Paraventricular nuclei (PVN) of the hypothalamus are the autonomic center forcoordination of functions and consists of a number of neuronal populations - sub-nuclei,which differ in structural and functional features and the nature of nerve connectionswith various parts of the nervous and neuroendocrine systems. When studying stressreactions and actions of the stress-limiting factors, it is important to study subpopulationsof PVN neurons of the hypothalamus, synthesizing stress-releasing hormones, whichinitiate stress reactions of the body. One of the main factors that show a pronouncedeffect in the regulation of ACTH secretion is corticotropin-releasing factor (CRF). TheCRF-immunoreactive tracer was found mainly in the medial small-cell subnucleus of theparaventricular nuclei of the rat hypothalamus. Despite deep and comprehensive studiesof the hypothalamus, until now there are no unifed ideas about its individual reactivityand the degree of involvement of these structures into the stress reaction caused by theprolonged change in the lighting regime.Purpose - to fnd out the effect of different illumination regimes on the morphometriccharacteristics of the medial small-cell subnuclei of the paraventricular nuclei of thehypothalamus of mature and old rats.Material and methods. The experiments were conducted on 72 nonlinear white malerats, divided into 6 series of studies, in each of which the biomaterial sampling wascarried out at 14.00 and 02.00 h using morphometric and statistical research methods.Results. Morphometric assessment of the medial small-cell sub-nuclei of theparaventricular nuclei (msPVN) of the hypothalamus of rats revealed the diurnaldynamics of indices. It has been established that in both old and mature rats, theiraverage volume (p <0.05) signifcantly decreases at 2.00 compared to 14.00 undernormal lighting. The nuclear-cytoplasmatic ratio at 02.00 increases in comparison with14.00 respectively.When the animals were kept under constant illumination, more pronounced changesin the structures under study were established. In particular, the volume of neurons inthe msPVN of the hypothalamus decreases both mature and old rats compared withrats under standard illumination and even more in comparison with rats under lightdeprivation. In accordance with this, the NCR also changes. The average number ofneurons on the standard plane of the histological section also decreases in comparisonwith other illumination regimes. It is also necessary to point out that under conditionsof light stimulation in mature rats in msPVN, the average volume of neurons (p <0.05)noticeably decreases at 02:00 in comparison with 14:00, when the volume of theirnuclei does not change on average during the indicated study periods. This is reflected,respectively, in the NCR change, which increases at 02.00 in comparison with 14.00.As to the old rats, the average neuron volume does not decrease at 2.00 in comparisonwith 14.00 in msPVN neurons at light stimulation, as well as the volume of their neucleichanges, on average, is barely noticeable ,and according to it, NCR at 02.00 remainsstable in comparison with 14.00, and the average quantity of neurocytes in msPVN doesnot change on a standard plane of the histological section during various periods ofthe twenty four hours. At the same time, with various modifcations of the light regime,cytometric parameters of neurons under study in old rats, on average, are signifcantlylower (p <0.001) than those in mature rats.Conclusions. 1. The duration of the photoperiod signifcantly affects the 24 hoursactivity of msPVN in the hypothalamus of mature and old rats. In particular, in bothinvestigated groups of rats, the average volume of neurons (p <0.05) reliably decreasesat 2.00 under normal illumination in comparison with 14.00, and, accordingly, the NCR at 02.00 increases compared to 14.00. All cytometric parameters of their neurons in oldrats are, on average, signifcantly lower (p <0.001) than in mature rats.2. Constant illumination causes more pronounced changes in the morphometricparameters of the msPVN of the hypothalamus than light deprivation. Thus, in maturerats in the msPVN, the average volume of neurons (p <0.05) decreases noticeably at02:00 in comparison with 14:00, which is reflected, respectively, in the change in NCR,which increases at 02:00 compared to 14:00. For old rats, under light stimulation, allthe studied cytometric parameters of their neurons, on average, are signifcantly lower(p <0.001) than those in mature rats.

Abstract Brain metastases from esophageal cancer is rare and the incidence has been reported at approximately 5%. We report a case of brain metastases with repeated bleeding from Esophageal carcinoma. The case is a 76-year-old man. Three years ago he was diagnosed with small cell carcinoma of the esophagus by endoscopic biopsy. Metastasis was found only in the cervical lymph node, but the condition was stable by chemoradiotherapy and no metastases were found throughout the body before 1 month. He was admitted to the hospital because of a sudden convulsion, and CT scan revealed cerebral hemorrhage in the right frontal lobe. We performed conservative treatment, but rebleeding was observed from the same site repeatedly after 1 month and 2 months. Due to the influence of bleeding, it was difficult to distinguish cerebral hemorrhage from brain tumor by contrast MRI. After surgery, the cause of bleeding was diagnosed as metastatic brain tumor of esophageal small cell carcinoma. Postoperative radiation therapy was performed in another hospital, but rebleeding was observed 3 months after the operation. A reoperation was performed at another hospital, and a recurrence of metastatic brain tumor was diagnosed. In the case of highly malignant metastatic brain tumors, it was considered necessary to frequently follow the images.

While the organs and tissues of the young rat are known to increase in size with age (Donaldson, 1924), little is known of the role played by the component cells in this increase. There is evidence that cells enlarge (Levi, 1906; Plenk, 1911) and new cells are added (Strasburger, 1893), but we do not know to what extent the enlargement and proliferation of the cells cause the growth of organs and tissues. The present work is an attempt to clarify this problem. In the past, the growth of organs and tissues has often been measured by weight gain (Donaldson, 1924). However, this approach might be misleading, since the body-weight may increase in the absence of growth, for instance as a result of fat-storage in old rats, of pregnancy in females, and even of changes in room temperature.

Neuroendocrine cells are widespread throughout the body and can give rise of neuroendocrine tumors due to abnormal growth of the chromaffin cells. Neuroendocrine tumors divide into many subtypes based on tumor grade (Ki-67 index and mitotic count) and differentiation. These tumors can be further divided into secretory and nonsecretory types based on the production of peptide hormone by tumor cells. Poorly differentiated small-cell-type neuroendocrine tumors are one of the subtypes of neuroendocrine tumors. These tumors are less common; however, they tend to be locally invasive and aggressive in behavior with poor overall median survival. Treatment of the nonsecretory small-cell type is modeled to small-cell lung cancer with a regimen consisting of platinum-based chemotherapy and etoposide with variable response. Here, we present a case of poorly differentiated small-cell neuroendocrine tumor originating from the prostate.

Urine is a very good source for biomarker discovery because it accumulates changes in the body. However, a major challenge in urinary biomarker discovery is the fact that the urinary proteome is influenced by various elements. To circumvent these problems, simpler systems, such as animal models, can be used to establish associations between physiological or pathological conditions and alterations in the urinary proteome. In this study, the urinary proteomes of young (two months old) and old rats (20 months old; nine in each group) were analyzed using LC-MS/MS and quantified using the Progenesis LC-MS software. A total of 371 proteins were identified, 194 of which were shared between the young and old rats. Based on criteria of a fold change ≥2,P< 0.05 and identification in each rat of the high-abundance group, 33 proteins were found to be changed (15 increased and 18 decreased in old rats). By adding a more stringent standard (protein spectral counts from every rat in the higher group greater than those in the lower group), eight proteins showed consistent changes in all rats of the groups; two of these proteins are also altered in the urinary proteome of aging humans. However, no shared proteins between our results and the previous aging plasma proteome were identified. Twenty of the 33 (60%) altered proteins have been reported to be disease biomarkers, suggesting that aging may share similar urinary changes with some diseases. The 33 proteins corresponded to 28 human orthologs which, according to the Human Protein Atlas, are strongly expressed in the kidney, intestine, cerebellum and lung. Therefore, the urinary proteome may reflect aging conditions in these organs.

At present, special attention is drawn to the study of the adaptation of aquatic organisms to a complex of environmental factors precisely at the cellular level. It is very important to study the structural and functional features of the kidney, which not only plays a key role in osmoregulation, but also makes a significant contribution to maintaining homeostasis at the level of functioning of a single nonspecific defense system of the body. In this aspect, the study of species belonging to a unique ancient group, united in the order salmoniformes, is highly relevant. Using the methods of light and electron microscopy, we investigated the morphology and ultrastructure of the mesonephros of the population of the semianadromous ecological form European smelt Osmerus eperlanus inhabiting the Gulf of Finland of the Baltic Sea and performing spawning migrations in the Luga River of the Leningrad Region. The general scheme of the trunk kidney organization is given, the structural features and the ratio of leukocytes and structures of the smelt nephron are revealed. It is shown that the development of hematopoietic tissue in the mesonephros, the number of mature forms of granulocytes are systematic signs which do not depend on the ecology of the species. The ratio of leukocytes, the width of the cisterns of the rough endoplasmic reticulum of plasma cells, the structure and number of granules in granulocytes are associated with the peculiarities of the functioning of the cellular link of the immune system under certain conditions of the species habitat. The ultrafine structure of the ion-transporting interstitial cells, as well as the ultrastructural features found in the smelt nephron, can be considered cytological markers of smelt adaptation to a semianadromous lifestyle.

The purpose of this work was to study changes in the level of cell-free DNA (cfDNA) in the blood of young and old rats in the normal state and with induced benign prostatic hyperplasia (BPH). Male Wistar rats were divided into 4 groups—young (3 months), old (20 months), intact, or with testosterone-induced BPH. Groups with BPH were subjected to surgical castration and administration of testosterone esters at a dose of 25 mg/kg for a total of 7 injections for 20 days. In intact animals, the level of cfDNA in old rats (2.00±0.14 ng/μl) was significantly higher than that in the young (1.02±0.30 ng/μl). The body and the prostate weights of old rats were 1.6 and 1.4 times larger than those of the young, without an increase in the prostate index (PI). The testosterone level in the blood of young rats was 1.6 times higher than that of old (6.20±0.93 nmol/l vs. 3.77±0.55 nmol/l; NS). In animals with BPH, the level of cfDNA in old rats (3.14±0.76 ng/μl) was significantly higher than that in young rats (0.80±0.14 ng/μl). The body and the prostate weights in old rats were 1.8 and 2.3 times larger, than those in young rats, with an increase in the PI. The level of testosterone in the blood of young (15.76±0.51 nmol/l) and old (16.99±1.1 nmol/l) rats was not significantly different. Morphological signs of BPH were observed in the prostate of both young and old rats. During the induction of BPH in the experiment, according to the level of cfDNA, cell death processes have not changed significantly in young rats but significantly increased in old rats. A similar trend was observed in the group of intact animals. The obtained data indicate that apoptosis processes are enhanced during the development of BPH despite the growth of tissues in the prostate itself.

ABSTRACT Mammalian orthoreovirus attachment to target cells is mediated by the outer capsid protein σ1, which projects from the virion surface. The σ1 protein is a homotrimer consisting of a filamentous tail, which is partly inserted into the virion; a body domain constructed from β-spiral repeats; and a globular head with receptor-binding properties. The σ1 tail is predicted to form an α-helical coiled coil. Although σ1 undergoes a conformational change during cell entry, the nature of this change and its contributions to viral replication are unknown. Electron micrographs of σ1 molecules released from virions identified three regions of flexibility, including one at the midpoint of the molecule, that may be involved in its structural rearrangement. To enable a detailed understanding of essential σ1 tail organization and properties, we determined high-resolution structures of the reovirus type 1 Lang (T1L) and type 3 Dearing (T3D) σ1 tail domains. Both molecules feature extended α-helical coiled coils, with T1L σ1 harboring central chloride ions. Each molecule displays a discontinuity (stutter) within the coiled coil and an unexpectedly seamless transition to the body domain. The transition region features conserved interdomain interactions and appears rigid rather than highly flexible. Functional analyses of reoviruses containing engineered σ1 mutations suggest that conserved residues predicted to stabilize the coiled-coil-to-body junction are essential for σ1 folding and encapsidation, whereas central chloride ion coordination and the stutter are dispensable for efficient replication. Together, these findings enable modeling of full-length reovirus σ1 and provide insight into the stabilization of a multidomain virus attachment protein. IMPORTANCE While it is established that different conformational states of attachment proteins of enveloped viruses mediate receptor binding and membrane fusion, less is understood about how such proteins mediate attachment and entry of nonenveloped viruses. The filamentous reovirus attachment protein σ1 binds cellular receptors; contains regions of predicted flexibility, including one at the fiber midpoint; and undergoes a conformational change during cell entry. Neither the nature of the structural change nor its contribution to viral infection is understood. We determined crystal structures of large σ1 fragments for two different reovirus serotypes. We observed an unexpectedly tight transition between two domains spanning the fiber midpoint, which allows for little flexibility. Studies of reoviruses with engineered changes near the σ1 midpoint suggest that the stabilization of this region is critical for function. Together with a previously determined structure, we now have a complete model of the full-length, elongated reovirus σ1 attachment protein.

Lung cancer is the second most common type of cancer in the UK. It is termed ‘primary’ if it originates in the lungs. and ‘secondary’ if it manifests elsewhere in the body but then spreads to the lungs. The main types of primary lung cancer are small cell lung carcinoma and non-small cell lung carcinoma. Bronchial carcinoids account for up to 5% of lung cancer. These are generally small when diagnosed and occur most commonly in people under 40 years of age. Unrelated to cigarette smoking, carcinoid tumours can metastasize and a small proportion of these tumours secrete hormone-like substances that may cause specific symptoms related to the hormone being produced. Carcinoids generally grow and spread more slowly than bronchogenic cancers, and many are detected early enough to be amenable to surgical resection. Mesothelioma is a rare type of cancer that affects the pleura.

The chronic effects of estradiol (E2) on energy balance have been investigated in ovariectomized rats with hypothalamic paraventricular nuclei (PVH) lesions. Body weight and food intake were monitored throughout the E2 treatment, which lasted 26 days. At the end of this treatment, rats were decapitated, and their carcasses were processed to determine the body contents in energy, fat, and protein. Plasma adrenocorticotropic hormone (ACTH) and corticosterone were determined by radioimmunoassay and protein-binding assay at the end of the study. Regardless of whether they were sham- or PVH-lesioned, E2-treated rats ate, expended, and gained significantly less energy than untreated animals. In addition, E2-treated rats deposited less fat and protein than the rats not receiving E2. In contrast to the E2 treatment, PVH lesions accelerated the gains in energy and fat regardless of whether the rats were treated with E2 or with a placebo. There were no interaction effects of PVH lesions and the E2 treatment on energy or fat gains. Plasma levels of corticosterone and ACTH were higher in E2-treated rats than in animals receiving the placebo treatment. The present results provide evidence that the hypothalamic PVH is not an essential neuroanatomical structure in the effects of E2 on energy and fat balances.

Background. Clinically, scar related complications are observed to be dissimilar in different regions of the body. Unequal distribution of dermal collagen and elastic fibres in different orientations could be one of the multifocal causes of scar related complications, for which this evaluating study has been taken up. Materials and Method. 300 skin samples collected in horizontal and vertical orientations were studied histomorphometrically. This study involved image analysis of specially stained histological section using tissue-quant software. The outcome result was termed as quantitative fraction. From the result, various ratio values were also calculated for the ratio analysis. Results. The differences in the quantitative fraction of dermal elastic content between 2 directions were statistically significant at joint areas (shoulder joint, wrist, and ankle) (P<0.001) but for collagen, significant difference was observed at shoulder joint and wrist only. Dermis of the forearm and thigh did not show any differences in their collagen content, but for elastic, thigh did show a significant difference while forearm had no change between 2 directions. Conclusion. Analysis of unequal content of dermal element in two directions under the perspective of wound healing consequences is subjective depending upon the anatomical position and functional status of the areas.
