). It was not found gonadotoxical impact bass GdVO4, as in the seminiferous tubules of the main animals present full pool of germ cells spermatogenesis. Morphometric parameters of glands histological structure indicate the activation of spermatogenesis, which may be due to increasing activity of hormone-producing testes of animals after chronic use NPs GdVO4.

This study investigated the effect of Eulophia macrobulbon (EM) extract on sexual performance in aged-related erectile dysfunction (ED) rats. The ethanol EM extract at the doses of 15, 150, and 450 and sildenafil citrate at the dose of 5 mg/kg body weight (BW) were administered orally to the aged male rats once daily for 21 days. Mating parameters and intracavernosal pressure (ICP) were measured to evaluate their sexual and erection functions. Numbers of sperm and sperm motility as well as the diameter of seminiferous tubules were observed. The serum testosterone and 3’,5’-cyclic guanosine monophosphate (cGMP) concentration in the rat penile tissue were analyzed. The results showed the significant increased sexual motivation, copulatory performance, and ICP of aged rats treated with sildenafil citrate and all doses of EM extract as compared to control aged rats. Moreover, their serum testosterone levels were slightly increased and significant increase in penile cGMP concentration was observed in these aged rats treated with sildenafil citrate and EM extract. The results suggest that treatment with EM could inhibit activity of PDE5 in penile tissue resulting in the increased cGMP level and bring to the improvement of erectile function and sexual performance.

The present study investigated the effects of a standardized methanol extract of E. longifolia Jack containing the major quassinoid constituents of 13α(21)-epoxyeurycomanone (1), eurycomanone (2), 13α,21-dihydroeurycomanone (3) and eurycomanol (4) on the epididymal spermatozoa profile of normal and Andrographis paniculata induced infertile rats. The standardized MeOH extract at doses of 50, 100 and 200 mg/kg, the EtOAc fraction (70 mg/kg), and standardized MeOH extract at 200 mg/kg co-administered with the EtOAc fraction of A. paniculata at 70 mg/kg were each given orally to male Sprague-Dawley albino rats for 48 consecutive days. The spermatozoa count, morphology, motility, plasma testosterone level and Leydig cell count of the animals were statistically analyzed by ANOVA with a post-hoc Tukey HSD test. The results showed that the sperm count of rats given the standardized MeOH extract alone at doses of 50, 100 and 200 mg/kg were increased by 78.9, 94.3 and 99.2 %, respectively when compared with that of control (p < 0.01). The low count, poor motility and abnormal morphology of the spermatozoa induced by the A. paniculata fraction were significantly reversed by the standardized MeOH extract of E. longifolia (p < 0.001). The plasma testosterone level of the rats treated with the standardized MeOH extract at 200 mg/kg was significantly increased (p < 0.01) when compared with that of the control and infertile animals. The spermatocytes in the seminiferous tubules and the Leydig cells appeared normal. Testosterone level was significantly higher in the testes (p < 0.01) than in the plasma after 30 days of oral treatment with the standardized MeOH extract. Interestingly, eurycomanone (2) alone was detected in the rat testis homogenates by HPLC-UV and confirmed by LC/MS, and may have contributed towards the improvement of sperm quality. Thus, the plant may potentially be suitable for the management of male infertility.

Depression and anxiety are recognized as public health problems. Epidemiological studies have shown that depression and anxiety often occur during reproductive ages between 20 and 60 years of age in males. Trazodone is one of the most frequently prescribed drugs in the treatment of depression and anxiety. Drugs used in repeated doses also play a role in the etiology of infertility. In our study, it was aimed to identify the possible toxic effects of trazodone on male rats and elucidate the underlying mechanisms. Vehicle or trazodone (5, 10, and 20 mg/kg/day) was administered to rats for 28 consecutive days (n=8 per group). At the end of that period, sperm concentration, motility, morphology, and DNA damage were determined and testicular morphology was assessed histopathologically in rats. Additionally, we investigated hormonal status by determining serum testosterone, FSH, and LH levels and oxidative stress by determining glutathione and malondialdehyde levels in testicular tissue to elucidate mechanisms of possible reproductive toxicity. According to our results, sperm concentration, sperm motility, and normal sperm morphology were decreased; sperm DNA damage was increased in trazodone-administered groups. Degenerative findings on the testicular structure were observed after trazodone administration in rats. Additionally, serum FSH, LH, and testosterone levels were elevated in the trazodone-administered groups. Increased MDA levels were the signs of enhanced oxidative stress after trazodone administration in testis tissues. Thus, we concluded that trazodone induced reproductive toxicity in male rats; this reproductive toxicity was accompanied by oxidative stress and hormonal changes, which are considered as important causes of reproductive disorders.

This study was conducted to evaluate the modulatory effect of aqueous extract of Curcuma longa (L.) against γ-irradiation (GR), which induces biochemical disorders in male rats. The sublethal dose of GR was determined in primary hepatocytes. Also, the effect of C. longa extract was examined for its activity against GR. In rats, C. longa extract was administered daily (200 mg/kg body mass) for 21 days before, and 7 days after GR exposure (6.5 Gy). The lipid profile and antioxidant status, as well as levels of transaminases, interleukin-6 (IL-6), and tumour necrosis factor α (TNFα) were assessed. The results showed that in hepatocytes, the aqueous extract exhibited radioprotective activity against exposure to GR. Exposure of untreated rats to GR resulted in transaminase disorders, lipid abnormalities, elevation of lipid peroxidation, trace element alterations, release of IL-6 and TNF, and decrease in glutathione and protein level of superoxide dismutase-1 (SOD-1) and peroxiredoxin-1 (PRDX-1). However, treatment of rats with this extract before and after GR exposure improved antioxidant status and minimized the radiation-induced increase in inflammatory cytokines. Changes occurred in the tissue levels of trace elements, and the protein levels of SOD-1 and PRDX-1 were also modulated by C. longa extract. Overall, C. longa exerted a beneficial radioprotective effect against radiation-induced oxidative stress in male rats by alleviating pathological disorders and modulating antioxidant enzymes.

Several conditions such as chemotherapy and toxins can interfere with spermatogenesis and reduce sperm quality and production. In the case where natural antioxidant response cannot manage oxidative stress and free radical overload, oxidative damage occurs and this begins the genesis of many diseases of which diabetes is one of them. This has awakened the interest of researchers to the use of an alternative source of medicine and herbal medicine. Medicinal use of Curcuma longa dates back to ancient China and India; its constituents are stated to have anti-hepatotoxic, anti-inflammatory, stimulant, and antioxidant and used since ancient time as medicinal and nutritive origins knowing to possess androgenic activities and have well effect in diseases treatment in more countries world-wide. As an antioxidant Curcuma longa possible has a useful effect on spermatogenesis and sperm parameters. Wistar male rats (n=24) were allocated into six groups, positive control (n=4), diabetic control (n=4) and experimental groups (n=20), that subdivided into groups of 4 that received treatment of Curcuma longa rhizome powder with or without STZ-induced diabetes in the dosages (25 and 100mg/kg/day) for 21 consecutive days. In twenty-second day, the testes were removed and semen was collected from epididymis and prepared for analysis. The percentage of sperm viability and motility in the treatment groups increased mildly with a significance of: (p<0.05) in comparison to control group and with the diabetic group being critically lower than those in control group. This suggested that Curcuma longa may be promising in enhancing sperm health parameters.Keywords: Turmeric, oxidative stress, semen, STZ-induced diabetes, antioxidant

Rutin, a food derived-polyphenolic bioflavonoid, has been acknowledged for several health benefits. This study aims to explore the ameliorative effects of rutin against carbon tetrachloride (CCl4) toxicity in male rats. Adult male rats were given either CCl4 (30% in olive oil, 3 ml/kg b.w. intraperitoneally) alone or in combination with rutin (70 mg/kg intragastrically) twice a week for 4 weeks. Our data showed that rutin mitigated CCl4 hepatorenal damage, as indicated by diagnostic markers (i.e., transaminases, alkaline phosphatase, total bilirubin, total protein, albumin, urea, uric acid and creatinine), and histopathological findings. In addition, CCl4 induced profound elevation of free radical generation and oxidative stress, as evidenced by increasing lipid peroxidation and reducing catalase, superoxide dismutase and glutathione peroxidase activities in liver, kidney and testicular tissues; these effects were suppressed by coexposure with rutin. Moreover, the increase in the levels of serum triglycerides, cholesterol, low-density lipoprotein cholesterol, and very-low-density lipoprotein cholesterol induced by CCl4 was effectively counteracted by rutin. The decrease in the level of high-density lipoprotein cholesterol in the CCl4 group was also counteracted by rutin treatment. Interestingly, the decreased levels of hormonal mediators associated with sperm production, including serum testosterone, luteinizing hormone and follicle-stimulating hormone, and the impaired sperm quality induced by CCl4 were reversed by rutin. Data from the current study clearly demonstrated that rutin supplementation could at least partly overcome CCl4-induced hepatotoxicity, nephrotoxicity and reproductive toxicity by antioxidant and antidyslipidemic effects.

Twenty-fourSprague-Dawleymale rats (7–9 weeks old, 200–250 g) were divided into Nicotine (N) (0.5 mg/100 g body weight (BW), Nicotine Control (NC) (saline, 0.1 mL/100 g BW),Habbatus saudaoil (HS) (6.0 μL/100 g BW), andHabbatus saudaControl (HSC) (corn oil, 0.1 mL/100 g BW) groups and treated for 100 days. Sperm parameters and seminiferous tubules measurements were evaluated. The N showed a significantly lower sperm motility (1.03±0.05×106 sperm/mL) and percentage of normal (82.61±0.03%) and live (93.88±0.01%) sperm, higher value for the seminiferous tubule (253.36±1.83 μm) and lumen (100.15±2.38 μm) diameters and spermatogonia (19.85±0.39 μm) and spermatocytes (33.37±0.59 μm) layers, and thinner spermatid-sperm layer (22.14±0.71 μm) than the NC (P<0.05). The HS had significantly higher sperm motility (1.49±0.04×106 sperm/mL) and percentage of normal (90.61±0.01%) and live (96.98±0.01%) sperm, smaller lumen diameter (67.53±2.34 μm) and thinner spermatogonia (17.67±0.32 μm) and wider spermatid-sperm (36.95±0.79 μm) layers than the HSC (P<0.05). This research confirmed that nicotine reduced sperm motility and morphology of normal and live sperms and also affected the testis histology, whileHabbatus saudaoil increased sperm quality and gave better testis histological features.
