Introduction: Muntingia calabura is used for many medicinal advantages. So far, limited study has been done for the bioactivities of M.calabura fruit. The study aimed to investigate the enzyme inhibitory, antioxidant, and antibacterial activities of M.calabura fruit. Methods: Ethanol extract of M.calabura fruit was tested for its inhibitory enzyme activities against key enzymes linked to human pathologies, such as diabetes (α-glucosidase and α-amylase), hyperuricemia (xanthine oxidase), and obesity (lipase). The antioxidant properties were investigated using different in vitro assays (DPPH, CUPRAC, reducing power, phosphomolybdenum, metal chelating and DNA-Damage protection assays). The fruit was also evaluated for its antibacterial activity against several gram positive and negative bacteria. Results: The total phenolic and flavonoid contents of the extract were 10.85 mgGAE/g and 3.30 mg QE/g, respectively. The fruit extract showed good inhibition against α-glucosidase and α-amylase (IC50 16.74 and 46.49 µg/ml, respectively), with activities stronger than acarbose (100.38 and 152.46 µg/ml, respectively). It exhibited weak inhibitory activity against xanthine oxidase (IC50 0.91 mg/ml) and lipase (IC50 16.48 mg/ml), weaker than the references used for respective test (IC50 allopurinol 5.31 µg/ml and orlistat 0.17 µg/ml). The extract showed antibacterial activities againts Chromobacterium violaceum, Staphylococcus aureus, Streptococcus mutans, Staphylococcus epidermidis, and Escherichia coli. The ethanol extract showed weaker antioxidant activities, when compared to ascorbic acid and BHT. However, the extract was able to protect DNA-damage. Conclusions: The study concludes that M. calabura fruit exhibits antioxidant, antibacterial, and enzyme inhibitory properties, thus can be a good source for pharmacological uses.

To evaluate possibility as a skin whitening agent of Sorghum bicolor (S. bicolor), its antioxidant activity and anti-melanogenic effect on 3-isobutyl-1-methylxanthine (IBMX)-induced melanogenesis in B16/F10 melanoma cells were investigated. The result of total phenolic contents (TPC) indicated that 60% ethanol extract of S. bicolor (ESB) has the highest contents than other ethanol extracts. Antioxidant activity was evaluated using the 2,2’-azino-bis-(3-ethylbenzothiazolin-6-sulfonic acid) diammonium salt (ABTS)/1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activities and malondialdehyde (MDA) inhibitory effect. These results showed ESB has significant antioxidant activities. Inhibitory effect against tyrosinase was also assessed using L-tyrosine (IC50 value = 89.25 μg/mL) and 3,4-dihydroxy-L-phenylalanine (L-DOPA) as substrates. In addition, ESB treatment effectively inhibited melanin production in IBMX-induced B16/F10 melanoma cells. To confirm the mechanism on anti-melanogenic effect of ESB, we examined melanogenesis-related proteins. ESB downregulated melanogenesis by decreasing expression of microphthalmia-associated transcription factor (MITF), tyrosinase and tyrosinase-related protein (TRP)-1. Finally, 9-hydroxyoctadecadienoic acid (9-HODE), 1,3-O-dicaffeoylglycerol and tricin as the main compounds of ESB were analyzed using the ultra-performance liquid chromatography-ion mobility separation-quadrupole time of flight/tandem mass spectrometry (UPLC-IMS-QTOF/MS2). These findings suggest that ESB may have physiological potential to be used skin whitening material.

Summary Prunella vulgaris ( Labiatae family) or self-heal is traditionally used for different ailments such as eye pain and inflammation, headache, dizziness, sore throat and wound healing. Total phenolic and total flavonoid contents of extracts (methanol, ethanol and aqueous) were determined by a spectrophotometer. The antimicrobial activity was evaluated by micro broth dilution assay. The total phenolic content of P. vulgaris extracts were higher in aqueous extract (156.5 mg GAC/g) followed by ethanol extract and methanol extract. The TFC content of P. vulgaris methanol extract (82.8 mg QE/g) was higher than ethanol extract (22.7 mg QE/g) and aqueous extract (16.2 mg QE/g). The antimicrobial activity of methanol or ethanol extracts was higher than aqueous extract from P. vulgaris. The sensitivity of microorganisms to different extracts is related to type of pathogens. There is no positive relation between total phenolic content and its antimicrobial activity. Prunella vulgaris ethanolic extract as a source of phenolic and flavonoid contents can be used as an antimicrobial agent.
