Diabetes mellitus is a metabolic disorder that often affected fertility health, specifically in men. Gynura procumbens (G. procumbens) has been used as traditional medicine due to its ability in treating various types of illness. Hence, this study was carried out to determine the potential beneficial effect of G. procumbens as anti-hyperglycaemia, pro-fertility and libido agent towards diabetes-induced male rats. A total of 42 male Sprague Dawley rats were randomly assigned into six groups; normal, negative, and positive control, and three treated groups of different dosages of G. procumbens aqueous extract (GPAE); 150 mg/kg, 300 mg/kg and 450 mg/kg. Each group was given treatment via oral gavage for seven consecutive days. The rats were sacrificed on day eighth for further analysis. The fasting blood glucose (FBG) level in all GPAE treated group showed significant decrease, and 450 mg/kg treated group showed significant reduction to a normal blood glucose level compared to all groups. There were significant increases in the sperm quality of GPAE treated groups after seven days of treatment compared to the negative and positive control group, specifically in 450 mg/kg treated group. Testis histology showed that GPAE treated groups produced a significant result whereby the seminiferous tubules were seen packed with sperm and successive stage of spermatogenesis were shown compared to control groups. Hormone analysis suggested that the luteneizing hormone (LH), follicle stimulating hormone (FSH) and testosterone hormone of the treated groups were elevated after seven days of treatment compared to the negative and positive control groups. Libido analysis of seven days treatment showed that GPAE treated groups, significantly improved in 450 mg/kg dose compared to other groups, with the highest number of mounting frequency and shortest mounting latency. Fertility test revealed a significant increase in number of implantation sites produced in GPAE treated groups compared to positive and negative control groups. In conclusion, GPAE exhibited a potential beneficial effect as an anti-diabetic and also as fertility agent in diabetic-induced male rats.

PURPOSE: Aloe vera is a tropical plant popularly known in Brazil as babosa. We have investigated the effect of aqueous extract of Aloe vera on the biodistribution of Na99mTcO4 and laboratorial parameters in Wistar rats. METHODS: Twelve animals were divided into treated and control groups. In the treated group, Aloe vera was given by gavage (5mg/mL/day) during 10 days. The control group received sorbitol by the same way and period. One hour after the last dose, we injected 0.1mL of Na99mTcO4 by orbital plexus. After 60 min, all the animals were killed. Samples were harvested from the brain, liver, heart, muscle, pancreas, stomach, femur, kidneys, blood, testis and thyroid and the percentage of radioactivity (%ATI/g) was determined. Biochemical dosages were performed. RESULTS: There was a significant increase of %ATI/g in blood, femur, kidneys, liver, stomach, testis and thyroid and also in blood levels of AST and ALT. A significant decrease in levels of glucose, cholesterol, triglycerides, creatinine and urea occurred. The statistical analyses were performed by Mann-Whitney test and T-Student test (p<0.05). CONCLUSION: The aqueous extract of Aloe vera facilitated the uptake of Na99mTcO4 in organs of rats and it was responsible to a high increase of levels of AST and ALT.

Purpose This paper aims to design to evaluate the protective effect of vitamin E to ameliorate the disturbances in testosterone pathway and sperm quality of male rats induced by the glycosides vicine (V) and convicine (C) of Vicia faba. Design/methodology/approach Forty male albino rats were divided into five equal groups; control, paraffin oil, V (400 mg/kg) C (150 mg/kg)-treated group, vitamin E (100 mg/kg) + VC-treated group, and vitamin E (200 mg/kg) + VC-treated groups which injected intraperioneally (IP) with 0.5-ml saline, 0.5-ml paraffin oil,V (400 mg/kg) and C (150 mg/kg) of Vicia faba, vitamin E (100 mg/kg) + VC-treated groups, and Vitamin E(200 mg/kg) + VC-treated groups, respectively. Blood and testicular tissue were obtained after one month of the study. The male genital organs were calculated. Testosterone (Ts), luteinizing hormone (LH), follicle stimulating hormone (FSH), dehydroepiandrosterone sulfate (DHEA-SO4), sex hormone binding globulin (SHBG),?-glutamyl transpeptidase (?-GT), glucose-6-phosphate dehydrogenase (G6PD), 3ß-hydroxysteroid dehydrogenase (3ßHSD), lactate dehydrogenase (LDH), spermatozoa concentration, percent of mortality and abnormal sperms were evaluated. Findings The VC-treated group showed significant decrease (p < 0.01) in Ts, DHEA-SO4, G6PD, spermatozoa number and mortality percent, as well as, the male genital organs (testes, epidydemis, seminal vesicle, prostate and vasa deferentia) while significant increase (p < 0.01) was found in LH, FSH, SHBG, LDH, ?-GT, sperms monoclonal Ki-67, and abnormal spermatocytes levels compared with control group. Vitamin E co-injection with VC-treated group returned all these parameters to the normal values. The higher dose of vitamin E (200 mg/kg) was more effect than the lower dose (100 mg/kg). Originality/value Vicia faba contains V and C glycosides. The V and C glycosides in Vicia faba are hydrolyzed by intestinal microflora to aglycones divicine and isouramil, respectively. Divicine and isouramil are highly reactive compounds generating free radicals where divicine and isouramil are the main factors of favism. The V and C glycosides induced disturbances in testosterone pathway and sperm quality of male rats and vitamin E ameliorates these disturbances.

This study investigated the reproductive toxicity of methylmercury (MeHg) and Aroclor (Sigma-Aldrich), alone or in combination, following exposure of prepubertal male rats considering the chromatoid body (CB) as a potential target. The CB is an important molecular regulator of mammalian spermatogenesis, primarily during spermatid cytodifferentiation. Male Wistar rats were exposed to MeHg and/or Aroclor , according the following experimental design: control group, which was administered in corn oil (vehicle) only; MeHg-treated group, which was administered 0.5mg kg−1 day−1 MeHg; Aroclor-treated group, which was administered 1mg kg−1 day−1 Aroclor; Mix-LD, group which was administered a low-dose mixture of MeHg (0.05mg kg−1 day−1) and Aroclor (0.1mg kg−1 day−1); and Mix-HD group, which was administered a high-dose mixture of MeHg (0.5mg kg−1 day−1) and Aroclor (1.0mg kg−1 day−1). MeHg was diluted in distilled water and Aroclor was made up in corn oil (volume 1mL kg−1). Rats were administered the different treatments from PND23 to PND53 by gavage, . The morphophysiology of CBs was analysed, together with aspects of steroid hormones status and regulation, just after the last treatment on PND53. In addition, the long-term effects on sperm parameters were assessed in adult animals. MeHg exposure increased mouse VASA homologue (MVH) protein levels in seminiferous tubules, possibly affecting the epigenetic status of germ cells. Aroclor produced morphological changes to CB assembly, which may explain the observed morphological defects to the sperm flagellum and the consequent decrease in sperm motility. There were no clear additive or synergistic effects between MeHg and Aroclor when administered in combination. In conclusion, this study demonstrates that MeHg and Aroclor have independent deleterious effects on the developing testis, causing molecular and morphological changes in CBs. To the best of our knowledge, this is the first study to show that CBs are targets for toxic agents.

Several conditions such as chemotherapy and toxins can interfere with spermatogenesis and reduce sperm quality and production. In the case where natural antioxidant response cannot manage oxidative stress and free radical overload, oxidative damage occurs and this begins the genesis of many diseases of which diabetes is one of them. This has awakened the interest of researchers to the use of an alternative source of medicine and herbal medicine. Medicinal use of Curcuma longa dates back to ancient China and India; its constituents are stated to have anti-hepatotoxic, anti-inflammatory, stimulant, and antioxidant and used since ancient time as medicinal and nutritive origins knowing to possess androgenic activities and have well effect in diseases treatment in more countries world-wide. As an antioxidant Curcuma longa possible has a useful effect on spermatogenesis and sperm parameters. Wistar male rats (n=24) were allocated into six groups, positive control (n=4), diabetic control (n=4) and experimental groups (n=20), that subdivided into groups of 4 that received treatment of Curcuma longa rhizome powder with or without STZ-induced diabetes in the dosages (25 and 100mg/kg/day) for 21 consecutive days. In twenty-second day, the testes were removed and semen was collected from epididymis and prepared for analysis. The percentage of sperm viability and motility in the treatment groups increased mildly with a significance of: (p<0.05) in comparison to control group and with the diabetic group being critically lower than those in control group. This suggested that Curcuma longa may be promising in enhancing sperm health parameters.Keywords: Turmeric, oxidative stress, semen, STZ-induced diabetes, antioxidant

The aim of the present study was to compare the effect of Phyllanthus amarus extracts and its fractions on urinary excretion. Aqueous extract of Phyllanthus amarus was prepared by decoction of the whole plant and lyophilized. Ethanolic fraction and chloroformic fraction of Phyllanthus amarus were obtained from aqueous extract. Animal were divided into 5 groups of 6 rats and placed individually in metabolic cages. The control group received normal water. A positive control group received furosemide (5 mg/kg, i.p.), used as a reference loop diuretic drug. Three other groups were treated with aqueous extract or ethanolic fraction or chloroformic fraction of Phyllanthus amarus by intraperitoneal injection at the same dose of 40 mg/kg. Urine volumes were collected each 2 h during 8 h period. The diuretic action was obtained by a ratio of urinary excretion of treated group and that of control group. Phyllanthus amarus extracts increased urinary excretion. This effect was time dependant and significant, compared to control group (p < 0.001). During all experiments, ethanolic fraction increased urinary excretion, more than other extracts. After 8h period, it had eliminated about 2.44 ± 0.27 mL, however this value remained less than that of furosemide (3.01 ± 0.17 mL). The urinary excretion induced by furosemide was significantly high (p < 0.05), compared to ethanolic fraction, but the ratio was similar. This study showed that, like furosemide, ethanolic fraction seemed to be the most potent extract for diuresis. Further studies might be carried out to identify the actives molecules and its mechanisms.

AbstrakTujuan penelitian ini adalah untuk menentukan efek pemberian minyak buah merah (pandanus conoideus lam) terhadap kadar glukosa darah dan malondialdehid (MDA) serum mencit yang diinduksi dengan aloksan. Induksi aloksan ini menyebabkan hiperglikemia yang dapat mencetuskan stres oksidatif. Minyak buah merah yang mengandung banyak antioksidan dapat mengendalikan stres oksidatif yang akhirnya dapat menurunkan kadar glukosa darah dan MDA serum.Penelitian eksperimental ini menggunakan 15 ekor mencit jantan yang diseleksi secara random. Hewan coba dibagi menjadi 3 kelompok masing-masing terdiri 5 ekor mencit, yaitu; kelompok kontrol negatif yang hanya diberikan makan dan minum, kelompok kontrol positif yang diinduksi aloksan, dan kelompok perlakuan (diberi minyak buah merah oral) yang diinduksi aloksan. Setelah perlakuan hari ke-14, darah mencit diambil untuk diperiksa kadar glukosa darah dan MDA serum.Hasil penelitian menunjukkan rerata kadar glukosa darah kelompok kontrol negatif adalah 141,20 ± 13,10 mg/dl. Rerata kadar glukosa darah kelompok kontrol positif adalah 266,00 ± 111,10 mg/dl. Rerata kadar glukosa darah kelompok perlakuan adalah 194,80 ± 27,00 mg/dl. Rerata kadar MDA serum kelompok kontrol negatif adalah 0,91 ± 0,10 nmol/ml. Rerata kadar MDA serum kelompok kontrol positif adalah 1,49 ± 0,34 nmol/ml. Rerata kadar MDA kelompok perlakuan adalah 0,97 ± 0,14 nmol/ml. Perbedaan bermakna kadar glukosa darah hanya antara kelompok kontrol negatif dengan kelompok perlakuan (p<0.05). Analisis statistik kadar MDA serum menunjukkan terdapat perbedaan bermakna antara kelompok kontrol positif dengan kelompok perlakuan (p< 0,05).Kesimpulan dari penelitian ini ialah minyak buah merah tidak mampu menurunkan kadar glukosa darah pada kelompok perlakuan, tetapi minyak buah merah mampu menurunkan kadar MDA serum pada kelompok perlakuan.Kata kunci: minyak buah merah, glukosa darah, MDA serum, mencit, aloksanARTIKEL PENELITIAN80AbstractThe objective of this studi was to determine the effect of red fruit oil (pandanus conoideus lam) on blood glucose and serum malondialdehyde in alloxan induced diabetic mice. Hyperglycemia in this mice could lead to oxidative stress. Red fruit oil contains high antioxidants that have capability to control oxidative stress, so blood glucose and serum MDA is reduced.This experimental study had been carried out to 15 rats (randomized group design) which divided into three group of five rats each, i.e. negative control, positive control (alloxan induced), and the group was treated by alloxan induced which followed by red fruit oil orally for 14 days. The result was analyzed by using one way Anova with confidence interval 95%.The result show that blood glucose in negative control was 141.20 ± 13.10 mg/dl. Blood glucose in positive control was 266.00 ± 111.10 mg/dl. Blood glucose in the group was treated by alloxan induced which followed by red fruit orally is 194.80 ± 27.00 mg/dl. The MDA level in negative control group is 0.91 ± 0.10 nmol/ml, positive control group is 1.49 ± 0.34 nmol/ml, and the group was treated by alloxan induced which followed by red fruit orally is 0.97 ± 0.14 nmol/ml. Blood glucose was significantly different between negative control and treated group (p<0.05). Serum MDA was significantly different between positive control group and the group was treated by alloxan induced which followed by red fruit orally (p<0.05).The conclusion is red fruit oil cannot reduce blood glucose level in the group was treated by alloxan induced which followed by red fruit orally, but it has capability to quench serum MDA level in this group.Key word: red fruit oil – blood glucose – serum MDA – mice – alloxan

To assess the effects of Curcuma longa aqueous extract (CE) on fertility in male rats exposed to Aluminum chloride (AlCl3), Twenty four male rats, 3 months old, divided into 4 groups (n=6) were used. Animal from the first group received, by the oral route, water-containing AlCl3 at the dose of 34 mg/kg body weight (BW). Rats from the second group received both AlCl3 at the same dose in combination with oral treatment of CE at a dose of 200 mg/kg BW. The third group received only an oral administration of CE with the same dose. Rats from the fourth group, without any treatment, served as control. After 4 weeks of experimentation, AlCl3 exposure showed a significant decrease in sperm concentration (4.58±0.65 × 106 cells /ml), and percentage of viability (61.53±23.63 %), and an increase of morphological abnormalities (26.11±17.84 %). A significant decreased in serum testosterone levels (0.31±0.26 ng/ml) and an increase of testicular malondialdehyde (MDA) level (0.16±0.015 μM/g) were also observed. Histological examination of the testes showed degeneration of the seminiferous tubules, germ line cells, and interstitial cells. However, CE treatment concomitant to AlCl3 showed that the rate of morphological abnormalities (19±2.65 %) is significantly decreased compared to AlCl3 group, with a significant increase in serum testosterone (1.17±0.24 ng/ml) and a significant decrease in MDA (0.11±0.003μM/g) level. Microscopic examination revealed a significant regeneration of seminiferous tubules and interstitial cells. This study demonstrated an ameliorative effect of Curcuma longa aqueous extract in testicular tissue and sperm quality. The study was conducted in Basrah University – veterinary medicine college, used about (60) sixty male rats divided into two groups randomly, control group and nicotine treated group with 0.5 mg / kg / BW, the result showed no significant difference in growth rats compared to control group also significant increase of liver enzymes compared to the control groups.

Abstract Tramadol is a synthetic opioid analgesic used for moderate-to-severe pain structurally related to codeine and morphine, where their analgesic mechanism is a result of opioid and non-opioid mechanisms. This study was designed to evaluate the effects of Moringa oleifera leaves extract (MLE) on tramadol-induced testicular toxicity, sperm changes, testicular damage, and oxidative stress in male rats. Forty male albino rats were divided into four groups and treated for 4 weeks (group 1, as control; group 2, MLE; group 3, tramadol; group 4, MLE + tramadol). The relative body weight, relative testes weight, serum total testosterone, luteinizing hormone, follicle-stimulating hormone, sperm counts, vitality, total sperm motility, catalase, and superoxide dismutase activities were significantly decreased in tramadol-treated group when compared with the control group. In contrast, sperm abnormality, immotile sperm percent, testicular injury, and TBARS concentration in testes were significantly increased in the tramadol-treated group. In addition, histopathological examination for the tramadol-treated group has shown incomplete spermatogenesis, moderate degeneration in some seminiferous tubules with a significant decrease in the number of spermatogenic cells and depletion of Leydig cells. The administration of MLE with tramadol ameliorates the testicular toxicity, injury, sperm count, abnormalities, and oxidative stress induced by tramadol.

AbstrakDiabetes Melitus telah dikategorikan sebagai penyakit global yang prevalensinya terus meningkat dan penyebab utama morbiditas dan mortalitas. Latihan fisik merupakan salah satu tatalaksana untuk mengontrol glukosa darah secara nonfarmakologis. Tujuan dari penelitian ini adalah untuk mengetahui gambaran kadar glukosa darah pada tikus Wistar diabetes melitus yang diinduksi aloksan setelah pemberian latihan fisik. Penelitian eksperimental ini menggunakan 18 ekor tikus Wistar dengan berat badan 150-200 gram yang dibagi menjadi tiga kelompok, yaitu 6 ekor kelompok kontrol negatif (K-), 6 ekor kelompok kontrol positif (K+) dengan induksi aloksan tanpa pemberian latihan fisik dan 6 ekor kelompok perlakuan (P) dengan induksi aloksan dan diberi latihan fisik selama 4 minggu. Pengukuran kadar glukosa darah dilakukan dengan glucometer merek Accu-Check. Hasil penelitian menunjukkan peningkatan rata-rata kadar glukosa darah puasa secara bermakna pada kelompok kontrol positif (K+) dan perlakuan (P) setelah periode induksi yaitu, pada kelompok kontrol negatif 84,83±6,88 mg/dl, kelompok kontrol positif 220,80±12,29 mg/dl, dan kelompok perlakuan 248,50±94,55 mg/dl (p<0,05). Setelah periode latihan fisik, terdapat penurunan rata-rata glukosa darah puasa secara bermakna pada kelompok kontrol positif (K+) dan Perlakuan (P), namun penurunan pada kelompok perlakuan (P) lebih besar secara bermakna dibandingkan dengan kelompok kontrol positif (K+), yaitu rata-rata glukosa darah puasa kelompok kontrol positif 192,00±12,00 mg/dl dan kelompok perlakuan 163,00±20,26 mg/dl (p<0,05).
